    Secondo il tribunale l’immotivato rifiuto della banca di perfezionare un contratto di conto corrente con un trustee può legittimare il rimedio risarcitorio per violazione dei doveri di buona fede e correttezza nella fase precontrattuale, ma non il rimedio “reale”, cioè la costituzione del rapporto su ordine giudiziale. Una banca non è soggetta all’obbligo di contrarre l’apertura di conti correnti e, anzi, tale obbligo è escluso dal principio costituzionale di libertà economica, dall’art. 2597 cod. civ., nonché dal fatto che il trustee non necessita di un conto dedicato per attuare la segregazione patrimoniale.     Investigating facilitators and barriers of university-based administrative staff, residents and researchers to implement an active breaks intervention: a mixed-methods study

    Objective: This study aims to analyse the barriers and facilitators towards an implementation of active breaks (ABs) intervention inside the workplace. Methods: This study used a mixed-methods approach, incorporating virtual focus groups and an online questionnaire, conducted among the workers of Alma Mater Studiorum - University of Bologna (Italy). A total of 30 participants (N=30), including administrative and academic staff, PhD candidates, specialised trainees and research fellows, were involved in the study. Key themes and subthemes related to the barriers and facilitators of ABs interventions at the university were identified and categorised according to the three levels of the socio-ecological model. Results: The most commonly reported barriers are: lack of time, lack of motivation (intrapersonal level); lack of influence, lack of awareness (social level); physical space, organisational support (environmental level). The most commonly reported facilitators are: access to information, flexible work hours (intrapersonal level); group support, awareness programmes (social level); adequate spaces, organisational structure (environmental level). A significant majority (81.48%) recognised the problem of sedentary time at work, with strong interest (92.59%) in workplace interventions to reduce it. Flexibility in timing for ABs (44.44%) was preferred over fixed schedules, with preference for dedicated areas (37.04%) and outdoor spaces (29.63%). The majority (62.96%) preferred taking ABs in small groups open to all, with a qualified kinesiologist (40.74%) leading the management. Conclusions: This study identified barriers and facilitators to an ABs intervention from the perspective of administrative staff, residents and researchers. These findings could help institutions foster an environment that promotes physical activity, reduces sedentary behaviour and promotes the well-being and safety of workers through an integrated and individual-centred approach

    Arzanol Inhibits Human Dihydroorotate Dehydrogenase and Shows Antiviral Activity

    : Human dihydroorotate dehydrogenase (hDHODH), catalyzing the rate-limiting step of the pyrimidine biosynthesis pathway (PBP), is a drug target extensively investigated for various diseases including cancer, autoimmune disorders, and viral infections. We present evidence that the heterodimeric phloroglucinylpyrone arzanol potently inhibits hDHODH by competing with ubiquinone for binding to the lipophilic patch (LP) of the enzyme. Co-crystallization experiments on the enzyme-arzanol complex provided further insights into the binding pocket of hDHODH, revealing detailed features that could inspire the design of innovative inhibitors. The cellular translation of these enzymatic and biochemical data was validated in antiviral assays on SARS-CoV-2 infected cells. Taken together, these results exemplify the potential of natural products to explore novel areas of the protein druggable space and provide information relevant to multiple critical areas of drug discovery

    In situ sonoporation to enhance the tumour uptake of silicon phthalocyanine and improve PDT effectiveness in a triple negative breast cancer murine model

    The effectiveness of photodynamic therapy (PDT) has been well demonstrated in vitro, but in vivo studies have only shown a delay in tumour growth. Tumour recurrence is often reported in clinical trials and is usually associated with limited tumour uptake of the photosensitiser (PS). In this study, sonoporation (SNP) is utilised as a physical targeting tool to enhance the uptake of an untargeted PS in cells and tumour tissues. Using a chemometric approach, we identified the optimal sonoporation stimulus to maximise uptake and cell viability (frequency 1.05 MHz, tON (percentage of positive signal in a single pulse): 50 %, DC: 90 %, burst: 1 s, sonoporation time 1 min). We achieved a viability of 83.9 % ± 10.3, and an uptake in live cells of 53.1 % ± 6.6. The combination of sonoporation and photodynamic therapy resulted in a significant reduction in cell viability. In a triple-negative breast cancer model, sonoporation combined with photodynamic therapy significantly inhibited tumour growth. For the first time, our results highlight the potential of sonoporation as a non-invasive method to increase the intratumour uptake of photosensitisers, offering a promising strategy to improve PDT efficacy

L’operazione, di rimeditazione della geografia del conflitto sindacale in uno con la valorizzazione dell’autodeterminazione dei lavoratori, oltre la conflittualità organizzata, si rivela idonea a realizzare un effetto trasformativo profondo, verso un modello partecipativo di relazioni sindacali atto a coinvolgere quanto più possibile i lavoratori, in prospettiva preventiva, nelle diverse decisioni aziendali

    Perceived difficulty of a mathematical task: do in-service and preservice teachers have a common view?

