TATA UPACARA PERKAWINAN DAN HANTARAN PENGANTIN BEKASRI LAMONGAN
ABSTRAK Bekasri Lamongan merupakan pengantin Tradisional yang merupakan warisan budaya lokal Lamongan yang tercermin pada tata rias, tata busana, dan penataan rambut serta rangkaian prosesi pernikahan diantaranya dengan berbagai macam jenis hantaran. Penelitian ini bertujuan untuk mendiskripsikan (1) tata upacara perkawinan Bekasri Lamongan serta maknanya, dan (2) jenis hantaran dan makna hantaran pengantin pada upacara perkawinan Bekasri Lamongan. Jenis penelitian ini adalah deskriptif kualitatif dengan teknik pengumpulan data menggunakan wawancara, dokumentasi dan observasi. Analisis yang digunakan adalah analisis deskriptif dalam bentuk narasi yang dilakukan secara mendalam terhadap tata upacara perkawinan dan hantaran pengantin Bekasri Lamongan. Hasil penelitian menunjukkan bahwa (1) tata upacara perkawinan Bekasri Lamongan terdiri dari tata upacara perkawinan pranikah, tata upacara perkawinan menjelang pernikahan dan tata upacara pasca nikah. (2) hantaran diberikan pada 3 tahapan yaitu pada tahap pranikah yaitu saat lamaran, berupa alat sholat oleh calon mempelai wanita kepada mempelai laki-laki yang bermakna supaya calon mempelai laki-laki rajin beribadah, sedangkan makanan yang rekat bermakna agar merekatkan kedua belah pihak. Calon mempelai laki-laki membalas lamaran dengan hantaran berupa pakaian “sak pengadek” yang merupakan simbolis keikhlasan lahir batin untuk memberi pada calon istri. Pada tahap menjelang pernikahan yaitu prosesi srah-srahan, hantaran berupa bahan makanan. Sedangkan pada tahap pasca nikah diberikan pada saat temu manten yaitu tikar dan bantal. ABSTRACK Bekasri Lamongan is traditional bride forms a local inheritance culture of Lamongan focuses in make up, dress-making, and hair styling and also a series of wedding procession which are with some kinds of wedding gift. The purpose of this research is to describe (1) wedding ceremony of Bekasri Lamongan and its meaning, (2) kinds of wedding gift and its meaning in the wedding ceremony of Bekasri Lamongan. The kinds of this research is qualitative description with collecting data technique which is used descriptive analysis in narration form which was done deeply about wedding ceremony and wedding gift of Bekasri Lamongan. The result of this research shows that (1) wedding ceremony of Bekasri Lamongan consists of wedding ceremony before marriage, wedding ceremony towards the wedding and wedding ceremony after marriage (2) wedding gift is given in three steps. They are on prewedding step, that is at engagement, consists of prayer equipment and some foods from the woman which have meaning in order to her fiance always pray frequently while some foods have meaning together each other. The fiance replies engagement with wedding gift contains cloths full of body that forms symbols of the sincere with all his heart and soul for his wife. On the towards wedding step is take and give procession, the gift contains of food material. While on the after marriage step is given in brade meeting which are pillow and bolster as a symbols. A Conserved GA Element in TATA-Less RNA Polymerase II Promoters
Initiation of RNA polymerase (Pol) II transcription requires assembly of the pre-initiation complex (PIC) at the promoter. In the classical view, PIC assembly starts with binding of the TATA box-binding protein (TBP) to the TATA box. However, a TATA box occurs in only 15% of promoters in the yeast Saccharomyces cerevisiae, posing the question how most yeast promoters nucleate PIC assembly. Here we show that one third of all yeast promoters contain a novel conserved DNA element, the GA element (GAE), that generally does not co-occur with the TATA box. The distance of the GAE to the transcription start site (TSS) resembles the distance of the TATA box to the TSS. The TATA-less TMT1 core promoter contains a GAE, recruits TBP, and supports formation of a TBP-TFIIB-DNA-complex. Mutation of the promoter region surrounding the GAE abolishes transcription in vivo and in vitro. A 32-nucleotide promoter region containing the GAE can functionally substitute for the TATA box in a TATA-containing promoter. This identifies the GAE as a conserved promoter element in TATA-less promoters
Ultrastructural characterisation of Bacillus subtilis TatA complexes suggests they are too small to form homooligomeric translocation pores
Tat-dependent protein transport permits the traffic of fully folded proteins across membranes in bacteria and chloroplasts. The mechanism by which this occurs is not understood. Current theories propose that a key step requires the coalescence of a substrate-binding TatC-containing complex with a TatA complex, which forms pores of varying sizes that could accommodate different substrates. We have studied the structure of the TatAd complex from Bacillus subtilis using electron microscopy to generate the first 3D model of a TatA complex from a Gram-positive bacterium. We observe that TatAd does not exhibit the remarkable heterogeneity of Escherichia coli TatA complexes but instead forms ring-shaped complexes of 7.5–9 nm diameter with potential pores of 2.5–3 nm diameter that are occluded at one end. Such structures are consistent with those seen for E. coli TatE complexes. Furthermore, the small diameter of the TatAd pore, and the homogeneous nature of the complexes, suggest that TatAd cannot form the translocation channel by itself. Biochemical data indicate that another B. subtilis TatA complex, TatAc, has similar properties, suggesting a common theme for TatA-type complexes from Bacillus

La diffusione delle ICT e dei social media ha determinato un cambiamento sociale nel modo in cui le persone interagiscono e comunicano. Ci troviamo di fronte a una democratizzazione dell’informazione, che trasforma gli utenti da semplici fruitori passivi a produttori, editori e condivisori di contenuti.
