    Specifity and genetic control of B cell reactivity in experimental arthritis

    Collagen-induced arthritis (CIA) and pristane-induced arthritis (PIA) are animal models for human rheumatoid arthritis (RA), and can be induced in susceptible mouse and rat strains. CIA is induced by immunization of type II collagen (CII), a cartilage specific protein together with Freund’s adjuvant. PIA is induced by intradermal injection of pristane (2,6,10,14-tetrametylpentadecane). In rats, induction of CIA is both T and B-cell dependent, whereas PIA induces a chronic T-cell dependent arthritis. In this study we focused on the role of B-cells in both the CIA and the PIA model. In rats, immunization of CII induces a strong B-cell response against CII, characterized by rapid IgG production. The B-cell recognition of rat CII was analyzed for epitope specificity and V gene selection. We were able to characterize several epitopes on the CII molecule. The recognition tended to be directed against the CB-11 fragment of CII. Interestingly, some of the selected epitopes were shared between species. The major epitope, as recognized by several different antibodies, was shared with the mouse, as previously defined. One epitope, defined from the rat response, was also predominantly recognized in RA patients. Analyze of the V genes showed a biased usage for specific epitopes, The VHQ52 and Vk12/13 families was predominantly used. Individual V genes displayed germline characteristics, The genetic control of B-cell responses was analyzed in a cross between E3 and DA. In the F2 progeny, we investigated the levels of rheumatoid factor, total immunoglobulin and isotype production. A total genome scan revealed linkages to previously defined disease loci. Rheumatoid factor production was controlled by a major locus (Rf1) on chromosome 1

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Popular Abstract in Swedish Immunsystemet är vårt skydd mot främmande organismer såsom bakterier och virus. När vi får en förkylning, eller en infektion, tar immunsystemets celler hand om och dödar de invaderade mikroorganismerna. I normala fall märker vi endast av detta i form av att vi känner oss sjuka med eventuell feber som följd. Det ställs stora krav på immunsystemet. Det ska inte bara kunna skydda oss från farliga mikroorganismer utan även kunna känna igen vad som inte utgör något hot. Ett stort antal olika organismer lever i symbios med vår kropp och ska därför inte angripas eller dödas. Vanligtvis ofarliga mikroorganismer kan dock utgöra ett hot mot vår existens om de befinner sig på fel plats eller får chansen att få övertaget genom obegränsad tillväxt. Immunförsvaret ska också kunna känna igen vad som tillhör vår egen kropp (kroppseget) och vad som är främmande (icke kroppseget). Denna egenskap är inlärd. Vi skyddas från farliga ”immunsystemceller” genom att de som kan känna igen kroppsegna strukturer tidigt selekteras bort. Autoimmuna sjukdomar antas uppstå när vårt immunförsvar tar fel på vad som är kroppseget och vad som är icke kroppseget. Detta leder till att immunsystemet attackerar och förstör de ämnen eller strukturer som det anser vara främmande. Två vanliga autoimmuna sjukdomar är reumatoid artrit, många gånger även kallad för ledgångsreumatism och multiple sclerosis. Reumatoid artrit kännetecknas av inflammation i lederna med stelhet och värk som följd. Det finns flera olika sjukdomsförlopp, alltifrån snabbt insjuknande med stadigt förvärrade symtom, till ett långsamt förlopp med återkommande perioder av besvär. De olika typerna av förlopp kan vara en följd av att reumatoid artrit egentligen utlöses av olika orsaker. Multiple sclerosis är en sjukdom där immunsystemet angriper strukturer i centrala nervsystemet med muskelförsvagningar, motoriska störningar och i värsta fall med förlamning som följd. Förloppet är många gånger periodiskt. Ett par procent av befolkningen i västvärlden lider av dessa sjukdomar. Hur vanligt förekommande de är i tredje världen är inte lika känt eftersom tillgång till tillförlitlig statistik saknas. Då det många gånger inte är möjligt att studera orsaker till autoimmuna sjukdomar hos människor kan djurmodeller som liknar sjukdomarna användas. Fördelarna är många. I en djurmodell kan sjukdomen sättas igång under kontrollerande former och hela sjukdomsprocessen detaljstuderas. Djurmodeller för reumatoid artrit är kollagen-inducerad artrit och pristan-inducerad artrit. Kollagen, eller mer specifikt kollagen typ II, är ett äggviteämne (protein) som är vanligt förekommande i broskvävnad och anses vara ett kandidatprotein som kan känns igen av immunsystemet. En ledinflammation kan framkallas genom att kollagen typ II tillsammans med en olja sprutas in underhuden. Pristan är en olja som ensamt kan framkalla en ledinflammation. Djurmodellen för MS kallas för experimentell autoimmun encefalit. Gemensamt för dessa experimentella djurmodeller är att det finns djurstammar, vanligtvis möss eller råttor, som är känsliga för sjukdomen och djurstammar som inte är känsliga. I en djurstam är alla djuren genetiskt lika varandra genom inavel. Genom att avla ihop känsliga djur med okänsliga djur kommer avkomman att slumpmässigt få egenskaperna från föräldrarna. Denna princip kan utnyttjas för att undersöka olika hur olika regioner av kromosomer bidrar till sjukdomen. Denna avhandling beskriver hur B-celler i råtta känner igen kollagen typ II och på vilket sätt produktionen av antikroppar genetiskt är styrd. B-celler producerar antikroppar som kan binda till och känna igen olika strukturer, vanligtvis främmande antigen såsom bakterier. Dessa antikroppar kan fungera som signalflaggor som talar om för immunsystemet att det finns främmande ämnen som måste tas omhand. Vi har detaljstuderat hur antikroppar binder in till kollagen typ II. Det visar sig att B-celler i både möss, råttor och i människor via sina antikroppar kan känna igen samma strukturer på kollagen typ II. Att djur och människor under likartade sjukdomsbetingelser producerar antikroppar mot samma strukturer stärker bilden av viktiga immundominanta bindningsställen och påvisar likheter mellan djurmodellen och den mänskliga sjukdomen. Vi har även studerat den genetiska styrningen, eller kopplingen till kromosomregioner, för produktionen av en speciell typ av antikroppar, s.k. reumatoid faktorer. Reumatoid faktorer är antikroppar som binder till andra antikroppar och är vanligt förekommande hos patienter med reumatoid artrit. Tidig förekomst anses kunna förutsäga hur sjukdomen kommer att utvecklas. Detta studerades genom att mäta halten av antikroppar i 153 råttor vars föräldrar, eller förfäder, antingen var resistenta eller var känsliga för artrit. Genom att undersöka kromosomerna i ett antal mätpunkter, och i dessa ta reda på från vilken förälder arvsmassan har kommit, kan man få fram en bild av den genetiska kopplingen. Vi har kunnat visa att produktionen av reumatoid faktorer styrs från en region på kromosom 11 och att den genetiskt styrs från den råtta som inte utvecklar sjukdomen. Vi har även visat att det finns regioner på andra kromosomer som kan styra olika typer antikroppsproduktioner. Denna forskning är ett led i sökandet av hur B-celler och antikroppar är involverade i autoimmuna sjukdomar.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
