Structural mapping of oligomeric intermediates in an amyloid assembly pathway - dataset
Transiently populated oligomers are commonly formed in the early stages of amyloid assembly. Determining the structure(s) of these species and defining their role(s) in assembly are key to developing routes to prevent or delay disease. Here, using a combination of chemical kinetics, NMR spectroscopy, native mass spectrometry and other biophysical methods, we identify and structurally characterise the oligomers required for amyloid assembly of the naturally occurring variant of β2-microglobulin, DN6. The results reveal a remarkably specific assembly pathway involving the formation hexamers that are non-toxic and retain a native-like immunoglobulin fold, but in which the C-terminal strand is unfurled ready to initiate the transition into the amyloid fold. The results suggest new avenues to combat disease by specific targeting of the early intermediates in amyloid assembly
Nanoparticle loaded hydrogel for the triggered release of liposomal antimicrobial peptides
This data contains information regarding the development of a liposome encapsulating an antimicrobial peptide (2a-3b) and the protection the liposome provides against proteolytic degradation (4). The data also described the photothermal properties of a gel containing peptide liposomes and Auɴʀs (5a, 5b) as well as their release profiles (5d) and toxicities of the gel and components (6a, 6b). Thedata also describes the use of the gel to treat S. aureus (7a, 8) and P. aeruginosa (7b). The remaining data describe the supplementary materials (S1-S5)
Public perceptions of how to reduce carbon footprints of consumer food choices (dataset)
Carbon footprints associated with consumer food choices substantially contribute to climate change. Life cycle analyses from climate and environmental sciences have identified effective rules for reducing these food-related carbon footprints, including eating seasonal produce and replacing dairy and red meat with plant-based products. This dataset was obtained in a national UK survey conducted in January 2018, where we explored how many and which rules N = 627 participants generated for reducing carbon footprints of food (Carbon_footprint_recoded_July2019.RData). The sample was recruited via the UK-based survey company ResearchNow and the survey was programmed in the survey software Qualtrics. Participants were randomly allocated to one of the three food groups produce, dairy, and protein-rich products. In an open-ended question, we asked them to first list the characteristics (or ‘rules’) they consider as typical for produce /dairy/ protein products with a low carbon footprint. They were then asked to rate how effective they perceived each of their generated rules to be, on a 1-7-scale. Participants then made numerical estimates of carbon footprint reductions associated with four pre-selected rules in their assigned food group, expressed in either grams or percentages. They were asked how confident they were into each of their estimates, on a 1-7 scale. Subsequently, we measured their pro-environmental worldviews (Dunlap et al., 2000; 15 items), their Climate Change Knowledge (Shi et al. 2015; 30 items), their levels of Numeracy (Cokely et al. 2012; adaptive version), and their willingness to think about complex problems (‘Need for Cognition’; Cacioppo et al. 1984; 18 items. All survey items are described in the Supplemental Materials of Kause, Bruine de Bruin, Millward-Hopkins & Olsson 2019, and in the document Survey_CarbonFootprint_Food.docx). In a set of linear and logistic regression models (AnalysisRQ1a-2_RevisionJune2019.R and README) we found that participants generated few and relatively less effective rules, including ambiguous ones like ‘Buy local’. Furthermore, participants’ numerical estimates of pre-selected rules were less accurate when they assessed carbon footprint reductions in grams rather than in percentages, as compared to estimates we obtained from life cycle analyses from climate and environmental sciences, conducted by other authors from those disciplines (see Supplemental Materials of Kause, Bruine de bruin, Millward-Hopkins & Olsson, 2019). Surveys for Urban Equity
This dataset contains results and documentation from three cross-sectional urban household surveys done in Kathmandu (Nepal), Dhaka (Bangladesh) and Hanoi (Vietnam) in 2017 and 2018. The surveys primarily aimed to test the feasibility of using new urban household survey methods that try to better cover/capture informal/slum settlements using sampling frame data generated from random forest models that incorporate census data (which is often outdated and inaccurate) with multiple remotely-sensed covariates, such as urbanisation and infrastructure data. Additionally, the surveys also aimed to gather data on a range of topics including many that are not commonly collected in household surveys, particularly of urban areas: A) basic socio-demographic details of household members, B) household characteristics, assets, income and expenses, C) household migration and social capital, D) household member injury and injury related death, and, for one individual per household, E) migration, social capital and depression/mental health. Antibacterial properties and regenerative potential of Sr2+ and Ce3+ doped fluorapatites; a potential solution for peri-implantitis
Scaffolds and implants in orthopaedics and regenerative dentistry usually fail because of bacterial infections. A promising solution would be the development of biomaterials with both significant regenerative potential and enhanced antibacterial activity. Working towards this direction, fluorapatite was synthesised and doped with Sr2+ and Ce3+ ions in order to tailor its properties. After experiments with four common bacteria (i.e. E. Coli, S. Aureus, B. Subtilis, B. Cereus), it was found that the undoped and the Ce3+ doped fluorapatites present better antibacterial response than the Sr2+ doped material. The synthesised minerals were incorporated into chitosan scaffolds and tested with Dental Pulp Stem Cells (DPSCs) to check their regenerative potential. As was expected, the scaffolds containing Sr2+-doped fluorapatite, presented high osteoconductivity leading to the differentiation of the DPSCs into osteoblasts. Similar results were obtained for the Ce3+-doped material, since both the concentration of osteocalcin and the RUNX2 gene expression were considerably higher than that for the un-doped mineral. Overall, it was shown that doping with Ce3+ retains the good antibacterial profile of fluorapatite and enhances its regenerative potential, which makes it a promising option for dealing with conditions where healing of hard tissues is compromised by bacterial contamination

This archive contains the dataset associated with the publication entitled ‘High-Speed Modulation of a Terahertz Quantum Cascade Laser by Coherent Acoustic Phonon Pulses'. Fast modulation of lasers is a fundamental requirement for applications in optical communications, high-resolution spectroscopy and metrology. In the terahertz-frequency range, the quantum-cascade laser (QCL) is a high-power source with the potential for high-frequency modulation. However, conventional electronic modulation is limited fundamentally by parasitic device impedance, and so alternative physical processes must be exploited to modulate the QCL gain on ultrafast timescales. Here, we demonstrate an alternative mechanism to modulate the emission from a QCL device, whereby optically-generated acoustic phonon pulses are used to perturb the QCL bandstructure, enabling fast amplitude modulation that can be controlled using the QCL drive current or strain pulse amplitude, to a maximum modulation depth of 6% in our experiment. We show that this modulation can be explained using perturbation theory analysis. While the modulation rise-time was limited to ~800 ps by our measurement system, theoretical considerations suggest considerably faster modulation could be possible.
