Circulating tumor DNA for predicting recurrence in patients with operable breast cancer: a systematic review and meta-analysis

The incorporation of circulating tumor DNA (ctDNA) into the management of operable breast cancer (BC) has been hampered by the heterogeneous results from different studies. We aimed to assess the prognostic value of ctDNA in patients with operable (non metastatic) BC.This work was supported by Lombardy Region Italy [grant agreement number RR33] as part of the project ‘I controlli periodici (follow-up) dopo la diagnosi e le terapie in pazienti liberi da malattia e asintomatici: verso una person- alizzazione delle strategie di follow-up’.Medicin

Structural attributes and macrofaunal assemblages associated with rose gorgonian gardens (Leptogorgia sp. nov.) in Central Chile: opening the door for conservation actions

Gorgonians (like corals) are important habitat- forming organisms that support a diversity of macrofauna. This study explored structural attributes of gorgonian gardens formed by rose gorgonians (Leptogorgia sp. nov.) and associated macrofaunal assemblages in Caleta Pichicuy (Central Chile). Hierarchical sampling was conducted at 20 m depth (maximum colony abundances) in order to assess spatial variability in abundance and colony attributes at two spatial scales (among sites and rocky walls). The abundance and composition of the associated vagile and sessile macro- fauna were also examined using univariant (Taxa richness and Shannon index (H’e)) and multivariant approaches and were compared with adjacent bare rocky habitats. Our results showed a high abundance of gorgonians (ca. 28.9– 36.5 colonies m −2 ) compared to other gorgonian gardens in the world. For structural attributes, our results showed smaller colonies with thicker holdfasts in more exposed sites, suggesting the influence of hydrodynamic forces on the colony morphology. Taxa richness and H’e of vagile fauna showed threefold and twofold, respectively, higher values in gorgonian gardens compared to bare walls, but no differences were observed for sessile fauna. In addition, PCoA and PERMANOVA evidenced a distinctive assemblages’ composition between habitats for both vagile and sessile fauna. Correlation analyzes and dbRDA showed, however, little association between structural attributes and associated faunal assemblages (R2 = 0.06, and ca. 3–9.4% of the total variation explained, respectively). Our results constitute the first assessment of structural habitat complexity and accompanying fauna in these gorgonian gardens and establish the baseline for understanding possible future changes associated to human activities.Open Access funding provided thanks to the CRUE-CSIC agreement with Springer Nature. Although these stages have been largely studied in native ecosystems, very little is known about larval patterns in smaller-scale, invaded, estuarine environments such as those available in the Mediterranean Sea. The general aim of this study is to assess the distribution and abundance of early life stages of C. sapidus in the communication channels of the Albufera lagoon with the Mediterranean Sea, and in adjacent coastal waters. Different sampling methods were employed to target each life stage. (1) For zoeae, planktonic tows were conducted using a 300 μm manta net along 200 m transects parallel to the coastline at distances of 300, 1000 and 3000 m from the shore. (2) A small and manageable passive collector model was used to capture megalopae and juveniles in estuarine areas. This system was first tested from 2020 to 2021 in a single channel, with a weekly sampling frequency, and two more channels were added in 2022. Additionally, intensive daily sampling was conducted in one of the channels from September to November 2022 during the settlement peak. Results showed that zoeae of C. sapidus were more abundant in August 2021, at 1000 m from the coast, and appear to have a disjoint temporal distribution with zoeae of other crab species. Megalopae and juveniles were more abundant from September to November at all sampling sites and times. In 2022, collectors featured a dominance of megalopae at two of the sites (ca. 73%), while the site with the lowest salinity showed a dominance of juveniles (84%). For megalopae, comparative results from the intensive and the regular sampling campaigns showed that time of residence within collectors is just a single day. For juveniles, higher numbers were detected with increased deployment time, suggesting a need for enhanced habitat complexity.Ciencias del Ma

Stress-Related Chronic Fatigue Syndrome: A Case Report with a Positive Response to Alpha-Methyl-P-Tyrosine (AMPT) Treatment

