    Detecting DNA mismatches with metallo-insertors: A molecular simulation study

    Molecules that selectively recognize DNA mismatches (MMs) play a key role as nucleic acids probes and as chemotherapeutic agents. Metallo-insertors bind to the minor groove (mG) of double strand (ds) DNA, expelling the mismatched base pairs and acting as their π-stacking replacement. In contrast, metallo-intercalators bind to the major groove (MG) of ds DNA and π-stack to adjacent base pairs. In this study we focused on structural and energetic properties of Δ−[Rh(bpy)2(chrysi)]3+ (1), Δ−[Ru(bpy)2(ddpz)]2+ (2), and Δ-[Ru(bpy)2(eilatin)]2+ (3) as prototypical examples of metallo-insertors and intercalators. For all molecules we characterized both insertion and intercalation into a DNA dodecamer via force field based molecular dynamics (MD) and hybrid quantum-classical (QM/MM) MD simulations. A structural analysis of the 1–3/DNA noncovalent adducts reveals that the insertion provokes an untwist of the DNA, an opening of the mG and of the phosphate backbone in proximity of the mismatch, while the intercalation induces smaller changes of these structural parameters. This behavior appears to be correlated with the size of the inserting/intercalating ligand in proximity of the metal coordination site. Moreover, our simulations show that the different selectivity of 1 toward distinct MM types may be correlated with the thermodynamic stability of the MMs in the free DNA and with that of the corresponding insertion adduct. Understanding the factors which tune a specific insertion is of crucial importance for designing specific luminescent probes that selectively recognize MMs, as well as for developing more effective anticancer drugs active in MM repair of deficient cells lines

    Partnership in Teacher Education: A Theoretical and Practical Analysis

    The aim of this study is to discuss theoretical and practical issues of the partnership in teacher education. A brief background of the debate in the last years is presented, questions about the partnership concept are introduced, the experience of the Brazilian university performing the inter-institutional work is analyzed and an overview is given of how the issue has been addressed in Québec. The current study seeks to note controversies and possible achievements of teachers in their struggles for the greater social approbation of the teaching profession. The research process benefited from documentary analysis, on-site observations and interviews; the last ones carried out in Brazil. The concept of partnership shows ambiguity. On the one hand, it is a strong ally for the implementation of official educational reforms, which puts it in scene between universities and schools. On the other hand, as a collaborative practice between the researchers and the school teachers, it helps to create new theoretical and practical basis for the initial and extended formation of the teaching professionals, in which academic knowledge and experience are articulated. In the case here analyzed, in the Brazilian scenario, the partnership helps to introduce innovations through institutional arrangements with other rationality, by which teachers are valued as collaborators in the processes of teacher training. Thus, we can affirm that the partnership is a relatively recent practice in the field of education, which involves tensions between official interests; and objectives of both collaborative work and collective struggles for the appreciation of the teaching profession of researchers at universities and teachers at schools

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     Il saggio presenta i risultati di un approfondimento empirico volta sull’esperienza degli assistenti sociali chiamati ad attuare le misure che hanno istituito il Reddito di Cittadinanza. Abbiamo ipotizzato che gli operatori possano fare fronte ad alcune criticità segnalate in letteratura facendo ricorso alla capacità di mobilitare e combinare risorse di diversa natura: persone, relazioni, competenze, saperi, dispositivi normativi etc. Il lavoro empirico si è basato su focus group condotti in Sardegna e Veneto. Le risultanze corroborano l’idea che la capacità combinatoria sia un elemento chiave dell’agire professionale e indicano alcuni fattori di contesto che possono promuovere od ostacolare la capacità combinatoria degli operatori.The paper presents the results of an empirical investigation of the actual experience of social workers who are called to implement the measures that have introduced the Citizenship Basic Income (“Reddito di Cittadinanza”). We hypothesized that social workers cope with some criticalities reported in literature by mobilizing and combining resources of different nature: people, relationships, skills, knowledge, regulatory devices etc. The empirical work was based on focus groups conducted in Sardinia and Veneto. Evidence corroborates the idea that combinatory capacity is a key element of professional action. It also indicates which contextual factors can promote or hinder practitioners’ combinatory capacity
