Screening of potent phytochemical inhibitors against SARS-CoV-2 protease and its two Asian mutants
Background: COVID-19, declared a pandemic in March 2020 by the World Health Organization is caused by Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2). The virus has already killed more than 2.3 million people worldwide. Object: The principal intent of this work was to investigate lead compounds by screening natural product library (NPASS) for possible treatment of COVID-19.Methods: Pharmacophore features were used to screen a large database to get a small dataset for structure-based virtual screening of natural product compounds. In the structure-based screening, molecular docking was performed to find a potent inhibitor molecule against the main protease (Mpro) of SARS-CoV-2 (PDB ID: 6Y7M). The predicted lead compound was further subjected to Molecular Dynamics (MD) simulation to check the stability of the leads compound with the evolution of time.Results: In pharmacophore-based virtual screening, 2,361 compounds were retained out of 30,927. In the structure-based screening, the lead compounds were filtered based on their docking scores. Among the 2,360 compounds, 12 lead compounds were selected based on their docking score. Kazinol T with NPASS ID: NPC474104 showed the highest docking score of -14.355 and passed criteria of Lipinski's drug-like parameters. Monitoring ADMET properties, Kazinol T showed its safety for consumption. Docking of Kazinol T with two Asian mutants (R60C and I152V) showed variations in binding and energy parameters. Normal mode analysis for ligand-bound and unbound form of protease along with its mutants, revealed displacement and correlation parameters for C-alpha atoms. MD simulation results showed that all ligand-protein complexes remained intact and stable in a dynamic environment with negative Gibbs free energy.Conclusions: The natural product Kazinol T was a predicted lead compound against the main protease of SARSCoV-2 and will be the possible treatment for COVID-19
Combined Effect of Biochar and Plant Growth-Promoting Rhizbacteria on Physiological Responses of Canola (Brassica napus L.) Subjected to Drought Stress
Biochar (BC) and plant growth-promoting microbes (PGPR) could represent a suitable agronomical strategy to mitigate the impacts of drought in arid agro-environmental conditions. However, there is currently little understanding of the synergistic benefit of combining BC and PGPR to increase drought tolerance in oilseeds. In this study, the physiological response of two water-stressed canola (Brassica napus L.) plants subjected to the application of BC obtained from waste wood of Morus alba applied solely or in combination with PGPR strains (Pseudomonas sp.) was evaluated. The experiment consists of two genotypes and nine treatments [(C-Control, T1-15 days drought (15DD), T2-30 days drought (30DD), T3-15 days of drought + PG (15DD + PG), T4-30 days of drought + PG (30DD + PG), T5-15 days drought + biochar (15DD + BC), T6-30 days drought + biochar (30DD + BC), T7-15 days drought + biochar + PG (15DD + BC + PG), T8-30 days drought + biochar + PG (30DD + BC + PG)]. Drought stress decreased emergence energy (EE), leaf area index (LAI), leaf area ratio (LAR), root shoot ratio (RSR), moisture content of leaves (MCL), percent moisture content (%MC), moisture content of shoot (MCS) and moisture content of root (MCR), and relative water content (RWC) in both varieties of Brassica napus L., which in contrast, it is increased by the collective application of both biochar and PGPR. In both varieties, N, P, K, Mg, and Ca concentrations were highest in all the biochar and PGPRs separate and combined treatments, while lowest in 15 and 30 days drought treatments. Osmolyte contents like Glycine betaine (GB) and sugar remarkably increased in the stress condition and then reduced due to the synergistic application of biochar and PGPR. Drought stress has a repressive effect on the antioxidant enzymatic system like Peroxidase (POD), Superoxide dismutase (SOD), and glutathione reductase (GR) as well as total flavonoids, phenolics, and protein content. The antioxidant enzymes and phenolic compounds were dramatically increased by the combined action of biochar and PGPRs. A significant increase in EE, LAR, RSR, and RWC under 15 and 30 days drought conditions, evidently highlighting the synergistic effect of BC and PGPR. The results conclude a substantial and positive effect of the combined use of BC and PGPR strains on canola's response to induced drought stress, by regulating the physiological, biochemical, and agronomic traits of the plants.[GRAPHICS]

