論文内容の要旨
主要組織適合性複合体 class IIB（major histocom-
patibility complex，MHC class IIB，IIB）は，微生物
などに由来する外来性抗原由来のペプチド断片を提示し
てヘルパーＴ細胞を活性化し，B細胞による抗体産生を
促す。IIB のペプチド結合領域（peptide binding re-
gion，PBR）は多型性に富み，種として認識しうるペ
プチドの多様性を増す方向に正の選択圧を受けている。
ニワトリ（Gallus gallus，Gg）では GgIIBハプロタイ
プと各種疾患感受性との関係が示唆されているが
GgIIBの近傍にはMHC class I（GgI）をはじめ免疫応
答やストレス応答など基本的生命現象に関与する遺伝子
がクラスターをなして存在し（MhcB領域），しかもこ
れら遺伝子が互いに強い連鎖不平衡関係に在るため，疾
患感受性と関わる遺伝子を明確にした報告は少ない。
これに対し，ニホンウズラ（Coturnix japonica，Cj）
はニワトリと同じキジ科に属し両者間でキメラおよび
F1の作出が可能であることから，ニホンウズラ MHC
領域，MhcCoja領域はニワトリMhcB領域と酷似して
いることが予想された。MhcCoja領域の遺伝子の構成
はMhcB領域と類似していたが，class IIB（CjIIB）が
7座位も存在するなど，直接抗原認識に関与する遺伝子
座が著しく重複していた。さらに，CjIIB領域の 5種類
のハプロタイプ（CjIIB-HT）間で，cDNAのクローニ
ングにより機能的な CjIIBの遺伝子構成を比較解析し
たところ，Copy number variation（CNV）が確認され
た。しかし，CjIIB領域は複数の遺伝子座を含む数 kb
にわたる類似した配列が複雑に重複しているため，サブ
クローニングに頼る従来の解析法では，CjIIB遺伝子構
成の全容を明らかにすることは困難であった。
そこで，本研究ではまず CjIIB 領域のハプロタイプ
を多型マーカーにより特定し，ついで次世代シーケン
サー（NGS）による網羅的な解析により機能的 CjIIB
が極めて多様性に富むことを明らかにした。
1． MhcCoja領域の DNA多型解析
1）MhcCoja領域の 7マーカーのアレル解析
本研究室にて飼育管理している A，B，ND，K，Pお
よび Y 系の 6 系統，270 個体のニホンウズラゲノム
DNA を供試した。MhcCoja 領域の上流より，HEP21
（①），TRIM39.2（②）および BTN2（③）（以上 TRIM
領域），DBB1（④ : Class IIB領域），ならびに DMB1
（⑤），DMB2（⑥）および TAP2（⑦）（以上 Class I
領域）の 7マーカーについてタイピングした。各マー
カーには 4∼6個のアレルが検出され，また何れの系統
もすべてのマーカーについて複数のアレルが検出され
た。B系は 7マーカーの平均アレル数（5.0）がもっと
も多く，逆に K 系のそれ（2.3）がもっとも少なかっ
た。また B系の①，②および③では低頻度ながら他系
統に認められないアレルが検出されたが，他のマーカー
および系統に特異的なアレルは認められなかった。
2）MhcCoja領域の 7マーカーによるハプロタイプ解析
ハプロタイプ推定プログラム PHASEを用いて，1）
で決定した各個体のアレルの組合せからなるハプロタイ
プを推定した。
（1）隣接する 2つのマーカーによるハプロタイプ : ②
& ③，③ & ④および④ & ⑤が特定のアレル以外とハプ
ロタイプを形成した割合（それぞれ 4.7%，13.1%およ
び 13.1%）は，① &②，⑤ &⑥および⑥ &⑦が特定の
アレル以外とハプロタイプを形成した割合（0.2%，0.2
%および 1.9%）に比べて高値を示した。

（2）3つのマーカーによるハプロタイプ : ③，④ &⑤
のハプロタイプの種類は 44種類ともっとも多く，つい
で①，② & ③のハプロタイプの種類は 20種類であり，
⑤，⑥ & ⑦のハプロタイプの種類は 8種類ともっとも
少なかった。また，Y系および B系は，他の系統に比
べて③，④ & ⑤および①，② & ③のハプロタイプの種
類が多く，逆に K系はもっとも少なかった。
（3）全 7マーカーよるハプロタイプ : 75種類のハプ
ロタイプが検出されたが，このうち 55種類は系統に特
異的なハプロタイプであった。
以上のことより HEP21 と TRIM39.2 との間（① &
②），ならびに DMB1，DMB2 および TAP2 との間
（⑤，⑥ & ⑦）に強い連鎖関係（ハプロタイプブロッ
ク）が存在することが示唆された。一方，② & ③，③
& ④および④ & ⑤は特定のアレル以外とハプロタイプ
を形成した割合が高かったため，TRIM39.2 と BTN2
との間（② &③），BTN2と DBB1の間（③ &④），な
らびに DBB1 と DMB1 との間（④ & ⑤）にそれぞれ
組換え部位が存在することが示唆された。したがって，
組換えハプロタイプを活用することで各領域の機能を独
立して解析できることが確認された。
2． 機能的MhcCojaIIB（CjIIB）の多様性解析
1）454GS Juniorによる各ハプロタイプの機能的 CjIIB
の同定
1．でハプロタイプを推定した個体から，Class IIB
領域内の DBB1 に検出された 6種類のハプロタイプ
（CjIIB-HT*01∼*06）をカバーするように 21個体を選
抜し，さらに詳細なハプロタイプ情報を得るため
BTN1（TRIM 領域），Tapasin（Class IIB 領域），な
らびに CD1.1および CD1.2（CD1領域）をタイピング
した。ついで，これらの個体の末梢リンパ球から全
RNAを抽出し，鋳型とした。CjIIBのもっとも多様性
に富む Ex2は，抗原ペプチドと直接結合し 18個のアミ
ノ酸残基により構成される PBRを含む。この Ex2をカ
バーする Ex1の一部から Ex3の一部まで（411bpまた
は 441bp）を共通で増幅するプライマーペアを用いた。
このプライマーにアダプター配列などを付加したフュー
ジョンプライマーにて，鋳型 RNA を RT-PCR 増幅し
た。454 GS Juniorが産出するリード数は平均 1×105で
あるが，PCR産物の推定サイズをカバーしうるリード
長を得ることが可能である。そこで本項では各 CjIIB-
HTの機能的 CjIIB遺伝子の種類を特定することを目的
とした。
（1）CjIIB-HT*01の機能的 CjIIBの検出 : Class IIB
領域のハプロタイプは，いずれも CjIIB-HT*01のホモ
接合体だが，TRIM領域および Class I領域以降のハプ
ロタイプは互いに異なる 5個体を供試した。Class IIB
領域以外の領域のハプロタイプが異なっているにも関わ
らず，これら 5個体の機能的 CjIIB配列は一致してい
た。また，既知のコンティグマップに認められる 7種類
の CjIIB配列は既報のサブクローニングにより検出さ
れた各配列のクローン数の頻度とほぼ一致する出現頻度
で認められた。さらにいずれの個体においても 5種類の
新規 CjIIB配列が認められた。したがって本法はサブ
クローニング法より高感度に機能的 CjIIB配列を検出
しうること，ならびに CjIIB-HT*01の機能的な CjIIB
は，いずれも Class IIB領域（BTN2∼Tapasin∼DBB1
近傍）に座位することが示唆された。
（2）機能的 CjIIBの塩基配列の同定 :（1）をふまえ，
他の CjIIB-HTをもつ個体を加えて，同様の手順にて機
能的 CjIIB 配列を解析したところ，22 種類の既知
CjIIB配列および 22種類の新規 CjIIB配列の計 44種類
の配列を確認した。なお，新規 CjIIB 配列のうち 2種
類は，特定の CjIIB-HTの半数未満の個体のみで検出さ
れた。
（3）機能的 CjIIBの系統樹解析 : DBB1*02と 6種類
の類似配列（DBB1*02グループ）は，いずれも CjIIB-
HT*02に検出され，クラスターを形成した。同様に，
DGB1と 5種類の類似配列に DDB1および DEB1を加
えた計 8種類の塩基配列（DGB1 グループ），CjIIB16
と 3種類の類似配列（CjIIB16 グループ），ならびに
DCB1 の 5アレル（DCB1 グループ）も，それぞれク
ラスターを形成した。
（4）各 CjIIB-HTにおける機能的 CjIIB配列数の多型
（CNV）: 44種類の CjIIB配列は，9種類の出現頻度の
高い主働 CjIIB（10%以上）と 35種類の低い微働 CjIIB
（5% 以下）に大別された。各 CjIIB-HT の CjIIB 配列
数は，主働 CjIIB（1∼3種類）および微働 CjIIB（0∼
10種類）ともに大きなハプロタイプ間差を示した。
（5）機能的 CjIIB 配列の重複と共有 : CjIIB-HT*01
および *06は DGB1グループの塩基配列を 5種類およ
び 2種類含み，また，ともに CjIIB16 グループの塩基
配列を 2種類ずつ含んでいた。さらに DBB1*02グルー
プの塩基配列は，いずれも CjIIB-HT*02に含まれてい
た。各グループの塩基配列は，グループ内の相互の類似
性の高低差から複数回に亘る遺伝子重複の結果，生じた
ことが示唆された。
塩基配列の類似性から，DBB1（または DAB1）およ
び DCB1 以外に CjIIB-HT*01∼*06のすべてに共通に

