A comparative study of bioactive and volatile components in cold-pressed apricot and peach kernel oils: implications for nutritional, nutraceuticals and cosmetics functional properties
This study provides a comparative analysis of the bioactive and volatile components in cold-pressed apricot (Prunus armeniaca L.) and peach (Prunus persica L.) kernel oils, emphasizing their potential applications in the food, nutraceutical and cosmetic industries. Both non-commercial cold-pressed oils and commercial samples were evaluated for their fatty acid profiles, volatile organic compounds, phenolic content, tocopherols, and other bioactive molecules. GC-FID analysis confirmed that both oils are rich in oleic and linoleic acids essential for their nutritional and skin-care properties, with varying levels of antioxidants, including tocopherols and polyphenols, which contribute to their health-promoting properties. The analysis of volatile organic compounds, using HS-SPME-GC-MS, revealed distinct aroma profiles, with benzaldehyde as a key marker with a concentration in the range of 0.010–16.370 mg/kg, a wide range, that can be influenced by different factors including the botanical source as well as the processing and storage conditions. Furthermore, acrolein, a marker of lipid spoilage, was only detected in commercial oil samples, ranging from 0.005 to 0.385 mg/kg for apricot and 0.017–0.097 mg/kg for peach, respectively. Finally, thermogravimetric assessments showed non-significant differences between the samples, demonstrating significant stability of cold-pressed oils, while HPLC-ESI-MS identified 9 polar phenolic compounds. Cyanogenic glycoside analysis verified the safety of the oils, with undetectable or trace amounts in all samples. Thanks to these analytical methods, it was possible to identify the qualities of the oil and to assess whether non-commercial cold-pressed oils were more suitable for cosmetic or food use. Therefore, the results suggest that non-commercial apricot kernel oil, with its favorable PUFA/SFA ratio, is ideal for food applications, and both non-commercial oils can be used in cosmetic formulations

Efficacy of myo-inositol and zinc on insulin resistance in a paediatric population with obesity
Aim: To assess the efficacy of the combined administration of myo-inositol and zinc, a mineral involved in the insulin pathway, in paediatric obesity with insulin resistance on HOMA-IR, glucose-insulin metabolism, and lipid profile. Materials and Methods: Double-blind, randomized, placebo-controlled study conducted in North Italy. Fifty-six patients (10–18 years, Tanner stage ≥3) with obesity and insulin resistance were randomized to myo-inositol (2000 mg), zinc gluconate (5 mg), and galactooligosaccharides (GOS) from plant-based origin (1000 mg) (TRT) or placebo (PLC) containing only GOS from plant-based origin (1000 mg). All patients received an isocaloric diet following the Mediterranean diet style. Data were collected at baseline (V0) and after 3 months (V1). The primary outcome was the insulin resistance index (HOMA-IR). Results: Fifty out of 56 recruited subjects completed the study. TRT improved HDL cholesterol level compared to PLC (p = 0.05) but not insulin resistance. A stratified post hoc analysis was performed by sex, BMI, and subgroups of adherence to the Mediterranean diet. Subjects were divided for obesity grade, fasting insulin (p = 0.0137) and HOMA-IR (p = 0.0273) were lower in TRT than in PLC patients, with a greater effect on severe obesity. No adverse events were detected. Conclusion: Three months of supplementation with myo-inositol and zinc were beneficial on lipid profile and in managing obesity complications at least in subjects with severe phenotype. Thus, myo-inositol and zinc could be used as non-pharmacological agents. This work suggests a long-term study with a larger sample size to enrich the findings

Impairment of the T cell memory response in chronic lymphocytic leukemia patients after SARS-CoV-2 vaccination
: CLL patients face increased vulnerability to COVID-19 because of weakened immune systems from comorbidities and treatments. Therefore, the need for these patients of vaccination is of outermost importance. In our study we have evaluated T cell-mediated responses to COVID19 vaccines by performing the activation-induced markers (AIM) assay which allows to determine spike-specific CD4+ and CD8+ T cell responses. A CD4+ T cell memory response was registered in all healthy control (HC) (responders), while 28.60 % of CLL patients did not respond to the stimulation (non-responders). CD8+ T cell memory response was impaired in 61.90 % of CLL patients and in 33.33 % of HC. In addition, CLL responders showed a significant impairment of the magnitude of memory response in CD8 subset. Interestingly, impairment of the CD4+ AIM+ memory was associated to a more severe COVID-19 infection. Ibrutinib therapy had negative impact on IL-2 production by CD8+ cells, while the duration of the treatment positively affected the memory response. The majority of CLL patients don't respond well to vaccination, leaving clinicians in need of a reliable way to identify non-responders and assess the protection levels of those who do. Our findings suggest the AIM test as a promising method for screening and categorizing patients, potentially addressing this need
