    IDENTIFICATION OF THE ANTIMICROBIAL ACTIVITY FROM OCTOPUS VULGARIS HAEMOCYTES.

    The marine environment comprises complex ecosystems and many of organisms are known to possess bioactive components as a common means of self-defence or for the protections of eggs and embryos. Lately, many bioactive compounds have been extracted, characterized and purified from various marine animals like tunicates, sponges, corals, and molluscs (Rajasekharan et al. 2011). Among cephalopods, the only studies involving bioactive peptides with antimicrobial activity (AMPs) are those conducted in squids and cuttlefish. They leave their eggs to fend for themselves and interestingly they do not get any infections. The eggs release polypeptides, sexual pheromones and AMPs trapped in the egg capsule, mainly expressed by female accessory sex gland that confers to them an efficient protection against microorganisms. Cephalopods, unlike other molluscs, have a closed circulatory system. The blood consists of haemolymph, haemocyanin and haemocytes (Novoa et al 2002). In this study we have focused the attention on the morphological and functional characterization of the haemocytes, from Octopus vulgaris, as the first responsible of the innate immunity. Three major haemocytes types were identified by light microscopy (hemoblast-like cells, hyalinocytes and granulocytes) based on nucleus/cytoplasm ratio and the presence or absence of cytoplasm granules. The haemocytes were cytochemically characterized for the presence of different activities responsible of the humoral response (phenoloxidase, peroxidise, phagocytosis). We have evaluated the bactericidal activity of haemocytes in the presence of different bacteria strains, using the method of the plate radial diffusion (Bauer et al 1996). We have also evaluated the activity of a methanolic acid extract of these cells, using the disc diffusion method and the minimum inhibitory concentration (MIC), in order to investigate the presence of soluble biotic compounds responsible of the antibacterial activity.     L'organo olfattorio di O. vulgaris è presente in due piccole fossette, difficilmente discriminabili, poste ai lati della testa posteriormente e ventralmente agli occhi in prossimità del margine mantellare. L???epitelio olfattivo ricopre il lume della fossetta all???interno della quale vi è una sorta di protuberanza mobile che le conferisce, in sezione, un aspetto a ?? che ne aumenta la superfice funzionale. Un nostro primo studio morfologico su tale organo ci ha consentito di evidenziare le tipologia cellulari presenti in tale tessuto e di compararle alle strutture olfattive già descritte di altri cefalopodi, ma ad oggi sono davvero pochi studi funzionali sull???olfatto di questo taxon e non vi è alcun dato riguardante O. vulgaris. Mediante metodiche di immunoistochimica classica, abbiamo localizzato a livello sia del lobo olfattorio che dell???organo olfattorio una serie di peptidi dal noto ruolo funzionale: GnRH; FMRFammide; NPY; APGWammide; G??-olf; G??-q; OMP; PCNA. Sia per quanto riguarda il GnRH sia per APGWammide cellule immunoreaative sono state osservate nel lopo olfattorio mentre nell???organo olfattorio sono state riscontrate solo fibre e terminazioni sinaptiche che terminano in prossimità sia delle ring cells sia dei neuroni olfattivi sensoriali (NOS). Una diffusa immunoreattività a livello citoplasmatico è stata riscontrata nei NOS sia per FMRFammide sia per NPY, quest???ultimo risulta marcare un numero maggiore di cellule in animali che avevano subito un periodo di starvazione. G??-olf; G??-q; OMP sono tutti peptidi coinvolti nelle reazioni biochimiche relative all???olfatto, materiale immunoreattivo per queste molecole è stato riscontrato in diverse popolazioni di NOS distribuite all???interno dell???epitelio sensoriale. La PCNA, noto marche di proliferazione, ci mostra un continuo rimaneggiamento dell???epitelio olfattivo a partire dalle zone periferiche verso il centro. L???analisi complessiva delle immunoreattività osservate ed il ruolo biologico dei peptidi analizzati, evidenziano una chiara modulazione dell???olfatto sia a livello centrale che periferico, e sottolineano il ruolo convergente che tali molecole hanno avuto nel corso dell???evoluzione

