Yüksek doz fruktoz diyetinin karaciğer antioksidan enzimleri üzerine etkilerinin araştırılması ve mirna moleküllerinin düzenleyici görevlerinin belirlenmesi
Tez No: 855305Son yıllarda diyetle alınan fruktozun kullanımı oldukça yaygınlaşmıştır. İnsülinden bağımsız olarak karaciğere alınan fruktoz, insülin direnci, abdominal obezite, inflamasyon, alkole bağlı olmayan karaciğer yağlanması ve metabolik sendrom gibi patolojilere sebep olabilmektedir. İnsidansı gittikçe artan sağlık problemlerinden biri olan metabolik sendromda reaktif oksijen türlerinde artışa bağlı olarak oksidatif stresin geliştiği çeşitli çalışmalarda gösterilmiştir. Bu tez çalışması kapsamında yüksek doz fruktoz diyetinin oluşturduğu metabolik sendromda oksidatif belirteçlerin incelenmesi, antioksidan enzim sistemlerinin nasıl düzenlendiğinin ortaya çıkarılması, bu süreçte görevli olabilecek bazı miRNA'ları etkileri ve probiyotik takviyelerinin incelenen parametreler üzerine düzenleyici görevlerinin ortaya çıkarılması amaçlanmıştır. Tez çalışmasında erkek Wistar sıçanlarına içme suyu içerisinde %20'lik fruktoz çözeltisi 15 hafta süreyle uygulanarak hayvanlarda metabolik sendrom modeli oluşturulmuş, kefir, Laktobasillus helveticus, Laktobasillus plantarum probiyotikleri altı hafta boyunca günde bir kez gastrik sondayla uygulanmıştır. Karaciğer dokularında; antioksidan enzimlerin aktiviteleri, protein ekspresyonları ve gen ekspresyonları analiz edilmiştir. Ayrıca total tiyol, nativ tiyol, disülfit, GSH/GSSG oranı, malondialdehit, protein karbonil, total antioksidan ve oksidan seviyesi gibi bazı oksidan ve antioksidan parametrelerin bu sendromdan nasıl etkilendiği belirlenirken, antioksidan enzimleri düzenleyen potansiyel miRNA'ların da ekspresyon analizleri yapılmıştır. Bulunan sonuçlar doğrultusunda karaciğer dokularında antioksidan sistemlerdeki değişiklikler miRNA-mRNA-protein-aktivite düzeyleri ile ilişkilendirilmiş, fruktozun patolojisindeki moleküler etki mekanizmasının açığa çıkarılması ve bu süreçte probiyotiklerin etkileri hakkında önemli veriler elde edilmiştir.In recent years, the use of dietary fructose has become quite widespread. Fructose, taken into the liver independently of insulin, can cause pathologies such as insulin resistance, abdominal obesity, inflammation, non-alcoholic fatty liver, and metabolic syndrome. Assorted studies have shown that oxidative stress develops due to an increase in reactive oxygen species in metabolic syndrome, one of the health problems with an increasing incidence. Within the scope of this thesis study, it is aimed to examine oxidative markers in metabolic syndrome caused by high-dose fructose diet, to reveal how antioxidant enzyme systems are regulated, to reveal the effects of some miRNAs that may be involved in this process, and to reveal the regulatory functions of probiotic supplements on the examined parameters. In the thesis study, a metabolic syndrome model was created in the animals by administering a 20% fructose solution in drinking water to male Wistar rats for 15 weeks, and kefir, Lactobacillus helveticus, Lactobacillus plantarum probiotics were administered by gastric tube once a day for six weeks. In liver tissues, antioxidant enzyme activities, protein expressions and gene expressions were analyzed. In addition, while determining how some oxidant and antioxidant parameters such as total thiol, native thiol, disulfide, GSH/GSSG ratio, malondialdehyde, protein carbonyl, total antioxidant and oxidant level are affected by this syndrome, expression analyzes of potential miRNAs that regulate antioxidant enzymes were also performed. In line with the results, changes in antioxidant systems in liver tissues were associated with miRNA-mRNA-protein-activity levels, and important data were obtained in revealing the molecular mechanism of action in the pathology of fructose and the effects of probiotics in this process
Examining the mediating role of sensation seeking in the relationship between basic psychological needs and digital game addiction in adolescents
