    Latar belakang: Asam lemak trans umumnya terbentuk dari proses hidrogenasi dari makanan produk yang diolah menggunakan minyak yang telah dihidrogenasi. Penelitian ini mengukur kadar nitric oxide (NO) yang merupakan mediator penghambat remodeling dan peningkatan resorpsi tulang akibat pemberian tinggi asam lemak trans, yang meningkatkan kadar osteokalsin. Tujuan penelitian: Penelitian ini untuk membuktikan bahwa pemberian tinggi asam lemak trans meningkatkan kadar NO dan kadar osteokalsin darah tikus Sprague Dawley (SD). Metode: Penelitian ini merupakan penelitian eksperimental laboratorik, dengan Randomized Pre & Post –test control group design. Perlakuan yang digunakan adalah pemberian asam lemak trans 10% pada tikus dengan keluaran perbedaan kadar NO dan osteokalsin. Hasil: Asam lemak trans secara bermakna meningkatkan kadar NO serum dan kadar osteokalsin serum. Hubungan kadar NO dan osteokalsin digunakan uji Pearson correlation, diperoleh nilai p=0,002 dan koefisien korelasi yang diperoleh adalah 0,848. Simpulan: Pemberian pakan tinggi Asam Lemak Trans 10% dari energi total pada tikus SD, meningkatkan kadar NO dan osteokalsin secara bermakna. Ada hubungan positif kuat dan bermakna antara peningkatan NO karena pemberian asam lemak trans dosis tinggi dengan peningkatan osteokalsin.     Quantitative Analysis of Potassium Concentrations in Hair of Breast Cancer Patients Using Atomic Absorption Spectrometry (AAS)

    Breast cancer, a leading cause of cancer-related mortality, is associated with genetic mutations influenced by endogenous factors, such as DNA repair errors, and exogenous factors, including environmental pollutants and heavy metals. Potassium, an essential trace element, has emerged as a potential factor in cancer biology, although its role remains underexplored. This study aims to evaluate potassium concentrations in hair samples from breast cancer patients and healthy individuals using Atomic Absorption Spectrometry (AAS), a sensitive and cost-effective technique for trace element analysis. Hair, a non-invasive biological sample, was selected for its ability to reflect long-term exposure to environmental and metabolic changes. Results indicate significantly elevated potassium levels in breast cancer patients (mean: 226.65 ppm) compared to healthy controls (mean: 151.79 ppm), with levels ranging from 30.38 to 725.56 ppm in patients and 42.97 to 403.16 ppm in controls. These findings suggest a potential correlation between potassium accumulation and breast cancer, highlighting its possible role in cancer pathophysiology

    Relationship between the BRAF V600E and tumor size, lymph node, and distant metastasis in papillary thyroid carcinoma

    Aim – This study aimed to analyze the association between B-Raf proto-oncogene, serine/threonine kinase (BRAF) V600E and tumor size, lymph node, and distant metastasis (TNM) status on patients with papillary thyroid carcinoma. Material and Methods – This research is a cross-sectional study. A total of 40 samples were papillary thyroid carcinoma tissue paraffin blocks from thyroidectomy surgery at State General Hospital (RSUP) Dr. Kariadi Semarang. Assessment of BRAF V600E mutation protein expression through immunohistochemical methods using antibody BRAF V600E was conducted. Results – The mean age of patients with papillary thyroid carcinoma with a positive BRAF V600E mutation was 49 years. The highest incidence was 71.4% in women. A significant difference between BRAF V600E and tumor size (p=0.03) was observed with as many as 53.2% more cases at size > 4 cm. A significant difference in the expression of BRAF V600E with enlargement of the lymph node (p=0.011) by 60.7% was observed compared to those without enlarged lymph nodes. On the other hand, no significant relationship between BRAF V600E expression and distant metastasis (p=0.652) was observed. The prevalence ratio in the positive BRAF V600E case at tumor size is more than 4 cm, and enlargement of lymph is 9.533 and 7.727. Conclusion – There is a significant relationship between BRAF V600E expression and tumor size and enlarged lymph nodes, while there is no significant relationship between BRAF V600E expression and distant metastases
