The Forsterite-Anorthite-Albite system at 5 kb pressure
This study looked at three components of a typical syenite composition: forsterite (Mg2SiO4), anorthite (CaAl2SiO8), and albite (NaAlSi3O8). Various proportions of forsterite and either a pure albite, An48, or An66 feldspar composition were mixed together to simulate a number of possible parent magma compositions. The compositional evolution of these parent magmas was experimentally investigated using a piston-cylinder apparatus in order to simulate the conditions at 5 kbar pressure and across a range of possible temperatures. After holding the sample for 2-4 hours at the desired temperature and pressure in the piston-cylinder, each sample composition was polished, carbon coated, and analyzed using a scanning electron microscope. Glass and mineral compositions were then plotted on a Fo-An-Ab ternary diagram to better constrain the cotectic at 5 kbar. One of the main purposes of this study was to constrain the influence of Mg-rich mafic minerals (such as forsterite, the Mg end-member of olivine) on feldspar compositions at 5 kb pressure. Plotting the compositions of each sample mixture resulted in some interesting trends. Feldspar-rich samples that qualitatively appear to be 100% glass do not plot on the expected bulk compositions. Instead, there is a distinct Na-rich nature to these glasses. As expected, any feldspar crystals that formed had a composition richer in Ca than that of the melt, resulting in glass compositions that are richer in Na and plot to the left of expected bulk compositions for that glass. Not all crystals that were seen compositionally in this way were physically evident on the SEM as Ca-rich crystals settled to the bottom of the less dense, Mg-rich magma. As these crystals were separated iii by only a few millimeters from the top of the graphite chamber, the bulk composition of the system as a whole remained constant. Differences exist between this study\u27s Fo-An-Ab ternary diagram at 5 kbar with the Fa-An-Ab system at the same pressure by Morse et al. (2007). The addition of a more Mg-rich mafic mineral (forsterite) in place of fayalite pushed the cotectic closer to the feldspar join. Consequentially, the forsterite-saturated plagioclase compositions can exist across a wider temperature range (approximately 250°C) than fayalite-saturated plagioclase (27°C) (Morse et al., 2007)

PDBe-KB: collaboratively defining the biological context of structural data
The Protein Data Bank in Europe – Knowledge Base (PDBe-KB, https://pdbe-kb.org) is an open collaboration between world-leading specialist data resources contributing functional and biophysical annotations derived from or relevant to the Protein Data Bank (PDB). The goal of PDBe-KB is to place macromolecular structure data in their biological context by developing standardised data exchange formats and integrating functional annotations from the contributing partner resources into a knowledge graph that can provide valuable biological insights. Since we described PDBe-KB in 2019, there have been significant improvements in the variety of available annotation data sets and user functionality. Here, we provide an overview of the consortium, highlighting the addition of annotations such as predicted covalent binders, phosphorylation sites, effects of mutations on the protein structure and energetic local frustration. Identification of complex biological network classes using extended correlation analysis
Modeling and analysis of complex biological networks necessitates suitable handling of data on a parallel scale. Using the IkB-NF-kB pathway model and a basis of sensitivity analysis, analytic methods are presented, extending correlation from the network kinetic reaction rates to that of the rate reactions. Alignment of correlated processed components, vastly outperforming correlation of the data source, advanced sets of biological classes possessing similar network activities. Additional construction generated a naturally structured, cardinally based system for component-specific investigation. The computationally driven procedures are described, with results demonstrating viability as mechanisms useful for fundamental oscillatory network activity investigation
Efeito citotóxico do óxido nítrico e do taxol em células linfoblásticas leucêmicas: envolvimento do fator de transcrição NF-kB e papel da glutationa

Programa de Pós-Graduação em Farmacologia.O presente trabalho avaliou, através de técnicas moleculares, bioquímicas e farmacológicas, o efeito citotóxico do óxido nítrico (NO) e do taxol sobre células linfoblásticas leucêmicas. Foi investigado o envolvimento do NF-kB e a importância da glutationa na citotoxicidade causada pelo NO e pelo taxol. Os resultados obtidos, demonstraram que o efeito citotóxico do NO e do taxol sobre células leucêmicas linfoblásticas parecem atuar em vias muito semelhantes, ou seja, ambos apresentaram ações independentes de alterações no citoesqueleto, inibiram a atividade do NF-kB e depletaram os níveis de glutationa (GSH). Além disso, quando associados, apresentaram efeito citotóxico sinérgico. Sendo assim, concluímos que o NO e o taxol, além de diminuírem os níveis de glutationa intracelular, o que sensibiliza as células aos danos oxidativos e nitrosativos, também impedem o restabelecimento dos níveis normais de GSH intracelular, através da inibição da atividade do NF-kB
Intergranular Diffusion in Metamorphic Rocks
