Intestinal histostructure and immune protection activity of age -1+ carp after consumption of a prebiotic feed additive
Currently, it is important in aquaculture to study and use products and additives, which have positive effects on the digestibility of nutrients by helping to normalize the intestinal microflora, thereby modulating fish immune response. The aim of the study was to assess the effect of a prebiotic based on mannan oligosaccharides produced from the outer walls of Saccharomyces cerevisiae cells on the histological structure, morphometric state of the intestine and activity of cellular and humoral components of nonspecific resistance of age -2+ carp. The experiment was conducted in four analogous ponds, which were stocked with age-1 scaly carp with an average weight of 55–58 g with a stocking density of 1000 fish/ha. Carp had been fed balanced compound feed for 60 days. Experimental groups of fish were fed with a prebiotic as a feed additive in the amount of: 0.025 % – Experiment 1, 0.05 % – Experiment 2 and 0.075 % – Experiment 3. Use of the studied prebiotic did not damage the intestinal histological structure of carp of the experimental groups. The villi height and intestinal crypt depth of carp in Experiment 1 (P < 0.05) and Experiment 2 (P < 0.01) were larger compared to the control group. Experiment 3 showed destructive changes in intestinal mucosa. The results of histological examination indicated the effectiveness of the use of the studied prebiotic in the amount of 0.025 and 0.05 % in the diet of age -1+ carp due to the absence of pathological changes and active morphofunctional state of the intestine, which, in turn, had a positive effect on feed absorption and metabolic processes in fish body. Analysis of the indicators of cellular and humoral links of natural resistance showed that in carp of the second and third experimental groups lysozyme activity of serum was 5 and 11 % (P < 0.001) higher than in the control group. Similar changes, though found in a greater extent, were recorded in the study of bactericidal activity of blood serum, especially in individuals of the second and third experimental groups (by 6.8 (P < 0.01) and 15.2 %). While with carps of the third experimental group, which used a prebiotic drug in the amount of 0.075 %, the content of CEC was 20.5 % higher than in the control group, which indicates an additional antigenic load on the body and is an unfavorable diagnostic factor. At the same time, with the fish of the second experimental group, which received, respectively, 0.05 % of the supplement of the drug “Actigen” to the main diet, probably higher phagocytic activity of blood neutrophils was recorded. A direct dependence of the growth of the phagocytic index and the phagocytic number of neutrophils on the dose of the studied prebiotic was detected. Immunological studies indicated a dose-dependent effect of the prebiotic on the activity of cellular and humoral components of non-specific resistance of carp. Given this, for the effective functioning of innate protective mechanisms, for the increase of the body's immune potential and productivity, it is most appropriate to apply to the diet of carp prebiotic supplement “Actigen” in the amount of 0.025 and 0.05 %
The comparative evaluation of complex drugs based on fipronil for ectoparasitosis of dogs and cats
The article presents data on the effectiveness of new domestic antiparasitic drug “Insectostop for dogs and cats” in comparison to the reference drugs “Burdi Fipro for dogs” and “Burdi Fipro for cats” that is used for therapeutic and prophylactic purposes against ectoparasitosis of dogs and cats of different breeds. According to the results of the research, the parasitism of fleas of Ctenocephalus canis, C. felis and Pulex irritans species and parasitiform mites of the Ixodes ricinus and Dermacentor reticulatus species and sarcoptiform mites of the species Otodectes cynotis were detected on experimental animals. Clinically, this was inspected by redness, inflammation of the skin, itching reflex, the emergence of papules on the skin, scales. Based on the results, it was found that 8 hours after usage of the experimental drug “Insectostop for dogs and cats” its effectiveness in syphonapterosis of dogs was 87.9 %, and after usage of the reference drug “Bourdie Fipro for dogs” – 86.9 %, and in experiments on cats, the effectiveness of the experimental drug for siphonapterosis in cats was 92.4 %, and the reference drug “Bourdie Fipro for cats” – 90.3 %. Starting from the first and third days after usage of drugs on the fur of dogs and cats of the experimental and control groups, parasitological studies did not reveal fleas of the species Ctenocephalus canis, C. felis and Pulex irritans. So starting from the first day, the experimental and reference drugs showed 100 % effectiveness during the siphonapterosis in dogs and cats. During the ixodidosis of dogs, it was found that at the 8-th hour after usage of the experimental drug “Insectostop for dogs and cats” its effectiveness was 82.9 %, and after usage of the reference drug “Bourdie Fipro for dogs” – 81.3 %. At the 24-th hour after usage of the experimental drug, its effectiveness was 97.6 %, and after usage of the reference drug – 95.9 %. At the 72nd hour of the experiment, no adult representatives of the species Ixodes ricinus and Dermacentor reticulatus were found on the fur of dogs of these groups. Thus, at 72 hours after usage of the experimental and reference drug, they showed 100 % efficiency on adult Ixodes mites. During the Otodectesis of dogs it was found that on the 7th day, effectiveness of the experimental drug was 75.6 %, on the 14th day – 98.1 % and after usage of the reference drug “Bourdi Fipro for dogs” its effectiveness on the 7th day was 73.5 %, and on the 14th – 98.3 %. At 21-st days after treatment of animals with drugs as a result of clinical examination and parasitological examination of dogs mites of the species Otodectes cynotis were not detected. In experiments on cats, on the 7th day after usage of experimental drug, its effectiveness during Otodectesis of cats was 76.9 %, and after usage of reference drug – 77.1 %. As a result of clinical examination and parasitological study of the experimental and control groups of animals on the 14th day after treatment of the auricles of cats with mites of the species Otodectes cynotis was not detected. Thus, usage of both drugs promotes the release of Otodectesis in dogs from parasites on the 21st day of the experiment, and cats – on the 14th day. Tests have shown that the experimental drug “Insectostop for dogs and cats” (100 ml of the drug contains the active substance: fipronil – 10 g) does not cause skin irritation, dermatitis, seborrhea, allergic and other side effects, that the drug is well tolerated by dogs and cats and doesn’t give any side effects and changes of clinical condition of animals
Histostructure of the gray matter of the spinal cord in cattle (Bos Taurus)
The scientific article presents the results of investigating the spinal cord`s morphology of a domestic bull (Bos Taurus). Data on the histo- and cytostructure of the spinal cord are given according to the results of histological, neurohistological, and morphometric studies. For their implementation, the selected material (spinal cord n = 8) was subjected to fixation in 10–12 % neutral formalin solution, followed by pouring into paraffin. Histological sections were made from paraffin blocks on a sliding microtome MS-2 with a thickness of not more than ten μm. Staining of sections with hematoxylin and eosin, according to Van Gieson's methods, as well as neuro-histological methods of impregnation of nerve tissue with silver nitrate according to the Bilshovskym-Gross method, was used for the morphometric studies, investigating the morphology of the cell, conducting and obtaining the review histological preparations. The histostructure of the spinal cord, the localization of neurons in the gray matter, and morphometric studies of structural elements were examined on histological specimens by light microscopy. The entire experimental part of the research was conducted following the requirements of the international principles of the "European Convention for the Protection of Vertebrate Animals for Experimental and Other Scientific Purposes" (Strasbourg, 1986). The spinal cord, medulla spinalis, an organ of the central nervous system of vertebrates, is located in the spinal canal. The spinal cord is protected externally by soft, arachnoid, and hard meninges. The space between the membranes and the spinal canal is filled with cerebrospinal fluid. It is well known that groups of multipolar nerve cells with the same functional value form the nuclei of the gray matter of the spinal cord. According to the results of our histological studies, a pronounced differentiation of nerve cells, which have different shapes and sizes. Among them are large, medium, and small nerve cells. The shape of nerve cells is different, which, in turn, depends on their location in certain areas of the gray matter of the spinal cord and the size of the cell. In general, there are multifaceted, stellate, spindle-shaped, elongated, rounded, and oval neurons. Small nerve cells have an oval or round, less often – irregularly rounded shape, medium – round, oval, spindle-shaped. Large nerve cells are dominated by a multifaceted shape with distinct processes. The nuclei of large nerve cells, in most cases, have a rounded shape, less often – oval, mostly in the center of the cells, seldom – eccentrically. According to the results of morphological studies, it is noted that the neurons of the gray matter of the spinal cord have different shapes and sizes. Consequently, in the gray matter, small cells are the highest quantity (47.91 ± 0.32 %) of the total number of nerve cells. The second place is occupied by average neurons (33.70 ± 0.46 %). The large cells are detected in the smallest amount (18.37 ± 0.50 %)