存在する遺伝子座は確認されなかった。したがって，各
ハプロタイプ特有の遺伝子欠損が存在する可能性が示唆
された。一方，CjIIB-HT*04には DCB1遺伝子座の塩
基配列が検出されなかったが，ゲノム DNAによる解析
で DCB1*04が確認されていることから，DCB1*04は
末梢リンパ球以外で発現している機能遺伝子であるか，
あるいは偽遺伝子化している可能性が示唆された。
また，CjIIB-HT*06 に検出された 2種の主働 CjIIB
の塩基配列は，それぞれ CjIIB-HT*01 の主働 CjIIB :
DBB1*01または CjIIB-HT*03の主働 CjIIB : DABI1L
*03の塩基配列と一致した。さらに，CjIIB-HT*01 と
CjIIB-HT*05 との間で，微働 CjIIB : DFB1 の塩基配
列が一致した。したがって，これら CjIIB-HT間でアレ
ルを共有していることが示唆された。
（6）CjIIB-HTの機能的多様性 : 微働 CjIIBが検出さ
れなかった CjIIB-HT*04以外のハプロタイプでは，微
働 CjIIBの Ex2全体の塩基配列（270bp），Ex2がコー
ドする b1ドメインのアミノ酸配列（89aa），ならびに
b1ドメインに含まれる PBRの 18個のアミノ酸残基の
類似性（73∼100%，60∼100%および 6∼100%）は，主
働 CjIIB の類似性（80∼92%，62∼88% ならびに 22∼
72%）に比べてより大きな多様性を示した。
主働 CjIIBおよび微働 CjIIBの PBRのアミノ酸残基
の組合せは，それぞれ 9種類および 21種類であった。
また，PBRの 18ヶ所各々のアミノ酸の種類を平均する
と，主働 CjIIBでは 1.5種類，微働 CjIIBでは 2.4種類
であった。さらに PBRのアミノ酸残基のうち 14ヶ所に
延べ 23種類の特異的なアミノ酸残基が確認され，その
うち 9種類は主働 CjIIBに，残りの 14種類は微働 CjIIB
に認められた。
主働 CjIIBが 1種類しか検出されなかった CjIIB-HT
*02と CjIIB-HT*05 の主働 CjIIB の PBR のアミノ酸
残基は化学的性質が類似しており，特異的なアミノ酸残
基は認められなかった。一方，CjIIB-HT*02 および
CjIIB-HT*05では，微働 CjIIB配列に特異的なアミノ
酸残基が 1種類および 2種類検出された。
主働 CjIIB が 2種類検出された CjIIB-HT*01，*03
および *06は（5）に記載したように，各々いずれか一
方の主働 CjIIB を共有していた。これらのうち DAB1
*01の PBRに 4個の特異的なアミノ酸残基が認められ
た。これら 3種類の CjIIB-HTの微働 CjIIBを比較する
と，CjIIB-HT*01および CjIIB-HT*03では特異的なア
ミノ酸残基を有する微働 CjIIB 配列が 1種類および 2
種類検出された。しかし CjIIB-HT*06では，PBRに特
異的なアミノ酸残基を有する微働 CjIIBが認められな
かった。
微働 CjIIB は検出されなかったが，主働 CjIIB が 3
種類検出された CjIIB-HT*04において，これら 3種類
の CjIIBの PBRのアミノ酸残基は，CjIIB-HTの中で
もっとも多様であり，いずれも特異的なアミノ酸残基が
認められた。
したがって，微働 CjIIB の PBRs は，各ハプロタイ
プの抗原ペプチドに対する親和性の幅を広げるととも
に，各ハプロタイプ間の抗原ペプチド親和性の多様性を
高めることにより，限られた遺伝子座によって構成され
る主働 CjIIBの抗原ペプチド親和性を補完しているこ
とが示唆された。また，微働 CjIIBはリンパ球の種類，
分化・成熟状態に特異的な発現を示す可能性も考えられ
る。
2）MiSeqによる各ハプロタイプにおける機能的 CjIIB
のmRNA転写頻度の確認
1）で供試した 21個体に 9個体を加えた，計 30個体
を供試した。MiSeqで決定しうるリード長は 300bp未
満と短いが，産出するリード数は 5×107に達する。そ
こで，454 GS Juniorで同定した CjIIB遺伝子の塩基配
列をリファレンスとして，MiSeqで産出した各遺伝子
のリード数から，出現頻度を推定した。44種類の CjIIB
リファレンス配列は，454 GS Juniorで推定した各々の
CjIIB-HT に検出され，それらの出現頻度も 454 GS
Juniorのアンプリコンシーケンシングにより産出した
出現頻度に準じた。
3）HiSeq2500による機能的 CjIIBの確認
アンプリコン解析では，CjIIB ユニバーサルプライ
マー，ならびに NGS : Roche 454 GS Juniorでユニバー
サルプライマーに結合するアダプター配列などが，各々
の CjIIB由来 mRNAの PCR増幅の効率に影響してい
る（PCR バイアス）可能性がある。そこで，新たな 4
個体を供試し HiSeq 2500を用いた RNA-Seqにより決
定した全 mRNA の配列を，1）で決定した CjIIB 配列
（リファレンス）にマッピングした。その結果，供試し
た CjIIB-HT*01，*03，*04および *05の主働 CjIIBは
6種類すべて，ならびに微働 CjIIBは 25種類のうち 22
種類が検出された。また各々の出現頻度は，454 GS
JuniorならびにMiSeqのアンプリコンシーケンシング
により産出した出現頻度に準じた。したがって，アンプ
リコンシーケンシングにおいて機能的 CjIIBの出現頻
度に対する PCRバイアスの影響は小さいものと考えら
れる。

結 論
MhcCoja領域に構築した 7つの DNA多型マーカー
を用いたハプロタイプ解析により，CjIIB領域は，その
上流 TRIM領域，下流 CjI領域との間で，組換えが生
じていることを示唆した。一方，各々の領域内は連鎖不
平衡状態にありハプロタイプブロックを形成しているこ
とが示唆された。NGSによるアンプリコンシーケンシ
ングにより各 CjIIB-HTの機能的 CjIIB遺伝子は連鎖し
ており，いずれも CjIIB領域内あるいはそれと連鎖す
る領域近傍に存在することが示唆された。
CjIIB-HTの CNVは，遺伝子の重複，融合，（機能的）
欠損，共有などダイナミックな遺伝子座の再編成によっ
てもたらされたことが示唆された。また，この CjIIB
遺伝子の CNVは，PBRにアミノ酸置換とともに遺伝
子数の多寡による多様性を付与し，抗原ペプチドに対す
る親和性に CjIIB-HT間差異をもたらし，種として認識
しうる抗原ペプチドの多様性を生み出すメカニズムとし
て機能していると考えられた。
多様なハプロタイプを形成する多重遺伝子族の機能的
遺伝子の解析において，リファレンス配列を整備するた
めには，NGSを活用したアンプリコンシーケンシング
による解析が有効であることを示した。また，リファレ
ンス配列が整備されれば，RNA-seqが PCRバイアスの
影響を最小限に抑えて，より網羅的に各遺伝子の転写量
を解析しうることを示した。
審査報告概要
本研究はニホンウズラMHC（MhcCoja）領域のハプ
ロタイプ解析をおこなうとともに，次世代シーケンサー
（NGS）を用いて機能的な CjIIB を同定しそれらの
CjIIB-HT間における多様性を明確にすることを目的と
した。MhcCoja 領域の 7つの DNA 多型マーカーを用
いたハプロタイプ解析により，CjIIB領域と，その上，
下流の領域との間で組換えが生じていること，各々の領
域内は連鎖不平衡状態にあることを示唆した。また，
NGSにより各 CjIIB-HTの機能的 CjIIB遺伝子は連鎖
しており，いずれも CjIIB領域近傍に存在することを
示唆した。さらに CjIIB-HTの CNVは，遺伝子座の再
編成によってもたらされたことを示唆した。CjIIBの複
雑な遺伝子構造の一端を明らかにするとともに，多重遺
伝子族の機能的遺伝子の解析法を確立した本研究の成果
は，今後のMhcCoja領域の多様性解析を一段と進展さ
せることで，家禽の抗病性育種に大いに貢献するものと
判断される。
よって，審査員一同は博士（畜産学）の学位を授与す
る価値があると判断した。

