    The Role of Heat Shock Proteins in Insect Stress Response, Immunity, and Climate Adaptation

    Heat shock proteins (HSPs) play a key role in enhancing insect resilience to abiotic and biotic stresses by preserving cellular integrity and modulating immune responses. This review summarizes the main functions of HSPs in insects, including protein stabilization, interaction with antioxidant systems, and involvement in the innate immune response. The expression of HSPs under environmental conditions reflects their evolutionary adaptation to various stressors, including thermal changes, chemical exposure, and pathogens. Future research should focus on the interaction between HSPs and other stress response systems to improve our understanding of insect adaptation. Furthermore, in the context of global climate change, HSPs emerge as a crucial resilience factor and potential biomarkers for environmental monitoring

    KRAS/NRAS variants and copy number alterations prognostically stratify patients with sinonasal melanoma. Pathologica

    Sino-nasal mucosal melanoma (SN-MM) is an aggressive and rare form of melanoma arising from mucosal melanocytes with pathogenesis unrelated to sun exposure. Conversely to cutaneous melanoma (CM), the molecular bases underling SN-MM development and progression are unclear, and no molecular predictive markers have been identified yet. To better define the molecular landscape of SN-MM, a retrospective series of 37 SN-MMs from 31 patients was analysed for both somatic mutations and cytogenetic alterations. The somatic mutation analysis identified the presence of a driver gene pathogenic variant in 54% of cases. In detail, mutually exclusive NRAS mutations were found in 42% of cases, KRAS mutations in 6%, and KIT mutations in 6% of cases. Remarkably, no BRAF mutations were detected. Patients with NRAS-mutated/KRAS-wild type (wt) melanomas showed better outcome than patients with NRAS-wt/KRAS-mutated melanomas, which were associated with multiple recurrences at local or regional sites. On the other hand, focusing on genomic alterations, copy number variants (loss of 1p36, loss of 3p/3q) were identified in 19% of SN-MMs, which showed poor overall survival and short disease-free survival with early metastatic dissemination. This work describes a new integrated characterization of both single nucleotide variants and, for the first time, genomic alteration in SN-MM, providing a new insight into molecular bases of these neoplasms and prompting further efforts for personalized clinical protocols according to tumour aggressiveness

    Dopo aver illustrato il fondamento giuridico della solidarietà fra Stati in materia sanitaria in tempo di crisi, il contributo si sofferma sulle iniziative adottate dall’Unione europea per promuovere l’accesso alle prestazioni sanitarie transfrontaliere da parte dei cittadini dell’Unione e sugli strumenti di cui l’Unione europea dispone per sostenere gli Stati nell’affrontare situazioni di emergenza sanitaria (FSUE, SEAR, rescEU). Una seconda parte del contributo è dedicata all’analisi dell’esperienza maturata dall’Unione europea in occasione della recente pandemia da SARS-Cov-2 e di come l’Unione europea abbia successivamente provveduto, con un approccio solidaristico, a dotarsi di strutture in grado di promuovere una migliore preparazione degli Stati membri nell’affrontare future emergenze sanitarie (HERA) ovvero a stimolare forme di collaborazione fra Stati al fine di prevenire o superare le disugualianze in ambito sanitari

    The rapid increase of human activities is threatening the ocean biodiversity, with marine vertebrates, particularly elasmobranchs, facing higher extinction risks. Among them, the whale shark (Rhincodon typus), an endangered migratory filter-feeding species, is threated by multiple anthropogenic pressures, including marine pollution. This study focuses on determining the presence and levels of legacy contaminants (PCBs, DDT) and trace elements (TEs) in baitfish, a primary prey species of whale sharks in Madagascar, and assessing the potential risks to whale sharks and humans through the consumption of contaminated baitfish (Sardinella gibbosa). Results indicate that while levels of DDT were below environmental safety thresholds, Hg levels in baitfish exceeded the acceptable limit, posing significant bioaccumulation risks to higher trophic level organisms, including whale sharks. Furthermore, comparisons with established maximum allowable limits for foodstuffs consumption revealed that Sardinella gibbosa may pose both non-carcinogenic and carcinogenic health risks to humans, particularly for pollutants like PCB, Cd, Ni, and Hg. This research highlights the importance of considering baitfish consumption as a significant pathway for pollutant uptake in whale sharks, suggesting that their exposure may be far higher than previously estimated through zooplankton alone. These findings underscore the critical need for continued monitoring of chemical pollution in coastal ecosystems, which are vital hotspot for global marine biodiversity, particularly in regions like Madagascar, where pollution from industrial, agricultural, and mining activities exacerbates the risk to both marine biodiversity and human health