Şehrî ve Gülî, klasik İran edebiyatının önemli şairlerinin edebî meclislerde anlamı üzerinde tartışılmış ve nazım sebebi sohbetlere konu olmuş beyitlerini, manzumelerini ya da anlaşılmasında zorluk çekilen beyitlerdeki mazmunları bazen hikâyelerini naklederek izah eden bir eserdir. İçeriği ile Farsça edebî eserler üzerine Anadolu?da oluşmuş şerh geleneğinden farklı bir örnek olan bu eser, Nevâdirü?l- Emsâl adlı Farsça-Türkçe deyimler sözlüğünün müellifi olan Mîrek Muhammed tarafından yazılmıştır. Taşkentli olan Mîrek Muhammed, şair Muhlisî-i Bedahşânî?den eğitim almış, hiciv şairi olarak bilinen Müşfikî-i Buharâyî ile görüşmüş ve Şiraz, İstanbul ve Zigetvar?da bulunmuştur. Bu çalışmada müellif ve eseri hakkında bilgi verilecektir.Shahrî ve Gulî is a collection of couplets and poems selected from important poets of classic Persian literature, which were difficult to interpret and discussed in literary congresses, sometimes presented with the stories of the couplets and poems. Having different qualities from the commentary tradition formed in Anatolia on Farsi literary works, it was written by Mîrak Muhammad, who is known as the writer of Farsi-Turkish idioms-dictionary named Navâdiru‟l-Emsâl. Mîrak. Muhammad, who received education from Muhlisî Badahşânî, and conferred with Muşfikî Buharâyî known as the satirical poet, visited some cities like Shiraz, Istanbul and Szigetvar. Traversable Wormhole Solutions Admitting Noether Symmetry in f(R,T2) Theory
This paper uses the Noether symmetry approach to examine the viable and stable traversable wormhole solutions in the framework of the f(R,T2) theory, where R is the Ricci scalar and T2=TμνTμν is the self-contraction of the stress–energy tensor. For this purpose, we consider a specific model of this modified theory to obtain the exact solutions of the Noether equations. Further, we formulate the generators of the Noether symmetry and first integrals of motion. We analyze the presence of viable and stable traversable wormhole solutions corresponding to different redshift functions. In order to determine whether this theory provides physically viable and stable wormhole geometry or not, we check the graphical behavior of the null energy constraint, causality condition and adiabatic index for an effective stress–energy tensor. It is found that viable and stable traversable wormhole solutions exist in this modified theory
Bostanzâde Yahyâ Gül-i Sad-Berg (analysis-critical text)
Bu çalısma 17. yüzyıl Osmanlı âlimlerinden Bostanzâde Yahyâ Efendi'nin hayatı, eserleri, kisiliği ve Gül-i Sad-Berg adlı eserinin tenkitli metni ile incelemesinden olusmaktadır. Bu çalısma ile adı geçen yazarın hayatının, eserlerinin ve kisiliğinin ortaya çıkarılması ve eserin sağlam bir metni olusturularak divan nesri içindeki yerinin belirlenmesi amaçlanmıstır. Bunun için dönemin biyografi kaynakları incelendi, yazarla ilgili biyografik bilgiler tespit edildi. Eserleri incelenerek edebi kisiliğine iliskin ipuçları belirlendi. Gül-i Sad-Berg adlı mensur eserinin el yazması 7 nüshası tespit edilerek tenkitli metni ortaya kondu. Elde edilen bu metin üzerinden eser içerik ve dil-anlatım bakımlarından incelendi. Sonuç olarak yazarın, çağına ve çevresine duyarlı bir Osmanlı aydını olduğu; Gül-i Sad-Berg adlı eserinin de Hz. Muhammed'in mucizelerini içeren ve Osmanlı estetik nesrinin özelliklerini tasıyan olgun bir metin olduğu belirlendi.This study consists of one of the Ottoman scholar in the 17th century, Bostanzade Yahya Effendi's life, his works, his personality and critical analysis of his work of Gul-i Sad-Berg. It aims to reveal the author's life, works, and personality anddetermine the place of the work mentioned before in the collected poems by making up a solid text. For this reason, biographical sources of the period were examined and biographical information about the author was ascertained. Tips on hispersonality were determined examining the literary text. The critical analysis was stated identifying seven copies of the manuscript work of Gul-i Sad-Berg. Through the text obtained, the work text was analysed in terms of content and language and expression. As a result, it is determined that the author is an Ottoman intellectual, sensitive to his environment and his age; and his work, Gul-i Sad-Berg is an mature text which consists of the miracles of Muhammad and has the characteristics of the Ottoman aesthetic

Investigating Urinary Pathogens and Their Antibiotic Resistance: A Cross-Sectional Urine Culture Study.

Females are more affected than males. The primary culprit is Escherichia coli. Multiple research investigations have documented widespread antimicrobial resistance in uropathogens, sparking global concerns, especially regarding the rise of multidrug resistance (MDR).
Methodology: This cross-sectional study was conducted from December 2023 to March 2024. A non-probability purposive sampling technique was employed to select participants, and informed consent was obtained from them. Data were extracted from the culture and sensitivity reports of these patients. The collected data were meticulously entered into IBM SPSS Statistics for Windows, Version 21 (IBM Corp., Armonk, NY). The findings were then presented using a blend of percentages and numerical figures, offering a clear and concise representation of the data.
Results: Our study of 313 participants showed a higher prevalence of UTIs in females (219, 70%) compared to males (94, 30%). E. coli and Citrobacter were the predominant pathogens, with E. coli and Citrobacter more common in females, while Enterobacter and Staphylococcus were more prevalent in males. Antibiogram analysis revealed sensitivities to specific drugs like nitrofurantoin and meropenem, while resistance was observed against others, including polymyxin B and ampicillin. These findings stress the need for tailored UTI treatment approaches.
Conclusions: In conclusion, our research highlights a concerning trend of escalating antibiotic resistance among Pakistani patients with UTIs. Tobramycin B, ticarcillin-clavulanic acid, ampicillin, and clotrimazole exhibited the highest resistance rates, while imipenem, meropenem, nitrofurantoin, sulfonamides, and tigecycline demonstrated notable sensitivity. These findings emphasize the urgent need for the exploration of alternative treatment options to combat rising resistance levels effectively
