    HPLC Detection of Aflatoxin in Meat, Poultry, and Fish and their Products and Detoxification by Gamma Radiation

    One of the most desired and promising diets in Egypt is beef products. It is an enriched media for mycotoxins. The occurrence of aflatoxigenic Aspergillus species is inspected in commercial beef products, HPLC-quantitative amount of aflatoxin B1, B2, G1, and G2 excesses, and genetic identification of aflatoxin regulatory gene (aflR1) by conventional PCR. Two hundred and forty commercial products (minced meat, beef kofta, beef sausage, beef burger, beef luncheon, frozen meat, beef frozen liver, chicken luncheon, chicken burger, chicken frozen liver, mloha, and fesikh; n=20 for each) were collected from different markets at Aswan City, Egypt. Enumeration, isolation, and identification of mold species were carried to each sample. The amount of aflatoxins was measured using HPLC. Genetic identification of the aflR1 gene in Aspergillus was performed using PCR. Mloha samples recorded the highest total mold count whereas the beef luncheon recorded the lowest mould count. Four fungal genera were identified and Aspergillus spp recorded the main with an incidence of 25.8%. By PCR, the aflR1 gene was productively augmented in all the tested Aspergillus spp. The findings illustrated that among the samples that were examined; the prevalence of AFB1 was 65%, followed by AFG2 at 63%, AFB2 at 40%, and AFG1 at 30%. Additionally, mloha (724.2±14.6), poultry frozen liver (288±6.7), and beef frozen liver (91.6±12.2) had higher mean values of total aflatoxins contamination than other samples. Every sample that has been analyzed shows a positive correlation between the amount of reduced total mycotoxins found in the samples and the increased dose of gamma irradiation used to treat the samples. Conclusion: Aflatoxin is frequently linked to meat, poultry, and fish, as well as the products made from these foods. The production of aflatoxin in meat, as well as the products made from it, creates a danger to the public’s health. Thus, the most effective way to prevent aflatoxigenic mould contamination during the product’s production stages is to apply stringent hygienic standards when processing meat products and to use high-quality flavoring agents as spices.     Ameliorative Effect of L-Carnitine against Hematological and Hepatorenal Alterations Induced by Cefquinome Sulfate in Male Albino Rats

    The present study was conducted to evaluate the ameliorative effect of L-carnitine (LC) against possible deleterious effects of cefquinome sulfate (CS) on hematological, hepatorenal parameters and histopathological changes of some internal organs in male rat's model. Therefore, sixty mature male rats were equally assigned into four groups as following: The control group of rats was administered with two subcutaneous (SC) injections of physiological saline (vehicle of other drugs) (2ml/kg. b.wt once daily for 5 successive days and 2ml/kg. b.wt once daily for 56 successive days), the second group was injected subcutaneously with LC (200 mg/kg b.wt. once daily for 56 successive days), the third group was administered with SC injection with CS (50 mg/kg. b.wt once daily for 5 successive days) and the fourth group was injected subcutaneously with CS concomitantly with LC by the same doses at the same treatment periods as mentioned in 2nd and 3rd groups. The obtained results showed that administration of CS induced a significant (P≤ 0.05) increase in WBCs count and serum levels of alanine amino transferase, alkaline phosphatase and creatinine with significant (P≤ 0.05) decrease in PCV value and serum levels of albumin. Moreover, CS induced histopathological changes on liver and kidney tissues. Conversely, administration of LC concomitantly with CS ameliorates all previous hematological, biochemical, and histopathological alterations induced by CS. It could be concluded that CS induces mild to moderate troubles on blood picture, hepato-renal indices and histopathological findings which could be ameliorated by L-carnitine in male albino rats

    Genetic Diversity of Multiple Antibiotic Resistance Streptococcus agalactiae Isolated from Bovine Mastitis and Retail Markets Milk by Enterobacterial Repetitive Intergenic Consensus PCR (ERIC-PCR)

