Caractérisation et modulation de la résistance au témozolomide chez le glioblastome multiforme
No full text"Le glioblastome multiforme (GBM) est la forme de tumeur cérébrale la plus commune et l'approche thérapeutique à ce jour implique une combinaison de chirurgie, radiothérapie et de la chimiothérapie avec le témozolomide (TMZ). De nombreux mécanismes contribuant à une résistance innée ou acquise à cet agent thérapeutique ont été identifiés et peuvent mener à un échec du traitement. Cette étude entreprend une approche métabolomique pour caractériser des profils métaboliques observés chez des cellules GMB sensibles ou résistantes au TMZ. Cette approche fut aussi utilisée pour explorer les changements métaboliques observés suite au traitement avec le TMZ et lomeguatrib, un inhibiteur de la MGMT avec un potentiel sensibilisateur au TMZ. Des métabolites ayant déjà été identifiés comme ayant des rôles dans la résistance à la chimiothérapie incluant le glucose, la citrate et l'isocitrate ont démontré des niveaux élevés chez les cellules résistantes au TMZ. De plus, une signature de métabolites comprenant l'alanine, la choline, la créatine et la choline phosphorylée a été identifiée comme élevée dans la lignée U373S suite à tous les traitements étudiés. Ces résultats présentent les profils métaboliques associés à la réponse au TMZ dans des modèles de lignées cellulaires GBM et proposent une suite intéressante qui pourrait influencer le développement d'outils thérapeutiques ou diagnostiques pouvant avoir un impact sur la réponse au TMZ ches les GBMs. Des analogues structuraux du lomeguatrib ont aussi été synthétisés afin de mieux comprendre le mécanisme d'inhibition et les paramètres structuraux nécessaires à une bonne inhibition de cette protéine. Malgré la variabilité de les GI₅₀ de ces molécules, trois d'entre elles semblent avoir un impact lors des traitements combinatoires avec le TMZ. Il concorso promosso dalla società dei Trasporti Pubblici Friburghesi (TPF) propone una riflessione sul futuro sviluppo del borgo di Chatel Saint Denis a partire dalle opportunità offerte dalla costruzione della nuova stazione. Il progetto presentato fa parte di una riflessione più generale sul tema della densificazione dei centri esistenti a ridosso delle linee di forza del trasporto pubblico: una strategia di trasformazione che nel territorio Svizzero ha prodotto recentemente diversi interventi nei centri urbani appoggiati alla fitta rete ferroviaria, ridefinendo la geografia e la gerarchia delle aree metropolitane alla scala terrritoriale. Patrick Procktor: Art and Life (Unicorn Press, 2010)
Keith Vaughan: The Mature Oils 1946-1977, (Sansom & Co., 2012)
The research is on two artists, Patrick Procktor (1936-2003), and Keith Vaughan (1912-1977). The monograph on Procktor – previously one of the least documented of the generation of artists who came to prominence in London in the Sixties – positions him in a history of art from which he had been notably absent. The research on Vaughan asserts a new reading of his work, one that is both deeper and more nuanced in its analysis of the ways in which personal experience and sexuality are encoded autobiographically within his work. Crucially, in both artists biography and work are symbiotically linked; the research therefore examines the links between life and art.
Revisionary in intent, the work examines trajectories of experience of gay British (or rather, English) artists in the twentieth century, artists who sought to express themselves and forge careers within the constraints of a heteronormative society, albeit one in which attitudes to sexuality were undergoing change. As gay men, both were constrained by the social mores of their times, and each used painting as a means to affirm personal and sexual identities. A key research interest is in the ways in which sexuality and persona are reflected in critical responses to the artist’s work: in Vaughan, Procktor and other gay male artists of the period. The writing on both Procktor and Vaughan examines the relationship between their personal and professional/artistic lives, framed within a broader socio-political and art historical context. It asserts the place of biography as a means to understand and form new readings of the work. The work adds substantially to the literature and wider discourse on post-war British painting and social history
