Software updates: the "unknown unknown" of the replication crisis

The replication crisis is largely concerned with known problems, such as the lack of replication standards, non-availability of data, or p-hacking. One hitherto unknown problem is the potential for software companies' changes to the algorithms used for calculations to cause discrepancies between two sets of reported results. Anastasia Ershova and Gerald Schneider encountered this very problem in the course of their own replication test, and argue that software developers should take more responsibility for their role in the strengthening of replication standards

Genome Mining for Unknown-Unknown Natural Products

    No full text
Genome mining of biosynthetic pathways with no identifiable core enzymes can lead to discovery of the so-called unknown (biosynthetic route)-unknown (molecular structure) natural products. In this work, we focused on a conserved fungal biosynthetic pathway (ank) that lacks a canonical core enzyme, and used heterologous expression to identify the associate natural product to be a highly modified cyclo-arginine-tyrosine dipeptide (cRY). Biochemical characterization of the ank pathway led to identification of a new arginine-containing cyclodipeptide synthase (RCDPS), which was previously annotated as a hypothetical protein (HP) and has no sequence homology to nonribosomal peptide synthetase (NPRS) or bacterial cyclodipeptide synthase (CDPS). RCDPS homologs are widely encoded in fungal genomes and we showed other members of this family can synthesize diverse cyclo-arginine-Xaa dipeptides. Characterization of a cyclo-Arg-Trp (cRW) RCDPS showed the enzyme is aminoacyl-tRNA dependent, and represents the first report of such CDPS-like enzyme from fungi. Further characterization of the biosynthetic pathway anchored by the cRW synthase led to discovery of new compounds of which the structures would not have been predicted without knowledge of RCDPS function

There is provided a buffered rotary valve in which a valve is additionally installed in a rotary valve for supplying a fluid of high pressure and a fluid of low pressure being input from a compressor, thereby reducing the amount of fluid being supplied and unnecessarily consumed, and enhancing the efficiency of a freezing system

Going Beyond Counting First Authors in Author Co-citation Analysis

The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

Defining the Attributes of Good In-Service Portland Cement Concrete Pavements, December 2004

Much of the current research in portland cement concrete (PCC) pavements deals with the analysis of early pavement life failures and seeks to find ways to prevent those from reoccurring. The Long Term Pavement Performance (LTPP) portion of the Strategic Highway Research Program (SHRP) has identified some of the key factors in designing and building new PCC pavements. This statement will build on the Iowa Highway Research Board (IHRB) project TR-463, Field Performance Study of Past Iowa Pavement Research: A Look Back. In Iowa and across the nation, there are multiple pavements that were built more than 20 years ago that have been and are continuing to provide very good service to the public. They are found on both state and local routes and in both low and high traffic volume areas. There is a need to learn what went into those pavements, from the subgrade through the surface, that makes them perform so well. The purpose of this research project was to conduct a scoping study that could be used to evaluate the need for additional research to study the attributes of well-performing concrete pavements. The concept of zero-maintenance jointed plain concrete pavements” was iterated in this study for long-lasting, well-performing portland cement concrete pavement sections. The scope of the study was limited to a brief literature survey, pavement performance data collection from many counties, cities, and primary and interstate roads in Iowa, field visits to many selected pavement sites, and analysis of the collected data. No laboratory orfield testing was conducted for this phase of the project. A problem statement with a research plan was created that could be used to guide the second phase of the project

    Variations on the Author

“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
