    Screening of infection conditions for brain microvascular endothelial cells infected by Streptococcus suis

    Streptococcus suis is a pathogen that causes swine meningitis, sepsis, and other diseases. There are 34 serotypes, of which type 2 is the most pathogenic. During the infection process of Streptococcus suis, several major virulence factors are involved and play a different roles. Streptococcal meningitis is caused by the bacteria’s ability to cross the blood-brain barrier and enter the central nervous system. Therefore, studying the interaction between Streptococcus suis and cerebral microvascular endothelial cells will help reveal meningitis's pathogenic mechanism. When studying the interaction between bacteria and cells, the number of infected bacteria and the time of infection are very important. In this study, Streptococcus suis serotype two was made into bacteria liquid and counted. Then bacteria were used to infect mouse brain microvascular endothelial cells with different multiplicity of infection (1,10,100 and 200). Cells were harvested at six h, nine h,12h,18h, and 24h after infection. The total RNAs of harvested cells were extracted, and the concentration of RNA was detected. The OD260/OD280 was between 1.8~2.4, OD260/OD230 was 1.5~2.4, and the concentration was greater than 100ng/µL. Total RNAs were reverse transcribed to cDNAs used to perform quantitative PCR to detect the mRNA expression of IL-18, IL-1beta, IL-6, and IL-10. The results showed that each MOI group's mRNA expression is higher than the control group with different infection times. When the multiplicity of infection is at 1, each group's relative expression of cytokines reaches a peak at 18hrs after infection. When the multiplicity of infection is at 10, each group's relative expression of cytokines reaches a peak at 12hrs after infection. When the multiplicity of infection is at 100, the relative expression of cytokines reaches a peak at 12hrs after infection. When the multiplicity of infection is at 200, the relative expressions of each cytokine reach a peak at 6hrs after infection. Based on the mRNA relative expression of each cytokine under different conditions, the optimal multiplicity of infection was 100, and the optimal infection time was 12h. The result provides a basis for the study of the pathogenic mechanism of meningitis

    Pharmacological studies of the veterinary medicinal product “Dibutalastin Ointment”

    The study of pharmacological studies of the veterinary medicinal product for external use in the form of ointment – “Dibutalastin Ointment” is a mandatory stage of preclinical research of the drug, which is a new development of PP “Biopharm” and LLC “DEVIE”. In a scientific experiment on laboratory animals, it is possible to assess the drug's safety for different terms of use to determine the zone of toxic action and doses that do not cause harmful effects on health. Therefore, the work aimed to carry out pharmacological studies of the drug with methyl salicylate and dimethyl sulfoxide in the form of an ointment for treating European fallow deer, deer, dogs, and cats with mastitis, arthritis, myositis, and injuries of various origins. The toxicological characteristics of the studied ointment “Dibutalastin” based on methyl salicylate and dimethyl sulfoxide were studied in an "acute" study on warm-blooded animals. The average lethal dose (LD50) for intragastric administration could not be established, as the volume exceeded the permissible level. It was established that according to the “average lethal dose when entering the stomach” indicator, the veterinary medicinal product belongs to relatively non-toxic substances (toxicity class IV). The results of pharmacological studies indicate the absence of resorptive-toxic effects, sensitizing properties, and irritating effects on the mucous membranes of the eyes and skin. Pathomorphological studies of animal organs when studying the acute toxicity of the drug showed that the developed ointment with methyl salicylate and dimethyl sulfoxide does not cause pathological changes in internal organs. From the point of view of veterinary toxicology, this drug is safe. Further studies will be the next stage of pre-registration tests aimed at studying the embryotoxic effect of “Dibutalastin Ointment”, which is mandatory material of the “Safety and residue studies” section of the dossier for this medicinal product

