    The surface expression of the IFNGR1 and IFNGR2 was determined using flow cytometry in the TL-1 cell line compared to Jurkat T cells, an IFN-γ responsive cell line. Representative flow cytometry histograms of (A) IFNGR1 and (B) IFNGR2 and (C) median fluorescence intensity (MFI) of IFNGR1 and IFNGR2 in TL-1 and Jurkat T cells are shown.     Jurkat T cells (A) or IL-2-starved TL-1 cells (B) were treated with 5000 U/mL IFN-β or water (vehicle control) for the indicated time. TL-1 cells were also treated with 200 U/mL IL-2 as a positive control for induction of p-STAT1. p-STAT1, total STAT1, and β actin were measured using western blot. (C) Jurkat or TL-1 cells were treated with 5000 U/mL IFN-β or water for 6 h prior to RNA extraction. Induction of IFN-γ and STAT1 transcripts was quantified using qPCR.     Sensitivities for coherent elastic scattering of solar and supernova neutrinos with future NaI(Tl) dark matter search detectors of COSINE-200/1T

    We investigate the prospects for measuring the coherent elastic neutrino-nucleus scattering of solar and supernova neutrinos in future NaI(Tl) dark matter detection experiments. Considering the reduced background and improved light yield of the recently developed NaI(Tl) crystals, more than 3σ\sigma observation sensitivities of the supernova neutrino within the Milky Way are demonstrated. In the case of the solar neutrino, approximately 3 observations are marginal with a 1 ton NaI(Tl) experiment assuming an order of magnitude reduced background, five photoelectron thresholds, and 5-year data exposure

    Thallium-doped sodium iodide (NaI(Tl)) crystals are widely used in radiation detection applications, from gamma-ray spectroscopy to particle dark matter searches. However, if the crystal is exposed to relative humidity of even a few percent, its light emission degrades, making the crystal impractical as a detector. Surrounding the crystal with organic scintillators not only protects the surface of the crystal from humid air but also offers a new capability to tag backgrounds such as external gamma rays and surface contaminations. We developed a detector that is constructed by fully encasing a NaI(Tl) crystal in a plastic scintillator and then immersing the plastic-crystal assembly in liquid scintillator. Using data collected from this triple phoswich detector, a pulse shape analysis is able to identify the various radiation signals from the three scintillators. Additionally, we find that the crystal's emission quality is maintained for a month.Comment: 14pages,7figure

    Tangle-bearing neurons survive despite disruption of membrane integrity in a mouse model of tauopathy

    Neurofibrillary tangles (NFTs) are associated with neuronal loss and correlate with cognitive impairment in Alzheimer disease, but how NFTs relate to neuronal death is not clear. We studied cell death in Tg4510 mice that reversibly express P301L mutant human tau and accumulate NFTs using in vivo multiphoton imaging of neurofibrillary pathology, propidium iodide (PI) incorporation into cells, caspase activation, and DNA labeling. We first observed that in live mice, a minority of neurons were labeled with the caspase probe or with PI fluorescence. These markers of cell stress were localized in the same cells and appeared specifically within NFT-bearing neurons. Contrary to expectations, the PI-stained neurons did not die during a day of observation; the presence of Hoechst-positive nuclei in them on the subsequent day indicated that the NFT-associated membrane disruption, as suggested by PI staining, and caspase activation do not lead to immediate death of neurons in this tauopathy model. This unique combination of in vivo multiphoton imaging with markers of cell death and pathological alteration is a powerful tool for investigating neuronal damage associated with neurofibrillary pathology

    © 2022 IOP Publishing Ltd and Sissa Medialab.The scintillation characteristics of a thallium-doped sodium iodide (NaI(Tl)) crystal with dimensions of 0.6 cm× 0.6 cm× 2 cm were studied by attaching a silicon photomultiplier (SiPM) directly to the crystal over a temperature range of 93-300 K. The scintillation light output and decay time were measured by irradiating 59.54 keV γ-rays with a 241Am source. We observed an approximately 20% increase in the light yield at 230 K compared with that at room temperature. Under these conditions, NaI(Tl) crystals with SiPM readout can be suitable for future dark matter search detectors.11Nsciescopu

    Phosphorylation of SU(VAR)3-9 by the chromosomal kinase JIL-1

    The histone methyltransferase SU(VAR)3-9 plays an important role in the formation of heterochromatin within the eukaryotic nucleus. Several studies have shown that the formation of condensed chromatin is highly regulated during development, suggesting that SU(VAR)3-9's activity is regulated as well. However, no mechanism by which this may be achieved has been reported so far. As we and others had shown previously that the N-terminus of SU(VAR)3-9 plays an important role for its activity, we purified interaction partners from Drosophila embryo nuclear extract using as bait a GST fusion protein containing the SU(VAR)3-9 N-terminus. Among several other proteins known to bind Su(VAR)3-9 we isolated the chromosomal kinase JIL-1 as a strong interactor. We show that SU(VAR)3-9 is a substrate for JIL-1 in vitro as well as in vivo and map the site of phosphorylation. These findings may provide a molecular explanation for the observed genetic interaction between SU(VAR)3-9 and JIL-1

    Bu çalışmada, içsularımızda son yıllarda hızlı artış gösteren gümüş balığı (Atherina boyeri Risso, 1810) üretiminin Türkiye balıkçılığı ve ülke ekonomisine etkisi araştırılmıştır. Ülkemizde yakın tarihe kadar ekonomik öneme sahip olmayan ve değerlendirilmeyen bu tür, son yıllarda değer kazanmış iç tüketimin yanında başta AB ülkeleri olmak üzere dış piyasaya taze ve işlenmiş olarak sunulmuştur. Gümüş balığı Türkiye de avcılığı yapılan toplam içsu ürünleri üretiminin ilk üç sırasında yer almakta ve toplam avcılık üretiminin % 16.1’lik kısmını oluşturmaktadır. Gümüş balığı avcılığı yıllar itibariyle sürekli artış göstermekte ve toplam üretim miktarında yükselen bir trend görülmektedir. Üretim 1986 yılında 255 ton iken 2000 yılında 1.538 tona çıkmış günümüzde ise 2008 yılı verilerine göre 6.630 tona yükselmiştir. Bu üretim sonucu toplam iç su ürünleri üretiminin % 10,2’lik bölümü kadar (13.260.000 TL) ülke balıkçılığına ve ekonomisine katma değer sağlamıştır

    A Study of NaI(Tl) crystal encapsulation using organic scintillators for the dark matter search

However, the hygroscopicity of NaI(Tl) crystals makes the construction of crystal detectors in these experiments challenging. Therefore, the crystal requires a tight encapsulation to prevent from air contact. More importantly, in a low radioactivity measurement, identification of external radiations and surface contamination is crucial to characterize the origin of total crystal radioactivities. Studies for the NaI(Tl) crystal encapsulation with active organic scintillator vetoes have been performed to mitigate the above-mentioned issues simultaneously. A bare crystal is directly coupled with liquid and plastic scintillators to tag external radiations that penetrate from the outer part of the crystal.
