Exploring conformational dynamics and equilibria of ABC transporters
ATP-Bindungskassetten-Transporter bilden eine bedeutende Superfamilie von Membranproteinen, die Energie aus der Hydrolyse von ATP nutzen, um Substratmolekle über die Zellmembran zu transportieren. Ein detailliertes Verständnis der Transportmechanismen, sowie ihrer Inhibition ist notwendig, um Krankheiten zu bekämpfen, die durch Fehlfunktionen, Mutationen oder Überexpression von ABC-Transportern verursacht werden. Verschiedene biochemische Werkzeuge wie Elektronenmikroskopie oder Röntgenkristallographie haben tiefe Einblicke in die Struktur und Funktion von verschiedenen ABC-Transportern gegeben, jedoch liefern sie nur statische Einsichten in den Transportzyklus. Dazu sind zusätzliche Methoden nötig, die die Dynamik und die Zwischenzustände des Systems beschreiben können. Elektronenspinresonanz (ESR/EPR)-Spektroskopie in Kombination mit ortsspezifischer Spinmarkierung bietet leistungsstarke Werkzeuge um genau diese Dynamiken zu untersuchen.ATP binding cassette (ABC) transporters form a significant super family of membrane proteins coupling the energy of ATP hydrolysis to substrate transport across the cell membrane. Unraveling transport and inhibition mechanisms is crucial to address several diseases caused by malfunction, over expression and mutation of ABC transporters. Several biochemical tools including electron microscopy and X-ray crystallography have aided the mechanistic characterization of many ABC transporters in static states of the transport cycle. While providing unprecedented insight into the mechanism, static states cannot explain the complete machinery. To achieve a full understanding, all the intermediate states as well as the collective behavior of transporters throughout the cycle need to be studied. Site-directed spin labeling and electron paramagnetic resonance (EPR) techniques provide invaluable information on dynamics, structure and function of ABC transporters under physiological conditions

From Kuṭa Rāja to Singhasāri: Towards a Revision of the Dynastic History of 13th Century Java
In attempting to reconstruct Javanese dynastic history of the first half of the 13th century, scholars have in the past been forced to rely almost entirely upon later works of literature, notably the Deśawarn M ana and Pararaton. Recently discovered epigraphical data demands a re-evaluation of these literary accounts. From an examination of the new evidence provided by the 1255 inscription of Mūla-Malurung, the author concludes that the term Singhasāri as it is currently applied to define 13th century Java is something of a misnomer. The accession of the king Wis J n J uwardhana in around 1250 needs to be viewed as a definitive landmark separating two distinct chapters, represented by the ages of Kut M a Rāja and Singhasāri proper. It is proposed that this new perspective can serve as a more accurate framework upon which to construct a revised history of the Rājasa dynasty.De Kuṭa Rāja à Singhasāri : vers une révision de l’histoire dynastique de Java au XIIIe siècle
Afin de tenter de reconstruire l’histoire dynastique javanaise de la première moitié du XIIIe siècle, les chercheurs ont été contraints, jusqu’à présent, de s’en remettre presque entièrement à des sources littéraires plus tardives, notamment le Deśawarn J ana et le Pararaton. Des données épigraphiques découvertes récemment réclament un réexamen de ces sources littéraires. À partir des éléments nouveaux livrés par l’inscription de Mūla-Malurung, datée de 1255, l’auteur conclut que le terme Singhasāri, tel qu’il est couramment utilisé à présent pour définir Java au XIIIe siècle, est inapproprié. L’arrivée au pouvoir du roi Wis M n M uwardhana vers 1250 doit être considérée comme un événement décisif séparant deux chapitres distincts représentés par les âges de Kut J a Rāja et de Singhasāri à proprement parler. When there is combustion there are three main components that cause the fire which is commonly called the Fire Triangle. The fire triangle itself consists of unified and balanced Heat, Fuel and Oxygen that will cause fires to the point where workers are also very detrimental to the company if this happens if it is not immediately addressed. The author in collecting the data needed until the completion of writing this scientific paper, uses the method of approach and data collection by Observation and Literature Study on the MT ship. Katomas which is an oil product cargo ship which is a flammable liquid, the best extinguishing agent is foam fire extinguisher. Fire fighting equipment is also not only a supporting factor but also an indispensable factor and is part of the ship and its crew. Safety concerns the safety of the cargo, crew and ship itself. Then the damage to the fire extinguisher and the decline in the work function of the tool is due to lack of maintenance and attention to the extinguisher which causes the work of the device to be less than optimal.