I social network, in particolare, sono oggi visti come ambienti per archiviare fotografie, condividendole con altre persone con interessi comuni. Le interazioni e la costruzione di relazioni si basano ormai spesso sulla creazione di una memoria e una conoscenza condivise e accessibili, fondate su fotografie, più o meno personali.
Il contributo presenta una riflessione sul bisogno di memoria, e mantenimento della stessa, manifestato dagli abitanti della città dell’Aquila dopo il terremoto del 2009, che ha provocato lo svuotamento del centro storico della città, e la conseguente rottura tra l’identità dei luoghi e la dimensione immateriale, intesa come spazio di vita quotidiano
REKAYASA TATA RIAS WAJAH PENGANTIN ARAB MODERN
Abstrak Tata rias wajah pengantin Arab memiliki karakteristik riasan yang tajam, glamour dan cenderung menor. Namun adanya tren tata rias wajah pengantin diharapkan depat menjadi inspirasi tata rias yang dapat mengesankan lebih anggun dan cantik. Penelitian ini bertujuan untuk mengetahui penilaian responden terhadap hasil perwujudan tata rias wajah pengantin Arab modern. Jenis metode penelitian yang digunakan adalah jenis metode dekriptif kualitatif. Dengan metode pengumpulan data yang digunakan yaitu angket. Teknik analisis data menggunakan rumus mean/rata-rata. Hasil penelitian ini menunjukkan bahwa hasil perwujudan rekayasa tata rias wajah pengantin Arab modern mendapat respon dan penilaian sangat baik dan dapat diterima oleh panelis. However, the bridal makeup make up trend is expected to be a makeup inspiration that can impress more elegant and beautiful. This study aims to determine respondents assessment of the results of the embodiment of modern Arab bridal makeup. The type of research method used is the type of qualitative descriptive method. The data collection methods used are interviews, observation and questionnaire. Data analysis techniques using the mean / average formula. The results of this study indicate that the results of the embodiment of modern Arab bridal make-up engineering received very good responses and ratings and were accepted by panelist. PENINGKATAN KETERAMPILAN TATA RIAS WAJAH MELALUI PELATIHAN TATA RIAS PANGGUNG BAGI TIM PADUAN SUARA GEREJA KRISTEN NAZARENE FILADELFIA YOGYAKARTA
Abstrak: Pelatihan keterampilan tata rias panggung diberikan pada Tim Paduan Suara di Gereja Kristen Nazarene Filadelfia Yogyakarta untuk meningkatkan keterampilan tata rias yang digunakan untuk penampilan tim paduan suara. Penelitian ini bertujuan untuk mengetahui: 1) pengelolaan pelatihan keterampilan tata rias panggung, 2) aktivitas peserta pelatihan, 3) hasil pelatihan keterampilan tata rias panggung, 4) respons peserta pada pelatihan tata rias panggung. Penelitian ini merupakan penelitian pra-eksperimen dengan rancangan penelitian pre-test and posttest group design. Data yang didapat berupa hasil pengelolaan pelatihan, aktivitas peserta pelatihan, hasil pretest dan posttest keterampilan tata rias panggung, dan respon peserta pelatihan. Subyek penelitian adalah 26 orang anggota Tim Paduan Suara Gereja Kristen Nazarene Filadelfia Yogyakarta. Metode pengumpulan data yang digunakan adalah metode observasi, tes kinerja, dan angket. Metode analisis menggunakan rata-rata untuk pengelolaan pelatihan dan aktivitas peserta, hasil pelatihan menggunakan uji t dan respon peserta menggunakan persentase. Berdasarkan hasil analisis data, pengelolaan pelatihan tata rias panggung memperoleh penilaian dalam kriteria sangat baik. Aktivitas peserta pelatihan memperoleh penilaian dengan kriteria baik. Data hasil tata rias panggung yang diperoleh dari nilai pretest dan posttest menunjukkan perbedaan yang signifikan dan peningkatan setelah dilakukannya pelatihan pelatihan keterampilan tata rias panggung dengan metode demonstrasi dan panduan handout. Hasil akhir yang diterima menunjukkan terdapat peningkatan keterampilan tata rias panggung sesudah dilakukan pelatihan bagi Tim Paduan Suara Gereja Kristen Nazarene Filadelfia Yogyakarta. Respon peserta terhadap pelatihan keterampilan tata rias panggung menunjukkan persentase rata-rata sebesar 100% dengan kriteria sangat baik.