1． 研究の背景および目的
外来種アライグマ（Procyon lotor）は，北米原産の
食肉目に属する中型哺乳類である。1970年代に放映さ
れたアニメの影響により人気を博し，日本にペット目的
で大量に輸入されたが，飼いきれなくなって捨てられた
り，脱走したりする事例が多発し，1980年代には日本
各地で多発的に野生化し，社会問題化した（Ikeda et
al., 2004）。そのため，アライグマは「特定外来生物に
よる生態系等に係る被害の防止に関する法律（外来生物
法）」の制定と同時に特定外来生物に指定された。現在，
全国で 447のアライグマ防除実施計画が策定され（2014
年 4月現在），日本各地で防除対策がとられている。外
来種の防除を行う際は，計画的・順応的に実施する
PDCAサイクルと取り入れた順応的管理を用いること
が推奨されており（環境省自然環境局外来生物対策室，
2014），これを実行するためには，対象動物がその対象
地域のどこに分布し，どのくらいの個体数がいるのかと
いう情報が必須である。アライグマにおいては分布確認
や個体密度を調べる方法がいくつか考案されている。防
除計画の立案者は，この中から目的を達成するための最
適な調査手法を選択する必要がある。そこで本研究で
は，アライグマの分布確認手法，密度指標の開発といっ
た防除事業に係る技術開発を行い，さらにそれらの技術
を用いて分布域と相対密度の経時的な変化を調べること
により，防除事業の効果を検証した。
2． 分布確認手法の比較
アライグマの分布を把握することは，防除計画の立案
や捕獲の実施場所，侵入を警戒する場所を決定するため
に必須な情報である。アライグマの分布調査において
は，捕獲記録や目撃記録，アンケートが良く用いられて
いる（神奈川県，2005 ; 環境省自然環境局生物多様性セ
ンター，2007 ; 大阪府立環境農林水産総合研究所，
2013）。しかし，これらの手法は，山林などの人間活動
の少ない地域において動物種の分布を過小評価する可能
性がある（環境省自然環境局生物多様性センター，
2004）。分布調査で用いられる痕跡調査や自動撮影調査
は，捕獲記録やアンケートによる調査と比較して高コス
トであるが，人間活動が少ない地域でも安定した調査精
度で分布把握が可能である。中でも自動撮影調査は現
在，哺乳類における生息状況の調査法として主流となっ
ている（安田ほか，2008）。東京農業大学野生動物学研
究室では痕跡調査の足跡トラップの一つとして，安価で
入手しやすい材料であること，機材の作成や調査手順が
簡便であること，得られる足跡が鮮明であることをコン
セプトとしたスタンプ板を開発した。
そのため，アライグマの分布確認を目的として，足跡
トラップの一種であるスタンプ板と，哺乳類相調査で主
流となっている自動撮影カメラの調査効率を比較した。
アライグマの出現頻度は自動撮影調査の方が有意に高く
（P＜0.01），検知されるまでに要した稼働夜数（Latency
to first detection : LTD）も有意に早かった（P＜0.01）。
自動撮影カメラ調査から得られたスタンプ板周辺の行動
についてみると，スタンプ板の周囲に現れた 8.5%のア
ライグマは板の脇をすり抜けていた。しかし，アライグ
マはタヌキやアナグマよりも板の上を通過する例が有意
に多かった（P＜0.05）。
これらのことからアライグマの分布確認としては，自

動撮影カメラ調査の方がスタンプ板調査よりも調査効率
が高かった。しかし，スタンプ板はアライグマに忌避さ
れている割合が他種よりも低いことから，有効であると
考えられた。さらにスタンプ板調査は自動撮影調査にな
い特性を持っていることから，両者の使い分けが重要で
あると考えられた。
3． 密度指標の開発
外来種密度の把握は，防除事業の成果を把握する上で
必須である。従って，アライグマの防除事業を評価する
ためには，アライグマ密度の経時的な変化を把握するこ
とが必須である。しかし，捕獲努力量あたりの捕獲頭数
（捕獲効率 Catch per unit effort : CPUE）は，警戒心の
強い個体の存在により防除効果を過大評価する場合があ
る（松田，2012）。そのため，捕獲とは独立した密度指
標による評価が必要である。一方，自動撮影調査から得
られたカメラの稼働日数あたりの動物撮影回数（撮影効
率）は食肉目の調査で用いられる密度指標の一つである
（金子ほか，2009 ; O’Brien，2011）。アライグマにおい
ては自動撮影カメラによる撮影率（アライグマ撮影回数/
全有効撮影回数）と除去法による推定生息密度には有意
な正の相関があると報告されている（安藤ほか，2003）
が，撮影効率と捕獲効率による検証は行われていない。
そこで，アライグマの防除事業による密度指標と自動撮
影調査による密度指標の関係を調べた。また，自動撮影
データから得られた密度指標の有効性を確認するため，
自動撮影データと既にアライグマの密度指標として有効
であるアライグマの防除事業から得られた捕獲効率との
関係を調べ，自動撮影調査から得られたアライグマ密度
指標の有効性を検証した。
その結果，得られた密度指標を地域間で比較すると，
撮影効率，撮影メッシュ率，捕獲メッシュ率は，捕獲効
率と同様の傾向を示したが，LTDと LTC（アライグマ
を捕獲するまでに要した期間 : Latency to first catch）
は捕獲効率とは異なる傾向を示した。一般化線形混合モ
デルにより解析したところ，撮影効率は捕獲効率に対し
て有意な正の関係がみられたが（P＜0.01），LTDにお
いてはみられなかった（P＝0.08-0.83）。
これらのことからアライグマの密度指標として，撮影
効率は有効であった。さらに本研究では撮影効率から捕
獲効率を推定する回帰式が得られたことから，捕獲が行
われていない地域においても自動撮影調査が行われてさ
えいれば，撮影頻度から捕獲効率を補完することが可能
となった。
4． 分布域と相対密度の経時的な変化
神奈川県では 2005年の外来生物法の制定を受けて，
2006年に神奈川県アライグマ防除実施計画を策定し，
本格的な防除活動を開始した（神奈川県，2006）。この
防除計画は 5年ごとに現状を踏まえて見直すことになっ
ており，2011年には第 2次アライグマ防除実施計画に
移行し，全国に先駆けてメッシュ単位でのアライグマ管
理が導入された（神奈川県，2011）。外来種の根絶を達
成するためには，捕獲などにより個体を除去すること
で，分布域の縮小化や生息密度の低密度化を実現する必
要がある。神奈川県では防除を計画的に行っていくため
に，アライグマの防除記録や目撃情報から分布域と捕獲
効率のモニタリングを行い，分布域と生息密度の把握を
行っている（神奈川県，2011）。しかし，防除記録や目
撃情報のみの分布域の把握では，アライグマへの関心が
低い地域や人の居住が少ない地域での分布情報が少な
く，その様な地域において分布域を過小評価する可能性
がある。また生息密度についても，第 1次計画内におい
て一部地域において低密度化が報告されているが，第 2
次計画以降の推移は不明である。そのため，神奈川県に
おけるアライグマの分布域と相対密度の経年的変化を調
べ，アライグマ防除事業の効果を検証した。分布域調査
として痕跡調査や自動撮影調査，聞き取り，文献調査，
捕獲記録により分布域を把握した。3次メッシュによる
アライグマの分布メッシュ率は 65.2%となり，県内の
平野部のほぼ全域に広がっており，丹沢山地や箱根山地
においても複数箇所で分布が確認された。相対密度調査
として神奈川県における 2005年から 2013年の捕獲記録
を，3年ごとに防除事業前期，防除事業中期，防除事業
後期に分け，3次メッシュごとに捕獲効率を算出した。
防除時期における捕獲効率の変化の比較には，ポアソン
分布を仮定した一般化線形混合モデル（GLMM）によ
り調べた。防除実施前の分布域と防除策定以降の分布図
を比較すると，防除計画策定以降は分布域が 1.6倍に拡
大していた。地域ごとに相対密度の経年的変化をみる
と，捕獲効率の減少がみられたのは横浜市と湘南地域の
みで，他地域では密度減少はみられなかった。
これらのことから，神奈川県において分布域の縮小と
生息数の減少はみられず，神奈川県アライグマ防除事業
は目標を達成できていないと考えられた。その原因とし
ては第 2次計画から取り入れられたメッシュ管理が十分
に機能せず，捕獲努力目標を達成できたメッシュが少な
かったことが要因の一つであると考えられた。