Saat terjadi pembakaran ada tiga komponen utama yang menyebabkan terjadinya api yang biasa disebut Segitiga Api. Segitiga api itu sendiri terdiri dari Heat, Fuel dan Oxygen yang bersatu dan seimbang yang akan menimbulkan kebakaran hingga membahayakan para pekerja juga sangat merugikan Perusahaan apabila hal ini terjadi jika tidak segera ditanggulangi. Penulis dalam mengumpulkan data yang diperlukan hingga selesainya penulisan karya tulis ilmiah ini, menggunakan metode pendekatan dan pengumpulan data dengan Observasi dan Studi Pustaka pada kapal MT. Katomas yang merupakan kapal muatan Oil Product yang merupakan zat cair yang mudah terbakar, maka zat pemadam yang paling baik adalah jenis pemadam api foam. Peralatan pemadam kebakaran juga bukan hanya faktor pendukung tetapi juga merupakan salah satu faktor yang sangat diperlukan dan menjadi bagian dari kapal dan awaknya. Keselamatan yang menyangkut keselamatan pada muatan, awak kapal, dan kapalnya itu sendiri. Maka kerusakan alat pemadam api dan menurunnya fungsi kerja dari alat tersebut dikarenakan akibat kurangnya perawatan dan perhatian terhadap alat pemadam tersebut yang menyebabkan kerja dari alat tersebut menjadi kurang optimal.

General distribution. Bangladesh, Far East, India, Malaysia, Pakistan, Sumatra (Perveen 1991). Comments. Moreover, one damaged specimen of Homoeocerus sp. was found from Saravan (Sistan & Baluchestan province). On the basis of preliminary identification by the first author and also I.M. Kerzhner, it is probably H. lacertosus Distant 1889 with distribution in India and Pakistan. Mode of Interaction of the Signal-Transducing Protein EIIA<sup>Glc</sup> with the Maltose ABC Transporter in the Process of Inducer Exclusion

IIAGlc (EIIAGlc) of the phosphoenolpyruvate
phosphotransferase system for the uptake of glucose in Escherichia
coli and Salmonella inhibits the maltose
ATP-binding cassette transporter (MalE-FGK2) by interaction
with the nucleotide-binding and -hydrolyzing subunit MalK, a process
termed inducer exclusion. We have investigated binding of EIIAGlc to the MalK dimer by cysteine cross-linking in proteoliposomes.
The results prove that the binding site I of EIIAGlc is
contacting the N-terminal subdomain of MalK while the binding site
II is relatively close to the C-terminal (regulatory) subdomain, in
agreement with a crystal structure [Chen, S., Oldham, M. L., Davidson, A. L., and Chen, J. (2013) Nature 499, 364−368]. Moreover, EIIAGlc was found to bind to the
MalK dimer regardless of its conformational state. Deletion of the
amphipathic N-terminal peptide of EIIAGlc, which is required
for inhibition, reduced formation of cross-linked products. Using
a spin-labeled transporter variant and EPR spectroscopy, we demonstrate
that EIIAGlc arrests the transport cycle by inhibiting
the ATP-dependent closure of the MalK dimer
Intraglandular toxoplasmosis of the parotid gland pre- or postoperative diagnosis?
Toxoplasmosis is an ubiquitous protozoan infection, caused by the coccidian Toxoplasma gondii. It usually presents clinically with lymphadenopathy, mainly in the head and neck region. However, in the English literature, very scarce cases of intraparotid Toxoplasma exist, none having been diagnosed preoperatively. In this article, we present 2 cases of intraglandular parotid toxoplasmosis, one diagnosed postoperatively and the other preoperatively. Is demand for polluting goods manageable? an econometric study of car ownership and use in Mexico
Charging for social marginal costs is efficient regardless of price elasticities, but the importance of getting prices"right"is greater the more manageable, or elastic, the demand. In efficient pollution control programs, options to make cars cleaner are combined optimally with demand conservation. The roles played by"cleaner cars"as compared with"fewer trips"are determined by empirical parameters: cheap, clean technologies would imply a great role for cleaner cars, while high demand elasticities lead to a greater role for demand reduction. In seminal research, evisence was found to support the hypothesis that demand for commodities such as gasoline should have lower price elasticities and higher income elasticities in developing than in industrial countries. The authors estimate a model of gasoline demand and car ownership in Mexico, using a panel of annual observations by state. Key features they introduce are instrumental variables on different data and the treatment of (1) possible dynamics, (2) measurement errors in the data, and (3) unobserved characteristics in individual states. They use tests of serial correlation in the residuals to model the dynamics properly. The resulting model is one of almost immediate adjustment, with a short-term price elasticity for gasoline close to the long-term estimate of -0.8. The model displays elasticities that are lower (for income) and higher (for price) than those hypothesized, and are within the range of elasticities found in industrial countries. Byproducts of the model: The elasticity of car purchases with respect to gasoline prices is positive. Scrappage decisions are affected by income and by car and gasoline prices. And these elasticities are not significantly different in the richer states. For policy purposes, these findings do not support"elasticity pessimism"The use of car services is sensitive to pricing, which suggests that consumers, for some of their demand, have reasonably good alternatives to car services.