Abstract: Stage makeup skills training is given to the member of choir team in the Church of Philadelphia Nazarene at Yogyakarta to improve their skills of makeup that used for performance. The purpose of this research is to find out about: 1) the execute of stage make up training, 2) the activities of participant, 3) the result of stage makeup skills training, 4) participant response about stage make up training. This research is a pre-experimental research that use pre-test and posttest group. The data result constitute the product of training execute, the activities of participant, the results of pretest and posttest stage makeup skills, and the participant response. The subjects of this research are 26 member of choir in the Church of Philadelphia Nazarene at Yogyakarta. The collection data method that used in this research is observation method, assignment tests, and questionnaire. Analysis method that use average to find out about training execute and the activities of participant. Ttest is use to find out about the training results and percentages is use to find out about the participant response. According to the data analysis, stage make up training execute was got score in very good criteria. The activity of participant was got score in good criteria. The result of stage make up from pretest score and posttest score shows significancy difference and increase after the stage make up training with the method of demonstration and handouts. The final result shows that stage make up skill is increase after executed training for the choir in the Church of Philadelphia Nazarene at Yogyakarta. The response of participants about stage make up training showsthe average percentage of 100% in the very good criteria.

The twin arginine translocase pathway : a functional and structural study in Escherichia coli and Synechocystis sp. PCC6803
The bacterial twin-arginine translocase (Tat) pathway is able to export large folded proteins across the cytoplasmic membrane. It derives its name from the almost invariant twin-arginine motif present in the signal peptide of Tat substrates.
Components of the Tat pathway have been identified in many bacteria, and in Escherichia coli one of these essential proteins, TatA, forms large complexes of variable size. The research presented in this thesis sought to gain insight into the Tat pathway in E. coli and the cyanobacterium Synechocystis sp. PCC6803, mainlyfocusing on the TatA component of the E. coli Tat pathway and how its various domains contribute to function and complex formation.
Green fluorescent protein tagged with a Tat signal peptide was used to compare the Tat pathway in E. coli and Synechocystis, using biochemical and bioimaging techniques. Substitution of the twin-arginine motif with lysine residues highlighted differences between the specificity of the two systems.
The transmembrane (TM) domain and amphipathic helix (APH) of E. coli TatA were analysed using synthetic peptides and the TOXCAT assay. Investigation of the TM domain demonstrated it is α-helical and spontaneously inserts into membranes. It interacts relatively weakly and substitution of a glutamine does not affect these interactions or function of the protein at high expression levels. In contrast, the APH only forms an α-helical secondary structure in high detergent concentration or in the presence of negatively charged lipids, and does not insert into lipid bilayers. Analysis of the unstructured C-terminus of TatA led to the identification of an acidic motif just after the APH. Removal of this motif has a severe effect on function and complex formation of TatA. Removal of the C-terminus does not affect the proofreading abilities of TatA.