    Erythron and protein system in piglets blood under stress influence

    The article presents the results of research on the development of adaptive syndrome in the system of erythron and protein of piglets at the effects of weaning stress. After one (anesthetic stage) and seven days (beginning of G.Selle's resistance stage), after the echoing, the adaptation of the erythron system was shown by a decrease in the number of red blood cells on the contrary of increasing the mean corpuscular volume and hemoglobin indicating the inhibition of erythrocytosis with simultaneous macrocytosis, and a decrease in mean Corpuscular hemoglobin concentration for 14.0–13.0 % that indicated hypochromia. In these stressful periods, the catabolism of protein substrates was discovered, the redistribution of albumins between blood and tissues in the form of a decrease in the total protein content by 6.1–7.1 % due to albumin by 14.8–10.0 %, and the deficit of the humoral link of the immune response due to reduction of γ-globulins by 29.9–18.7 %. In the later stages of the resistance stage (after 20 and 60 days after weaning), there was a stabilization of erythrocytic indices and the development of anisocytosis in the form of an increase in red cell distribution width by 19.0 %, with a simultaneous decrease of 6.1 % of the total protein content, redistribution of its fractions in the side of the growth of γ-globulins by 18.4 %. A practical, prolonged way of preventing disorders in the erythron and protein of extramarital piglets was feeding the additive “Biovir”, which stimulated the intensity of oxidative-reducing processes and the exchange of proteins

    Study of the activity of sunflower honey against a mixed microbial association isolated from bees with signs of dyspepsia in laboratory conditions

    Beekeeping is one of the essential branches of agriculture in Ukraine, the main activity of which is aimed at breeding bees and obtaining honey and other products beneficial for human health. Maintaining the physiological state of bee colonies at the proper level is one of the essential tasks of veterinary medicine doctors and beekeepers. Compliance with veterinary and sanitary requirements at bee farms is the basis of preventing infectious pathologies in bee colonies. Since the body of insects has a fast metabolism, any infectious pathology quickly acquires a massive manifestation. One indicative symptom of infectious damage to the “gut” of bees is dyspepsia. Diarrhea, anorexia, and loss of productivity are observed in bees in this condition. The market of means to prevent such pathological manifestations in beekeeping is relatively narrow. The healing and favorable properties of honey, manifested in pain-relieving, antimicrobial and anti-inflammatory effects, have been known since ancient times. Sunflower honey differs from other types in a wide range of components, particularly phytoncides, which inhibit the growth of pathogenic microorganisms. Therefore, the main goal of the experiment was a laboratory study of the effect of different concentrations of sunflower honey sieve on the mixed microbial association isolated from bees with signs of dyspepsia. The activity of sunflower honey in laboratory conditions was studied by the disco-diffusion method in Petri dishes on MPA medium (meat-peptone agar). To obtain syrup from sunflower honey, sunflower honey was diluted with distilled water in a ratio of 1:1, working solutions were prepared from sunflower honey syrup in dilutions of 1:2, 1:4, 1:10, 1:100. The data analysis indicates the bacteriostatic activity of the syrup from sunflower honey in all studied concentrations concerning the mixed microbial association. Moreover, the largest diameter of the growth inhibition zone of the studied microorganisms was noted when the syrup from sunflower honey was diluted in a ratio of 1:10 (19.2 ± 0.42 mm). In turn, the antagonistic effect of the syrup from sunflower honey was observed in all dilutions concerning the mixed microbial association, where the largest diameter of the antagonism was 19.6 ± 0.27 mm when the disc was impregnated with the native working solution. Thus, it is promising to accumulate and identify a pure culture antagonist concerning a mixed culture of bacteria isolated during bee diarrhea to further create a pharmacological agent for preventing infectious pathologies of bees in unhealthy apiaries

    Comparative assessment of reproductive capacity of boar-sires depending on the breed and season