Light-scattering properties of microorganisms Desulfuromonas acetoxidans by influence of silver
The article deals with the concentration changes and relative content of bacterial cells of Desulfuromonas acetoxidans in the intervals of sizes 0.2–2.0 µm under the influence of nano silver particles. Correlation between these changes of light-scattering properties of bacterial cells and growth abilities of bacteria Desulfuromonas acetoxidans under inflence of silver nanoparticles and ions has been shown. The purpose of the work was to research the intensity of processes the change of indexes of the antioxidant system the cells of Desulfuromonas acetoxidans at influence of silver nanoparticles and silver nitrate. The influence of various concentrations of silver nanoparticles and silver nitrate on enzymatic activity of catalase and reduced glutathione synthesis by Desulfuromonas аcetoxidans cells under their cultivation with fumarate addition and with absence of sulphur has been determined. Specific catalase activity increased with enhancing of concentration and duration of bacterial cultivation under the addition of this salt. The highest specific catalase activity was determined on the second day of bacterial growth under the influence of all concentration range of investigated metal salt. The reduced glutathione content under silver nitrate and silver nanoparticles exposure varied depending on the cultivation time and metal concentration. The maximum reduced glutathione content has been observed. The result of catalase activity changes and glutathione content changes of sulfur-reducing D. acetoxidans bacteria cell-free extracts and has been investigated under the influence of different concentrations of Ag nanoparticles during four days of cultivation has been investigated
Features of the macro and microscopic structure of the lungs of a mature rabbit – Oryctolagus cuniculus L. 1758
The macro- and microscopic structure of the lungs of mature rabbits – Oryctolagus cuniculus L., 1758 – European rabbit was studied. This work was performed using comprehensive research methods: anatomical, organometric, histological and histometric. The research was carried out in the conditions of the laboratory of pathomorphology in the Department of Anatomy and Histology. The scientific work was performed on the subject of research work of the Department of Anatomy and Histology of Polissya National University on the theme: “Development, morphology and histochemistry of animal organs in normal and pathological”, according to the state registration number – № 0113V000900. The morphological studies followed the basic rules of Good Laboratory Practice GLP (1981), the provisions of the “General Ethical Principles of Animal Experiments”, adopted by the First National Congress of Bioethics (Kyiv, 2001) and the requirements for the “Rules for carrying out work with experimental animals”, approved by the order of the Ministry of Health № 281 of November 1, 2000 “On measures to further improve the organizational forms of work with the use of experimental animals”. The material for histomorphological studies were pieces of lungs selected from clinically healthy adult rabbits (n = 5). Slices were made from paraffin blocks on a sled microtome MS–2. The thickness of histological sections did not exceed 8–12 μm, which were then stained with hematoxylin and eosin and Van Gizon to study the morphofunctional activity of cells and tissues of the respiratory tract. Histometric studies of histological specimens were performed using a microscope “Micros” with a digital camera at a magnification of × 100–1000. Studies have shown that macroscopically the lungs of mature rabbits have a reflection of the shape of the thoracic cavity, and gradually expands ventrally. The right and left lungs are surrounded by pleural right and left sacs. The pleural cavities of both lungs in the test animals are not interconnected. Organometry showed that the absolute lung mass of rabbits is 18.05 ± 1.32 g, relative to 0.624 ± 0.013 %. The histoarchitectonics of the lungs is formed by the lung lobes. The respiratory department of the lungs is represented by a set of acinuses. The latter are represented by respiratory bronchioles, alveolar passages, alveolar sacs and alveoli that form the alveolar tree. The results of morphometric studies showed that the respiratory (respiratory) part of the lungs of rabbits occupies 39.6 ± 0.62 %, and the connective tissue base, respectively, 58.5 ± 1.27 %. The average volume of alveoli, which are divided into small, medium and large, is 42.3 ± 4.35 thousand μm3. Thus, studies of the macro- and microscopic structure of the lungs of experimental animals are of great general biological importance, as they are an important criterion for objective assessment for the differential diagnosis of diseases of various origins