Dünyada giderek artan dijital oyun kullanımı farklı yaş gruplarında çok fazla kullanıcı sayısına ulaşmıştır. Dijital oyunların aşırı ve zorlayıcı kullanımlarının bireylerde farklı şekillerde olumsuzluklar ortaya çıkardığı düşünülmektedir. Bu araştırmada ergenlerde temel psikolojik ihtiyaçlar ile dijital oyun bağımlılığı arasında heyecan aramanın aracılık rolünün incelenmesi amaçlanmıştır. Araştırma grubu 2022-2023 eğitim öğretim yılında Türkiye’de bir ilde öğrenimine devam eden 266’sı kız (%50,1) ve 265’i erkek (%49,9) toplamda 531 lise öğrencisinden oluşmaktadır. Veriler Dijital Oyun Bağımlılığı Ölçeği, Ergenler İçin Kısa Heyecan Arayışı Ölçeği, Temel Psikolojik İhtiyaçlar Ölçeği-Lise Formu ve Kişisel Bilgi Formu ile elde edilmiştir. Katılımcıların temel psikolojik ihtiyaçları ile dijital oyun bağımlılığı puanları arasında heyecan aramanın aracılık rolü yapısal eşitlik modeli ile incelenmiştir. Araştırmanın değişkenleri arasındaki ilişkiler incelendiğinde, oyun bağımlılığı ile heyecan arama arasında pozitif yönde ve anlamlı bir ilişki elde edilmiştir. Bununla birlikte oyun bağımlılığı ile temel psikolojik ihtiyaçlar alt boyutları arasında negatif anlamlı ilişkiler elde edilmiştir. Yapısal eşitlik modeli bulgularına göre ergenlerde temel psikolojik ihtiyaçlar ile dijital oyun bağımlılığı arasında heyecan aramanın aracılık rolüne sahip olduğu tespit edilmiştir. Daha büyük örneklemler için yapılan bootstrapping yönteminde ergenlerin temel psikolojik ihtiyaçlarının heyecan aramanın aracılığı ile dijital oyun bağımlılığı düzeylerini dolaylı olarak yordadığı sonucuna ulaşılmıştır. Ergenlerde temel psikolojik ihtiyaçlar arttıkça heyecan arama seviyeleri azalmakta ve dijital oyun bağımlılığı seviyeleri azalmaktadır.The increasing use of digital games in the world has reached a large number of users in different age groups. It is thought that the excessive and compulsive use of digital games causes negativities in individuals in differently. In this study, it was aimed to examine the mediating role of sensation seeking between basic psychological needs and digital game addiction in adolescents. The research group consists of a total of 531 high school students (266 girls (50.1%) and 265 boys (49.9%) continuing their education in a city in Turkey in the 2022-2023 academic year. The data were obtained with the Digital Game Addiction Scale, the Adolescents Brief Thrill-Seeking Scale, the Basic Psychological Needs Scale-High School Form, and the Personal Information Form. The mediating role of sensation seeking between the basic psychological needs of the participants and the scores of digital game addiction was examined with the structural equation model. When the relationships between the variables of the study were examined, a positive and significant relationship was obtained between digital game addiction and sensation seeking. In addition, negative significant relationships were found between digital game addiction and basic psychological needs sub-dimensions. According to the structural equation model findings, it was determined that sensation seeking has a mediating role between basic psychological needs and digital game addiction in adolescents. n the bootstrapping method conducted for larger samples; it was concluded that the basic psychological needs of the adolescents indirectly predicted the levels of digital game addiction through sensation seeking. As basic psychological needs increase in adolescents, levels of sensation seeking decrease and levels of digital game addiction decrease
MODELING THE FACTORS INFLUENCING SECONDARY STUDENTS’ PERFORMANCE IN STEM SUBJECTS
STEM education plays a key role in influencing and orienting students’ interests towards STEM fields and careers. This influence is multifaceted, involving not only STEM content and experiences but also teachers, schools, and personal factors. The purpose of this study is to explore the factors impacting the academic performance of 951 tenth grade students in STEM subjects (physics, chemistry, biology, mathematics, and information technologies) who are attending public and private schools in two provinces of Turkey. Using a correlational research model, the study assessed how students’ personal characteristics, teacher effectiveness and school characteristics affect achievement in STEM courses. Data were collected