Identification of cryptic species of rodents (Mastomys, Aethomys, Saccostomus) in the Kruger National Park
Mastomys natalensis, Aethomys chrysophilus and Saccostomus campestris are three common and widespread rodent species in the Kruger National Park. Chromosomal and protein electrophoretic investigations reveal that these species are in fact complexes of morphologically similar, though genetically distinct, species. Their respective distributions in the Kruger National Park are reported and species diagnostic characters (diploid chromosome number, genitalia, spermatozoa and electromorphs) are presented for the practical identification of the cryptic species. The value of a genetical approach to resolving cryptic species is emphasized with respect to ecological studies

Advanced-stage CRC incidence patterns following the phased implementation of the CRC screening programme in the Netherlands
Background and aims: From 2014, the Dutch colorectal cancer (CRC) faecal immunochemical testing-based screening programme was gradually rolled out by birth cohort. We evaluated changes in advanced-stage CRC incidence by timing of invitation to further strengthen the evidence for the effectiveness of CRC screening. Methods: Data on advanced-stage CRC incidence in the period 2010-2019 by invitation cohort were collected through the Netherlands Cancer Registry. Crude rates of advanced -stage CRC incidence and cumulative advanced-stage CRC incidence were calculated. Observed advanced-stage CRC incidence and cumulative advanced-stage CRC incidence were compared with expected advanced-stage CRC incidence and cumulative advanced-stage CRC incidence by invitation cohort using trend lines extrapolating data prior to the introduction of screening. Results: For the invitation cohort that was first invited for screening in 2014, advanced-stage CRC incidence increased before the introduction of screening from 94.1 to 124.7 per 100,000 individuals in the period 2010-2013. In 2014, the observed increase was higher than in preceding years, to 184.9 per 100,000 individuals. Hereafter, a decrease in incidence was observed to levels below expected incidence based on trends before the introduction of screening. A similar pattern was observed for invitation cohorts in subsequent years, coinciding with the first invitation to the screening pro-gramme. In 2019, the observed incidence for all invitation cohorts remained below expected inci-dence. The cumulative advanced-stage CRC incidence in the 2014-2016 invitation cohorts was significantly lower than the expected cumulative CRC incidence in the period 2010-2019. Conclusions: In the period 2014-2019, an increase in advanced-stage CRC incidence was observed for all invitation cohorts first invited for screening, followed by a decrease below expected incidence, following the pattern of the phased implementation. The cumulative advanced-stage CRC inci-dence in invitation cohorts invited for screening multiple times was lower than expected based on trends from the pre-screening era. These findings support a causal relationship between the intro-duction of the Dutch screening programme and a decrease in advanced-stage CRC incidence.(c) 2022 The Author(s). An evaluation of group counselling as a therapeutic intervention for patients suffering from a chronic illness with special reference to rheumatoid arthritis.
Freedom to obey : the obedience of Christ as the reflection of the obedience of the Son in Karl Barth's 'Church dogmatics'
This thesis argues that Barth’s asymmetrical structuring of the Trinity in I/1, his doctrine of election in volume II, his concept of the humanity of Christ as the imago Dei in III/2 and his account of the obedience of the Son being reflected in his incarnate life, as detailed in IV/1 and IV/2, are not just coherent but mutually reinforcing. The thesis demonstrates that Barth uses a nexus of crucial terms, including ‘correspondence’ [Entsprechung], ‘reflection’ [reflex/Abbildung] and ‘overflowing’ [Ueberstroemen], to express that God’s actions and relationships ad extra reveal who God is. The concept of ‘correspondence’, tentatively present in the first two volumes, gathers pace through III/2 and achieves full force in volume IV, where the obedience of Christ in IV/2 ‘reflects’ or ‘mirrors’ the obedience of the Son in IV/1. Crucially, the fact that the economic Trinity ‘reflects’ the immanent Trinity, or (differently stated) that the immanent Trinity ‘overflows’ into the economy, establishes a direction, an asymmetry, to the relationship of ‘correspondence’.
In ch. II of the thesis we argue that the asymmetry developed in the doctrine of the Trinity in I/1 is the basis for this asymmetric correspondence. Barth describes the triune life as one of giving and receiving existence, suggesting a divine order with an irreversible direction, an asymmetric order. This is shown to be particularly evident in Barth’s defence of the filioque clause which enables him to claim that the Spirit is the one in whom the ruling Father and obedient Son are united ad intra.
On this basis we argue, in ch. III, that, when Barth revises his doctrine of election, he comes to see it as the event of triune reflection: the Father, Son and Spirit electing to reflect who they are with a direction of determination, an asymmetry, which is irreversible. In this respect we argue against Bruce McCormack, who sees election as the event in which God elects triunity.