    Public health is at risk because Streptococcus agalactiae is increasingly linked to incidences of bovine mastitis in Egypt. In this study, enterobacterial repetitive intergenic consensus PCR (ERIC-PCR) were used to explore the genotyping of several antibiotic resistant Streptococcus agalactiae isolated from bovine mastitis and retail milk. Also, antimicrobial resistance genes were detected. Two hundred and sixty-six (46.7%) strains were isolated from samples of milk obtained from cases of bovine mastitis in dairy farms and retail markets representing 34 (5.96%) Streptococcus agalactiae and 232 (40.7%) other Streptococcus species strains based bacterial identification. By using of disc diffusion assays, it was examined the susceptibility of all Streptococcus isolates to twelve antimicrobial agents. The   highest prevalence of resistance of Streptococcus species was observed against ampicillin (65.5%), amoxicillin (56%), tetracycline (52.5%), ofloxacin (47.8%), and nitrofurantoin (46.9%). High proportion of the S. agalactiae isolates were resistance to amoxicillin (83%), followed by tetracycline (82.4%), nitrofurantoin (64.7%), azithromycin (61.8%), and ampicillin (50%). Most (88.2%) of S. agalactiae showed multiple antibiotic resistances (MAR) phenotypes with MAR index of 0.1-0.7 and 28 different MAR patterns. The results of genetic antimicrobial resistance of S. agalactiae strains revealed amplification of blaTEM (23%) and blaCTX (26%) genes in β-lactam-resistant strains, erm(B) gene (20%), mef(A) (35%) in macrolides resistant isolates, and tet(M) in  (44%) tetracycline resistant strains. Using ERIC-PCR, the present study showed the genetic diversity and heterogeneity among S. agalactiae strains (n=34) that were classified into 19 distinct ERIC-PCR groups (A–S).  Between them, ERIC O (20.6%, 7) was the most widespread group. These results indicated that milk samples served as source of MAR S. agalactiae, consequently posing threats to public health, so the improvement of the hygiene regimen on the farms and promotion of the wise use of antimicrobials are necessary. The obtained findings showed that milk samples were a source of MAR S. agalactiae, endangering public health. As a result, it is essential to enhance farm hygiene practices and promote the responsible use of antimicrobials. Additional research on the epidemiology of S. agalactiae is required to add bacterial genetic information in order to help in rational vaccine strategy in the future

    Prevalence of Multidrug-resistant Enterotoxigenic Staphylococcus aureus in Pagrus and Saurus Fish Intended for Human Consumption

    Staphylococcus aureus (S. aureus) is considered the most important cause of food borne intoxication, which occurs due to ingesting of food contaminated with enterotoxin of S. aureus. In this study, S. aureus from saurus, and pagrus fish species were isolated, then identified by morphological and biochemical examination. S. aureus coagulase, and D-Nase +ve were detected in 16 from 25 (64%), and 17 from 25(68%) of the examined samples of saurus, and pagrus, respectively. Total S. aureus counts were 5.14±0.06 in saurus, and 5.02±0.03 log 10 cfu/g in pagrus fish, respectively. Detection and typing of enterotoxin by Reverse Passive Latex Agglutination technique "RPLA" revealed the detection of staphylococcal enterotoxins (SEs) SEA, SEC, and SED at 18%, 6%, 0% in saurus and 5%, 0%, and 11%, at pagrus, respectively. S. aureus was tested for antimicrobial susceptibility. The recovered S. aureus coagulase, and D-Nase +ve in the current study showed resistance to kanamycin, clindamycin, nalidixic acid, and sulphamethoxazole at 100%, 87.9%, 84.8%, and 81.8%, respectively. The isolates showed sensitivity to amikacin, imipenem, meropenem, oxacillin at 90.9%, 87.9%, 84.8%, and 81.8% respectively. It could be concluded that multidrug resistant enterotoxigenic strains of S. aureus could be isolated from saurus and pagrus spp. Therefore, it is highly recommended to adopt strict hygienic measures and efficient cooking during before consumption of such fish species

    Molecular Identification of Dermo-Mycotic Infection and the Effect of Dietary-Essential Oils on Broiler Chickens in Upper Egypt