5． 総合考察
本研究は，アライグマの防除事業の評価に係る技術と
して，新たに開発したアライグマの分布確認手法の調査
効率を検証するとともに，密度指標の開発を行った。さ
らに，これらの調査技術を用いて神奈川県のアライグマ
の分布域と相対密度の経年的変化を調べ，防除事業の効
果を検証した。アライグマの分布確認手法としては，本
研究で扱ったスタンプ板や自動撮影カメラ以外にも複数
の調査手法があるが，それぞれ特性がある。アライグマ
の防除事業の立案単位は，通常，市町村や都道府県単位
と面積が広域であり，その中にも人間活動の濃淡や環境
の違いが存在する。そのため，調査対象全域の分布域を
単一の手法で把握することは困難である。正確な分布域
を広域的に収集するには，各手法のメリットを活かして
複数の調査手法を組み合わせて実施した方が現実的であ
る。そのため，アライグマ防除事業の立案者は，単一の
調査手法に囚われることなく，複数の調査手法を組み合
わせて分布調査を行う実施体制が望まれる。アライグマ
の相対密度としては，自動撮影調査から得られた撮影効
率が有効であることが分かった。捕獲効率はトラップ
シャイ個体による影響やアライグマの防除事業において
は捕獲条件の統一の困難さから，個体数減少を検証する
唯一の指標とするべきではない。一方，自動撮影カメラ
から得られる撮影効率は特別な調査が必要であるが，相
対密度を得られる手法としては，種同定の精度が高く，
調査に係るランニングコストが低く抑えることが可能で
あり，手法としての捕獲行為と完全に独立した調査であ
るため，捕獲効率とのクロスチェックとしても適してい
ると考えられた。分布域と相対密度の経年的変化から神
奈川県の防除事業を評価した結果，神奈川県内のアライ
グマ分布は防除実施計画策定後の分布が拡大しており，
相対密度も減少したのは一部の地域のみであった。この
ことから防除事業が上手く機能していなかったことが分
かった。防除事業の評価としては生物的側面の他に社会
的側面がある。社会的側面は生物的側面である分布域や
個体数を抑制できなかった原因を理解する上で有用であ
る。今後は生物的・社会的の両側面から防除事業を評価
することで，防除事業を改善するためのより有効な情報
が得られると考えられる。
審査報告概要
外来種アライグマ（Procyon lotor）は，日本各地で
野生化し大きな社会問題となっている。この防除事業を
効果的に進めるためには，本種の分布域と個体密度を継
続的にモニタリングすることが必要である。本研究で
は，本種の分布確認手法および密度指標を開発し，神奈
川県における分布域と相対密度との経時的な変化を明ら
かにした。分布確認手法では自動撮影調査と新たに開発
したスタンプ板調査を比較し，両調査法が有効であるこ
とと，自動撮影調査の調査効率が高いことを示した。ま
た，自動撮影調査の撮影効率は，防除事業データから得
られた捕獲効率との間に有意な正の相関を認めたことか
ら，密度指標として有効であることを示した。本研究で
はアライグマ防除事業に貢献する新たな知見を得た。
よって，審査員一同は博士（畜産学）の学位を授与す
る価値があると判断した。

Sirtuin, often referred as the longevity gene, was
first identified as SIR2 in yeast and its homologous
genes have been widely found in both eukaryotic and
prokaryotic organisms. In addition, the main enzyme
activity of sirtuin proteins were proven that it was a
NAD+-dependent deacetylase of which the substrates
are histones, p53 and other acetylated proteins in the
nucleus or cytoplasm. In prokaryotes, sirtuin gene
was identified as cobB, encoding a cobalamin process-
ing enzyme, and later its involvement in regulating
metabolic enzymes, transcription factors, chemotactic
proteins and others as NAD+-dependent deacetylase.
On the other hand, little is known about its roles in
lactic acid bacteria (LAB).
LAB are widely consumed by human as food fer-
mentation starters and as probiotics. Probiotics are
live bacteria that are thought to be beneficial in
preventing several health conditions. According to the
2002 definition by the World Health Organization
(WHO), probiotics are “live microorganisms which,
when administered in adequate amounts, confer a
health benefit on the host.” Therefore, those probiotics
bacteria must be alive in probiotics product before they
were administered to the host. In addition, the im-
portant function of the probiotics was known to
regulate the functions of intestine and immunopoten-
tiative actions. Therefore, the author assumed that it
is important to increase stress tolerance, intestinal
tract adhesion and the useful substance produced by
LAB in order to confer more beneficial health effects
on the host. In this study, the author intended to
analyze the role of sirtuin in LAB.
In the chapters 1 and 2, the author analyzed sirtuin
homolog genes of LAB and analyzed enzyme kinetics
of the recombinant L. paracasei sirtuin proteins. In
the chapter 3, the author identified several candidate
target proteins both in vivo and in vitro by using the
recombinant LpSirA protein and sirtuin inhibitor
nicotinamide (NAM). One of the target was identified
as 30S ribosomal protein S4. In the chapter 4, the
author analyzed intracellular localization of sirtuin
using immunofluorescent staining and LpSirA-Venus
fusion protein in L. paracasei.
Chapter 1. Identification of sirtuin genes in the
genomes of the L. paracasei strains and its ho-
mology to sirtuins of related bacteria
The author demonstrated that almost all LAB have
homologs of eukaryotic sirtuin. Interestingly, the
author was not able to find the gene in Lactococcus spp
and Melissococcus spp. In a previous study, the
author’s group determined draft genome sequences of
three L. paracasei strains NRIC 0644, NRIC 1917 and
NRIC 1981 (Shiwa et al., 2015). The sirtuin genomes
of three strains identified highly homologous (99.9%)
to those of L. paracasei BL23. The author tentatively
designated the gene as sirA (indicating the first sirtuin
homolog of L. paracasei). Interestingly, there was
another sirtuin isozyme found in the genome of strain
NRIC 1981, which was hit by BLAST for L. rhamnosus
GG sirtuin with 78% identity. We designated this
second gene as sirB, which was not found in the ge-

nomes of the strains NRIC 0644, and NRIC 1917.
LpSirA encoded by sirA (693 bp) gene was shown to
have two conserved active histidine residues and the
NAD+-binding motif as reported in human SIRT1. On
the other hand, the LpSirB encoding by sirB (726bp)
gene contained similar NAD+ -binding domains, but
only one conserved active histidine residue (his 79)
site. When comparison was made using LALIGN
program, the LpSirA sequence was 26.2% homologous
to yeast Sir2 (BL23 LpSirA residues 8-226 vs S.
cerevisiae Sir2 residues 251-518) and 29.4% homolo-
gous to human SIRT1 (BL23 LpSirA residues 3-208 vs
SIRT1 residues 245-469). The amino acid sequence
homology of LpSirA proteins with sirtuins of E. coli,
Salmonella enterica serovar Typhimurium LT2 and B.
subtilis were 29.3%, 29.5% and 31.1% respectively.
The amino acid sequence of LpSirB protein showed
only 80% homology to other reported L. rhamnosus
sirtuins which share 94-97% homology with each
other.
Chapter 2. Enzyme kinetics of the recombinant
LpSirA protein
Expression and purification of recombinant L.
paracasei sirtuin was conducted with E. coli expres-
sion system. The sirA and sirB were cloned into bac-
terial protein expression vector pET-15b. The con-
structed pET-15b plasmid was transformed into E. coli
BL21 (DE3) to express recombinant LpSirA and
LpSirB. The recombinant proteins with histidine tag
were expressed and purified on a Nickel-affinity col-
umn. However, the recombinant LpSirB protein was
found in the inclusion body of E. coli BL21 (DE3).
Therefore, for the purification, the protein was dis-
solved in the presence of 8 M urea. Recombinant
proteins were detected in single bands as apparent
molecular sizes of 29kDa for LpSirA, and 34kDa for
LpSirB on SDS-PAGE.
Kinetics studies were performed using Flour de
Lys® fluorimetric activity measurement kit. The
relative fluorescence unit was measured using Nano-
Drop 3300Fluoropectrometer. The substrate used was
an acetylated peptide comprising amino acids 379-382
of human p53. The control recombinant SIRT1 protein
was provided in the same kit. The apparent Km and
Vmax of SIRT1 and LpSirA from NRIC 0644, 1917 and
1981 were calculated from Line weaver Burk plot
obtained in the presence of 3mM NAD+ (fixed) and 0.2
µg of each enzyme protein in the reaction mixtures.
The standard curve was calculated from using de-
acetylated fluorescent peptide substrate (Flour de
Lys® deacetylated standard) provided in the kit. In
addition, the author tested SIRT1 activator resvera-
trol and SIRT1 inhibitor suramin. The apparent Km
values were determined to be 130.2µM, 186.3µM, 180.1
µM and 130.1µM for human SIRT1, L. paracasei NRIC
0644 LpSirA, NRIC 1917 LpSirA and NRIC 1981
LpSirA, respectively. The Vmax values were 257.5
nmol/min/mg, 160nmol/min/mg, 170nmol/min/mg and
212.5nmol/min/mg for SIRT1, NRIC 0644 LpSirA,
NRIC 1917 LpSirA and NRIC 1981 LpSirA proteins,
respectively. The optimal temperature for the enzy-
matic reaction of LpSirA proteins displayed higher
optimal temperature (45-50℃) than SIRT1 (37℃). In
addition, Resveratrol decreased in the apparent Km
values of human SIRT1, but the conclusion has not
been reached yet on the resveratrol effects to apparent
Km values of NRIC 0644 LpSirA. Therefore, in ad-
dition to examine the reproducibility, the author
thinks it is important to search for new activator
which strongly activate the enzyme activity of LpSirA.
Suramin inhibited deacetylase activity of both human
SIRT1 (IC50 : 18µM) and NRIC0644 LpSirA (IC50 : 359
µM). The results suggests the similarity and the
difference of enzyme property between human SIRT1
and LpSirA.
Chapter 3. Identification of the sirtuin-target
acetylated proteins in L. paracasei BL23
Hereafter, instead of the three strains, a widely
studied standard strain, L. paracasei BL23 was used.
The putative target proteins of sirtuin in L. paracasei
BL23were first screened by inhibiting sirtuin deacety-
lase using NAM in the culture medium. In parallel,
cell extracts (from cells cultured in MRS supplemented
with 50mM NAM) containing 100µg protein was
treated in vitro with 10µg purified recombinant
LpSirA in the presence of 10mM NAD+ to maximize
NAD+-dependent deacetylation. From each of the in
vivo and in vitro target samples obtained above, 100

