    Background: There is a need for new approaches in agriculture to improve safety of final products as well as to increase environmental acceptability. In this paper, the biodynamic preparation 501 (horn silica) was sprayed on Vitis vinifera (L.) cv Garganega plants in two vineyards located in Veneto region, North-East Italy. Leaf samples were collected on the day of 501-treatment and 11 days later, and berries were sampled at harvest time. Leaves and berries samples were analysed combining targeted and untargeted measurements related to primary metabolism (pigment, element and amino acid contents) and to secondary metabolism. Chlorophyll content in leaves, and amino acid and element (C, N, S) analysis in berries were combined with untargeted UPLC-QTOF metabolomics.Results: The discriminant compounds related to the 501-treatment were annotated on the basis of accurate MS and fragmentation and were identified as secondary metabolites, namely phenolic constituents belonging to the shiki-mate pathway. The level of most of the identified compounds increased in plants treated with 501 preparation.Conclusions: Results highlight the prominent value of the metabolomic approach to elucidate the role of the 501 applications on grapevine secondary metabolism

    Preliminary evaluation of Quince (Cydonia oblonga Mill.) fruit as extraction source of antioxidant phytoconstituents for nutraceutical and functional food applications

    Background Quince (Cydonia oblonga) fruits can be considered as starting material for the extraction of health-promoting phytochemicals, to be exploited in food and nutraceuticals. In the present work, LC-DAD-MSn analysis allowed to study the phytochemical composition of quince fruits and to compare it with those of six commercial apple varieties. Results Distribution and quantification of secondary metabolites in peel and pulp was studied and compared with six commercial apple varieties. Furthermore the in vitro antioxidant activity was determined by DPPH assay. Quince fruit showed significant amounts of shikimic and quinic acid derivatives, as well as flavonoids and procyanidins. Compared with apple, quince composition is charachterized by the presence of 4-caffeoylshikimic acid, 4-caffeoyl quinic acid, quercetin-3,7-diglucoside, kaempferol-3-O-rhamnoside and kaempferol-7-O-glucoside, while the dihydrochalcones are not detectable. Considering fruit parts,3-O-Caffeoylquinic acid is the most abundant compound both in peels and the pulp. Pearson correlation index indicated a positive relationships between the amount of phenolic constituents and the radical scavenging activity toward DPPH• both for peels and pulps extracts. Conclusions This study highlighted the presence of significant amounts of valuable secondary metabolites in quince and in particular procyanidins and caffeoyl esters with shikimic and quinic acid. Notably, due to the higher content in phenolic compounds and the stronger antioxidant capacity compared to the other considered fruits, the use of C. oblonga as source of antioxidant can be valuable in nutraceuticals , showing new possible applications of quince fruit

    A new step on the chemical profiles and pharmacological effects of three Scorzonera species (S. hieraciifolia, S. hispanica and S. tomentosa)

    The genus Scorzonera is one of the most important genera in the Asteraceae family, and members of the genus have great potential in traditional medicine systems. In this article we aimed to determine skin protective and antimicrobial effects of different extracts from the roots and aerial parts of three Scorzonera species (S. hieraciifolia, S. hispanica, S. tomentosa). Antimicrobial properties were evaluated against different pathogen bacterial and fungal strains, whereas protective effects were studied in isolated mouse skin specimens exposed to hydrogen peroxide. In this biological model, the levels of l-dopa and prostaglandin E2 (PGE2) were evaluated, as markers of tyrosinase activity and inflammation, respectively. All extracts were effective in blunting the increased hydrogen peroxide-induced l-dopa turnover and PGE2 level; thus, suggesting skin protection that could be partly related to the content of flavonoids, among which rutin, in the tested extracts. Additionally, all extracts were effective as antibacterial and antifungal agents; intriguing results were observed against Trichoderma viride, which was particularly sensitive to S. tomentosa root extracts. Therefore, the present study strengthens the importance of investigating herbal preparations obtained from plants belonging to the genus Scorzonera, for the management and prevention of inflammatory and infectious skin conditions

    MUCEET 2009 is organized by Malaysian Technical Universities Network (MTUN) comprising of Universiti Malaysia Perlis (UniMAP), Universiti Tun Hussein Onn (UTHM), Universiti Teknikal Melaka (UTeM) and Universiti Malaysia Pahang (UMP), 20th - 22nd June 2009 at M. S. Garden Hotel, Kuantan, Pahang.This paper presents the possibility of generating electricity for a single house in Malaysia using solar updraft tower (SUT). This paper starts with the short introduction of SUT and the advantages. Then followed with a table that shows the temperature readings in Subang Jaya, Malaysia to support the idea that SUT can be a suitable way to generate electricity in Malaysia because of the weather conditions. After that, the design and construction of SUT and the components are presented. Finally the results that have been obtained on several experiments with the solar updraft tower are also presented

    Tebu produk rekayasa genetika SUT (PRG SUT) merupakan tebu overekspresi gen SoSUT1 yang diketahui mampu meningkatkan produksi Sucrose trasporter (SUT). SUT merupakan protein membrane yang berfungsi sebagai protein transporter sukrosa dari jaringan asimilasi ke jaringan penyimpanan. Peningkatan translokasi sukrosa harus di imbangi dengan peningkatan sintesis sukrosa di organ asimilasi. SoSPS1 merupakan gen pengkode Sucrose phosphate synthase yang merupakan enzim kunci dalam sintesis sukrosa di sitosol. Insersi gen SoSPS1 pada tebu Produk rekayasa genetika SUT dipandang perlu untuk mendapatkan tebu dengan sintesis sukrosa tinggi. Penelitian ini bertujuan untuk mendapatkan tanaman tebu produk rekayasa genetic SUT - SPS. Insersi gen SoSPS1menggunakan vektor Agrobacterium tumefaciens LBA4404 pembawa konstruk pKYS-SoSPS1. Parameter keberhasilan penelitian ini adalah terintegrasinya konstruk pKYS-SoSPS1 kedalam genon tanaman Tebu PRG SUT dengan uji ketahanan terhadap media seleksi antibiotik dan analisis PCR. Penelitian ini berhasil mendapatkan 10 tanaman tebu PRG SUT-SPS