    The article highlights the results of the reproductive capacity of different breeds of boar sires in the farms of eastern, southern, and central regions of Ukraine in dynamics over three years are given. It represented the influence of the year's season on sperm quality by estimating the ejaculate volume, concentration, and motility of sperm of boars-sires of the different breeds and their hybrids. Data were also calculated on the culling of ejaculates of boar sires of various breeds for the years 2019–2021 in the dynamics. In a comparative aspect, the indicators were analyzed by the following genotypes: Large White, Ukrainian Meat, Landrace, Duroc, Yorkshire, and synthetic line Makster. The reproductive capacity of boars was within the limits of the indicators for the elite and first classes. However, specific differences were observed between boars of different breeds, which were constant and took place during the three studied years. The difference in the concentration of sperm in the ejaculates of the Ukrainian Meat breed gradually decreased starting from 2019 and was the smallest in 2021. The highest ejaculate volume was found in boars of the Ukrainian meat breed (310.9 ± 113.2 cm3), and the sperm concentration was the highest in boar sires of the Large White breed (215.7 ± 83.5 million/cm3). On the other side, hybrids of the Large White breed had low indicators compared to other groups of animals (Large White × Yorkshire – 184.2 ± 68.5 million/cm3, Large White × Makster – 189.0 ± 59.7 million/cm3). Sperm motility did not undergo significant breed differences: the average annual indicator of motility was 7.6 ± 0.09 points, and it was the highest in the winter period (8.6 ± 0.11 points), however, for the established variances; the dilution was used 1.65–3.58 times and a high percentage of unusable sperm (15.4–29.4 % of the total amount) was established in a large number of samples. According to the results of the annual analysis of the sperm production of boars, a high degree of culling of ejaculates was established for various reasons (high level of sperm agglutination (52.3–74.0 %), low concentration of germ cells (14.5–29.8 %), presence of foreign impurities (5,8–12.4 %). The highest percentage of culling was in summer (20.3 %). Thus, the obtained results showed the breed and seasonal characteristics of the reproductive capacity of breeding boars

    Gross Anatomical Features of Tongue of Khaki Campbell Duck (Anas platyrhynchos domesticus) At Different Postnatal Ages

    The present study aims to investigate the gross morphological structure of the tongue of the khaki Campbell duck at different postnatal ages. The ducks were from four age groups (A total of 25 ducks): day one (1), one (1) month, two (2) months, three (3) months, and four (4) months, having five ducks in each group. The tongues were collected immediately after the ethical killing of the birds for gross anatomical observations. The tongue of the Khaki Campbell duck was located on the floor of the oropharynx. In all examined ages, the tongue did not extend to the total limits of the lower beak. The elongated shaped tongue was divided into three parts, i.e., apex, body, and root. The spatula-shaped apex of the tongue was smooth dorsally and devoid of lingual papillae. On the ventral surface of the apex, there was a flat, triangular, white plate of the lingual nail. The dorsal surface of the body of the tongue was divided into two symmetrical parts by a median groove, which became more prominent at the advancement of age. Large and small conical papillae were found symmetrically along both body edges. The root of the tongue was the smallest part which was located adjacent to the laryngeal prominence. Papillae were found on both sides of the root. A fold of membrane, the frenulum linguae present on the ventral aspect of the body. The length and width of the tongue increased with the advancement of age. The present study revealed the gross anatomical structure of the tongue of the Khaki Campbell duck at different postnatal ages and helped to compare the findings with other avian species

    Changes in the chemical composition of broiler meat when chelated compounds are added to the diet

    The paper considers the use of chelated forms of micronutrients for feeding broilers. The study aims to investigate the chemical composition of broiler meat in the case of broiler chickens of Cobb-500 cross, provided Zn, Cu, and Mn chelated forms are supplied to the diet. Experimental studies were conducted in 2020 on broiler chickens of Cobb-500 cross. Two groups of 20 birds were formed to study the chemical composition of poultry meat. The birds of the control group received an essential diet supplied with sulfates of Zn, Cu, and Mn. The birds of the experimental group received a diet enriched with chelated compounds of Zn, Cu, and Mn. The study has shown that introducing Zn, Cu, and Mn chelated compounds into the diet of broiler chickens has no adverse effect on the chemical composition of meat. It has also been determined that the meat of broilers eating feed supplied with chelated micronutrients contains significantly less cholesterin but more Ca, Zn, Cu, and Mn, and several essential amino acids. These indicators prove an increase in the health benefits of chicken meat

