A noninvasive molecular approach: exploiting species-locus-specific PCR primers in defeating numts and DNA cross-contamination of cercopithecidae

    No full text
The lack of a standardized, noninvasive molecular approach to studying genetic aspects of primates has made it hard for primatologists to decode the evolutionary history of these species. Researchers must optimize their own techniques to fully exploit the available samples. Lack of species-locus-specific primers also contributes to difficulties in using noninvasive genetic samples. Thus, the objectives of this study were to develop a standardized technique to collecting samples noninvasively, propose newly designed species-locus-specific primers, and optimize conditions for polymerase chain reaction (PCR) for Macaca fascicularis, M. nemestrina, Trachypithecus cristatus, and T. obscurus. Nine new species-locus-specific primers for three different loci of mitochondrial DNA, namely D-loop, cytochrome oxidase subunit I (COI), and cytochrome b, were successfully designed. These primers proved to be efficient in amplifying larger datasets (up to ~1,000 bp) of the targeted species in the optimized PCR conditions. The species-locus-specific primers are able to anneal to host DNA alone in highly contaminated feces of highlighted species. They can also offer alternatives measures in avoiding contamination related to nuclear insertion of mitochondrial pseudogenes (numts)

Melting heat transmission of Maxwell nanofluid flow caused due to a stretchable cylindrical pipe through Finite Element Technique

    No full text
The objective of current investigation is to examine Maxwell fluid flow generated by cylindrical stretching pipe. Thermophoresis and Brownian moment phenomena will also incorporate on the said fluid. Melting heat shifting phenomenon incorporated at the surface of the elongating cylinder. Mathematical formulation is supported by boundary layer assumption. Suitable similarity transformations diminishes model of nonlinear partial differential equations  into a model of nonlinear ordinary differential equations  that ultimately solved via numerical technique named Finite Element Technique. The properties of numerous effective variables are graphically displayed and examined. Further the physical quantities like velocity gradient, heat and mass fluxes are tabulated numerically and analyzed. The dimensionless temperature distribution rises for rising values of curvature parameter and Brownian motion parameter however an opposite behavior is observed for melting parameter and Prandtl number. Moreover, a reduction occurs in heat and mass fluxes for sufficiently high values of curvature parameter. The author (Taseer Muhammad) thanks the KKU research unit for the financial and administrative support under grantnumber 521 for year 44King Khalid University, Grant/AwardNumber: 52

Steady-state power distribution in VSC-based MTDC systems and dc grids under mixed P/V and I/V droop control

    Get PDF
[EN] This paper proposes a steady-state power distribution derivation method for voltage source converter (VSC)based multi-terminal HVDC (MTDC) systems and dc grids under mixed power/voltage (P/V) and current/voltage (I/V) droop control. P/V and I/V droop control are two commonly used control schemes, which can be combined to achieve co-regulation of powers & currents in MTDC systems and dc grids. The proposed method can be used to estimate the power distributions under different scenarios for MTDC systems and dc grids based on VSCs with mixed P/V and I/V droop control. After determining the initial operating point based on an estimation-correction algorithm, redistributed power due to power disturbances, current changes or converter outages is analyzed in detail considering converter overload. An excess power reduction strategy is further proposed to avoid violation of power limits after converter outage. The accuracy of the proposed method is validated through multiple scenarios in a modular multilevel converter (MMC)-based four-terminal dc grid. The comparison between the proposed method and other approaches in the current literature further demonstrates the advantages of proposed power distribution derivation method. The third author (Muhammad Khalid) would like to acknowledge the support from Deanship of Research Oversight and Coordination (DROC) at King Fahd University of Petroleum and Minerals (KFUPM) through project No. DF201011.Sun, P.; Wang, Y.; Khalid, M.; Blasco-Gimenez, R.; Konstantinou, G.
