Overexpression of cellular prion protein alters postischemic Erk1/2 phosphorylation but not Akt phosphorylation and protects against focal cerebral ischemia
Purpose: The physiological function of the cellular prion protein (PrP(C)) is still unclear. A growing body of evidence suggests that PrP(C) has neuroprotective properties and that its deletion increases susceptibility to focal cerebral ischemia. The purpose of this study was to elucidate the role of PrP(C) overexpression in ischemic brain injury in vivo. Methods: PrP(C) overexpressing (TG35) and wild type (WT) mice were subjected to a 90-minute transient focal cerebral ischemia followed by infarct volume analysis 24 hours after lesion. To identify effects of PrP(C) overexpression on signalling pathways important for the regulation of ischemic cell death, we studied postischemic activation and expression of Akt and Erk1/2 using quantitative Western Blot analysis. Results: TG35 mice displayed significantly smaller infarct volumes and showed reduced early postischemic Erk1/2 phosphorylation, a pathway known to exacerbate neuronal injury following transient cerebral ischemia. In contrast, PrP(C) overexpression did not change postischemic Akt phosphorylation, which acts anti-apoptotic and is reduced in PrP(C) knockout animals. Conclusions: These results demonstrate that PrP(C) overexpression reduces deleterious Erk1/2 activation but does not affect Akt activation after transient cerebral ischemia, suggesting a role for distinct cytosolic signalling pathways in PrP(C) mediated neuroprotection

Quantitatives Monitoring der Thrombozytenadhäsivität mit dem Platelet Adhesion Assay (PADA) bei Patienten mit akuter zerebraler Ischämie
Aktuell steht kein verlässlicher und breit anwendbarer Laborparameter zur Verfügung, der eine Vorhersage des klinischen Ansprechens auf eine Sekundärprophylaxe nach stattgehabter akuter zerebraler Ischämie mit einem Thrombozytenaggregationshemmer erlaubt oder mit dem sich die individuelle Gefahr von Blutungskompli kationen abschätzen lassen könnte. In der vorliegenden offenen, prospektiven kontrollierten Studie soll der Frage nachgegangen werden, ob der PADA-Test in der Diagnostik der akuten zerebralen Ischämie und in der Therapieplanung der Sekundärprophylaxe mit Thrombozytenaggregationshemmern bei Patienten mit akuter zerebraler Ischämie und TIA sinnvoll eingesetzt werden kann. Der PADA-Test untersucht den Funktionszustand d er Thrombozyten bezügli ch ihrer Fähigkeit zur Bindung an Fibrinogen über den GP IIb/IIIa -Rezeptor unter den Bedingungen von niedrigem Scheerstress. Insgesamt wurden 528 Patienten untersucht, von denen 401 der endgültigen Analyse zur Verfügung standen. Von diesen hatten 229 Patienten eine akute zerebrale Ischämie, 28 eine TIA und 7 Patienten eine ICB erlitten. Weitere 40 Patienten wurden wegen ander er neurologischer Erkrankungen behandel t und hatten länger als 6 Monate zurückliegend eine akute zerebr ale Ischämie erlitten. Weiterhin wurden 5 Patienten mi t einer gesicherten systemischen Vaskulitis und 3 Patienten mit einer SVT untersucht. Als Kontrollgruppe dienten 89 Patienten, die wegen ander er neurologischer Nichtgefäßerkrankungen stationär behandelt wurden. Bei 100 Probanden erfolgte zusätzlich eine Konstanzprüfung mittels Doppelbestimmung des PADA-AI. In der vorliegenden Studi e konnte erstmalig nachgewiesen werden, dass sowohl ältere Patienten, als auch Patienten mit niedrigerer Thrombozytenzahl und Patienten mit einem erhöhten Rezi divrisiko, wie es im „Essener Stroke Rezidiv Score“ erfasst wird, einen signifikant höheren PADA-AI aufweisen, und zwar unabhängi g von der zur stationären Aufnahme führ enden Erkrankung. Weiterhin wiesen Patienten mit einer extra- oder intrakraniellen Stenose der hirnversorgenden Gefäße, Patienten mit einer koronaren Herzerkrankung und Patienten mit arteriellem Hypertonus im Vergleich zu Patienten ohne diese Merkmale im Mittel signifikant höhere PADA-AI auf
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Zerebrale Physiologie und neurologische Funktion nach hypothermem Kreislaufstillstand und selektiver hypothermer Hirnperfusion: Untersuchungen im Großtierversuch
Der hypotherme Herz-Kreislaufstillstand und die selektive zerebrale Perfusion sind Verfahren der Neuroprotektion, die in der Herzchirurgie eingesetzt werden. Die vorliegende experimentelle Arbeit untersucht die Physiologie beider Techniken isoliert sowie in kombinierter Anwendung am Großtiermodell des Schweins in Form eines Langzeitversuchs. Zielgrößen sind die Parameter der globalen zerebralen Durchblutung (Blutfluss, Widerstand und Sauerstoffstoffwechsel des Gehirns) sowie der Blutfluss einzelner Hirnregionen, welche mittels Injektion fluoreszierender Mikrosphären bestimmt werden. Die neuroprotektive Effizienz beider Verfahren wird anhand eines postoperativen neurologischen Punktescores bewertet
Role of Müller cells in the onset and progression of neuronal degeneration after acute retinal ischemia