In ch. IV we read Barth’s III/2 account of the humanity of Christ as the imago Die, as an attempt to demonstrate that God’s economy of salvation corresponds to who he is. This theme comes into full focus in the first two part-volumes of volume IV, explored here in ch. V. The obedience of Christ reflects, corresponds to, the obedience of the Son. There is obedience in God. This concept, which so mystifies Paul Molnar and Rowan Williams, is shown to be theologically consistent with a doctrine articulated by Barth some thirty years previously: his asymmetrically structured doctrine of the Trinity
Designing an Optimal Kilonova Search Using DECam for Gravitational-wave Events
Indexación: Scopus.We address the problem of optimally identifying all kilonovae detected via gravitational-wave emission in the upcoming LIGO/Virgo/KAGRA observing run, O4, which is expected to be sensitive to a factor of ∼7 more binary neutron star (BNS) alerts than previously. Electromagnetic follow-up of all but the brightest of these new events will require >1 m telescopes, for which limited time is available. We present an optimized observing strategy for the DECam during O4. We base our study on simulations of gravitational-wave events expected for O4 and wide-prior kilonova simulations. We derive the detectabilities of events for realistic observing conditions. We optimize our strategy for confirming a kilonova while minimizing telescope time. For a wide range of kilonova parameters, corresponding to a fainter kilonova compared to GW170817/AT 2017gfo, we find that, with this optimal strategy, the discovery probability for electromagnetic counterparts with the DECam is ∼80% at the nominal BNS gravitational-wave detection limit for O4 (190 Mpc), which corresponds to an ∼30% improvement compared to the strategy adopted during the previous observing run. For more distant events (∼330 Mpc), we reach an ∼60% probability of detection, a factor of ∼2 increase. For a brighter kilonova model dominated by the blue component that reproduces the observations of GW170817/AT 2017gfo, we find that we can reach ∼90% probability of detection out to 330 Mpc, representing an increase of ∼20%, while also reducing the total telescope time required to follow up events by ∼20%. © 2024. Cytotoxic Action In Myoblasts And Myotubes (c2c12) And Enzymatic Characterization Of A New Phospholipase A2 Isoform (bj-v) From Bothrops Jararacussu Venom
A new PLA2 Bj-V from Bothrops jararacussu (14039.49 Da determined by MALDI-TOF mass spectrometry) was isolated in only one chromatographic step by HPLC ion-exchange and its purity was confirmed by reverse phase. Amino acid analysis showed a high content of hydrophobic and basic amino acids as well as 14 half-cysteine residues. The N-terminal sequence (DLWQFGQMIL KETGKIPFPY YGAYGCYCGW GGRGGKPKDG TDRCCYVHD . . .) showed a high degree of homology with basic D49 PLA2 myotoxins from other Bothrops venoms. Bj V showed discrete sigmoidal enzymatic behavior, with maximal activity at pH 8.4 and 35-40°C. Full PLA2 activity required Ca 2+ (10 mM) and there was little catalytic activity in the presence of 1 mM Ca2+.The addition of Mn2+ or Mg2+ (10 mM) in the presence of low (1 mM) Ca2+ slightly increased the enzyme activity, whereas Zn2+ and Cu2+ (10 mM) diminished the activity. The substitution of Ca2+ for Mg2+ or Cu 2+ also reduced the enzymatic activity. Bj V had PLA2 activity and produced cytotoxicity in murine C2C12 skeletal muscle myoblasts and myotubes. Aspects of registered psychaitric nurses' talk about their clinical judgement and decision-making.
Researchers have been investigating the clinical judgement and decision-making of nurses for several decades now. However, prior to the research described in this thesis, Registered Psychiatric Nurses (RPNs) working in the Irish Republic had not been the subject of a comprehensive study looking specifically at their clinical judgement and decision-making. As this is the first study conducted in this area, it takes an exploratory descriptive approach. With a comprehensive review of the literature and pilot study (n=7) as its basis, a novel mixed methods study was designed. Simulated cases presented in an audio-visual format were used to collect in-vivo and retrospective data in the form of narratives from participants (n=40) across the Irish Republic. The sample comprises RPNs across all levels of experience working in several sites, representing the full range of Irish mental health services. The data were analysed using comparative keyword analysis and conversation analysis informed discursive analysis. Quantitative and qualitative analysis reveals participants’ judgement and decision making to be routinised and habitual, hinging on reference to typicality grounded mainly in psychiatric diagnoses. The role of participants can be seen to represent the paternalism of the social order of which it is part. Participants express confidence and certainty in their judgement and decision-making, even where paraverbal and other discursive evidence points towards situations characterised by uncertainty. The study’s findings are of particular interest given the direction envisaged for the profession of psychiatric nursing by leading academics, health service providers and professional and statutory bodies. The findings of this study suggest that if psychiatric nursing in the Irish Republic is to proceed towards more person-centred, autonomous practice with a stronger therapeutic focus, dramatic restructuring of psychiatric nurses’ roles will be required. In conclusion, the thesis discusses this situation with reference to the challenges made evident by the study, along with the viable options available to address them
The geochemistry and geomicrobiology of relict hydrothermal sulphide deposits
