Diplomová práce je členěna do čtyř kapitol. První kapitola představuje teoretickou část, v druhé kapitole je zhodnocena pozice města Hodonín ve vztahu k trvalé udržitelnosti. Třetí kapitola zpracovává základní dokumenty Hodonína vycházející z koncepce trvale udržitelného rozvoje. Čtvrtou kapitolu tvoří tři projekty realizované městem Hodonín, které představují praktickou aplikaci principů TUR

    Tur\'{a}n problems for star-path forests in hypergraphs

    An rr-uniform hypergraph (rr-graph) is linear if any two edges intersect at most one vertex. Let F\mathcal{F} be a given family of rr-graphs. A hypergraph HH is called F\mathcal{F}-free if HH does not contain any hypergraphs in F\mathcal{F}. The Tur\'{a}n number exr(n,F){\rm{ex}}_r(n,\mathcal{F}) of F\mathcal{F} is defined as the maximum number of edges of all F\mathcal{F}-free rr-graphs on nn vertices, and the linear Tur\'{a}n number exrlin(n,F){\rm{ex}}^{\rm{lin}}_r(n,\mathcal{F}) of F\mathcal{F} is defined as the Tur\'{a}n number of F\mathcal{F} in linear host hypergraphs. An rr-uniform linear path PrP^r_\ell of length \ell is an rr-graph with edges e1,,ee_1,\cdots,e_\ell such that V(ei)V(ej)=1|V(e_i)\cap V(e_j)|=1 if ij=1|i-j|=1, and V(ei)V(ej)=V(e_i)\cap V(e_j)=\emptyset for iji\neq j otherwise. Gy\'{a}rf\'{a}s et al. [Linear Tur\'{a}n numbers of acyclic triple systems, European J. Combin., 2022, 103435] obtained an upper bound for the linear Tur\'{a}n number of P3P_\ell^3. In this paper, an upper bound for the linear Tur\'{a}n number of PrP_\ell^r is obtained, which generalizes the result of P3P_\ell^3 to any PrP_\ell^r. Furthermore, some results for the linear Tur\'{a}n number and Tur\'{a}n number of some linear star-path forests are obtained

    Cytokines Modulate Integrin Alpha(V)Beta(3)-Mediated Human Endothelial Cell

    Angiogenesis is a complex process regulated by the interactions of endothelial cells with cytokines and the adhesive protein matrix. The cytokines basic fibroblast growth factor (bFGF) and tumor necrosis factor- alpha (TNF- alpha) are two of the modulators of angiogenesis. One mechanism by which these cytokines induce their effects may be through theregulation of integrin adhesion receptor activity, in particular,alpha( v)beta(3). In this study, we examined the ability of these angiogenicfactors to modulate the adhesion of human umbilical vein endothelial cells( HUVECs) to immobilized disintegrins (i.e., rhodostomin and arietin), whichare specific in antagonizing integrin alpha(v )beta(3) in cells. As thesedisintegrins were immobilized as substrates, they acted as agonists to induceHUVEC adhesion in a dose- and alpha(v)beta(3)- dependent manner. In addition ,adhesion also triggered a sustained increase of intracellular free calcium.Furthermore, bFGF-primed HUVECs potentiated, but TNF-alpha primed cellsattenuated, about 50% adhesion events and calcium signaling triggered byimmobilized disintegrin compared to naive cells, respectively. The mechanismsof modulating alpha(v)beta(3)- dependent HUVEC adhesion by cytokines may berelated to changes of integrin alpha(v)beta(3) conformation, as demonstratingthe antagonistic effect of Mn(2+) on decreased adhesion by TNF-alphapretreatment, and confirmed with flow cytometric analysis probed byanti-LIBS1 mAb. However, cytokine pretreatment did not alter the expressionof this integrin on the cell surface, as determined by flow cytometry.Phosphoinositide-3 kinase may be one of the signaling molecules involved in the enhanced adhesion of bFGF-primed cells