µg cellular protein was subjected to 12.5% SDS-PAGE,
and blotted onto PVDF cellulose membrane. Western
blotting was done using anti-LpSirA primary antibody
or Acetylated-Lysine primary antibody, together with
donkey anti-rabbit IgG secondary antibody. The
results from each of the in vivo and in vitro samples
indicated that a 28kDa acetylated protein was the
target protein of sirtuin. Furthermore, the 28kDa
acetylated protein was purified using ammonium
sulfate precipitation (0-80%), Butyl-Toyopearl column
and DE52 column. The elution at 0.5 M NaCl from
DE52 column showed single band of an acetylated 28
kDa protein, and this protein was deacetylated by
LpSirA. The fraction containing 28kDa protein was
further concentrated by acetone precipitation (80%),
and subjected to SDS-PAGE followed by electro-
blotting onto FluoroTrans® PVDF Transfer Mem-
brane. Its N-terminal amino acid sequences were de-
termined by the Edman degradation method using a
peptide sequencer PPSQ30. The N-terminal Amino
acid sequence of the 28kDa target protein was deter-
mined to be SRYTGPRWKQ, which was perfectly
identical to that of 30S ribosomal protein S4 of L.
paracasei in the data bank. The function of 30S
ribosomal protein S4 was known to assemble the
rRNA together with 30S ribosomal subunit protein S5
and S12. Additionally, the ribosomal large subunit
component MRP10 of mitochondria was deacetylated
by mitochondrial sirtuin (SIRT3), and it was found to
decrease protein synthesis rate. Thus, the author
presumed that LpSirA is involved in the regulation of
protein synthesis in L. paracasei BL23.
Chapter 4. Intracellular localization of sirtuin
protein in L. paracasei BL23
First, the author analyzed intracellular localization
of sirtuin using anti LpSirA antibody in L. paracasei
BL23. Cells were fixed on cover glasses and treated
with 4% paraformaldehyde for 20min. Cells were
probed with a rabbit anti-LpSirA antibody (1 : 250
dilution) and anti-rabbit Alexa Fluor® 488 dye
antibody (1 : 2,000 dilution) for 1h at 37℃. The nu-
cleoid of the cells were stained with 5µg/ml Hoechst
33342 for 10min. Finally, a drop of SlowFade® Dia-
mond was applied to the sample and localization of
LpSirA was examined in L. paracasei BL23 by Con-
focal Microscopy. The result showed that LpSirA was
localized on the division plates and cellular poles
during cell division.
Next, the author analyzed intracellular localization
of sirtuin using LpSirA-venus fusion protein in the
living cells of L. paracasei BL23. The sirA gene se-
quence (693bp) was cloned into pCS2 Venus plasmid
equipped with an incorporated Venus gene. The sirA-
venus fusion gene was cloned into bacterial protein
expression vector pLPM11. The pLPM11 construct
(5µg) was transformed into competent L. paracasei
BL23 cells by electroporation. Transformant cells ex-
pressing LpSirA-Venus were inoculated into 5ml MRS
medium containing erythromycin and were grown for
12 hours at 37℃ in incubator. The 5ml culture was
washed with 0.86% saline and inoculated to 5ml of
GYP medium (where the glucose was replaced with
galactose). Finally, the localization of LpSirA in the
LpSirA-venus high expresser strain was examined by
confocal microscopy. The result indicated that LpSirA-
Venus protein was localized with a regular pattern,
such as a spiral in the cytoplasm. In Bacillus subtilis,
FtsZ in the Z-ring was shown to be localized to the
division plates during cell division. In the cellular
pole, there are RacA protein which promotes the
movement to the cell pole of the nucleoid, and the cell
division protein DivIVA which is connecting the RacA
to cellular poles. Further, the actin homolog MreB
protein and the MinC, D, E and J proteins were
localized as a spiral in the cytoplasm. Thus, the
author presumed that LpSirA is involved in cell
division in L. paracasei BL23.
In this thesis, the author found that the existence of
sirtuin gene in LAB and their deacetylase enzyme
activities in three strains of L. paracasei. In addition,
the author revealed that the target protein of LpSirA
in L. paracasei BL23 is 30S ribosomal protein S4which
is involved in protein synthesis. Using immunofluor-
escent staining, LpSirA protein was shown to be
localized to the division plates and cellular poles
during cell division. Additionally, using the LpSirA-
Venus high expresser strain, LpSirA protein was lo-
calized with a regular pattern, such as a spiral in the

cytoplasm of living LAB. In conclusion, the author
concluded that the function of sirtuin may likely be
involved in protein synthesis control and cell division
in L. paracasei. Thus, the author expects that sirtuin
may serve as a novel criteria to select LAB for
developing better probiotics to contribute for the pro-
motion of human health.

サーチュインは，代謝の適正化やストレス応答などに
かかわる長寿遺伝子として注目されている。その本体は
NAD+依存性のタンパク質脱アセチル化酵素であり，染
色体ヒストンタンパク質に作用して遺伝子発現を制御す
る。一方原核生物では未知の部分が多い。本研究では先
行研究例のない乳酸菌に着目し，プロバイオティクスと
して繁用される Lactobacillus paracaseiより遺伝子を
クローニングし，組換えタンパク質の産生と精製を経
て，脱アセチル化酵素活性を検出し，LpsirAと命名し
た。次に同菌種における標的アセチル化タンパク質の一
つを精製し，N 末端アミノ酸配列を決め，30S riboso-
mal protein S4であることを同定した。さらに，抗体と
蛍光タンパク質融合体を用いて，その細胞内局在が，細
胞極，細胞分裂面，らせん構造であることを示した。こ
れら新規の発見は，乳酸菌の生存やストレス応答機構の
解明に迫る貴重な一歩である。
よって，審査員一同は博士（生物産業学）の学位を授
与する価値があると判断した。

Spotted seal (Phoca largha) strongly depends on sea
ice because they breed on sea ice. Hokkaido is the
southernmost extent of their distribution. Their
habitat range is greatly influenced and changed by
sea-ice dynamics and the number of seals also strongly
affected by hunting pressure along the Hokkaido
coast. Due to these factors, in recent years, many
spotted seals had been observed in the Sea of Japan of
Hokkaido when seals did not distribute in the past and
thus, the ecological changes of spotted seals had been
reported in this region. The number of seals gradually
increased in the Sea of Japan year by year. Thereby,
their distribution in the Sea of Japan expanded
southward and the numbers of haul-out sites also
increased in this region. Under such conditions, the
number of regions in which harmful-animal controls
on spotted seals have been conducted with the aim to
mitigate damage to fisheries has increased in recent
years.
As the southernmost extent of the seals’ distribu-
tion, the coast of Hokkaido is more affected by en-
vironmental conditions than areas of the distribution
further north. Therefore, the effects of global warm-
ing are especially feared in recent years, confirming
the population dynamics of seals along the Hokkaido
coast is crucially important. Additionally, competition
for haul-out sites and food is expected to intensify with
the excessive density of seals in their habitat and by
the growing numbers of seals. Thereby, the popula-
tion structure of spotted seals is expected to change,
and damage to fisheries would also possibly increase.
The results of efforts to monitor spotted seals thus
provide important indicators verifying the soundness
of the whole marine ecosystem, because these seals are
a higher predator in the food chains of the marine
ecosystem.
Rebun Island is the northernmost island in the Sea
of Japan west of Hokkaido, and it is also the first haul-
out site in this region where spotted seals were ob-
served. This study therefore focuses on Rebun Island
with the aim to 1) confirm how the population struc-
ture of spotted seals on Rebun Island has changed, 2)
including the effects of harmful-animal controls that
have been conducted in recent years, and 3) identify
the breeding period and breeding area of seals on
Rebun Island and 4) the characteristics of behavioral
ecology within their life history.
Chapter 1. Changes in the population structure
In Rebun Island, a survey of the usage haul-out site
and the numbers of seals was conducted confirm the
population structure of spotted seals in a total of 187
days from December 2008 to December 2009. As a
result of my previous reports, before the 1990s, ap-
proximately 200 to 300 sub-adult seals had migrated to
only Todojima at Rebun Island in the Sea of Japan.
However, the usage numbers of seals have increased
since the late 1990s, and in May of the late 2000s, the