    The importance of searching for natural alternatives away from chemicals in poultry health and treatment has benefits for humans in many directions, as we control the bad effect of the accumulation of harmful chemicals in their meat, as well as reduce the risk of zoonotic infection and preserve the environment from chemical pollution. Enormous fungi induce a considerable level of annihilation in the poultry industry and human consumers due to their zoonotic implications. This study is designed to explore the effects of keratogenic and toxigenic skin fungal affection and the effects of dietary-essential oils in broilers -chicken (n-120). Skin scrapings and feather samples were examined mycologically in association with PCR sequencing for genomes of the culturally detected fungi (in South Korea) based on phylum tree and all Sequences data was deposited in GenBank and each was assigned an accession number. Sera samples of the tested broilers were examined by ELIZA against biogenic amine mainly histamine during the summer season, also a histopathological examination of skin sections before and after taking feed additives (essential oils) as anti-fungal for thirty days, the broiler-fed diet was supplemented with peppermint, thyme, and Carvacrol 70 mg/kg (w/w) in dietary feed. The isolated fungi were: Fifteen fungal species belonging to 9 genera of filamentous fungi which were isolated from skin scrapings and feathers of chickens. Aspergillus niger and A. flavus are the most prevalent species (20 samples representing 100% of total samples for each. Rhizopus oryze 20% and Fusarium oxysporum 15% were cultured from total samples respectively. Four fungal species appeared in 10% of the tested samples which are Aspergillus qudrilineatus, Paecilomyces variotii (Byssochlamys spectabilis), Scopulariopsis brevicaulis and Exserohilum rostratum. Finally, the other seven fungi presented as 5% from tested samples. The average level of serum histamine before treatment was 16.6 ng/ml and after feeding was 12.3 ng/ml (significant decrease, P < 0.05) referring to the significant role of the essential oils in broilers ration

    Cross Sectional, Seroprevalence Study of Peste des Petits Ruminants and the Related Risk Factors During Outbreak in Goats’ Farm in Egypt

    Peste des Petits ruminants' virus (PPRV) is a notifiable transboundary and economically significant viral disease that affects goats and sheep. The current study was conducted to identify the seroprevalences of PPRV in goat farm in Marsa-Matroh province, Egypt during an outbreak in 2022. Moreover, this work aims to study the relevant risk factors directly related to the virus infection and attempt molecular characterization and phylogenetic analysis of the circulating strain. The samples included 356 sera, 154 nasal swabs, and 10 tissue samples were collected for PPRV screening and molecular characterization. The seroprevalence percent was found of 42.69%.  It was observed that the prevalence rate, and case-fatality rate were higher under 6 months of age than in adults, and referring to sex, females had a more significant disease incidence than males. Based on phylogenetic analysis; the strains of the current study: PPR/AHRI-Matrouh1/Egy/2022 (accession number: OP881991) and PPR/AHRI-Matrouh2/Egy/2022 (accession number: OP881992) were identified as PPRV lineage IV, with 99.2% and 98.8% identity to the Ethiopian strain (Accession number MK571524) and Sudanese strain (Accession no HQ131931) respectively. An efficient PPR vaccination program with rigorous quarantine measures at the borders is advised to be implemented in the country to control the spread of the disease and avoid the entry of novel strains into the Egyptian governorates

    The Ameliorative Potential of Vitamin E and Selenium on the Possible Adverse Effects of Azithromycin

    Azithromycin, an antibiotic belonging to the macrolide group, is used not only for bacterial infections, but also for fungal and viral diseases. The main side effects of its use are hepatic and renal toxicities. The present study was carried out to evaluate the possible ameliorative effect of vitamin E and selenium against azithromycin-induced hepatic and renal toxicities. Fifty male albino rats used were allocated into 5 equal groups, each of 10 rats. All the treatments were taken orally for ten days in all groups.  Group I served as a normal control. Group II received olive oil. Group III received azithromycin (6mg/kg bwt). Groups IV and V received vitamin E and sodium selenite at doses of 100 mg/kg bwt and 0.3mg/kg bwt, respectively at two hours before azithromycin administration. Two blood samples were collected from each rat, one on heparin and the other without anticoagulant to obtain clear sera for hematological studies and biochemical analysis respectively. Liver and kidney tissues were collected for histopathological examination. Vitamin E or selenium administrations significantly decreased alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) and Gamma- glutamyle transferase (GGT) activities, creatinine, urea and uric acid levels, and MDA concentration in azithromycin-treated rats. Moreover, both Vitamin E and selenium significantly increased hematological parameters, protein profile (total protein, albumin, and globulin) and antioxidant enzyme activities (GPX, SOD, GSH, CAT) in rats treated with azithromycin with a significant improvement in the histopathological pictures of the hepatic and renal tissues. Based on these findings, vitamin E and selenium may be beneficial agents for protection against liver and kidney toxicities induced by azithromycin