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    Crotalin, a Vwf and Gp Ib Cleaving Metalloproteinase from Venom of Crotalus Atrox

    Binding of von Willebrand factor (vWF) to a variety of extracellular matrix (ECM) components and to platelet glycoprotein (GP) lb-IX-V complex is important in mediating platelet adhesion and aggregation in the early stage of hemostasis. We previously purified a potent antithrombotic protein, named crotalin, functionally acting as a GP Ib antagonist (1). In this study, we further characterized crotalin as a P-I metalloproteinase with a molecular mass of 25 kDa as determined by gel filtration and two- dimensional SDS-PAGE. Crotalin is a vWF binding and cleaving metalloproteinase. In addition, crotalin cleaved platelet GP lb as judged by flow cytometry and Western blotting. The multiple effects of crotalin on vWF and platelet GP Ib antagonized ristocetin-, but not collagen and thrombin- induced platelet aggregation, suggesting that its effect is specific. We also found that crotalin auto-proteolytically degraded to similar to 14 and similar to 10 kDa fragments in the presence of SIDS, Interestingly, both degradation fragments, intact and reduced crotalin were able to bind vWF , suggesting the binding of crotalin to vWF is conformation- independent. In conclusion. the results presented further explain the potent antithrombotic effect of crotalin in vivo . In addition , the multiple effects of crotalin may be used as a tool to determine the binding motifs that are responsible for the vWF-ECMs or vWF-GP Ib interaction

    Variations on the Author

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    What Have Snakes Taught Us About Integrins?

    Snake venoms contain unique components that affect cell- matrix interactions. Disintegrins represent a class of low molecular weight, Arg- Gly-Asp (RGD)-containing, cysteine- rich peptides purified from the venom of various snakes among the Viperidae and Crotalidae. They bind with various degrees of specificity to integrins alpha IIb beta 3, alpha 5 beta 1 and alpha V beta 3 expressed on cells. Snake venom metalloproteases ( high molecular mass haemorrhagins) also contain disintegrin-like domains, in addition to zinc- chelating sequences. Membrane-anchored ADAMs (A Disintegrin And Metalloprotease domain), multidomain molecules consisting of metalloprotease, disintegrin-like, cysteine- rich, and epidermal growth factor domains, a transmembrane domain and a cytoplasmic tail, are a new family of proteins. In the light of the large number and wide distribution of ADAMs, they may participate in cell-cell fusion events, including sperm-egg binding and fusion, myoblast fusion and other cell-cell and cell -matrix interactions. The structure- function relationship of these molecules is discussed

    La adecuada planificaci?n del uso del espacio f?sico en los destinos es un factor clave para prevenir conflictos de uso entre los actores involucrados en el desarrollo de las actividades tur?sticas y recreativas; m?s a?n, cuando de por medio existen recursos naturales y culturales que merecen especial atenci?n, cuidado y potencializaci?n. Este trabajo investigativo examina las potencialidades tur?sticas del cant?n Zamora. Con la participaci?n de actores locales se identifican tres zonas tur?sticas, se proponen cinco programas y quince proyectos que aportar?n al desarrollo tur?stico sostenible del cant?n Zamora. Esta propuesta se basa en la acci?n participativa y articula a todos los actores con una visi?n de desarrollo integral. Por otra parte, los resultados muestran la factibilidad de implementar dos nuevas rutas tur?sticas basadas en las caracter?sticas y potencialidades existentes en dos de las tres zonas identificadas, de tal modo que se dinamice la oferta tur?stica del cant?n. En tal virtud, la propuesta ?Ordenamiento del desarrollo tur?stico del cant?n Zamora?, es una herramienta de apoyo para el desarrollo tur?stico sostenible del sector y contribuye al desarrollo integral de la poblaci?n.Mag?ster en Planificaci?n Tur?stic
