    Interaction between ephrin/Eph and BDNF in modulating hippocampal synaptic transmission and synapse formation

    The hippocampus is a brain structure known to be critical for learning and memory consolidation. Abnormal development or damage to this structure is known to play a role in developmental or degenerative neurological disorders such as autism and Alzheimer's disease. In this thesis, I argue that an interaction between ephrin/Eph and BDNF signaling pathways is critical for the development of the selective connection of CA3 neurons to CA1 neurons in hippocampus. This claim was evaluated on the basis of electrophysiology evidence about the ephrin/Eph -- BDNF interaction in synaptic activity, and the effect of the interrupting Eph and BDNF signaling on the hippocampal projection specificity onto CA1 neurons by functional synapse identification via the combination of electrophysiology and immunocytochemistry. First, I confirmed that the primary hippocampal neuronal culture can be a model for studying the specificity of synaptic connection within the hippocampal circuitry. Functional synapses were characterized by recording from pairs of cells which we subsequently identified with immunocytochemical labeling. Most connections were unidirectional, and I found that when one of the cells was a CA1 neuron (identified by labeling with the CA1 marker anti-SCIP) it was predominantly the postsynaptic member of the pair, a result consistent with in vivo connectivity. Second, ephrin-A/EphA signaling was shown to produce a transient increase in synaptic transmission and be able to inhibit the effect of subsequent BDNF application on synaptic activity. These electrophysiological experiments were suggestive of possible interaction between ephrin-A/EphA and BDNF in modulation synapse formation. Third, interruption of the endogenous EphA signaling by the kinase dominant negative EphA constructs significantly changed the natural synaptic connection selectivity in the hippocampal circuitry and dramatically increased the bi-directional connections in the culture as a consequence. The empirical results presented in this thesis provide a valuable mechanism for hippocampal trisynaptic circuitry development and function through balancing opposite influences of various modulating factors at specific developmental phases.Ph.D.Includes bibliographical references (p. 94-112)

The integration of the three typical narrative types enables the important discourses on the history of the CPC to contain profound philosophical speculation, heavy historical connotation, and aesthetic and literary conception.     The Impact of Positive Psychology-based Classroom on English Speaking Teaching and Learning of EFL College Students

    Positive psychology (PP) insists that a positive environment can promote intrinsic motivation and self-construction. To explore and verify whether a positive classroom environment in English speaking teaching can enhance EFL college students’ classroom engagement and boost their speaking proficiency and examine the practice of applying a PP-based positive classroom environment to spoken language teaching of EFL college students, this research takes two English classes in Z University as the research objects, employing PP as the theoretical framework to create a positive classroom environment, adopting a combination of quantitative and qualitative research methods, and conducts a comparative experiment for one semester. The research finds that speaking teaching of EFL college students based on a positive classroom environment can enhance EFL students’ classroom engagement and boost their speaking proficiency. To go a step further, this research, based on the teaching practice, summarizes the methods and strategies of creating a positive classroom environment and provides a teaching sample for reference, thus offering theoretical support and practical recommendations for speaking teaching and learning of EFL college students

    Recurrent Neural Network Language Model with Part-of-speech for Mandarin Speech Recognition

    Recurrent neural network language models (RNNLMs) have been successfully applied in a variety of language processing applications ranging from speech recognition to machine translation. They can fight the curse of dimensionality by learning a distributed representation (word vector). The components of these vectors measure the co-occurrence of the word with context features over a corpus. However, RNNLMs ignore the fact that the meaning of word can vary substantially in different contexts (e.g., for polysemous words). In this paper, we investigate part-of-speech information to address this issue to some extent on the basis of information about the meaning of a word they could provide.     The Effect of Part-of-speech on Mandarin Speech Recognition

    This paper concentrates on the effect of part-of-speech on Mandarin speech recognition by incorporating it into language model and pronunciation dictionary. This work is motivated by the two benefits of part-of-speech, one is to reduce the lexical ambiguity in language model to some extent and the other is to provide some information about the pronunciation of heteronyms. The experiments conducted on two corpora, tagged manually or automatically, show that a 3% relative character error rate (CER) reduction is achieved.