    Cytoprotective Effects of Nigella sativa Seeds on Monosodium Glutamate Induced Seminal Vesicle Damages: Histological and Immunohistochemical Studies

    Monosodium glutamate (MSG) is a worldwide food flavour enhancer commonly used by the food industry. This feed additive may cause male infertility. Nigella sativa seeds (NSS) is a widely used in herbal medicine as it has many biological benefits and could provide a solution. This work was designed to investigate the histological effects of NSS on rats ingesting MSG. To achieve this aim, adult male albino rats (2- 3 months old) were randomly and equally assigned into three experimental groups. For a period of 21 days, control group received no treatment, MSG group received MSG as 30 g/kg feed, and MSG + NSS group received MSG as 30 g/kg feed and NSS as 30 g/kg feed. Seminal vesicle histopathology in MSG group showed mild seminal vesiculitis with degeneration of smooth muscle fibers in tunica muscularis. In addition, there was an increase in the amount of connective tissue and apoptotic cells count. Periodic Acid Schiff stain indicated irregular and interrupted epithelial basement membranes. Glutathione reductase (GR), superoxide dismutase 2 (SOD2), and caspase-3 immuno-expressions increased in MSG group. It was found that there was an increase in the number of apoptotic cells, intraepithelial lymphocytes and dendritic cells in MSG group. However, treatment with NSS ameliorated these disturbances. NSS mitigated MSG-induced seminal vesicle damage by its histoprotective, cytoprotective and anti-apoptotic activities

    Antimicrobial Resistant Profile of Bacterial Pathogen Isolated from Macaque species Rescued in the Center for Rescue, Conservation and Creature Development, Phong Nha-Ke Bang Nation Park, Vietnam

    Macaque species play important roles in the cultures, and religions of many societies. They are an essential component of tropical biodiversity, contributing to forest regeneration and ecosystem health. The close phylogenetic relationship between humans and macaque species also creates a high potential for pathogen exchange. A total of 228 macaques which belong to four species, including Macaca arctoides, Macaca leonine, Macaca assamensis, and Macaca mulatta, were rescued in the Center Rescue, Conservation and Creature Development, Phong Nha-Ke Bang National Park (PN-KB NP). Of 228 macaques, 149 (65.4%) individuals successfully reintegrated into the wild. The prevalence and the antimicrobial resistance (AMR) profile of Escherichia coli (E. coli), Salmonella, and Staphylococcus aureus (S. aureus) isolates from macaques rescuing in the Center were investigated. The fecal and nasal samples from 19 macaques were collected. These samples were positive for E. coli (73,7%), Salmonella (36.8%), and S. aureus (57.9%). All of the tested bacterial strains showed 100% resistance to penicillin and vancomycin. The multi-drug resistant (MDR) profile was observed in S. aureus (71,4%), E. coli (95,3%), and Salmonella (100%). This is the first report on the rescue and natural reintegration of the macaque species status in Vietnam and the prevalence of AMR in zoonotic bacterial pathogens isolated from these macaques. This result indicated that AMR of the zoonotic bacterial pathogens could colonize in macaques and may transmit to humans

    Self-presentation of the Veterinarian: Does it Affect Clients' Perception in Bosnia and Herzegovina?

    Parallel with professional skills, verbal and non-verbal communication should be mastered by veterinarians in order to improve the quality of the “veterinarian-owner-patient†relationship. Non-verbal communication implies facial expressions and attitude, but professional’s attire can be an influencing factor as well. The effect that veterinarian self-presentation has on clients’ perceptions of veterinarians was not investigated previously in Bosnia and Herzegovina (B&H). Therefore, the aim of this study was to examine the perception of animal owners from B&H regarding veterinarians’ verbal and nonverbal communication skills, presentation on social media and professional attire. The questionnaire was compiled in Google form and consisted of an introduction and 13 questions followed by an explanation. A total of 648 clients participated in the survey between February and April 2023. Obtained data were processed in Microsoft Excel using descriptive statistics. Results of our survey indicate that veterinarians’ attire, along with verbal communication, plays a significant role in gaining trust and comfort from animal owners in B&H. Given that it is currently not present, we recommend inclusion of verbal and non-verbal communication skills as a mandatory subject or module in the undergraduate study curriculum of veterinary medicine in B&H
