Solution Nuclear Magnetic Resonance Structure and Molecular Dynamics Simulations of a Murine 18.5 kDa Myelin Basic Protein Segment (S72-S107) in Association with Dodecylphosphocholine Micelles.
peer reviewedThe 18.5 kDa myelin basic protein (MBP), the most abundant splice isoform in adult mammalian myelin, is a multifunctional, intrinsically disordered protein involved in the development and compaction of the myelin sheath in the central nervous system. A highly conserved central segment comprises a membrane-anchoring amphipathic alpha-helix followed by a proline-rich segment that represents a ligand for SH3 domain-containing proteins. Here, we have determined using solution nuclear magnetic resonance spectroscopy the structure of a 36-residue peptide fragment of MBP (murine 18.5 kDa residues S72-S107, denoted the alpha2-peptide) comprising these two structural motifs, in association with dodecylphosphocholine (DPC) micelles. The structure was calculated using CS-ROSETTA (version 1.01) because the nuclear Overhauser effect restraints were insufficient for this protein. The experimental studies were complemented by molecular dynamics simulations of a corresponding 24-residue peptide fragment (murine 18.5 kDa residues E80-G103, denoted the MD-peptide), also in association with a DPC micelle in silico. The experimental and theoretical results agreed well with one another, despite the independence of the starting structures and analyses, both showing membrane association via the amphipathic alpha-helix, and a sharp bend in the vicinity of the Pro93 residue (murine 18.5 kDa sequence numbering). Overall, the conformations elucidated here show how the SH3 ligand is presented to the cytoplasm for interaction with SH3 domain-containing proteins such as Fyn and contribute to our understanding of myelin architecture at the molecular level

Spatial lymphocyte dynamics in lymph nodes predicts the cytotoxic T Cell frequency needed for HIV infection control
The surveillance of host body tissues by immune cells is central for mediating their defense function. In vivo imaging technologies have been used to quantitatively characterize target cell scanning and migration of lymphocytes within lymph nodes (LNs). The translation of these quantitative insights into a predictive understanding of immune system functioning in response to various perturbations critically depends on computational tools linking the individual immune cell properties with the emergent behavior of the immune system. By choosing the Newtonian second law for the governing equations, we developed a broadly applicable mathematical model linking individual and coordinated T-cell behaviors. The spatial cell dynamics is described by a superposition of autonomous locomotion, intercellular interaction, and viscous damping processes. The model is calibrated using in vivo data on T-cell motility metrics in LNs such as the translational speeds, turning angle speeds, and meandering indices. The model is applied to predict the impact of T-cell motility on protection against HIV infection, i.e., to estimate the threshold frequency of HIV-specific cytotoxic T cells (CTLs) that is required to detect productively infected cells before the release of viral particles starts. With this, it provides guidance for HIV vaccine studies allowing for the migration of cells in fibrotic LNs. © 2019 Grebennikov, Bouchnita, Volpert, Bessonov, Meyerhans and Bocharov

Investigation of the behavior of plutonium(V) in alkaline media
The stability of the plutonium(V) oxidation state in alkaline media was studied with respect to the neighboring Pu(IV) and Pu(VI) oxidation states. Tests were conducted in 1 M or higher NaOH solutions in the presence and absence of other components of Hanford Site high-level tank waste. Spectrophotometric techniques were found to be effective in studying the behavior of plutonium(V) in alkaline solution at plutonium concentrations above 10{sup -3} M. To this end, plutonium(V) and plutonium(VI) in NaOH were prepared and their spectra characterized. In alkaline solutions with NaOH concentration below 8 M, plutonium(V) was found to be unstable to disproportionation occurring according to the reaction 2 Pu(V)(aq) {yields} Pu(VI)(aq) + Pu(IV)(s). The disproportionation of Pu(V) is complicated by at least two simultaneous processes: (1) the sorption of a significant fraction of the Pu(V) onto the forming Pu(IV) hydrous oxide precipitate, and (2) partial reduction of Pu(VI) by water {alpha}-radiolysis products
Об обеспечении сопоставимости статистических показателей при изменении государственных границ
The paper discusses the author's proposals for addressing the problem of achieving the comparability of Russian socio-economic indicators related to the change in the borders of the state as a result of the special military operation. The experience of ensuring indicators comparability when changing state borders, namely during the unification of Germany in 1990 and reunification with Crimea in 2014, is considered. It is shown that, due to the long duration of the process of changing borders and the uncertainty of its results, the specificity of the discussed Russian episode of changing state borders makes it unique, i. e., having no direct analogues in recent decades for developed countries. The article discusses requirements for data comparability, which are imposed by the tasks for which they are used. The need to find such a solution to the problem, which would allow analyzing the dynamics of indicators for both a comparable and entire territories, is explained. The author proposes a simple and technologically advanced approach to solving the problem of comparability. Features of its solution for indicators of different types, different levels of aggregation, and with different time steps are considered. Recommendations for organizing the work related to ensuring the comparability of statistical indicators when changing state borders are proposed.Изложены авторские предложения по решению проблемы обеспечения сопоставимости российских социально-экономических показателей в связи с изменением границ государства в результате проведения cпециальной военной операции. Рассмотрен опыт обеспечения сопоставимости показателей при изменении границ, в частности при объединении Германии в 1990 г. и при воссоединении с Крымом в 2014 г. Показано, что связанная с большой длительностью процесса изменения границ и неопределенностью его результатов специфика обсуждаемого российского эпизода изменения границ, делает его уникальным, т. е. не имеющим прямых аналогов в последние десятилетия для развитых стран. Обсуждаются требования к сопоставимости данных, предъявляемые содержательными задачами, для решения которых они используются. Обосновывается целесообразность такого решения проблемы, которое позволило бы анализировать динамику показателей как по сопоставимой, так и по полной территориям. Предложен простой и технологичный подход к решению проблемы сопоставимости. Рассмотрены особенности ее решения для показателей разных типов, уровней агрегирования и с разными шагами по времени. Сформулированы рекомендации по организации работы, связанной с обеспечением сопоставимости статистических показателей при изменении государственных границ
Transformational Recession and Structural Changes in Russian Industrial Production
This work studies the transformational recession in Russian industrial production and its accompanying structural changes. It focuses on problems associated with measuring output behavior in the Russian transitional economy; constructing the indicators needed to analyze structural changes in industrial production; analyzing the course of transformational recession and discussing its causes and characteristics; studying the intensity, progress, and trends of structural change; analyzing the mutual impact of these changes and output behavior; discussing some general laws and their possible causes; and describing a series of new transformational effects. Finally, proposals for economic policy are formulated.