    Quality control of cooked sausages

    Despite the increased interest in the quality and safety of cooked sausages, the situation in the field of food is becoming more complicated and more threatening every year. As a result of research, it should be noted that the consumer market of the Rivne region is saturated with products of good quality, which are made mainly by mini-shops or private enterprises. When inspecting sausages, it was found that the city's stores in 2020 sold more than 50 tons of high-quality products in a wide range. These include boiled sausages, hot dogs and sausages, semi-smoked, boiled-smoked, raw-smoked, beef, pork and chicken delicacies, pâtés, loaves of bread, ham, and skewers. Experimental studies have shown that their name and manufacturer influence the evaluation of cooked sausages. The highest score was given to the sausage “Molochna” and “Teliacha z vershkamy”, which has become an authentic brand. Buyers of our country should prefer it. According to organoleptic and physical studies, further Examination of five samples of cooked sausages from different manufacturers showed that the samples meet the requirements of DSTU 4436: 2005. Microbiological tests revealed that the samples of cooked sausages met the requirements of regulatory indicators. Sausage “Likarska”, Yalovycha “Liuks” and “Z vershkamy” on two physical indicators: the content of mass fraction of moisture and mass fraction of salt, did not meet the requirements of DSTU 4436: 2005, this product is a counterfeit

    Protein-synthesizing function of the liver of cows at experimental fasciolosis

    Cattle fasciolosis is quite widespread in the world, including in Ukraine. Animal blood proteins, being in close functional connection with proteins of various tissues, reflect the changes that occur in the tissues and organs of the body during disorders of their metabolic processes caused by pathological factors. The aim of the study was to investigate the level of total protein and its fractions in the blood of cows in experimental fasciolosis. 12 cows of 4–5 years of age, black-spotted breed, were selected for experiment, out of which 2 groups were formed, six animals in each. Animals in the control group were clinically healthy. Animals of the experimental group were experimentally infected with adolescents. During the research, the rules of compulsory experiments were followed - selection and keeping of analogue animals in groups. The cows' diet was balanced in terms of nutrients and minerals. In the study of protein-sensitizing function of the liver of cows, it was found that during fasciolosis invasion, the content of total protein decreased by 13.8 %. These changes were due to a decrease in albumin and an increase in globulins. Thus, in fasciological invasion of cows, the level of albumin in the serum decreased by 24.4 %, and the level of globulins increased by 7.9 %. The value of the albumin / globulin ratio in the blood of infested cows with the causative agent of fasciolosis is quite indicative. It was found that the albumin-globulin ratio in the blood of cows of the experimental group was probably lower compared to the control group of animals. This value of the coefficient, of course, indicates the suppression of protein-synthesizing function of the liver in these periods of research. In the future it is planned to investigate the functional state of the liver of cows with experimental fasciolosis

    Amino acid composition of duck muscle tissue on using dietary supplements Activio

    The optimal amount of protein is necessary for intensive growth. Protein malnutrition is one of the leading causes of poor growth and increased care of birds. However, excessive feeding of poultry with protein is impractical because the protein, in this case, is not fully used and harms the body, delaying its development. The lack and the excess of amino acids are harmful to a bird. Due to the imbalance of amino acids consumed with food, about 30 % of their amount is not used by the body for protein synthesis and is lost converted into energy. When normalizing the amino acid needs of poultry, it is necessary to consider the interaction of amino acids in the body with vitamins, micro-minerals, energy levels in feed. The article presents a study of the effect of the biologically active additive Activio, a selected combination of natural standardized active substances isolated from aromatic herbs and spices concentrated in one microencapsulated particle on the amino acid composition of Peking duck meat and cherry valley cross. It was found that the use of Activio supplement in the ducks feeding has a positive effect on the digestibility of amino acids