The research presented here has led to the proposal of a new model for TatA complex formation in E. coli – the weak interactions of the TM domains bring the monomers together, whilst interactions between the acidic motif and other regions stabilise these complexes
MODIFIKASI TATA RIAS PENGANTIN PUTRI MUSLIM BLITAR KARTIKA RUKMI
Abstrak: Tata rias pengantin Blitar Kartika Rukmi dapat dipertahankan kelestariannya dan dikembangkan melalui cara modifikasi pengantin putri muslim. Modifikasi berarti memberi sentuhan baru untuk menghasilkan tampilan yang berbeda tanpa harus meninggalkan jejak asli dari karya tersebut. Modifikasi pengantin putri muslim Blitar Kartika Rukmi dapat dilakukan dengan penambahan kreasi jilbab, busana yang digunakan ( sebagai pendukung), dan tata rias wajah yang memadu padankan dengan corak busana serta aksesoris yang digunakan. Tujuan penelitian ini adalah 1) Mengetahui modifikasi tata rias wajah, penataan jilbab modern, tata busana dan askesorisnya pengantin putri muslim Blitar Kartika Rukmi. 2) Mengetahui respon panelis terhadap modifikasi tata rias pengantin putri muslim Blitar Kartika Rukmi meliputi tata rias wajah, penataan jilbab modern , tata busana beserta aksesorisnya. Jenis penelitian ini adalah penelitian deskriptif, yang bertujuan untuk mengetahui hasil modifikasi tata rias wajah, tata busana dan penataan jilbab. Data penelitian dikumpulkan dengan menggunakan teknik wawancara, observasi dan dokumentasi. Penelitian ini menggunakan sumber data primer dan sekunder. Hasil penelitian ini adalah: 1) dipilih 1 desain dari 2 desain yang telah disiapkan, berdasarkan masukan para ahli dipilih desain 2 yaitu: a) warna pada busana hijau botol percampuran kain bludru dan brokat, b) aksesoris pengantin blitar kartika rukmi, c) warna eyeshadow ( hijau tua di baurkan dengan hijau muda), d) warna foundation dan bedak tabur (kuning langsat), e)kain pada jarik tetap menggunakan jarik kawung wulan purnomo.2) hasil modifikasi tata rias pengantin putri muslim blitar kartika rukmi melalui tahapan persiapan area kerja, alat, bahan, lenan, dan kosmetik serta persiapan model. Modifikasi difokuskan pada tata rias wajah, penataan jilbab dan tata busana. 3)penilaian halayak terhadap hasil modifikasi menyatakan bahwa tata rias wajah baik, kreasi jilbab dinyatakan baik, tata busana beserta aksesoris dinyatakan baik. Sehingga hasil modifikasi dinyatakan baik dan layak untuk digunakan atau dipublikasikan.

PENGEMBAGAN MULTIMEDIA ELECTRONIC PORTFOLIO PADA TATA RIAS PENGANTIN TRADISIONAL SUKU TOLAKI SULAWESI TENGGARA
Abstrak: Tata rias pengantin tradisional merupakan salah satu bentuk kebudayaan di Indonesia yang harus dijaga kelestarianya, seperti tata rias pengantin suku Tolaki Sulawesi Tenggara. Cara yang dapat dilakukan untuk melestarikan kebudayaan tersebut adalah dengan membuat dokumentasi dengan materi tata rias pengantin tradisional suku Tolaki. Dokumentasi tersebut diwujudkan dalam bentuk aplikasi multimedia electronic portfolio yang dapat menyimpan dokumen dalam berbagai format seperti: teks, gambar, video, dan power point (PPT), dengan menggunakan adobe acrobat X pro.Penelitian ini bertujuan untuk menghasilkan multimedia electronic portfolio pada tata rias pengantin tradisional suku Tolaki Sulawesi Tenggara yang layak Penelitian ini dilakukan dengan menggunakan model penelitian reaserch and development (R&D), yang terdiri dari 6 tahapan yaitu, potensi dan masalah, pengumpulan data, desain produk, validasi desain, perbaikan desain, dan analisis dan pelaporan. Hasil penelitian menunjukkan nilai rata-rata hasil validasi kelayakan multimedia oleh 3 orang dosen ahli multimedia UNESA adalah 87,7%. Nilai rata-rata hasil validasi kelayakan materi oleh 1 orang dari Dinas Pariwisata Sulawesi Tenggara dan 2 orang perias pengantin tradisional suku Tolaki adalah 90,3%. Dari hasil penelitian respon yaitu mahasiswa S1 pendidikan tata rias UNESA dan perias pengantin tradisional suku Tolaki mendapat nilai rata-rata sebesar 87,6%. Sehingga dari hasil analisis yang didapatkan dari pengembangan multimedia electronic portfolio pada tata rias pengantin tradisional Suku Tolaki layak untuk digunakan dengan kategori sangat baik/ sangat layak.