    La neurogenesi adulta è il processo mediante il quale si generano e si differenziano nuove cellule nervose nel sistema nervoso di un organismo adulto. E??? ormai accertato che nel cervello adulto persistono cellule staminali, confinate in nicchie neurogeniche, che supportano la formazione di nuovi circui8 nervosi. Il processo è influenzato da una mol8tudine di faMori intrinseci ed estrinseci, tra i primi ricordiamo ormoni steroidei e faMori di crescita, tra i secondi abbiamo l???apprendimento, l???arricchimento dell???ambiente circostante e gli s8moli sensoriali. La neurogenesi adulta è stata riscontrata in organismi che presentano un???elevata complessità struMurale e funzionale del sistema nervoso, quali pesci, reOli, uccelli e mammiferi, e, tra gli invertebra8, crostacei decapodi ed inseO. Sia nei vertebra8 che negli invertebra8, la neurogenesi adulta si verifica in aree che presentano un elevato grado di plas8cità struMurale e che svolgono un ruolo fondamentale nell'apprendimento, nella memoria e nell???integrazione di s8moli sensoriali, ciò suggerisce che il processo sia indispensabile per il supporto delle capacità cogni8ve più avanzate.Octopus vulgaris presenta un sistema nervoso centralizzato con un???organizzazione struMurale e funzionale gerarchica, capacità cogni8ve avanzate e complesse abilità comportamentali. Inoltre, vi sono diverse corrispondenze funzionali e struMurali tra specifici lobi del cervello di Octopus e specifiche aree del cervello di mammiferi e inseO in cui ha luogo la neurogenesi, quali, rispeOvamente, ippocampo e mushroom bodies. Pertanto, O.vulgaris può essere considerato un oOmo candidato per lo studio della neurogenesi adulta. Gli esemplari u8lizza8 per la sperimentazione appartengono alla specie Octopus vulgaris. Gli esperimen8 sono sta8 effeMua8 indis8ntamente su individui di sesso maschile e femminile, il peso medio dei polpi seleziona8 è circa di 800g. Gli animali sono sta8 stabula8 presso il Dipar8mento di Biologia del Complesso Universitario di Monte Sant???Angelo, in vasche in PVC, alimentate con acqua di mare a circuito chiuso, a temperatura controllata. Sono sta8 quindi profondamente aneste8zza8 aMraverso l???u8lizzo di isoflurano (Polese e Di Cosmo, CIAC 2012) e in seguito soMopos8 a dissezione del cervello, condoMa in condizioni di sterilità. Dal 1° gennaio 2013 i cefalopodi sono soMo tutela della Comunità Europea, e il comitato di bioe8ca dell???Ateneo regolamenta la sperimentazione su Octopus vulgaris. I nostri esperimen8 sono sta8 condoO in conformità con i principi e8ci di Riduzione, Perfezionamento e Sos8tuzione (Russell e Burch, 1992), che consistono nell???u8lizzare qualsiasi approccio che evi8, allievi e riduca al minimo il dolore, reale o potenziale, angoscia e altri effeO nega8vi subi8 in qualsiasi momento durante la vita degli animali coinvol8, o che migliori il loro benessere. I cervelli sono sta8 incuba8 con BromodeossiUridina (BrdU), un intercalante nucleo8dico che si incorpora al DNA in sintesi, in vitro, in accordo con Di Cristo, Paolucci e Di Cosmo (2008). In seguito all???inclusione in paraffina, i tessu8 sono sta8 taglia8 al microtomo in sezioni da 7??m, reidrata8 ed incuba8 con an8corpo an8-BrdU a 4°C per tuMa la noMe. Dopo abbondan8 lavaggi, le sezioni sono state incubate con un an8corpo secondario bio8nilato per 1 h a temperatura ambiente, poi con streptavidina coniugata con perossidasi per 1 h a temperatura ambiente. Infine la reazione è stata rivelata usando la diaminobenzidina (DAB) come cromogeno. Le aree del sistema nervoso che risultano posi8ve all???an8-BrdU sono localizzate nella massa sopraesofagea. In par8colare, si osservano struMure riconducibili a nicchie di cellule in proliferazione che si approfondano nei neuropili dei lobi appartenen8 al sistema verFcale-frontale, sede dell???apprendimento e della memoria. Tali risulta8 confortano l0ipotesi che in Octopus vulgaris abbia luogo il processo della neurogenesi adulta

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     NEUROPEPTIDES ANALYSIS OF OCTOPUS VULGARIS TRANSCRIPTOME: OCT-PEDAL PEPTIDE EXPRESSION IN CNS LOBES INVOLVED IN MOTOR COORDINATION

    The discovery of novel peptides and hormones from the phylum Mollusca, enabled by the availability of whole genomes, transcriptomes and peptidomes, gives now ample tools for studying the structural evolution of peptides and hormones, and for redefining the phylogenetic relationships of key families of peptide and hormones across ecdysozoans, lophotrochozoans and chordates. In this frame, the genome sequence of the limpet Lottia gigantea (Veenstra, 2010) and the initiated large-scale molluscan transcriptome and genome projects (Moroz et al., 2011; De Lisa et al., 2011; Di Cosmo and Polese, 2012) offer an excellent opportunity to look at the neuropetides and neurohormone genes. Mollusks have several genes coding pedal peptides (PP), three in Lottia and four in Aplysia (Veenstra, 2010). Neuropeptides are the fastest evolving intercellular signals; in particular, cephalopods developed a number of specific innovations in their repertoire of secretory molecules that might supp ort their unique features (Moroz, 2011). Using neuronal transcriptome from Octopus vulgaris, we developed a specific probe to analyze expression of Oct-PP in O. vulgaris brain in horizontal sections throughout the suboesophageal and supraoesophageal masses, including optic lobes. Performing ISH analysis we found the expression of Oct-PP mRNA confined in the anterior suboesophageal mass, in the prebrachial lobe and in the middle suboesophageal mass, in all pedal lobes. The prebrachial lobe is involved in reflex coordination. The middle suboesophageal mass, representative of the pedal ganglionated cord of early mollusks, is involved in all actions in Octopus (Young, 1971). Our findings support the involvement of Oct-PP in reflex and motor coordination in Octopus