using the ‘Factors Affecting STEM Achievement Questionnaire (FA-STEM-A)’, a 20-item questionnaire divided into three sub-dimensions. Analyses focused on the relationships between student characteristics, teaching quality, school infrastructure and STEM performance. Structural equation modelling revealed that positive student characteristics, especially fondness for STEM subjects and confidence in STEM abilities, moderately increase achievement. In contrast, negative factors attributed to teachers and schools, such as inadequate classroom activities, lack of educator expertise, inadequate technological resources, and large class sizes, hindered student performance. These findings emphasize the important influence of both inherent student qualities and external educational conditions in determining STEM education. © 2024, Scientia Socialis Ltd. Search for supersymmetry in final states with disappearing tracks in proton-proton collisions at ?s=13 TeV
A search is presented for charged, long-lived supersymmetric particles in final states with one or more disappearing tracks. The search is based on data from proton-proton collisions at a center-of-mass energy of 13 TeV collected with the CMS detector at the CERN LHC between 2016 and 2018, corresponding to an integrated luminosity of 137 fb(-1). The search is performed over final states characterized by varying numbers of jets, b-tagged jets, electrons, and muons. The length of signal-candidate tracks in the plane perpendicular to the beam axis is used to characterize the lifetimes of wino- and Higgsino-like charginos produced in the context of the minimal supersymmetric standard model. The dE/dx energy loss of signal-candidate tracks is used to increase the sensitivity to charginos with a large mass and thus a small Lorentz boost. The observed results are found to be statistically consistent with the background-only hypothesis. Limits on the pair-production cross section of gluinos and squarks are presented in the framework of simplified models of supersymmetric particle production and decay, and for electroweakino production based on models of wino and Higgsino dark matter. Optimal Nodes Localization in Wireless Sensor Networks Using Nutcracker Optimizer Algorithms: Istanbul Area
Node localization is a non-deterministic polynomial time (NP-hard) problem in Wireless Sensor Networks (WSN). It involves determining the geographical position of each node in the network. For many applications in WSNs, such as environmental monitoring, security monitoring, health monitoring, and agriculture, precise location of nodes is crucial. As a result of this study, we propose a novel and efficient way to solve this problem without any regard to the environment, as well as without predetermined conditions. This proposed method is based on new proposed Nutcracker Optimization Algorithm (NOA). By utilizing this algorithm, it is possible to maximize coverage rates, decrease node numbers, and maintain connectivity. Several algorithms were used in this study, such as Grey Wolf Optimization (GWO), Kepler Optimization Algorithms (KOA), Harris Hawks Optimizer (HHO), Gradient-Based Optimizer (GBO) and Gazelle Optimization Algorithm (GOA). The node localization was first tested in Istanbul, Turkey, where it was determined to be a suitable study area. As a result of the metaheuristic-based approach and distributed architecture, the study is scalable to large-scale networks. Among these metaheuristic algorithms, NOA, KOA, and GWO have achieved significant performance in terms of coverage rates (CR), achieving coverage rates of 96.15%, 87.76%, and 93.49%, respectively. In terms of their ability to solve sensor node localization problems, these algorithms have proven to be effective. © 2024 The Authors
Earthquake-related trauma and smoking behavior: a cross-sectional study on Kahramanmaraş-Centered earthquakes, Turkey