Abstract: Traditional wedding make up is one of Indonesian culture that must be kept its at sustainability, such as traditional wedding make up of Tolaki ethnic of southeast Sulawesi. The way to conserve these culture is making documentation associated with subject of traditional wedding make up of Tolaki ethnic. The documentation can be made in the form multimedia application of electronic portfolio that can save documention in various format, such as text, picture, video, and powerpoint (PPT), by using adobe acrobat X pro. This research is due to produce feasible mulitmedia electronic portfolio in traditional wedding make up of tolaki ethnic of Southeast Sulawesi. This research using research and development (R&D), model which consists of 6 steps, namely: potency and problem, data collecting, product design, design validation, design improvements, and analysis and report. Experiment result shows average scores of validation result multimedia feasible by 3 lecture of UNESA multimedia master is 87,7%. Average score of subject feasible validation by one representative of tourism department of Southeast Sulawesi and two representative of traditional wedding beautycian of Tolaki ethnic are 90,3%. From respondent result, that is student of S1 cosmetology education of UNESA and tolaki ethnic beautician result average score as 87,6%. So, from the analysist result that is achieved from multimedia electronic portfolio development in traditional wedding make up of Tolaki ethnic is feasible by category very good/very feasible.

TATA RIAS DAN KOSTUM KARAKTER NYONYA LOVETT DALAM OPERA SWEENEY TODD PRODUKSI TEATER SENDRTASIK UNIVERSITAS NEGERI SURABAYA
Abstrak: Tata rias dan kostum merupakan salah satu kebutuhan dalam seni pertunjukan teater. Teater Sendratasik Universitas Negeri Surabaya mementaskan sebuah naskah legendaris Inggris berjudul Sweeney Todd ke dalam pertunjukan Opera. Pertunjukan tersebut membutuhkan proses analisis naskah untuk membuat konsep desain, penciptaan karya dan penyajian hasil tata rias dan kostum karakter Nyonya Lovett. Penelitian ini bertujuan untuk mengetahui 1) penokohan karakter berdasarkan analisis naskah 2) desain tata rias dan kostum 3) hasil penciptaan tata rias dan kostum, dan 4) penyajian karya tata rias dan kostum karakter Nyonya Lovett dalam Opera Sweeney Todd. Jenis penelitian yang dilakukan adalah penelitian deskriptif kualitatif dan kuantitatif. Data dikumpulkan dengan metode wawancara, dokumentasi dan observasi. Prosedur penelitian melalui tahap analisis naskah, perancangan desain, penciptaan, dan penyajian karya yang dapat dinilai langsung oleh penonton. Teknik analisis untuk data kualitatif melalui tahapan mengorganisasikan data, mengelompokan, dan menguji keabsahan data. Analisis data kuantitatif berupa rata-rata (mean) untuk menilai hasil tata rias dan kostum karakter Nyonya Lovett. Hasil analisis naskah menunjukkan penokohan Nyonya Lovett secara tiga dimensi (fisiologis, psikologis, dan sosiologis), yaitu seorang wanita berusia 40 tahun, janda pemilik kedai pai, bermuka dua, dan sadis. Berdasarkan analisis penokohan tersebut dihasilkan 3 konsep desain tata rias Nyonya Lovett. Hasil pemilihan desain disepakati desain nomor 2 yang akan diciptakan. Desain yang telah dipilih kemudian diciptakan dan ditampilkan pada pertunjukan Opera Sweeney Todd. Hasil penciptaan dinilai langsung oleh penonton dengan rata-rata 4,29, dengan predikat baik. Dengan demikian dapat disimpulkan bahwa hasil tata rias dan kostum karakter Nyonya Lovett telah sesuai dengan kebutuhan pertunjukan dan dapat diterima oleh penonton.

Abstract:Makeup and costumes is one of necessity in theater performing. Sendratasik Theatre of Universitas Negeri Surabaya staged a legendary British manuscript entitled Sweeney Todd into Opera performances. The performance need a analyzes the Sweeney Todd script to makes a design concept and present the makeup and costume character of Mrs. Lovett. This research aims is to know 1) characterizations of the characters based on analysis script 2) makeup and costume design 3) the result of the creation makeup and costume, and 4) the presentation of the results of the makeup and costume characters of Mrs. Lovett. Type of this research is a descriptive qualitative and quantitative. Data were collected by interview, documentation and observation. The research prosedure is a script analysis, designing, creating, and presenting of Mrs. Lovett’s makeup and costume that can be accepeted directly by the audience. Analysis techniques for qualitative data is organizing data, categorize, and test the validity of the data. Analysis of quantitative data such as average (mean) to assess the results of the Mrs. Lovett’s makeup and costume. The script analysis showed Mrs. Lovett’s three-dimensional characterization (physiological, psychological, and sosiological dimentions) is a 40 year old woman, widow of pie shop owner, a liar, and sadistic . The analysis has been produced 3 designs and the result is second design has been selected. The design has been created and used on Sweeney Todd Opera perfomances.