maximum haul-out number of seals was confirmed
1,000 or more. Also two haul-out sites were confirmed
at Rebun mainland as well as Todojima for the first
time in the late 1990s, besides, the 2 of new haul-out
sites were found in the late 2000s. Additionally, the
numbers of haul-out seals differed in the several haul-
out site, and, the adult seals were observed in all of
haul-out sites.
On the other hand, their period of stay in the late
2000s got longer than the late 1990s. In recent years,
these changes of the population structure have been
also seen in the others of haul-out sites in the Sea of
Japan. In addition, seals have been observed through
the year at Rebun. Spotted seals originally breed on
sea ice; however, it was found that the seals in Rebun
bred at the haul-out site (on land). These ecological
changes of seals that were seen only in Rebun were
suggested because it was not yet confirmed in other
haul-out sites in the Sea of Japan.
As describing above, the ecology of seals at Rebun
Island has been significantly changed from the late
1970s, and those changes have been confirmed at haul-
out sites in the Sea of Japan west of Hokkaido at the
first time. Therefore, Rebun was considered to be a
crucially important indicator for monitoring changes
in the ecology of spotted seals in the Sea of Japan.
Since Rebun’s position as the northernmost location of
haul-out sites in this region and its proximity to sea-
ice area further north that is assumed as their
breeding areas, it was consider that is easy for spotted
seals to migrate to their breeding sites and use haul-
out sites on Rebun.
Chapter 2. Changes in numbers and haul-out
use of seals by harmful-animal controls
Since 2010, harmful-animal controls on spotted seals
have been conducted each spring in the northern
Rebun due to an increase of the number of seals after
the late 1990s. To verify the influence on seals by the
harmful-animal controls, the usage haul-out sites and
haul-out seals were investigated before (between
December 2008 and November 2009) and after (be-
tween March2012 and February 2013) the harmful-
animal controls. As a result, after controls, the num-
ber of haul-out seals in the northern Rebun of the
controls area decreased greatly, and five of new haul-
out sites were confirmed in the northern Rebun. On
the other hand, the total number of seals in Rebun
changed little was indicated. Before controls, the peak
number of mean haul-out seals of control site in the
northern Rebun was in spring, however, the numbers
greatly decreased in spring, and the peak of number
changed to winter after controls. On the contrary,
after controls, the numbers in the southern Rebun as
non-control area increased. Additionally, the photo-
identification from the photos of the haul-out seals was
conducted and the changes in the haul-out use of each
seal between before and after controls were verified.
As a result, after controls, seals that used both the
northern and the southern areas increased, and the
most of them had used only the northern area before
controls. Furthermore, since more than 90% of seals
that used both the northern and the southern areas
after controls were observed through the year, it was
suggested that the staying period of many seals got
longer than before controls. Many seals that had used
only the northern area until controls avoided the con-
trols in the northern area and became to use the
southern area in spring. Thereby, after controls, there
was a possibility that their period of stay got longer
than before controls was considered.
From these results, on the controls area during the
control period, the controls at Rebun had a direct in-
fluence on reducing the numbers of seals in the con-
trols area mainly during the period of controls.
However, many seals that avoided the controls area
used other haul-out sites. Therefore, it was suggested
that the controls mostly had no an effect to keep the
seals away from Rebun. Additionally, based on the
ecology of seals that their behavioral range are widely,
it was also suggested that seals moving to the other
haul-out sites excluding Rebun also existed.
Chapter 3. Estimation of breeding period and
breeding area of seals
To identify breeding period and breeding area of
spotted seals in Rebun Island, location information
and the daytime and nighttime haul-out ratios of seals
were analyzed by attaching the satellite transmitters
with the captured seals. As a result, the breeding area

were cleared that the adult seals in Rebun breed on
the sea-ice area of the Tatar Strait between February
and April. This finding thus represents new knowl-
edge that is different from an earlier report by Naito
and Nishiwaki (1972) which stated that the breeding
site of seals migrating to the coast of Hokkaido was on
sea ice in the Sea of Okhotsk and the Nemuro Strait.
Their breeding period and molting period are
considered to conduct on the sea-ice area of the Tatar
Strait. On the other hand, sub-adults conduct their
molting at Rebun Island. The adult seals also stayed
there longer than the sub-adults. In addition, the
distribution of adult female and male on sea ice in the
Tatar Strait overlapped; however, the distribution of
sub-adult did not overlap with the adults. This result
corresponded with the previous reports. It was con-
sidered that the sub-adults avoided places that large
numbers of adults used during the breeding season or
had purposes, such as foraging, that were different
from those of the adults, though in the same sea-ice
area. From the above, these findings suggested anew
that adults are more dependent on sea ice than sub-
adults which do not participate in the breeding.
Chapter 4. Characteristics of diving behavior in
each life history of seals
To confirm how diving behavior and behavioral
range of spotted seals change with their life history or
seasonal change, the changes of diving behavior in life
history (season) of the seals regarding to age and sex
based on the breeding period that was specified in the
chapter 3 were analyzed. As a result, it was provided
that the adult seals were observed to conduct shallow
and short diving only during the breeding period and it
suggests that diving of adult seals behavior was sug-
gested that is a peculiar diving pattern to concentrate
for the breeding activity. Furthermore, just prior to
the breeding season adult female seals were observed
to conduct deeper and longer diving than adult males
in Tartar strait. From the Rebun Sea area to Tartar
strait prior to the breeding season it was showed that
their aim was to accumulate the energy that they
required for the breeding period. This diving behavior
has been reported in other phocid seals as well. It was
considered that females accumulate much more en-
ergy than males before the breeding period because
females have to engage in the breeding activity longer
than males which do not participate in giving birth
and nursing, and because they have to maintain their
own fetus growth. In addition, it is also suggested
with adults of grey seals that the amount of energy
storage of females differs from the one of males before
the breeding period. The report was corresponded
with the result of this study.
On the other hand, the foraging behavior of sub-
adults was considered to be carried out more actively
to maintain their own body temperature and to in-
crease their own subcutaneous fat during severe
winter. On the contrary, their foraging was showed to
become passive during the molting period.
The above findings indicate that the energy balance
of spotted seals changes seasonally in relation to their
life history and that internal (psychological) factors
determine their diving patterns and haul-out ratios.
Although the earlier reports as for the diving behavior
of spotted seals have been approached indirectly only
by the analysis of location information and feeding
habits of seals so far, this study was able to clear
directly the seasonal changes in their diving patterns.
As described above, the ecology of seals on Rebun
Island has been significantly changed from the past,
and those changes have been confirmed the first at
haul-out sites in the Sea of Japan west of Hokkaido
(Chapter 1). Therefore, Rebun was suggested that is
to be a crucially important indicator for monitoring
changes in the ecology of spotted seals in the Sea of
Japan. Furthermore, the breeding area of these seals
was found to be on sea ice in the Tatar Strait, and the
breeding period was between February and April
(Chapter 3). Since Rebun’s position as the northern-
most location of haul-out sites in this region and its
proximity to sea-ice area further north that is assumed
as their breeding areas, it was consider that is easy for
spotted seals to migrate to their breeding sites and use
haul-out sites on Rebun. Also, the identification of
their breeding period and breeding site has revealed
that the diving behavior of adult seals becomes passive
only during the breeding period. Additionally, among
the adult female, the diving behavior that the energy

demand is high from autumn in the Rebun Sea area
had been confirmed (Chapter 4). The Rebun sea
region has many haul-out sites for resting, a large
shallow area, and an abundant variety of food re-
sources in all four seasons. The Rebun sea region has
thus become a very important foraging area for seals
that need to satisfy their high demand for energy prior
to the breeding period.
The common changes in the Sea of Japan Sea,
including increases in the number of seals, the ex-
pansion of their distribution southward, and their
longer periods of stay (Chapter 1), quickly contributed
to greater severity in the conflicts between seals and
fisheries in this region during seasons when seals were
previously not there. Since fisheries in the Sea of
Okhotsk do not operate during the sea-ice season
while inshore fisheries continue to operate in the Sea
of Japan during the same period, it is possible that the
seals are attracted to the Sea of Japan as a foraging
site. Seals can become dependent on fisheries because
they can easily prey on fish caught in the nets and
consume less energy for foraging. Dependence on
fisheries enables more seals to survive rather than
succumb to the difficulties of foraging on their own. It
also means, however, that the number of seals that
survive could exceed the environmental carrying ca-
pacity of the sea in that region and thus upset the
balance of the marine ecosystem and lead to ecological
degradation. To block such problems, harmful-
animal controls on spotted seals have been conducted
each spring since 2010 on the northern side of Rebun.
As the results of this study of the effects of harmful-
animal controls on the spotted seals in Rebun Island,
the results therefore suggested that the controls had
little lasting effect on seals at Rebun. They also sug-
gested that seals moved to other haul-out sites away
from Rebun (Chapter 2). Since spotted seals are
known to move long distances over a large area, this
study therefore recommended that seals be managed
in the entire Sea of Japan region west of Hokkaido.
Based on this recommendation, “The Management
Plan of Seals at Hokkaido in 2015” was established to
reduce the numbers of seals in 2015 from summer to
autumn in the Sea of Japan, mainly at Rebun Island
(Hokkaido 2015). In the future, the effects of those
controls should also be continuously analyzed and
evaluated from scientific perspectives.
The above findings indicate that the energy balance
of spotted seals differs between age class and sex, and
changes seasonally in relation to their life history and
that internal (psychological) factors determine their
diving patterns and haul-out ratios (Chapter 4).
Therefore, the differences of seasonal changes in the
number of seals at each haul-out site in Rebun
(Chapter 1) are considered a possibility that is in-
fluenced by the internal factors. Global warming has
also been a factor in recent years. The possibility that
the distribution of seals will expand southward along
coastal regions of Japan cannot be denied, because
seals can possibly balance their energy needs more
efficiently by moving to warmer waters. The internal
factors of spotted seals, however, being adapted to cold
regions, may limit the extent of their southward
expansion. On the other hand, sea-ice areas and sea-
ice conditions are expected that will decline at the
same time. If spotted seals need to migrate further
north to find sea ice for giving birth and nursing, their
movement further south may be limited, as they would
need to migrate even greater distances between their
northern and southern habitats. Furthermore, declin-
ing sea-ice conditions would also reduce the numbers
of seals giving birth and weaken their abilities to give
birth and nurse. There is thus an increased risk of
pups being abandoned or births occurring under water
where they would be more exposed to natural pred-
ators. Since these advantages and disadvantages are
mixed, it is essential to conduct more detailed studies
on the seals’ behavioral ecology including their energy
balance.
The fact that previous research has not reported on
the migrating population to Hokkaido that breeding in
the Tatar Strait suggests the possibility that a popu-
lation was introduced from a new another region and
may become part of the population that migrates to
Hokkaido. Additionally, as described above, if seals
increasingly breed on land, the population structure
will probably change. In addition to factors such as
the rise in sea level and decreases in the sea-ice area,
increases in the number of seals and the longer periods
of stay are expected to impact and change population