This study aimed to examine the relationship between earthquake-related trauma severity and smoking status in individuals affected by earthquakes. The study sample consisted of 488 earthquake survivors (57.1% women) older than 18 years old who were placed in dormitories in an earthquake-safe area after the Kahramanmaraş-centered earthquakes in 2023. The study data were collected two months after the earthquakes using an information form and the Scale that Determines the Level of the Trauma after the Earthquake (SDLTAE). The data were analyzed using descriptive statistics and independent sample t tests, one-way ANOVA, and binary logistic regression at the significance level (p <.05). The findings showed that 75.2% of the individuals experienced severe trauma. According to the results of the present study, gender, the need for psychological support, earthquake-related losses, and smoking related to trauma. Being a woman, seeking psychological support, experiencing earthquake-related health problems, facing earthquake-caused material losses, and increased smoking after the earthquake were significant predictors of earthquake-related trauma. This study, conducted two months after the earthquake, aimed to measure the early effects of the earthquakes. Experimental and theoretical results of gamma shielding features for copper based shape memory alloys
In this study, the gamma ray attenuation characteristics for CuAlNi shape memory alloys with different proportions of Sn doping was investigated. We determined the mass attenuation coefficient (mu/rho) of CuAlNiSn alloys both experimentally and theoretically within an energy range of 59.5-1332.5 keV. The experimental measurements were made using a high purity Germanium detector (HPGe) and theoretical calculations were made using WinXCOM program. To evaluate the gamma radiation shielding abilities of the alloy samples, the obtained (mu/rho) values were used to determine the gamma protection parameters mu, HVL, TVL, MFP, and Z(eff). In addition, the radiation protection efficiency (RPE) parameter was determined using gamma ray intensities in the absence and presence of the attenuator. Further analysis of the samples was conducted using a Rigaku Miniflex 600 model computer-controlled X-ray diffractometer (CuK alpha & lambda = 1.5405 A0). The crystallographic structure of the alloys before and after irradiation was investigated. Other analysis which include EDX analysis (to investigate the chemical content) and SEM analysis (to investigate the microstructure) were conducted. According to result outcomes, gamma ray radiation did not affect the shape memory properties of structure. Another interesting observation is that, the radiation attenuation properties increase with increasing Sn concentration. Finally, we discovered that the CuAlNiSn4 alloy (which has the highest doping rate) provides good protection especially at low energy levels
Utilizing genetic programming to evaluate and predict roller-compacted concrete pavements reinforced with coal powder and basalt fiber
Roller-compacted concrete (RCC) pavement technology has drawn significant attention in recent years, thanks to its many advantages over conventional paving materials. The main benefits of RCC include decreased shrinkage, reduced life-cycle expenses, early access to traffic and a decrease in the urban heat island effect. This study proposed a cost-effective method to address the disposal of coal powder (CP), which is an important contributor of environmental problems, by integrating it into the manufacturing of RCC. Cement was replaced with CP at weight ratios of 5%, 10%, 15%, and 20%. CP was substituted for cement, with weight ratios of 5%, 10%, 15% and 20%. The albedo and mechanical properties of the obtained RCC mixtures were measured. The albedo of the RCC mixture with 20% CP decreased approximately 2.3 times in comparison to that of the reference mixture. After 28 days, RCC mixes had compressive strengths of 26-38 MPa and flexural strengths of 4.3-6.1 MPa. Flexural and splitting tensile strengths increased substantially with BF. RCC mixtures with 5% CP and 0.5% BF improved freeze-thaw resilience after 100 cycles. The genetic algorithm approach may improve RCC design optimisation. Investigation of antibacterial activities of copper based multinary sulfide alloys
