La sostenibilità nel servizio sociale. Il ruolo dei processi di co-creazione

    No full text

Development of PROTACs Targeting the Moonlighting Enzyme Nicotinamide Phosphoribosyltransferase (NAMPT) for Breast Cancer Therapy

: PROTACs (proteolysis-targeting chimeras) enable selective protein degradation through the ubiquitin-proteasome system and offer opportunities to target moonlighting proteins with nonenzymatic functions. We report the design, synthesis, and biological evaluation of NAMPT-directed PROTACs derived from our previously described inhibitor MV78 (7). A modular click chemistry strategy facilitated rapid assembly of a focused library by varying linker architectures and E3 ligase recruiters, with emphasis on the impact of a triazole unit. Structure-activity relationship studies revealed that eliminating the triazole from the linker and introducing an (S)-methyl group on the VHL ligand markedly enhanced degradation. The optimized degrader, U42, exhibited low nanomolar antiproliferative activity, robust intracellular and extracellular NAMPT degradation, excellent metabolic stability, favorable pharmacokinetics, and sustained efficacy in mammosphere models, three-dimensional breast cancer cultures not previously explored with NAMPT degraders. These findings highlight U42 as a lead compound and provide strong rationale for advancing NAMPT-directed PROTACs as a therapeutic strategy in breast cancer

    No full text

Case Report: Intensive multidisciplinary motor-cognitive rehabilitation treatment in Gerstmann–Sträussler–Scheinker syndrome

    No full text
Gerstmann–Sträussler–Scheinker syndrome (GSS) is a genetic, autosomal dominant prion brain disease that causes ataxia and slow cognitive decline. Herein, we describe a 34-year-old woman (S.C.B.) diagnosed with GSS (variant Pro102Leu). In 2018, she presented with gait instability without falls, early fatigue, and dizziness; after 3years, urinary incontinence, initial insomnia and rapid-eye movement sleep behavior disorder, writing difficulties, and occasional dysphagia had appeared. S.C.B. was admitted to the Neurorehabilitation Unit due to the progressive worsening of her balance and walking ability [Timed Up and Go (TUG): not evaluated; Berg Balance Scale (BERG): 5/56]. At admission, neurological, neuropsychological, physiotherapeutic, occupational, and speech assessments were performed. She was completely dependent in basic (ADL) and instrumental activities of daily living (iADL) (ADL: 5/6; iADL: 4/8; Barthel Index: 60); furthermore, moderate cognitive decline and depressive symptoms were observed. After 4weeks of intensive motor-cognitive rehabilitation treatment, an improvement was observed in all the intervention areas, leading to a global gain in autonomy (TUG: 1 min 50 s; BERG: 10/56; Barthel Index: 70). This case report highlights the importance of tailored motor-cognitive rehabilitation and how these interventions can enhance patients’ quality of life and their ability to cope with their symptoms. In conclusion, this case contributes to a broader understanding of treatment options that clinicians can propose to patients

L’ineguaglianza economica tra lavoratori autonomi e lavoratori salariati nell’Atene classica

    No full text

Under pressure: trade competition from low-wage countries and demand for immigrant labor in Italy

This study examines whether trade competition from low-wage countries (LWCs) influences immigration patterns in an advanced economy. We focus on Italy between 2003 and 2013, a period characterized by rising market pressure from China and Eastern Europe. Using census data on sectoral employment, administrative records on immigrants by nationality, and disaggregated bilateral trade data, we investigate whether heightened import competition acted as a pull factor for migrant workers by exploiting the differential exposure of local labor markets. To identify the exogenous component of these trade shocks, we adopt a shift-share instrumental variable strategy, while disaggregating immigrant data by nationality allows us to control in detail for the role of local networks and for bilateral push and pull factors. Our findings indicate that trade competition from LWCs significantly increased local immigrant shares. Exploring potential mechanisms, we find indirect evidence that firms under competitive pressure sought to cut labor costs by relying on a more flexible, lower-paid workforce, primarily composed of foreign workers

Gasotransmitters bridging tumor biology and immunity: from pathophysiological insights to therapeutic potential

The tumor microenvironment (TME) is a highly intricate and dynamic milieu, comprising neoplastic, immune and stromal cells in concert with extracellular matrix components, all engaged in continuous bidirectional crosstalk that critically orchestrates disease progression and therapeutic resistance. Beyond the local context, the TME is deeply shaped also by systemic influences, such as inflammatory mediators, metabolic cues and hematopoietic perturbations, collectively fostering a tumor-permissive macroenvironment. The interplay between local and systemic signals plays a pivotal role in modulating cellular differentiation, immune dynamics and stromal architecture, thereby sustaining malignancy. Among the myriad regulatory modulators involved in this complex network, endogenously produced gasotransmitters, namely carbon monoxide (CO), nitric oxide (NO) and hydrogen sulfide (H2S), have emerged as key modulators of tumor biology. These small, diffusible molecules exert a context-dependent spectrum of both pro-and anti-tumorigenic effects, influenced by their concentration, cellular source and tumor-specific microenvironmental conditions. Through the modulation of redox balance, metabolic signaling and epigenetic regulators, gasotransmitters impact immune cell functions, stromal remodeling and tumor cell behavior, thereby contributing to either immune evasion and therapy resistance or, conversely, to tumor suppression. Despite their growing relevance, the molecular mechanism governing these dualistic roles remain incompletely elucidated. This review provides a comprehensive overview of the current knowledge regarding the roles of CO, NO and H2S in shaping TME. We focus on their influence on immune, stromal and tumor cell differentiation, metabolism and function, and discuss how this understanding could inform novel therapeutic strategies aimed at reprogramming the TME to enhance clinical outcomes in cancer treatment