dynamics. The author therefore hopes to conduct
longer-term and more detailed monitoring on the
genetic structure of the populations and the habitation
status of the spotted seals. Monitoring the spotted
seals will provide important indicators that could
verify the soundness of the whole marine ecosystem,
because seals are a higher predator in the food chains
of the marine ecosystem.
審査報告概要
本論文は，未だに多くが未解明であり，漁業産業との
多大な軋轢が存在するゴマフアザラシの生態学的知見を
示すため，近年本種の生態変化が生じている北海道礼文
島の生息個体群に着目し，過去からの生息状況の変遷と
生活史ごとの行動生態を明らかにすることを目的として
遂行された。第 1章では，礼文島における本種の生息状
況は過去から大きく変遷し，礼文島が日本海側における
本種の生態変化の早期的指標となる場所であることが明
らかになった。第 2章では有害駆除による本種への影響
が初めて学術的に評価され，本種の個体数管理のために
極めて重要な資料が提示された。第 3章，第 4章では，
衛星発信器により，礼文利用個体が間宮（タタール）海
峡で 2月∼4月に繁殖していることが初めて直接的に明
らかにされ，既存報告とは異なる場所で繁殖していた。
それにより本種の生活史ごとの潜水行動の変動が初めて
直接的に示された。本研究の成果は，個体数増加による
漁業被害が急速に深刻化している昨今において，本種の
生態学的知見が盛り込まれた，重要な知見となる。
よって，審査員一同は博士（生物産業学）の学位を授
与する価値があると判断した。

現在，石油資源枯渇が危惧される中，産油国ではない
我が国はその持続的かつ安定的な供給が可能であるとは
言い難い。このようなエネルギー供給事情を背景に，持
続的かつ安定的な原料供給が可能な植物バイオマスを原
料としたエネルギー生産方式の構築が求められてきた。
特に植物バイオマスに含まれる炭化水素を発酵により燃
料に変換するバイオ燃料生産が注目されている。中でも
バイオエタノールにおいては大規模プラントでの生産が
行われ燃料としての商業的な利用も普及しつつある。し
かし現状では，バイオエタノール生産に利用されている
原料はトウモロコシ，サトウキビなどの可食原料であ
る。これら可食原料の燃料生産への利用によって食料と
の競合が生じ，燃料生産への利用の開始と同時にそれら
食料の市場価格は上昇した。このため，食料ではなく現
在廃棄物として処理されている未利用資源の有効活用が
求められる。米食を主とする我が国において稲わらはバ
イオ燃料生産に最適なバイオマスである。稲わらは米作
の副産物として生じ，その多くは農業廃棄物として処理
されている。稲わらはその乾燥重量の 70%程度が炭化
水素であり，特にグルコースが b1-4結合したポリマー
であるセルロースを主要構成成分とする。本研究ではセ
ルロースを主成分とする未利用資源の中でも特に稲わら
を基質として利用することで，カーボンニュートラルな
エネルギー生産を目指した。一方，生産するエネルギー
としてブタノールに着目した。ブタノールは C4のアル
コールであり，その熱量は 34MJ/kgでエタノールの熱
量 27MJ/kgに比べ高い。更に親水性が低いことから金
属腐食性も低いため，燃料として優れた性質を有してい
る。また，ブタノールは工業的にも利用価値が高く，塗
料，ゴム，吸水ポリマーなど化成品原料としての用途も
多岐に渡る。しかし，そのブタノールも原油からの化学
合成により製造されることから，化学工業においてもブ
タノールの安定的かつ恒久的な提供が可能なバイオマス
からの生産系の構築が期待されている。ブタノールは一
部の Clostridium 属細菌により発酵生産が可能である
が，高ブタノール生産性 Clostridium 属細菌において
はセルロース分解能を有する株は報告がない。また，セ
ルロースを高効率に分解する細菌にも高ブタノール生産
性を示す株も報告がない。そこで本研究では，高セル
ロース分解性 Clostridium 属細菌と高ブタノール生産
性 Clostridium 属細菌の混合培養によってセルロース
系の未利用資源を基質としてブタノールを高効率に生産
する並行複発酵系の構築を目指した。更に本混合培養系
においてセルロース分解性を向上させることでブタノー
ル生産性も向上させることが可能であることを見出した
ことから，ブタノール生産性 Clostridium 属細菌にお
ける異種セルラーゼ高分泌によるセルロース分解性向上
を目的として稲わらを基質とした際に利用されるシグナ
ルペプチドの同定を行った。また，培養温度を上昇させ
ることはセルラーゼ活性向上のもう一つの因子であるた
め，至適発酵温度が 30℃であるブタノール生産菌のよ
り高温域への培養温度の向上を試みた。
1． セルロース基質を用いた混合培養系の条件検討
Clostridium属細菌の中でも高いセルロース分解能が
報告されている Clostridium thermocellum NBRC103400
（以下 NBRC103400）を用い，純粋なセルロースである
結晶性セルロースを用いた混合培養系の条件検討を行っ
た。その結果，最適な条件は「基質濃度 4% で NBRC
103400を初発菌体濃度 OD600nm＝1.0で植菌し，60℃で
24時間培養後，ブタノール生産菌 Clostridium saccha-
roperbutylacetonicum N1-4（以下 N1-4）を初発菌体量