Effects of implant Suprelorin on cytological changes of vaginal epithelial cells in female dogs (4.7 mg deslorelin)
Diagnosis of the estrous cycle in female dogs is important in veterinary medicine. The estrous cycle is divided into the following stages: proestrus, estrus, diestrus, metestrus, and anestrus, which due to the action of hormones, replace one after another. This feature of the estrous cycle leads to some difficulties with reproduction. It is hard to obtain newborns because of prolonged anestrus. Vaginal cytology is a simple and available diagnostic method of the estrous cycle. It is based on the analysis of vaginal mucosa epithelial cells changes. This method is used to determine the time of mating and diagnose pathological conditions in the reproductive system of female dogs. There are various drugs in veterinary medicine for the correction of the estrous cycle in female dogs. We used implant Suprelorin® to estrus induction. There were described methods of estrus stimulation using a subcutaneous implant Suprelorin (4.7 mg, deslorelin) and changes in vaginal epithelial cells using the vaginal cytology method. The study was performed on twelve (n = 12) adult female dogs in the anestrus stage. Implants were injected subcutaneously in the groin area and removed at the beginning of the estrus
Assessment of ecological conditions for growing the market-size fish in the Kantivka fattening pond (Khmelnychchyna, Ukraine)
Abiotic (water quality) and biotic (Phyto-, zooplankton and benthos) factors in the Kantivka fattening pond the Khmelnitskrybhosp Private Joint-Stock Company were studied during the vegetation seasons in 2020. The aquaculture objects in the pond are cyprinid fish species (carp, grass carp, silver carp, pike). The ecological conditions of the pond were evaluated as good enough for the fish fattening purpose. The critical water quality parameters were following traditional values accepted for fish farming in Ukraine. The taxonomic composition of plankton and benthos was represented by widespread and Ubiquiti species mainly. The 124 species and intraspecies taxa of microalgae and the 34 species of zooplankton were identified in the Kantivka fattening pond. The mean abundance and biomass for the phytoplankton were 183.8 million cells/dm3 and 35.61 mg/dm3, respectively; for zooplankton, those were as 336600 ind/m3 and 14.83 g/m3, and for zoobenthos, those values were as 309.6 ind/m2, and biomass as 5.84 g/m2. Such parameters of natural forage base correspond to the optimal values for natural fish food according to fish farming standards in Ukraine. In the late summer, the free-living copepod stages and mature adult females of the parasitic copepod crustacean Ergasilus sieboldi Nordmann were registered in plankton. This copepod causes the invasion of fish diseases like infection and destruction of gill arches, which leads to a severe disturbance in fish respiration, emaciation, delaying in growth, and the potential death of commercial fish even. The development of natural fish food in the Kantivka fattening pond is shown an intensive level of biomass and abundance of phytoplankton, zooplankton, and zoobenthos. It is considered sufficient to meet the natural nutritional needs (in combination with artificial feeding) for older age groups of the cultivated cyprinid fish species
Antagonistic effect of Bacillus subtilis isolated and identified from different honey species against Klebsiella pneumoniae bee pathogens
The search for alternative methods for treating and preventing bee dysbacteriosis is a priority for beekeeping as a branch of veterinary medicine. Lime honey, buckwheat honey, flower honey, forest honey, and acacia honey were tested to evaluate their antagonistic effect against a test culture of enterobacteria of bees of Klebsiella pneumoniae species. The study was conducted in several stages: 1. Determine the activity of honey microflora against a pure culture of enterobacteria of bees of Klebsiella pneumoniae bee pathogens; 2. The identification and isolation of Bacillus subtilis – bacteria-antagonists against Klebsiella pneumoniae bee pathogens; 3. Determine the antagonistic effect of pure culture of Bacillus subtilis against a pure culture of enterobacteria of Klebsiella pneumoniae bee pathogens. The antagonistic action of honey microorganisms and the determination of the most effective honey species were established by the diffusion method in agar wells. Staining of typical colonies from different types