RESPON AKTIVITAS AMILASE DAN PROTEASE IKAN GURAMI Osphronemus gouramy Lac. TERHADAP PERBEDAAN TEMPERATUR AIR
Research to determine the activity of amylase and protease digestion gourami on various temperature has been carried out experimentally using a completely randomized design (CRD) with 3 treatments and 4 replications. The treatments were included gourami nurturing on water temperature 24oC (P1), gourami nurturing on water temperature 28oC (P2), and gourami nurturing on water temperature 32oC (P3). The results the amylase activity is showed no significant difference between treatments (P>0,05) where as protease activity showed there are significant differences between treatments (P0≤0,05). In conclusion, the difference in protease activity of gourami occured in water temperature 24oC (P1) and 28oC (P2) and the protease activity was highest at 28oC water temperature, but the amylase activity did not difference
Isolasi dan Uji Efektivitas Beberapa Isolat Dekomposer Lokal dalam Mendekomposisi Limbah Tanaman Jagung
Soil microorganisms are able to digest and degrade organic material present in the environment in to substances that available for plants to absorb. This research aimed to isolate and examine local bacteria that had the ability to decompose corn stover. Soil samples were taken from various natural habitats in which bacteria grow, including remains of corn planting area, pile of waste corncob and corn stalk, pile of rice husk and straw, and pile of used sorghum stalk. Which were collected from several locations across South Sulawesi. Bacteria was isolated by making soil suspension that being inoculated on to NA and CMC medium, and then incubated for 48 hours at room temperature. Bacterial colonies that grow were then incubated 50o C to obtain bacteria resistant to high temperature and able to produce cellulose. Bacterial isolates that produce cellulose was then examined for the effectiveness in decomposing corn stover waste. Isolates result were able to obtain 119 decomposers isolates with 29 isolates resistant to temperature above 50o C and had ability to produce cellulose. From 29 isolates, among 15 isolates collected from rice straw, 5 isolates from remains of corn planting area and 9 isolates collected from several natural habitats. Almost all isolates had good ability to decompose corn stover waste. From isolates tested in corn cob waste and corn leaves waste, only 16 bacterial isolates were proven to be effective as decomposer
KAJIAN TOKSISITAS DARI TUBUH BUAH Ganoderma lucidum DENGAN METODE BRINE SHRIMP LETHALITY TEST (BST)
Ganoderma lucidum is polyporus fungi from Basidiomycetes which can be used as traditional medicines. Toxicity test with Brine Shrimp Lethality Test (BST) method using Artemia salina was conducted to find out toxic effect of G.lucidum. An extract would have toxic effect if the LC50 < 1000 µg/ml. The aims of this experiment were to know about the toxicity level from G.lucidum with Brine Shrimp Lethality Test (BST) method and determined the concentration of fruiting body extract of G.lucidum which had the best toxicity effect in LC50. There were two extraction methods used in this experiment, first one stage extraction and then multilevel extraction with hexane, ethyl acetate, and ethanol. Each solvent was made in 1000 ppm, 500 ppm, 250 ppm, and 125 ppm. This process was repeated for three times. This experiment used G.lucidum extract which was tested to A.salina and secunder compound metabolit test from the most toxic G. lucidum was done with phytochemical analysis. The result indicated that fruiting body extract from G.lucidum could kill A.salina because all extract was positively contained alkaloid and terpenoid but negatively contained flavonoid. Fruiting body extract from G.lucidum which had lowest LC50 wass ethyl asetat extract with one stage extraction in concentration 53,70 ppm and highest LC50 was ethanol extract with multilevel extraction in concentration 501,18 ppm
PENGARUH PEMBERIAN MIKROBA EFEKTIF PRODUKTIF PLUS (MEP+) PADA MEDIUM BUDIDAYA IKAN NILA YANG DIBERI PAKAN FERMENTATIF TERHADAP KEPADATAN BAKTERI ASAM LAKTAT