OD600nm＝1.0で植菌し 30℃で培養」することであると
明らかにした。本混合培養系により 40g/Lの結晶性セ
ルロースを基質として 7.9g/Lのブタノールを生産可能
であり，グルコースを基質とした場合と同等程度の収量
を実現した。このことから本混合培養系によってセル
ロースから高ブタノール生産が可能であることを明らか
にした。
2． 混合培養による稲わらからのブタノール生産系の構
築
NBRC103400と N1-4の混合培養によりセルロースか
らの高ブタノール生産が可能であることが明らかになっ
たため，実用基質である稲わらを用いた時の混合培養系
での高ブタノール生産条件の探索を行った。稲わらのよ
うなバイオマスは結晶性セルロースのような純粋なセル
ロースとは異なり，複合多糖のヘミセルロース，フェ
ノール系ポリマーであるリグニン，灰分なども含んだ複
合基質である。特にリグニンはセルラーゼの阻害作用が
強いことが知られており，本混合培養系においても熱水
による蒸煮処理をおこなった稲わら 40g/L からは 2.4
g/Lのブタノールしか生産できなかった。そこで，アル
カリ処理による脱リグニン処理を施した結果，リグニン
含量は対乾燥重量で 24.7%から 5.5%に低下し，その脱
リグニン稲わら 40g/L を基質とした混合培養では 5.5
g/Lまでにブタノール生産量は向上した。しかしなが
ら，そのブタノール生産量は未だに結晶性セルロースの
7.9g/Lより比べて低かった。そこで，当培養系におい
てセルラーゼ活性が不足しているのではないかと考え，
Aspergillus niger由来の商用セルラーゼを培養系に添
加したところブタノール生産量は 1.3倍の 6.9g/Lに向
上した。一方，NBRC103400，N1-4 および商用セル
ラーゼのエンドグルカナーゼ，エキソグルカナーゼおよ
び b-グルコシダーゼの 3種類のセルラーゼの酵素活性
を測定し比較したところ，NBRC10400と N1-4に比べ
添加した商用セルラーゼはエキソグルカナーゼ活性と
b-グルコシダーゼ活性が高いことが明らかになった。
商用 b-グルコシダーゼのみを添加して混合培養を行っ
たが，ブタノール生産量の向上は確認できなかったこと
から，ブタノール生産量向上に重要な因子はエキソグル
カナーゼ活性の向上であることが明らかとなった。
3． エキソグルカナーゼ発現に向けた分泌シグナルの探
索
第 3章では N1-4におけるエキソグルカナーゼ高発現
のため，細胞内で生産された酵素を細胞外へと分泌する
ための分泌シグナルの同定を行った。N1-4をグルコー
ス，セロビオース，キシランおよび脱リグニン稲わらで
培養し，培養上清のタンパクを SDS-PAGEに供した。
その結果，いずれの炭素源を用いた場合も異なるバンド
パターンを示したことから，基質により分泌されるタン
パク質が変化することが明らかとなった。キシランおよ
び脱リグニン稲わら培養において高発現していたタンパ
ク質をアミノ酸シーケンス解析に供することで，N末
端のアミノ酸配列を解析した。その結果，特に稲わらを
基質にした際に誘導される Xylanaseのシグナルペプチ
ドを同定した。このシグナルペプチドはゲノム解析から
推定されている特徴的な 3つのドメイン（正に帯電する
アミノ酸から構成される n-domain，疏水性アミノ酸か
らなる h-domain，シグナルペプチド SPaseIが共通で
認識する c-domain）と高い相同性を示した。以上の様
に，稲わらを基質とした際に高分泌されるシグナルペプ
チドの同定に成功し，今後は本シグナルペプチドを用い
たエキソグルカナーゼの高分泌が期待される。
4． メタボローム解析を用いた N1-4における高温での
増殖，ブタノール生産低下原因の探索
一般的に，セルラーゼ活性は 50℃∼70℃のような高
温が至適温度であるものが多く，N1-4植菌後 30℃で 14
日間の培養期間に徐々にブタノール生産を行う本混合培
養系ではセルラーゼ活性は低い状態で並行複発酵をして
いることを示す。そのため，培養温度を向上させること
ができれば，培養系のセルラーゼ活性を向上させブタ
ノール生産量向上が可能と考えられる。しかしながら，
N1-4は温度感受性が高く，グルコース 40g/Lで単独培
養を行った場合においても，30℃から 37℃へ培養温度
を上げると菌体量（OD600nm）は 7.1から 3.6へ 50%低
下し，ブタノール生産量は 10.3g/Lから 2.1g/Lへ 80%
も低下する。これは混合培養においても同様であり，結
晶性セルロースでは 30℃から 34℃で 30%，37℃で 60%
低下した。このため，混合培養系の温度上昇には N1-4
の熱耐性の獲得が必要とされるが，その熱感受性機構は
不明であった。37℃では生育能が低下することから，
N1-4の 37℃培養では代謝経路の一部が停滞することに
よって代謝の流れが滞り増殖，ブタノール生産量の低下
を引き起こしていると仮定し，30℃または 37℃で培養
した N1-4のブタノール発酵初期，中期，後期で細胞内
代謝物量を測定後比較し，律速となる代謝経路を推定し
た。その結果，37℃培養においては解糖系をはじめとし
てアミノ酸や核酸などの生育に必須となる一次代謝の代
謝物量がブタノール発酵初期から総じて低下しているこ

とが明らかになった。さらに，37℃培養時に不足してい
るこれら一次代謝物であるアミノ酸，核酸塩基，ピリジ
ンヌクレオチドなどを培養液中へ添加し N1-4を 37℃で
培養したところ，核酸 5種類（アデニン，グアニン，チ
ミン，シトシンおよびウラシルそれぞれ 100mg/L）を
添加したものにおいてのみ菌体数とブタノール生産量が
共に向上することが明らかになった。そこで添加した核
酸塩基 5種類の培養上清における残存量を測定したとこ
ろ，アデニンのみが培養初期から急激に低下し，培養後
9時間で完全に消費した。そこで，添加した核酸塩基 5
種類から 1種類ずつ除いた 4種の核酸塩基を添加し
37℃で培養をしたところ，アデニンを添加しなかったサ
ンプルのみが無添加のサンプルと同等の低い生育量とブ
タノール生産量を示す一方，他の 4種類を除いたサンプ
ルは 5種類を添加したサンプルと同様に高いブタノール
生産量を示した。このことから，N1-4はアデニンの添
加によって 37℃における熱耐性を獲得することが明ら
かになった。また，その熱耐性の付与は最大 39℃まで
であり，40℃以上ではアデニン添加による増殖とブタ
ノール生産量の向上は確認できなかった。また，アデニ
ンはプリン代謝経路で合成される核酸塩基であるが，ア
デニンのみが特異的に作用するのか，あるいはプリン代
謝経路の他の代謝物でも可能であるかを検討した。そこ
で，プリン系の核酸塩基，ヌクレオシドおよびヌクレオ
チドを培地へ添加したところアデノシンにおいても同様
の効果が得られた。一方で，IMPやヒポキサンチンで
は効果はなく，特にアデニンを分子構造に有するものの
みが有効であることが明らかになった。以上のことか
ら，N1-4の熱感受性機構はアデニンを分子構造に有す
る化合物が低下することで引き起こされる可能性が示唆
された。
5． アデニン添加による N1-4の熱耐性獲得メカニズム
第 5章ではアデニン添加による熱耐性獲得のメカニズ
ムについて検討した。N1-4の耐熱性獲得にはアデニン
の増強が必要であることが明らかになったが，そのメカ
ニズムは不明である。多くの生物で細胞外のアデニンは
細胞内に取り込まれると AMPとして利用されるため，
アデニン添加によって N1-4の細胞内アデニンヌクレオ
チド量が増加している推察しアデニン添加時の ATP量
を測定した。30℃では ATP 量は 36時間まで培養時間
が進むにつれて増加したが，37℃ではアデニン非添加で
は 12 時間から ATP 量は減少した。しかしながら，
37℃培養時でもアデニン添加により 16時間までは 30℃
と ATP量は同等と ATPの減少は抑制されていたこと
からアデニン添加により ATP量が維持されることが明
らかとなった。次に ATP量の向上が細胞の恒常性を維
持していると推察し，生菌数の経時変化を CFU（Colony
forming Unit）測定にて評価した。その結果，アデニン
非添加の 37℃培養では 8時間から CFU が急激に低下
し，20時間でコロニーは形成されなくなった。一方，
アデニン添加では CFU低下の開始は 24時間まで抑え
られ，培養 48時間までコロニーの形成がみられた。
ATPは GTPの合成に関与し，GTPの減少は Bacillus
subtilisにおいて芽胞形成との関係が報告されている。
このため，37℃培養の N1-4においても ATPの減少に
よって芽胞を形成することにより CFU低下が生じてい
る可能性が考えられた。そこで，細胞内のゲノム DNA
を蛍光染色することにより芽胞形成の経時変化を観察し
た結果，アデニン非添加の 37℃培養では培養 12時間で
は芽胞形成率は 90%であり，アデニン添加時には芽胞
形成は 20%でありアデニン添加によって芽胞形成が抑
制されていた。以上の様に，アデニン添加によって
ATP 量と CFU の向上，芽胞形成率が低下することが
明らかとなった。
6． 総 括
本研究は以上の様にセルロース分解菌とブタノール生
産菌の混合培養による未利用資源である稲わらからのブ
タノール生産系を構築し，その生産性の向上を試みた。
C. thermocellum NBRC103400によるセルロースの糖
化，C. saccharoperbutylacetonicum N1-4 によるブタ
ノール発酵を組み合わせることにより，40g/Lの結晶性
セルロースから 7.9g/Lの高ブタノール生産に成功した。
また，未利用資源である稲わらにおいてもリグニン除
去，セルラーゼ増強を加えることで最大 6.9g/Lのブタ
ノール生産を実現した。また，その培養系におけるセル
ラーゼ活性増強の第一段階として，シグナルペプチドを
同定した。更に培養系の温度向上のため，熱ストレス下
における N1-4の代謝物解析を用いて，アデニンの増強
が熱耐性獲得に重要であり，アデニンの添加によって熱
ストレス下においても至適温度の 30℃と同等のブタ
ノール生産が可能であることを明らかにした。以上のよ
うに，未利用資源である稲わらからの簡便且つ低コスト
なブタノール生産系を構築し，また熱ストレスによる核
酸合成系の重要性を提唱した。

審査報告概要
本研究は，Clostridium属細菌を用い高温性セルロー
ス分解菌と中温性ブタノール生産菌の混合培養による植
物バイオマスからの高効率ブタノール生産系の開発を目
的とした。混合培養の条件を最適化後，植物バイオマス
である稲わらからも脱リグニン処理を施すことで効率的
にブタノール生産が可能であること，エキソグルカナー
ゼ活性を向上させることでさらなる高生産が可能である
ことを見出した。また，エキソグルカナーゼ活性向上を
目指して，その発現に必要とするシグナルペプチドの同
定，培養温度の向上についても検討した。メタボローム
解析と生理学的解析からブタノール生産菌は 37℃では
生育と発酵能が著しく低下するものの，不足代謝物であ
るアデニンを添加することで生育と発酵能を回復させる
ことが可能であり，アデニン添加は細胞内 ATP量の向
上と胞子形成の抑制に寄与することを明らかにした。こ
れらの知見は，植物バイオマスを原料とした安価なバイ
オブタノール製造技術に寄与するものである。
よって，審査員一同は博士（醸造学）の学位を授与す
る価値があると判断した。