of honey revealed bacillary colonies of Gram-positive bacilli with endospores. Isolation of clean culture was conducted by a method of Gold. The cultural, tinctorial, and morphological signs of bacteria have been consistently determined in all investigated kinds of honey and coincided with characteristics of the Bacellaceae family. Specific belonging of bacteria-competitors was identified by biochemical typing. After determining their physiological properties in reactions and tests (activity of catalase, oxidase, urease, the ability to grow at different temperatures and to ferment carbohydrate substrate), the distinguished stamms of microorganisms from the investigated kinds of honey belong to the type of Bacillus subtilis. The repeated estimation of antagonistic action of pure cultures of Bacillus subtilis (isolated from each type of honey) against a pure culture of enterobacteria of bees of Klebsiella pneumoniae species confirmed their high activity. This type of microorganisms can represent the alternative component in probiotics at the therapy of dysbiosis of bees
Isolation, identification and Analysis of Drug Resistance of Salmonella Pul-lorum
The article provides data on the isolation and identification of the pathogen S. Pullorum from pathological material of chickens. With further study of tinctorial, cultural-morphological and biological properties of the pathogen. The susceptibility of Salmonella pullorum to broad-spectrum antibacterial drugs such as cephalosporins and carbopenems was further studied to determine the drug of choice for improving treatment and prevention of avian bacterial diseases. In order to diagnose Salmonella pullorum (S. Pullorum) diarrhea accurately and analyze its drug resistance. In this study, the pathogen of a chicken suspected of S. Pullorum was isolation, PCR amplification and drug sensitivity analysis of the pathogen from in chicken farm in Xinxiang, north China. The results showed that the bacteria strain was diagnosed as S. Pullorum base on isolation and identification, Gram staining and biochemical identification of the bacteria. Antibacterial drugs sensitivity test confirmed that the bacteria was sensitive to ceftiofur, ceftriaxone, meropenem and kanamycin, and the effect of sensitive antibiotics was obvious in clinical treatment. Altogether, the present experiment revealed a detailed measure for S. Pullorum prevention and control and that achieved good clinical results, which laid a fundamental information for farmers and veterinary workers on eradication of S. Pullorum
Bioenergetic and ethological features of the first-calf heifers of different genotypes
The aim of this work was to study bioenergetic and ethological indicators in crossbred first-calf heifers compared to purebred first-calf heifers. The research was conducted on the first-calf heifers of Ukrainian Black-Pied dairy breed and first-generation crossbreeds obtained as a result of crossing with Brown Swiss breed, as well as, on the first-calf heifers of Ukrainian Red-Pied dairy breed and first-generation crossbreeds obtained as a result of crossing Ukrainian Red-Pied dairy with Montbéliarde breed. The purebred Black-and Red-Pied first-calf heifers prevailed over crossbreed first-calf heifers on the indicator of milk yield for 305 days of lactation by 106.45 and 218.36 kg, respectively. Accordingly, the indicators of average daily milk yield were also higher by 0.35 and 0.72 kg and the indicators of average daily milk yield adjusted by 4 % fat content per 0.15 and 0.16 kg. At the same time, the indicator of average fat content in milk prevailed in crossbreeds of Black-Pied and Brown Swiss breed by 0.08 % compared to purebred Black-Pied breeds, and in Red-Pied crossbreeds with Montbéliarde breed by 0.16 % compared to purebred Red-Pied breeds. The purebred Black-Pied first-calf heifers had an advantage by 2.04 MJ compared to crossbreeds, while the Red-Pied first-calf heifers had a slight advantage by 0.06 MJ compared to crossbreed first-calf heifers on the indicator of daily cost of exchange energy (EE). The purebred Black-and Red-Pied first-calf heifers had higher cost of exchange energy allocated with milk compared to crossbreed first-calf heifers by 0.42 and 0.82 MJ. They also had higher cost of exchange energy rates for heat products by 0.79 and 0.25 MJ. According to the energy index indicator, purebred Black - and Red-Pied first-calf heifers prevailed over crossbreeds first-calf heifers by 0.16 and 0.07 %, respectively. The same trend was observed in relation to the productive index indicator. The purebred Black-and Red-Pied first-calf heifers had a slight advantage by 0.001 and 0.002 kg adjusted by 4 % of milk fat content per 1 MJ. According to the indicators of daily behavioral reactions, no special differences between a purebred and crossbreed were found