M.P. designed, acquired, analyzed, and interpreted the data; M.P. and E.O. drafted the manuscript and critically revised the content; M.L. drafted figures and tables. All authors have read and agreed to the published version of the manuscript.Chronic fatigue syndrome (CFS) is a heterogeneous disorder with a genetically associated vulnerability of the catecholamine metabolism (e.g., catechol O-methyltransferase polymorphisms), in which environmental factors have an important impact. Alpha-methyl-p-tyrosine (AMPT; also referred to as metyrosine) is an approved medication for the treatment of pheochromocytoma. As a tyrosine hydroxylase inhibitor, AMPT may be a potential candidate for the treatment of diseases involving catecholamine alterations. However, only small-scale clinical trials have tested AMPT repurposing in a few other illnesses. The current case report compiles genetic and longitudinal biochemical data for over a year of follow-up of a male patient sequentially diagnosed with sustained overstress, neurasthenia, CFS (diagnosed in 2012 as per the Center for Disease Control (CDC/Fukuda)), and postural orthostatic tachycardia syndrome (POTS) over a 10-year period and reports the patient’s symptom improvement in response to low–medium doses of AMPT. This case was recognized as a stress-related CFS case. Data are reported from medical records provided by the patient to allow a detailed response to treatment targeting the hyperadrenergic state presented by the patient. We highlight the lack of a positive response to classical approaches to treating CFS, reflecting the limitations of CFS diagnosis and available treatments to alleviate patients’ symptoms. The current pathomechanism hypothesis emphasizes monoamine alterations (hyperadrenergic state) in the DA/adrenergic system and a dysfunctional autonomic nervous system resulting from sympathetic overactivity. The response of the patient to AMPT treatment highlights the relevance of pacing with regard to stressful situations and increased activity. Importantly, the results do not indicate causality between AMPT and its action on the monoamine system, and future studies should evaluate the implications of other targets. This research was funded by several economic organizations. The impacts of including the serogroup B meningococcus vaccine (MenB) into the national immunisation schedule on vaccination rates, co-administration rates, and timeliness were assessed using a population-based pre-funding (2022) and post-funding (2023) study design. MenB vaccination rates improved after funding and were in line with previously funded vaccines. Co-administration rates also increased significantly. Timely administration increased, protecting children at an early age. Public funding has a positive impact on vaccine accessibility and early protection. Consistent population characteristics highlight the role of funding.Medicin

The exemplarity of divine justice in behind in the portuguese version Vision of Tundal

In the imagination of medieval Christian society, earthly justice was not conceived as an action disconnected from divine justice, they coexisted in both the world of the living and the world of the dead. However, according to clerical discourses, God carried out true justice in the afterlife, which determines the sentences destined for souls in the Beyond structured in spaces associated with Christian moral conduct, Hell, Purgatory and Paradise. Visionary narratives became an essential instrument in the dissemination of this Christian rhetoric by providing a visual projection of a judging God who punishes, saves and rewards the elect and the damned, according to their merits, in the post-mortem. In this article, we analyse the descriptions of divine justice over souls in the places of the Afterlife in the Portuguese version of the Tundal Vision as exemplary elements that aim to instruct and advise medieval society on the behavioural actions of this world that lead to the path of salvation or damnation. An over-view of the applications of justice carried out on earth through the Portuguese king's actions of the Avis dynasty is also mentioned.Humanidade

Sensitivity of marine invertebrates to acidification of seawater by an atmosphere enriched with CO2

Ocean acidification, a consequence of increasing atmospheric CO2, results from oceans absorbing about one-third of CO2 emissions, leading to a decrease in pH. This phenomenon can cause sublethal or lethal damage to marine organisms some of which are crucial for coastal ecosystems and human economies. Understanding how to reproduce acidified seawater in a laboratory is essential for assessing potential impacts. Our protocol uses a CO2-enriched atmosphere to determine the pH tolerance of Mysidopsis juniae, exposed at pH 6.8 and 7.0, and larvae of Arbacia lixula, exposed at pH 7.2. M. juniae showed lethal responses at pH 6.8 ± 0.3, while A. lixula exhibited delayed larval development as a sublethal reaction at pH 7.2 ± 0.3. However, these pH levels are extreme and cannot be predicted for future oceanic conditions. Other studies have indicated negative sublethal effects on similar organisms at higher pH levels than those observed in this study. Continued experiments are necessary to prepare for and inform about the dangers of ocean acidification, and also to develop other types of studies.

Medicin
