Imaging the daily life of the G protein-coupled thromboxane A2 receptor : from dimerization to internalization

    No full text
The thromboxane A2 receptor (TP) is a seven transmembrane protein coded by a single gene that undergoes alternative splicing giving rise to the TPα and TPβ isoforms with identical N-terminal 328 aminoacids and different C-terminal tails of 15 and 79 aminoacids, respectively. Using confocal microscopy or wide-field microscopy combined with FRET analysis we have imaged TPα and TPβ, epitope-tagged or fused to fluorescent proteins (GFP, CFP, YFP), ectopically expressed in transfected cells. We show that at steady state TPα and TPβ form homo- and hetero-dimeric complexes at the cell surface. The architecture of homo- and hetero-dimers was predicted by an in silico approach to be essentially characterized by contacts between hydrophobic amino acids I25, W29, C35, V36, L39, L43, L44 and S47 in helix 1 from both monomers. Substitutions of these residues into alanines generated TPα and TPβ mutants unable to dimerize, thus conferming the theoretical predictions. When individually expressed in cells TPβ, not TPα, underwent endocytosis upon agonist stimulation via the clathrin-dependent route. However, TPα was capable of agonist-induced endocytosis when co-expressed with TPβ thus suggesting co-internalization of TPα-TPβ hetero-complexes. However, the intracellular route of TPα-TPβ dimer was different from that of TPβ, since it was not affected by co-expression of dominant negative mutants of dynamin and EPS-15, and showed less intense co-localization with transferrin receptor, Rab4, Rab5 vesicle markers

Austen(s) on the Screen: An intertextual reading of Becoming Jane, Miss Austen Regrets, and her Biographies

The cult of Jane Austen has extended well beyond literary manifestations and criticism, into the realm of the cinema. Besides all the filmic adaptations of Austen's novels and the whole series of Austen-themed adaptations, the famous English writer has become the protagonist of two recent biographical films: Becoming Jane (2007) and Miss Austen Regrets (2010). These two cinematic attempts at portraying Austen's life for the screen investigate two different periods of her life and appear to be very different in their approach. Beside making use of published material about the author such as biographical accounts, letters and memoirs, both try to reinvent the writer's life, as well as re-imagine and give voice to some of the most crucial and formative periods of her life. Austenian critics and enthusiasts recognize the fact that Austen is a notoriously problematic figure, whose biography brims with gaps and hazy details. In order to contribute to the ongoing debate on the biographical information on Jane Austen's life, this essay aims to outline the ways in which the cinematic medium has helped re-imagine the life of the author, and to assess whether such representations have opened up new perspectives or have merely recovered and recycled older, and already familiar, materials

“Alfonso de Ulloa protoispanista: la Introdutione che mostra il signor Alfonso di Uglioa a proferire la lingua castigliana”

numero monografico: “Le ragioni delle seconde lingue: un approccio grammaticografico” a cura di Félix San VicenteThe main purpose of the present paper is to discuss subsequent editions of Ulloa’s treatise on the pronunciation of the Spanish language. Inclusion of information about the author and his work in Venice will enable us to address the publication of the book in the wider context of the Venetian printing activities of the time. The treatise is intended for Italian readers and represents one of the first works in contrastive linguistics for the Spanish-Italian language pair. First published in 1553, the treatise went through four subsequent editions, each coming with modifications. While this paper concentrates on the very first edition – the only one written in both Spanish and Italian – later additions are also discussed. As will be seen, subsequent editions give evidence of Ulloa’s interest in providing better versions of the original publication through more precise wording and inclusion of useful information. Importantly, his ultimate goal was not only to assist the user, but also to prove his own proficiency

    P. R. Moore-Dewey's "Pregiudizio e Orgoglio": An Italian Re-Make of Austen's "Pride and Prejudicce"

Virtually since its publication in 1814, Jane Austen's most famous novel, Pride and Prejudice, has enjoyed great success and has been the object of countless re-workings, re-mediations and adaptations on global scale. This essay aims at discussing a special instance of this phenomenon: an Italian re-make entitled Pregiudizio e Orgoglio (“Prejudice and Pride”), and written by Austen enthusiast and admirer, P.R. Moore-Dewey (pseudonym of Patrizia Murreddu). Appeared in Italy in 2012, to coincide with the bicentenary celebrations of its model and source, this title constitutes a genuine literary effort that deserves the attention of Austen enthusiasts, fans, and also scholars for its many merits, in particular that of being an intricate patchwork of quotations and, thus, a thoroughly post-modern work. In this essay, I intend to examine Pregiudizio e Orgoglio's emblematic significance by outlining the numerous aspects that make it such a noteworthy adaptation of Austen. In particular, it is my intention to analyze the change of perspective adopted by the author, who decided to re-write Austen's tale through the point of view of Mr. Darcy and his sister Georgiana. Moreover, it is my intention to look at some of the many intertextual references present in Moore-Dewey's work (taken both from the whole Austen output, and from many other “classic” or “canonical” English writers) and consider the impact of such references and quotations on the reading experience. Finally, I will consider the extensive use of paratextual devices, which contribute to define this reworking a particularly relevant literary manifestation

A sweeping vision of the scientific landscape Lynn Margults Fritjol Capra is at forefront of the revolution in modern science which has challenges a conventional mechanistic view of the world based on the thinking of Descartes and Newton and brought us towards a holistic, ecological view. Here, Capra offers a brilliant, radical synthesis of such recent scientific breakthrough as the theory of recent scientific breakthroughs as the theory of complexity, Gara theory, cnaos theory and other explanation of the properties of organism, social systems and ecosystem. His findings-based on ten years of research and discussion with scientists from around the world-provide an extraordinary new foundation for ecological policies that will allow us to build and sustain communities without diminishing opportunities for future generations and come closer than ever before to answering the question, what is life? This stunning synthesis by an inspired scientist, draws scientist, experts decision maker and concerned citizens into broader multi-disciplinary networks and dialogues about human development and the future of all the on earth Hazel Henderson, author of the Politics of the Social Age and Paradigms in Progress

Capra aegagrus cretensis on Theodorou Island Wildlife Preserve, Greece, have seasonal and daily activity patterns that minimize their exposure to high ambient temperatures. There were significant differences in time spent lying, standing, feeding, and moving among subsets of the population classified by sex, season, and time of day. Agrimi usually were solitary, but they often formed small groups that varied in both age and sex composition. There was an age-and sex-specific dominance hierarchy with older, consequently larger, males being dominant. -from Author

The ecology of law ::toward a legal system in tune with nature and community /

    No full text
"At the root of many of the environmental, economic, and social crises we face today is a legal system based on an outdated worldview. In this groundbreaking book, bestselling author, physicist, and systems theorist Fritjof Capra and distinguished legal scholar Ugo Mattei show how, by incorporating concepts from modern science, the law can become an integral part of bringing about a better world. This is the first book to trace the fascinating parallel history of law and science from antiquity to modern times, showing how the two disciplines have always influenced each other - until recently. In the past few years, the scientific paradigm has shifted dramatically, from seeing the natural world as a kind of cosmic machine to understanding it as a network of fluidly interacting communities. But law is stuck in a mechanistic, seventeenth-century view that the world is made up of discrete individual parts. This has led to legal theory focusing on these parts and ignoring the bigger picture - for example, elevating the rights of individual property owners over the good of the community. But Capra and Mattei outline the basic concepts and structures of a legal order consistent with the ecological principles that sustain life on this planet. This is a profound and visionary reconceptualization of the very foundations of the Western legal system, with profound implications for the future of our planet."--Publisher's website
