Pressure-induced vascular oxidative stress is mediated through activation of integrin-linked kinase 1/betaPIX/Rac-1 pathway.

High blood pressure induces a mechanical stress on vascular walls and evokes oxidative stress and vascular dysfunction. The aim of this study was to characterize the intracellular signaling causing vascular oxidative stress in response to pressure. In carotid arteries subjected to high pressure levels, we observed not only an impaired vasorelaxation, increased superoxide production, and NADPH oxidase activity, but also a concomitant activation of Rac-1, a small G protein. Selective inhibition of Rac-1, with an adenovirus carrying a dominant-negative Rac-1 mutant, significantly reduced NADPH oxidase activity and oxidative stress and, more importantly, rescued vascular function in carotid arteries at high pressure. The analysis of molecular events associated with mechanotransduction demonstrated at high pressure levels an overexpression of integrin-linked kinase 1 and its recruitment to plasma membrane interacting with paxillin. The inhibition of integrin-linked kinase 1 by small interfering RNA impaired Rac-1 activation and rescued oxidative stress-induced vascular dysfunction in response to high pressure. Finally, we showed that βPIX, a guanine-nucleotide exchange factor, is the intermediate molecule recruited by integrin-linked kinase 1, converging the intracellular signaling toward Rac-1-mediated oxidative vascular dysfunction during pressure overload. Our data demonstrate that biomechanical stress evoked by high blood pressure triggers an integrin-linked kinase 1/βPIX/Rac-1 signaling, thus generating oxidative vascular dysfunction. © 2009 American Heart Association, Inc

Microstructural damage of the cortico-striatal and thalamo-cortical fibers in Fabry disease: a diffusion MRI tractometry study

    No full text
Purpose: Recent evidences have suggested the possible presence of an involvement of the extrapyramidal system in Fabry disease (FD), a rare X-linked lysosomal storage disorder. We aimed to investigate the microstructural integrity of the main tracts of the cortico-striatal-thalamo-cortical loop in FD patients. Methods: Forty-seven FD patients (mean age = 42.3 ± 16.3 years, M/F = 28/21) and 49 healthy controls (mean age = 42.3 ± 13.1 years, M/F = 19/28) were enrolled in this study. Fractional anisotropy (FA), axial (AD), radial (RD), and mean diffusivity (MD) maps were computed for each subject, and connectomes were built using a standard atlas. Diffusion metrics and connectomes were then combined to carry on a diffusion MRI tractometry analysis. The main afferent and efferent pathways of the cortico-striatal-thalamo-cortical loop (namely, bundles connecting the precentral gyrus (PreCG) with the striatum and the thalamus) were evaluated. Results: We found the presence of a microstructural involvement of cortico-striatal-thalamo-cortical loop in FD patients, predominantly affecting the left side. In particular, we found significant lower mean FA values of the left cortico-striatal fibers (p = 0.001), coupled to higher MD (p = 0.001) and RD (p < 0.001) values, as well as higher MD (p = 0.01) and RD (p = 0.01) values at the level of the thalamo-cortical fibers. Conclusion: We confirmed the presence of an alteration of the extrapyramidal system in FD patients, in line with recent evidences suggesting the presence of brain changes as a possible reflection of the subtle motor symptoms present in this condition. Our results suggest that, along with functional changes, microstructural damage of this pathway is also present in FD patients. © 2020, The Author(s)

The global research on meiofauna associated to seagrasses: a bibliometric network analysis

To better understand the state of the art of the research on meiofauna (small metazoan between 500 and 30μm) and seagrasses, a bibliometric network analysis was performed exploring the period from 1981 to 2021 from the Scopus database. The indexed global scientific literature on this topic was explored using VOSviewer software, which allowed us to obtain a comprehensive overview on these topics. A total of 101 publications were identified, revealing that scientific research on meiofauna related to seagrasses is still relatively limited. Results showed that the scientific documents were published by 15 countries, leaded by Australia, Italy, and USA, 50 researchers, and only 7 journals (with a maximum of 8 documents each). De Troch was the most productive author with 14 published documents. Following the most common keywords (meiofauna, seagrass, nematode, and copepoda), “community structure” occurred 25 times, suggesting that the main research field linked to meiofauna and seagrasses is community ecology. The analysis of co-authorship (among researchers and countries), the co-occurrence of keywords and cited journals showed an increasing attention to this topic worldwide and over time. Indeed, although meiofauna is still relatively poorly studied, the awareness of its crucial role in key coastal habitats, such as seagrass beds, is growing

From stage to screen: adapting a children's theatre production into a digital toy

This photo essay explores the adaptation of the Scottish theatre for early years (TEY) production White into a transmedia digital toy. In 2013, Catherine Wheels Theatre Company commissioned an app developer, Hippotrix, to create an app for mobile tablet computers and smartphones inspired by the world of White. The paper outlines the process of creating White The App, including wire-framing, asset capture, sound recording, coding and prototyping. It also explores the impact of design decisions on dramaturgy and performativity, noting that digital media offer new possibilities for embracing non-linear storytelling while retaining key aspects of the live aesthetic.Publisher PD

The paper consists of the project of a scenographic city, from a written script, the book "One hundred years of solitude" by Colombian author Gabriel Garcia Marquez. The city was designed with the purpose of serving as the setting for an animated film. In this context the reason for the city to exist is to portray a story, suffering influence of the urban transformation driven by the interactions present in the narrative. The book tells the story of the Buendia family, founder of the city of Macondo, and tells story of the life of these characters through seven generations, and tells the evolution of the city during that period of time. The history of the family and the city intertwines, where Macondo is at the same time the scenary of the history and the consequence of it. This paper deals with literature and cinema, applying concepts of architectural design and urbanism to build a unique and functional scenographic city for animation cinema.

2010 - 2011Over the last fifteen years, the Italian University was asked to make an internal change in the organization and teaching in line with the change of the Italian Welfare (Salvatore & Scotto di Carlo, 2005) and following the invitation of the European Union (EC; 2000, 2001, 2006, 2009), which calls on educational institutions to respond adequately to the "Modernising the European social model" (EC, 2000). These changes, which have found their implementation in a number of legislative reforms, the latter of which is the law Gelmini (204) of 30 December 2010. While respecting their official duties, the University has faced two major issues: the first is the inability to assimilate outright job knowledge to employment, the second one concerns the need to establish an efficient and effective dialogue between stakeholders same University (students, faculty, staff, families, employers) increasingly differentiated and heterogeneous. To meet this mandate, it has turned into a service that, as such, provides services and manufactures products defined in this case, the training of students. The service logic, is immediately associated with that of Quality, as a guideline for interpreting from one side to the needs, desires and demands of customers, and the other to provide services that meet those needs (Gentile & Cocozza, 2000). The University Quality of service is, therefore, in this view of the improvement guarantee of training and employability of people and therefore support the development of the productive system (EC, 2006, 2009). The declination of quality within the University has increasingly required a capacity to negotiate between the demand satisfaction of European standards and national and local demand, defined by users including students. These are the customers of the service and the product of the same, and am no longer regarded as passive consumers but as co-builders thought the service (Vairetti, 1995, Salvatore, 2001). .. ..
