Evaluation of spa-typing of methicillin-resistant Staphylococcus aureus using high-resolution melting analysis
No full textOBJECTIVE: spa-typing of methicillin-resistant Staphylococcus aureus (MRSA) has been widely used in clinical diagnostics and epidemiological studies. We aimed to evaluate high-resolution melting (HRM) as a rapid and cost effective method, to replace DNA-sequencing, for spa-typing in a global collection of 50 MRSA isolates. METHODS: The polymorphic X region of spa gene was amplified by colony PCR using SensiMix™ HRM kit and melting temperature (Tm) and melting curves of the amplicons was analyzed in close tubes using a Rotor-Gene 6000 instrument. RESULTS: Fifteen out of nineteen spa-types each had distinct Tm, which was sufficient to unambiguously type each of these spa-types. The remaining 4 spa-types cannot be separated by Tm alone: t008 and t2770 shared Tm (80.3°C) and t021 and t311 shared Tm (80.0°C). But, they can be separated based on the shapes of their melting curves. There are discrepancies between ours and previous studies, suggesting that standardization reminds a challenge for cross references. CONCLUSION: HRM-spa-typing is reproducible, simple, rapid and cost-effective. t037 is prevalent in Brazil and Sudan while diverse spa-types are found in Scotland and Saudi Arabia. Standardization is required for cross-references between labs globally

growth is exciting and provides new opportunities, it has also
provoked a considerable shortage of experienced spa therapists
and spa managers, and many spas are finding themselves
with inexperienced or under-qualified teams of people. The
recruitment and retention of qualified employees have become
one of the industry's greatest challenges. The diverse nature of
the industry, along with a lack of recognised standards and a
growing need for employees, has also created both opportunities
and challenges for education and training providers.
This chapter aims to give an overview of employment in the spa
industry, the range of spa education providers and the factors
that drive spa education, accreditation issues and the regulatory
environment for spa education along with future trends

Full text linkThe objective of the article is to identify and explain the relationship between spa services distribution and spa specialization in Kujawsko-Pomorskie, Pomorskie and Podkarpackie Voivodeships spa communes. Correlation and regression analysis were used based on data from the Local Data Bank and unpublished data sets from the Central Statistical Office of Poland. In order to explain the relation between spa services distribution and spa specialization, time-series analysis was used. In five of nine researched communes (Horyniec-Zdrój, Solina, Ustka, Ciechocinek and Inowrocław) there is a functional relationship between spa services distribution and spa specialization
The 2016 status of marine protected areas in the Mediterranean
Full text linkMedPAN and SPA/RAC are working alongside their partners (IUCN, WWF,
local NGOs, research organization, etc.) to establish an ecological network
of MPA to protect at least 10% of the marine and coastal waters which is
representative of the Mediterranean’s diversity and made up of ecologically
interconnected and well managed MPAs, in accordance with the latest
guidelines from the Convention on Biological Diversity and the Barcelona
Convention.
Every 4 years, MedPAN and SPA/RAC carry out the status of Mediterranean
MPA to evaluate the progress that has been made, since the first inventory
done in 2008, on the Mediterranean system of MPAs in view of the above
mentioned objectives: does the network cover 10% of the Mediterranean,
is it representative of the Mediterranean diversity, are MPAs well-connected
and well managed?
The main findings of the 2012 status of Marine Protected Areas in the
Mediterranean Sea were that the target of 10% protection was far from
being achieved, that the network was not yet coherent and that MPA
management was still insufficient.
This 2016 report has used the 2015-2016 inventory made on MPAs
(MAPAMED) and a survey questionnaire sent to managers not only to
assess the progress made since 2012 but also to identify the remaining
steps needed to achieve by 2020, the objectives set for the network of MPAs
by the Convention on Biological Diversity and the Barcelona Convention.peer-reviewe