Theoretical models of intergranular diffusion demonstrate that grain size and grain boundary width are the most important textural parameters influencing diffusion in metamorphic rocks. Taking diffusion in an aqueous solution as a limiting case for diffusion in the intergranular region of metamorphic rocks, it appears that the diffusivities of many species will be similar and relatively insensitive to variations in temperature and pressure over a wide range of metamorphic conditions. Observed variations in the mobility of chemical components during metamorphism may be related principally to variations in the ratio of the component density in the intergranular region to the component density in the minerals. Experimental studies of the growth of forsterite reaction zones around periclase crystals at 650° and 700°C and I kb water pressure provide an upper limit to the effective diffusivity of silica through polycrystalline forsterite: DSiO2 ~ I0- 9/a cm 2/s where a is the grain size in microns. This maximum diffusivity can be used to obtain minimum times for the growth of reaction zones in metamorphic rocks
Epstein-Barr virus latent proteins regulate expression of the anti-apoptotic cellular bfl-1 gene
The ubiquitous and oncogenic human herpes-virus Epstem-Barr virus (EB V) establishes a latent infection and promotes the long-term survival of the infected host cell by targeting the molecular machinery that controls cell fate decisions, including apoptosis, proliferation and differentiation. These host-virus interactions are likely to play a crucial role in the development of EBV-associated malignancies such, as Burkitt’s lymphoma, Hodgkin’s disease, nasopharyngeal carcinoma and tumours in lmmunosuppressed individuals. It has previously been shown in our laboratory that two EBV latent proteins, latent membrane protein 1 (LMP1) and EBV nuclear antigen 2 (EBNA2), which are major effectors of cellular phenotypic change, can independently regulate expression of the cellular bfl-1 gene Bfl-1 is an anti-apoptotic protein of the Bcl-2 family, whose preferential expression in hematopoietic and endothelial cells is controlled by inflammatory stimuli. In this study, it is reported that LMP1 and EBNA2 regulate bfl-1 activity through interactions with components of the NF-kB and Notch signalling pathways respectively NF-kB composed of p65 sub-units trans-activated the bfl-1 promoter m the EBV-negative cell line DG75, and an NF-icB-like binding site at position -52 to -43 relative to the transcription start site was essential for this effect. An RBP-Jk/CBF1 mutant blocked EBNA2-mediated trans-activation of bfl-1 in DG75 cells, indicating an important role for this DNA-binding protein in bfl-1 trans-activation by EBNA2. Although RBP-Jk/CBFI is also essential for signalling by the cellular equivalent of EBNA2, mtra-cellular Notch (NotchIC), this protein was not found to trans-activate the bfl-1 promoter. Both EBNA2 and LMP1 are expressed in EBVmfected cell lines, and EBNA2 is responsible for induction of LMP1 Blocking of either EBNA2- or LMP1-mediated signalling in EBV-mfected cell lines did not dramatically affect the level of bfl-1 promoter activity. However, when both EBNA2 and LMP1 signalling were blocked simultaneously, a significant decrease in the level of bfl-1 activity was observed. These data indicate a role for both EBNA2 and LMP1 in the regulation of the promoter for the bfl-1 gene m the context of the EBV-infected cell. These findings are relevant to our understanding of EBV persistence in the infected host, and its role in malignant disease
Structural analysis of Precambrian mylonite zones, Henry\u27s Lake Mountains, southwest Montana and Idaho
Mylonites, protomylonites, and cataclasites occur in thin, widely spaced, NE trending, NW dipping, SE verging (Fig. 5 - 7) shear zones in the Henrys Lake Mountains, the field area for the 2011 Montana, Big Sky Keck Project. Differences in protolith composition are interpreted to control position of shear zones and textural differences between mylonites, protomylonites, and cataclasites. Cataclasites likely have the highest concentrations of retrograde minerals because their highly fractured texture provided a conduit for water flow. Symplectite grew during shear, potentially as a means for volume reduction (Simpson and Wintsch, 1989). Mineral assemblages typical of greenschist facies and phengite equilibrium with k-feldspar, quartz and phlogopite suggests a minimum pressure during shear of 5 kb (0.5 GPa). As shear zones in the Henrys Lake Mountains are found to be the along strike equivalents of the 1.8 Ga Madison mylonite zone in the Southern Madison Range (Erslev and Sutter, 1990), it is believed that they are the result of shearing as part of a foreland thrust zone inboard of a major compressional orogen at 1.8 Ga (Erslev and Sutter, 1990). It is suggested that this event was the Big Sky Orogeny
The formation of large garnets at Gore Mountain, New York : experimental partial melting of amphibolite and meta-gabbro
This study examined the hypothesis that partial melting aided the growth of large garnets in the Barton Deposit of Gore Mountain, New York. Samples of meta-gabbro and garnet amphibolite from Gore Mountain were ground into powders. The powders were then subjected to temperatures and pressures resembling the peak conditions of the Adirondack Highlands (800°C and 8 kb) using a piston cylinder press. Water was added to half of the samples to determine if an influx of water had an influence on the melting or garnet growth. After each experiment, the nickel sample holder was mounted in epoxy and ground to expose the contents of the run products. Melt was not identified within the experimental samples. The results give no evidence that partial melting had an effect on garnet growth, as melt did not appear to form readily under the peak temperature conditions. Additionally, the results of this study show that the simple addition of water did not convert the meta-gabbro into the garnet amphibolite ore. The process must be more complex than purely hydrating the meta-gabbro to become amphibolite as garnets of the hydrated meta-gabbros differed in composition from garnets of the Gore Mountain garnet amphibolite
