A rapid method of Sertoli cell isolation by DSA lectin, allowing mitotic analyses
We have developed a rapid and convenient method of Sertoli cell preparation for studying the growth kinetics of these cells in in vitro culture. Datura Stramonium agglutinin (DSA)-coated dishes were used to rapidly purify single Sertoli cells from immature rat testis. We have monitored by immunohistochemical markers the degree of contamination of our Sertoli cell preparation by other cell types. The cell preparation is essentially free of germ cells and interstitial cells and contains a minimal percentage of myoid cells. Sertoli cells isolated with this method retain functional activities such as the FSH responsiveness in terms of cAMP production. In addition, we have studied the proliferative activity of Sertoli cells isolated by lectin binding from rats of different ages. Sertoli cells exhibited a characteristic pattern of proliferation which was a function of the donor animal age. The proliferative activity of isolated Sertoli cells decreased with age, being much higher in 3 day-old rats than in older animals. A similar pattern was observed when the mitotic activity of Sertoli cells in response to mitogens present in the testicular extracts from 5 day-old rats was evaluated. The method described here reduces or eliminates many of the drawbacks of the conventional procedures used to isolate Sertoli cells, thus providing a useful tool in studies of growth kinetics and regulation of cell proliferation in vitro. Short communication: The influence of vermicompost based substrates on basil growth and nutrient content

The influence of vermicompost based growth substrates on basil (Ocimum basilicum L.) growth and nutrient content was assessed. Treatments were: 1. 30% vermicompost, peat, sand, dolomite stone; 2. 25% vermicompost, peat, gravel, perlite; 3. 25% vermicompost, peat, gravel, light gravel, 4. commercially available growth medium, or 5. 20% vermicompost and organic matter rich clay soil. Basil growth was best with treatments 1 and 2. Nutrient content of basil was best in treatment 3. The best growth substrate for basil production is treatment 1, based on growth results, while based on nutrient analysis when basil is used in medicine, the right treatment is 3: 25% vermicompost, peat, gravel, light gravel

The experiments in the greenhouse were carried out in the spring 2014 at the Estonian Crop Research Institute. Treatments: 1) stabilized silicic acid treatment; 2) control. Si treatment (2 mL L–1 of silicic acid): First spray, when 1 true leaf was present; second spray was two weeks after spray 1; third spray was two weeks after spray 2. The pH of spray solution was 5.5. pH of spray solution was 5.5. Control plants were treated with water. The plants were 26% higher in Si variant. The width of leaf lettuce was 32% larger in Si variant. In leaf lettuce the phosphorus content was 14% higher, the content of calcium was 32% higher, the content of magnesium was 12% higher in Si variant than in control

Supervisor: Poul Hansen This study focuses on looking at differences and similarities in single households and life satisfaction between The United Kingdom, Sweden and Germany. To interpret the differences, we are investigating the applicability of Esping-Andersens theory on important parameters in the different welfare regimes, the liberal, social democratic and conservative for the respective country. We find that there is poor statistical support for differences between Sweden and Germany. Instead, results demonstrated a convincing difference between The United Kingdom and the other two countries in number of single households and in relation to age and gender. We can’t say with certainty where these differences are derived from, but we discuss if it can be factors related to the British welfare regim

Little is known about the ontogeny of steroidogenic enzymes and activities of steroidogenic pathways in the hFK at early pregnancy. Our aim was to explore steroidogenesis and the expression of steroidogenic enzymes in the hFK at gestational weeks (GW) 9-12. Steroids in the hFK were analyzed by gas chromatography/coupled to tandem mass spectrometry. The expression of steroidogenic enzymes in the hFK at GW 9-12 was investigated by qPCR, automated Western blotting and immunohistochemistry. We observed that the hFK produced substantial amount of steroids of the Delta(5) and Delta(4) pathways and several steroid precursors in the biosynthesis of DHT via the backdoor pathway but not DHT itself. The levels of steroids and expression of relevant steroidogenic enzymes (e.g., CYP17A1, HSD3B1, HSD3B2, CYP11B1 and AKR1C4) we significantly higher in the hFK at GW11-12 compared to GW9. We also found the expression of sex steroid receptors (e.g., AR, ER alpha and ER beta) in the hFK at GW9-12. No sex dependent differences in the levels of all identified steroids and expression of steroidogenic enzymes in the hFK from male and female fetuses were found. Altogether, our data indicate that the hFK at early pregnancy is steroidogenic organ with potential to synthesize multiple steroids that may play an important role in the formation and development of this organ in humans

nutrients, vitamins, growth hormones, enzymes such as proteases, amylases, lipase, cellulase and chitinase and immobilized microflora. The enzymes continue to disintegrate organic matter even after they have been ejected from the worms. Reduced use of water for irrigation, reduced pest attack, reduced termite attack, reduced weed growth; faster rate of seed germination and rapid seedlings growth and development; greater numbers of fruits per plant (in vegetable crops) and greater numbers of seeds per year (in cereal crops) are only some of the beneficial effects of the vermicompost usage in agricultural production. Earthworms and vermicompost can boost horticultural production without agrochemicals. In spite of the benefits associated with vermicompost, its use is not widespread yet. Short communication: The improvement of the quality, and nutrient content of leaf celery transplants by effective microorganisms

The purpose was to assess the influence of EM on the growth and nutrient content of leaf celery transplants. Two treatments were compared: 1) EM treatment comprised of seed soaking in EM 1:500 solution, sowing in EM treated peat and watering the plants weekly with EM solution. 2) Control treatment included similar treatments with water. Leaf celery transplants grown with EM were significantly shorter and had thicker stems than those grown without EM. The contents of nitrogen, phosphorus, potassium and calcium increased in transplants grown with EM compared to those grown without EM. The contents of nitrates and magnesium did not show statistically significant differences. Conclusion: EM improves the growth and increases nitrogen, phosphorus, potassium and calcium contents of leaf celery transplants