One of the nanomaterial-based antimicrobials, CuS, has emerged as a potential active therapeutic agent to combat biofilm infections, showing superior antibacterial activity with both photothermal and photodynamic effects under NIR illumination. To investigate the influence of transition metals on the antibacterial activity of CuxS, the antibacterial properties of shape- and phase- controlled copper-based multinary sulfide (M: CuxS (M: Mn, Zn and Ni)) nanostructures prepared by hot injection method were investigated against Gram-positive Staphylococcus aureus and Gram-negative Escherichia coli under NIR light irradiation. It has been shown that CuxS-based nanomaterials suppress bacterial growth under NIR light irradiation and that, in addition to their antibacterial activity, they also exhibit a remarkable ability to form biofilms. The antibiofilm efficacy of the M: CuxS (M: Mn, Zn and Ni) nanocomposites was significantly higher than that of bare CuS, resulting in biofilm inhibition with rates of 52.53 %, 62.24 % and 74.48 % on E. coli for Mn:CuxS, Zn:CuxS and Ni:CuxS, respectively. In addition, the biofilm of S. aureus was more affected by treatment with M:CuxS (M: Mn, Zn and Ni) nano- composites, with biofilm activity of 64.15 %, 68.42 % and 87.35 % for Mn:CuxS, Zn:CuxS and Ni:CuxS, respectively. Ni:CuxS showed the highest antibacterial activity, which is attributed to smaller crystal and particle size and energy band gap, resulting in a larger number of nanoparticles per unit volume, a larger surface area and a stronger tendency to electrostatically bind to bacterial cell membranes, a faster ability to penetrate channels and stronger absorption in the NIR range. Determination of student attitudes towards distance education platforms in science courses
Birçok alanda olduğu gibi uzaktan eğitim sistemini etkileyen teknolojideki gelişmeler uzaktan eğitim ortamında kullanılan platformlara yön vermektedir. Bu çalışmada uzaktan eğitim ortamında fen (fizik, kimya, biyoloji) derslerini öğrenmede kullanılan platformların öğrencilerin tutumları üzerindeki etkileri belirlenmiştir. Araştırmaya bir lisedeki 9. ve 11. sınıflardan toplam 79 öğrenci katılmıştır. Çalışmada nicel ve nitel veriler birlikte kullanılmıştır. Google Form ortamında karma yöntem anketi uygulanmış ve böylece nicel veriler bir ölçek üzerinden nitel veriler ise açık uçlu soruya verilen cevaplardan toplanmıştır. Verilerin analizinde betimsel istatistikler (aritmetik ortalama, standart sapma, yüzde ve frekans), kestirimsel istatistikler (Mann-Whitney U testi ve t testi) ve betimsel analiz uygulanmıştır. Analizler, öğrencilerin uzaktan eğitimde fen derslerini öğrenmek için kullandıkları platforma yönelik tutumlarının düşük düzeyde olduğunu göstermiştir. Platformun kullanımıyla ilgili sorun yaşamayan öğrenciler öğrenmede ve derslerden verim almada sorun yaşamışlar, kullanılan platformun öğrencilerin motivasyonlarına ve özgüvenlerine olumlu bir etkisi olmamıştır. Bu araştırmanın, uzaktan eğitim ve uzaktan eğitim platformlarının daha etkili olmasına yönelik katkı sunması beklenmektedir.As in many fields, developments in technology affecting the distance education system give direction to the platforms used in distance education environment. In this study, the effects of the platforms used in learning science (physics, chemistry, biology) courses in distance education environment on students' attitudes were determined. A total of 79 students from 9th and 11th grades in a high school participated in the study. Quantitative and qualitative data were used together in the study. A mixed-method questionnaire was applied in Google Form and thus quantitative data were collected through a scale and qualitative data were collected from the answers given to an open-ended question. In the analysis of the data, descriptive statistics (arithmetic mean, standard deviation, percentage, and frequency), inferential statistics (Mann-Whitney U test and t test) and descriptive analysis were applied. The analyses showed that students' attitudes towards the platform they used to learn science courses in distance education were low. Students not having problems with the use of the platform had problems in learning and getting efficiency from the lessons, and the platform did not have a positive effect on students' motivation and self-confidence.