    This qualitative study investigates how pre-service and in-service teachers perceive the difficulty of mathematical tasks and examines the factors that shape their evaluations. Using two tasks from the national large-scale assessments INVALSI, we aimed to identify whether perceived difficulties align with those of students and to explore any discrepancies between the two teacher groups. Our findings reveal significant differences between pre-service and in-service teachers, particularly in their handling of open-ended versus structured tasks. In-service teachers generally demonstrated alignment with students' evaluations for simpler tasks, while pre-service teachers exhibited emotions with respect to complex tasks. In our interpretation, these results highlight both the influence of teacher experience on perceived difficulty and the importance of addressing both cognitive and affective factors in teacher professional development

    Lo scritto propone una lettura integrata degli artt. 2407 c.c. e 25 octies CCII, muovendo dall’idea che la riforma della prima norma debba essere letta in raccordo con i doveri di segnalazione e vigilanza previsti dal Codice della crisi e in parallelo con gli obblighi di avviso e salvataggio sanciti dal Codice civile. L’assunto centrale è che la limitazione di responsabilità operi come incentivo “abilitante”: essa tutela i sindaci solo se attivi e diligenti, mentre l’omessa segnalazione può, de iure condito, configurare una presunzione juris tantum di dolo eventuale, con conseguente esclusione del cap. Con spirito di autocritica l’A. riconosce essere più ardita la proposta di configurare la segnalazione, ove ne ricorrano i presupposti, come porta d’ingresso al limite risarcitorio, bilanciando la funzione preventiva del controllo con l’esigenza di non scoraggiare l’assunzione della carica. Le conclusioni, volutamente interlocutorie, delineano un modello in cui diritto vigente e prospettive de iure condendo s’intrecciano, rafforzando la coerenza sistemica tra Codice civile, Codice della crisi e disciplina assicurativa

    Acute Myeloid Leukemia: A Key Role of DGKα and DGKζ in Cell Viability

    Acute myeloid leukemia (AML) is a heterogeneous disease with an unmet need for novel therapeutic drugs. Previous studies have reported the upregulation of diacylglycerol kinases (DGKs) in AML. This study investigated the effects of ritanserin, a DGKα-specific inhibitor, and DGKζ-IN4 or BAY 2965501, DGKζ-selective inhibitors, on a panel of AML cell lines. Ritanserin induced apoptotic cell death across all tested models, whereas DGKζ inhibitors triggered both apoptosis and necrosis to variable extents, with HL-60 cells being the most responsive to both compounds. Drug sensitivity did not correlate with DGKα or DGKζ expression levels, indicating that additional factors may influence cellular susceptibility. THP-1 proteomic profiling revealed that ritanserin broadly downregulated proteins involved in antigen presentation, cell cycle and metabolism, while BAY 2965501 affected a smaller and distinct but functionally similar protein subset, implying different mechanisms of action. Gene silencing confirmed AML cell line-specific dependence on DGK isoforms: HEL cells were sensitive to DGKα knockdown, HL-60 to DGKζ silencing, whereas K562 and THP-1 were resistant to both. These findings indicate that DGKs targeting can effectively reduce AML cell viability. However, AML heterogeneity and the limited selectivity of current inhibitors underscore the need for predictive biomarkers and combinatorial strategies to translate DGK inhibition into effective therapy

    Laparoscopic Versus Open Caudate Lobe Resection: A Systematic Review with a Meta-Analysis of Comparative Studies

    : Background: Liver resection of a caudate lobe is a challenging procedure in both open and minimally invasive approaches. The reason is mainly related to its anatomical position: segment 1 (S1) lies on the inferior vein cava, behind the main and the left portal veins, and below the hepatic veins. This meta-analysis aimed to assess the results of laparoscopic liver resection (LLR) versus open liver resection (OLR) for S1 resection. Methods: Available literature up to June 2024 was retrieved from the Medline and Embase databases. A systematic review with a meta-analysis was carried out to investigate the safety and efficacy of LLR for the S1 segment. Results: Six studies including 292 patients (LLR: n = 132; OLR: n = 160) were selected for the meta-analysis. The OLR cohort showed higher estimated blood loss (EBL) (MD: 140.1, 95% CI 49.3-130.8; p = 0.011) and longer length of hospital stay (MD: 3, 95% CI 1.8-4.2; p = 0.001). No differences in severe postoperative morbidity, overall morbidity, R1 resection rates, transfusion rates, operative time, and duration of Pringle maneuvers were shown. Conclusion: LLR for lesions located in S1 is safe and effective and may be associated with lower EBL and shorter length of stay than OLR. Further larger prospective studies are needed to confirm such results

    The interplay between childhood trauma, hopelessness, depressive symptoms, and mental pain in a large sample of patients with severe mental disorders: A network analysis

    Background: Mental pain represents a significant risk factor for suicidal behavior in severe mental disorders. The present study aims to investigate the interplay between childhood traumatic experiences, hopelessness, depressive symptoms and mental pain, using a network analysis approach in a large transdiagnostic sample of participants living in the community. Methods: The present investigation was conducted using data gathered in a multicentric observational cross-section study organized as a joint project, including different Italian research and clinical settings. Considering the assessment tools adopted in the study, 12 different variables were included as nodes in the EBICglasso network analysis. Stability of the edges and of centrality indices were assessed using bootstrap procedures, considering case-dropping and node-dropping procedures. Results: A total of 2147 participants were included in the analysis. Mental pain represents a central feature in a complex network of relationships, including traumatic experiences, hopelessness, and depressive symptoms. More in deatail, mental pain and, to a lesser extent, affective and cognitive depressive symptoms emerged as the most central and influential nodes of the network, highlighting the strong link existing between these aspects and their importance in the lives of people with mental disorders. Conclusions: Results confirm the importance of mental pain as a transdiagnostic feature, requiring careful assessment and consideration in all patients, beyond the diagnostic categories and regardless of suicide risk. Assessing and managing the presence and severity of mental pain should be taken into account in clinical practice, in the perspective of providing significant clinical benefits, as well as relevant research insight