Microbes Effective Productive Plus (MEP+) in fish culture has role as probiotics and decomposer. Application of MEP+ is done by adding MEP+ on culture medium of Tilapia and fish feed. Fish feed is fermentative feed with addition of different concentration of cassava peel flour. The aim of this research were to find out the influence of MEP+ administration in culture medium and in fermentative feed with addition cassava peel flour on the increasing density of lactic acid bacteria in culture medium and find out the highest density of lactic acid bacteria. The research was done experimentally, used Complete Randomized Design with treatment of MEP+ administration in culture medium with fermentative feed cassava peel flour addition of 25%, 50%, 75% and without MEP+ administration on culture medium with fermentative feed cassava peel flour addition of 25%. The data obtained were analyzed using a variety analysis. The result showed that MEP+ administration on culture medium and in fermentative feed cassava peel flour addition did not influence the increasing density of lactic acid bacteria and total density of lactic acid bacteria in culture medium was not different inter treatment
Karakter dan Potensi Inokulum Bakteri Fosfat dalam Melepaskan Fosfor
Soil bacteria group which can release soil P-adsorbed is phosphate bacteria (PB). PB can release soil P-adsorbed through: 1) acidification, 2) chelating 3) ligand exchange 4) mineralization and 5) compete colloid sorption site. PB have a potential to release soil P-adsorbed. This study was aimed to determining the character and PB inoculant potential in soil P-adsorbed release. The results of BLASTn PB showed that isolate 1 was Pseudomonas trivialis, isolate 5 was Pseudomonas putida and isolate 9 was Pseudomonas fluorescens. Based on the growth curve on day 5th (the end log. phase) population of Pseudomonas trivialis was 1010 CFU/mL, Pseudomonas putida was 1014 CFU/mL and Pseudomonas fluorescens was 1017 CFU/mL. Storage inoculant PB population decrease 97-84 % and 80-65 % PB lowering capabilities
Sexing pada Ikan Brek Puntius orphoides (Valenciennes, 1863) Menggunakan Metode Truss Morfometrics
The red chick barb (Puntius orphoides) is one of fresh water ichtiofauna included in family Cyprinidae found a lot in River Klawing, the biggest and richest river in Purbalingga with 18 species of fish. The aim of this study was to find out the differences between male and female red chick barb by truss morphometrics and to find out truss morphometrics distance. The material used were sex mature fish from River Klawing. The variables meassured included morphometry based on truss morphometrics point, number of male and female fish. Truss distance was then compared to the total length so that the truss distance ratio was determined. Next, a statistical test i.e. t test was performed between the male and female fish. The results of this study showed that the male and female red chick barb can be distinguished by truss morphometrics method. The distance ratio to be used as diagnostic character were 11 and 24 ration of truss morphometrics distance compared found in the head, body, and tail
KELIMPAHAN CHLOROPHYTA PADA MEDIA BUDIDAYA IKAN NILA YANG DIBERI PAKAN FERMENTASI DENGAN PENAMBAHAN TEPUNG KULIT UBI KAYU DAN PROBIOTIK
Chlorophyta is autotroph organisms which has an important role in fresh waters as one of the largest algae division. This study aimed to determine the abundance and similarity structure of Chlorophyta in Tilapia culture media which was given fermented feed with the addition of cassava peel flour and MEP+ probiotic. This study used four treatments with four replications: (A) fermented feed without addition of cassava peel flour or 0% and MEP+ probiotic administration in media, (B) 25%, (C) 50% and (D) 75%. Sampling was carried out 6 times at intervals of 2 weeks. The main parameters were the number of Chlorophyta species and individuals,while supporting parameters were physical (temperature and TDS) and chemical (DO, BOD, pH, NO3, NO2 and total of PO4). The abundance of Chlorophyta data were analyzed descriptively and its similarity structure were analyzed using Cluster analysis. Then, continued with SIMPER analysis to determine the contribution of species to abundance similarity with PRIMER-E v.5 software. Analysis results showed that the abundance of Chlorophyta consists of 33 species with the average number of 10.412 individuals/liter. Cluster analysis results based on Bray-Curtis similarity index had a quite high similarity and it ranged between 57.79% -68.84%. SIMPER analysis results showed that the species which given highest contribution were Kircheneriella lunaris (31,03%), Selenastrum sp. (21,69%), and Gonatozygon monotaenium (12,96%)