Müller cells (MCs), the most common retinal glial cell type, are responsible for the maintenance of retinal homeostasis under physiological conditions. After retinal damage, MCs become reactive and protect and support the neurons. However, this reactivity can lead to loss of basal functions and thus to impaired tissue homeostasis. The effect and timing of this "double faced" behavior of reactive glial cells has not been fully elucidated. The aim of this study was to investigate the time point for the transition of MC gliosis from neuroprotective to detrimental, and to investigate the effect of enhanced antioxidant activity of these cells on neuronal survival in vivo in a mouse model of acute ischemic/reperfusion injury. Fluorocitrate was injected into the vitreous of the eye to study the timing of gliosis. This led to a specific inhibition of the Krebs cycle in MCs and thus to a reduced cell metabolism. This transient inhibition allowed the identification of three phases of the gliotic cell response: an early neuroprotective phase, followed by the transition to a later deleterious phase beginning 12-18 hours after ischemia. One hallmark of ischemia is the increase in oxidative stress. Selective enhancement of MCs antioxidative response through overexpression of the mitochondrial enzyme frataxin resulted in increased neuronal survival. This correlated with increased expression of antioxidant enzymes and neurotrophic factors. Thus, a time-dependent influence or a functional support of MCs can increase the neuronal survival after retinal injuries. This is associated with decreased neuron-detrimental gliosis and enhancement of intrinsic protective mechanisms. This makes MCs a lucrative therapeutic target for treatment of ischemic retinal pathologies
Effect of sensory deprivation on the transcriptome of adult murine brain microglia
Microglia, the CNS’s resident macrophages, are involved in brain homeostasis, synapse formation, plasticity, and cognition. Their response to neuronal activity changes remains unclear, though sensory deprivation in young mice alters microglial morphology and increases neuronal phagocytosis. However, the molecular mechanisms driving these changes are unknown. This study investigated microglial responses to altered neuronal activity in the adult mouse visual cortex using a monocular deprivation (MD) model. We hypothesized that MD alters microglial function, reflected in specific signaling pathways. Using RNA-Seq, we analyzed the microglial transcriptome at 12h, 24h, 5d, and 10d post-MD in 3-month-old male and female Cx3cr1CreERT2/+;Rpl22HA/+ (RiboTag) mice. The RiboTag approach enabled the isolation of ribosome-bound mRNA, providing insights into microglial activity. We optimized the protocol to ensure high-quality RNA and used qPCR to assess plasticity-related genes. Microglia in the contralateral visual cortex (VL) showed stronger and earlier responses than in the ipsilateral cortex (VR), with the highest number of altered genes at 12h and 24h. Microglia upregulated G protein-coupled receptors and downregulated transcription factors regulating the cytoskeleton. We identified Sptbn4, Actn1, and Mical2 as novel players in microglial responses to sensory deprivation. Prolonged MD (5–10d) increased immediate early gene and plasticity-related gene expression, suggesting a homeostatic response. The VR showed minimal early changes, though some genes exhibited a conserved systemic response. Neuronal gene analysis revealed delayed Arc and Egr1 upregulation in the VR at 10d, suggesting late strengthening of the non-deprived cortex. In conclusion, microglia rapidly respond to sensory deprivation in the VL, with minimal VR changes. This study identifies novel pathways in microglial adaptation, offering targets for future research
Validierung der funktionellen Repräsentation des Nervus medianus im primären somatosensorischen Kortex SI am ARGOS 200 Vektor-MKG im Vergleich mit dem fMRT
Ziel der vorliegenden Arbeit war es, zum einen eine exaktere magnetenzephalographische Lokalisation der funktionellen Repräsentation des Nervus medianus im somatosensorischen Kortex SI Area 3b zu erreichen sowie zum anderen erstmals die radiale Dipolquelle der P22-Aktivität der somatosensorisch evozierten Potenziale (SEP) in Area 1 magnetenzephalographisch nachzuweisen. Hierfür wurden Messungen an dem 3 Raumrichtungen detektierenden Messsystem ARGOS 200 Vektor-MKG von der Firma ATB (Chieti, Italien) sowie an dem eine Raumrichtung detektierenden 2x31-kanaligen Doppeldewar-Kryostaten von der Firma Philips durchgeführt. In die Studie eingeschlossen wurden 11 Probanden. Zusätzlich wurde von jedem Probanden ein MRT des Kopfes einschließlich eines fMRTs angefertigt. Während aller Untersuchungen wurde der rechte Nervus medianus proximal des Handgelenks elektrisch gereizt. In die Auswertungen wurden nur Messungen mit ausreichend aufgeklärtem Feldbildmuster sowie ausreichender Aufklärung des gemessenen Feldes aufgenommen, wobei die Orts- und Orientierungsparameter der tangentialen und radialen Dipolquellen N20 und P22 betrachtet wurden. Hierbei zeigte sich für die Lokalisation der tangentialen Dipolquelle in Area 3b im Vergleich des Philips-Messsystems mit dem fMRT ein durchschnittlicher Dipolabstand von 12,98mm. Die entsprechende Untersuchung mit dem ARGOS 200 Vektor-MKG-Messsystem erbrachte einen durchschnittlichen Dipolabstand von 17,05mm. Zwischen den Dipollokalisationen beider Messsysteme an sich zeigte sich eine durchschnittliche Differenz von 15,02 mm. Somit konnte entgegen vorheriger Annahmen eine exaktere Lokalisation der tangentialen Dipolquelle der N20-Aktivität der SEPs durch den speziellen Messsensorenaufbau des ARGOS 200 Vektor-MKGs nicht bestätigt werden. Demgegenüber gelang es jedoch erstmals die radiale Dipolquelle der P22-Aktivität der SEPs magnetenzephalographisch nachzuweisen. Dabei konnten für alle Probanden die erwarteten Feldbilddarstellungen für die einzelnen Raumrichtungen aufgezeigt werden, sowie insgesamt für 5 Probandenmessungen im Rahmen der anatomischen Variation erwartete Werte für die Dipolabstände zwischen der tangentialen und radialen Dipolquelle sowie der dazwischen liegenden Winkel aufgezeigt werden