The diagenetic re-mineralisation of seafloor-sulphide deposits and the role of microbes in the metal-exchange processes were investigated in metalliferous sediments from the Alvin relict hydrothermal zone in the TAG area at 26º8'N (Mid-Atlantic Ridge). The solid-phase and concomitant pore water concentrations of AI, Si,Ca, Ctot, Corg, S, Fe, Mn, Cu, Zn, P, V, Co, U, Mo, Au, Ag and REEs were measured in a 230 cm long gravity core from the southern periphery of the relict vent field. These measurements were complemented by detailed analysis of bacterial abundance and specific activity. The altered sulphidic sediments are capped with a ~30cm thick layer of carbonate-rich (~60% CaCO3), Fe-stained sediments. Two distinct sulphide layers, interbedded with Fe-oxysilicates, and overlain by a thin layerof Fe/Mn oxyhydroxides, were found in this core. The dominant mineral-phase in both sulphide layers, which originate from mass-wasting of mound sediments, is pyrite with some goethite. Reaction of the exposed metal-sulphides in the upper sulphide layer with seawater has produced a thin layer of secondary atacamite, which is enriched in Au. Primary sphalerite is dissolved in the upper sulphide layer and re-precipitation as secondary sphalerite directly above and below. U continues to be scavenged from the porewater, producing marked enrichments on oxidised sulphide rims. The re-mineralisation processes identified in this core are in close analogy to the large-scale zone-refining that has been described for the active TAG mound and ancient ore-deposits. REE/Fe ratios clearly distinguish between plume derived sediments in the carbonate cap and slumped material from the hydrothermal mound. The REE signature of bulk sediments and clay phases imply multiple stages of alteration by diffuse fluids in the upper sulphide layer and intermediate layer, whereas the lower sulphide layer is not affected. Alteration by reactive low-temperature hydrothermal fluids is also inferred to be responsible for the observed diagenetic overprinting of trace-metal distributions in the upper sulphide layer. The intermediate layer is rich in nontronite, which has been precipitated in situ from diffuse fluids. The presence of Mn- and Fe-reducing bacteria coincide with elevated pore water concentrations of Mn and Fe, indicating direct involvement of bacteria in the cycling of these metals. Total counts of viable cells and general activity measurements show that although bacterial populations are relatively small, they are healthy and well adapted to this potentially toxic environment. The existence of active microbial communities in metalliferous sediments may therefore provide a continuum of bacterial populations between high and low temperature hydrothermal systems, thus representing an important transitional stage in the hydrothermal ecosystem. Microbial reduction and oxidation of S was observed throughout the core, indicating that microorganisms are particularly active in terms of S-cycling. For deep-sea sediments extremely high sulphate reduction rates (67 nmol/cm3/d) were measured in the iron-stained carbonate cap. In the absence of significant organic carbon (~0.2 %) this strongly suggests the synthesis of alternative electron-donors by chemolithotrophic bacteria to support the observed high rates of heterothrophic activity in these sediments
Second asymptomatic carotid surgery trial (ACST-2): a randomised comparison of carotid artery stenting versus carotid endarterectomy
Background: Among asymptomatic patients with severe carotid artery stenosis but no recent stroke or transient cerebral ischaemia, either carotid artery stenting (CAS) or carotid endarterectomy (CEA) can restore patency and reduce long-term stroke risks. However, from recent national registry data, each option causes about 1% procedural risk of disabling stroke or death. Comparison of their long-term protective effects requires large-scale randomised evidence. Methods: ACST-2 is an international multicentre randomised trial of CAS versus CEA among asymptomatic patients with severe stenosis thought to require intervention, interpreted with all other relevant trials. Patients were eligible if they had severe unilateral or bilateral carotid artery stenosis and both doctor and patient agreed that a carotid procedure should be undertaken, but they were substantially uncertain which one to choose. Patients were randomly allocated to CAS or CEA and followed up at 1 month and then annually, for a mean 5 years. Procedural events were those within 30 days of the intervention. Intention-to-treat analyses are provided. Analyses including procedural hazards use tabular methods. Analyses and meta-analyses of non-procedural strokes use Kaplan-Meier and log-rank methods. The trial is registered with the ISRCTN registry, ISRCTN21144362. Findings: Between Jan 15, 2008, and Dec 31, 2020, 3625 patients in 130 centres were randomly allocated, 1811 to CAS and 1814 to CEA, with good compliance, good medical therapy and a mean 5 years of follow-up. Overall, 1% had disabling stroke or death procedurally (15 allocated to CAS and 18 to CEA) and 2% had non-disabling procedural stroke (48 allocated to CAS and 29 to CEA). Kaplan-Meier estimates of 5-year non-procedural stroke were 2·5% in each group for fatal or disabling stroke, and 5·3% with CAS versus 4·5% with CEA for any stroke (rate ratio [RR] 1·16, 95% CI 0·86–1·57; p=0·33). Combining RRs for any non-procedural stroke in all CAS versus CEA trials, the RR was similar in symptomatic and asymptomatic patients (overall RR 1·11, 95% CI 0·91–1·32; p=0·21). Interpretation: Serious complications are similarly uncommon after competent CAS and CEA, and the long-term effects of these two carotid artery procedures on fatal or disabling stroke are comparable.