KAJIAN PENYAKIT YANG DISEBABKAN OLEH CENDAWAN PADA TANAMAN CABAI MERAH (Capsicum annum L.) DI PERTANAMAN RAKYAT KABUPATEN BREBES
This research aimed to determine the various diseases caused by fungi, to identify the fungi causing the disease, and to determine the most dominant disease in red chili crops (Capsicum annum L.) from smallholder agriculture located in Kaligiri Sirampog Brebes. The research was a survey applying purposive random sampling technique. This study collected samples from five locations, within five plots placed diagonally at each site. This study picked ten infected plants from each plot, identified the disease; isolated and identified the fungi causing the disease. This research also measured the environmental factors such as temperature, humidity, and soil pH. The results showed diseases in red chili plants (Capsicum annum L.) were the leaf spot caused by Cercospora sp., the anthracnose caused by Colletotrichum sp., and the fusarium wilt caused by Fusarium sp., and the most dominant were leaf spot disease
HUBUNGAN INTENSITAS PENYAKIT KARAT DENGAN PRODUKTIVITAS TANAMAN KEDELAI (Glycine max (L.) Merr.) PADA BEBERAPA VARIETAS BERBEDA
The severity of plant disease is closely related to the development of the disease itself, including rusts caused by pathogenic fungi. This research aimed to determine the difference of rusts disease severity and to determine the correlation between rusts disease severity with the productivity of different varieties of soybean. The material used was inoculum of rusts pathogenic fungi, Phakopsora pachyrhizi Syd., and four varieties of soybean, i.e., Slamet, Lokon, Bromo, and Ringgit. This research was a split-plot design experimental with subplot was four different varieties of soybean, and the main plot was the control without the treatment of inoculation (In0). The spores suspension density of inoculum of P. pachyrhizi Syd applied was 104 spores/ml (In1). The main parameters observed in this study were the height of plants, the number of pods per plant, the number of seeds per plant, the wet and dry weight of the plant, and the total weight of 100 grains seed. The supporting parameters were the disease severity, the soil acidity, the humidity, and the air temperature. The results showed the plant with lowest rusts disease severity was Slamet variety (18,38%), and the highest was Ringgit variety (35,92%). The correlation test showed the increment of rusts disease severity reduced the number of pods and dry weight of the plant in Lokon variety
PERKEMBANGAN OOSIT INDUK Osteochilus hasselti C.V. YANG DIBERI HORMON ESTRADIOL-17β DAN PAKAN DENGAN KADAR PROTEIN BERBEDA
The availability of fish seed is very important in fish propagation. Good quality of fish seeds were produced by a good brooder which characterized a large number of egg production at spawning. The aims of this research were to evaluate the oocytes development in the hard-lipped Barb (Osteochilus hasselti C.V.) given 17β-estradiol, different percentage of protein in the diet, and their combination. This research was conducted experimentally applying Factorial Completely Randomized Design. The first factor was percentage of protein in the diet consisted of 4 levels namely 25% (P1), 30% (P2), 35% (P3), and 40% (P4), the second factor was dose of 17β-estradiol benzoate (EB) consisted of 3 levels namely 0 µg/kg of body weight (D0), 126 µg/kg of body weight (D1), and 210 µg/kg of body weight (D2), thus there were 12 combinations of treatment with 3 replicates. The results showed that neither of protein proportion or 17β-estradiol affected the proportion of oocytes of any developmental stage (p>0,05) within the first two weeks post spawning. The treatments, however, significantly increased the proportion of oocytes at V3-V5, V6-V7, and post-vitellogenic stages (p 0.05). In conclusion, the percentage of protein in the diet and different doses of 17β-estradiol improved oocyte development of Hard-Lipped Barb (Osteochilus hasselti C.V.)
