ENPP1 gene, insulin resistance and related clinical outcomes

    No full text
Purpose of review Insulin resitance plays a significant role in both morbidity and mortality of the general population. Understanding the molecular mechanisms of insulin resistance would help the identification of at-risk individuals in the presymptomatic stage, and the discovery of novel and more effective treatments. The transmemebrane glycoprotein ectonucleotide pyrophosphates phosphodiesterse 1 (ENPP1) inhibits insulin receptor signalling and has recently emerged as a key player in the development of insulin resistance. This review will summarize data available on the relationship between ENPP1 and insulin resistance. Recent findings Overexpression of ENPP1 in insulin target tissues is an early, intrinsic defect observed in human insulin resitance. A missense ENPP1 single nucleotide polymorphism, K121Q, has been recently described with the Q121 variant being a stronger inhibitor than K121 of insulin receptor function. In addition, the Q121 variant has been repeatedly associated with insulin resitance and related abnormalities including body weight changes, type 2 diabetes and macrovascular complications, thus suggesting a pleiotropic role of the ENPP1 gene on several metabolic abnormalities. Summary A deep undestanding of ENPP1 mode of action and the mechanisms regulating its expression and function are likely to provide new tools for early identification and treatments of patients at risk for the devastating clinical outcomes related to insulin resistance

Role of relationship between HbA1c, fibrinogen and HDL-cholesterol on cardiovascular disease in patients with type 2 diabetes mellitus

Objective: To assess the impact of relationship between glycated hemoglobin (HbA1c), fibrinogen and HDL-cholesterol (HDL-c) on cardiovascular disease in type 2 diabetes. Methods: We investigated i) the relationship of HbA1c, fibrinogen and HDL-c in 375 coronary artery disease (CAD)-negative and 320 CAD-positive diabetic patients and ii) the association between clustering of these three factors and incident major cardiovascular events in 317/320 CAD-positive patients. Results: i) The relationships between HbA1c and both fibrinogen and HDL-c and between HDL-c and fibrinogen were significant only in CAD-positive patients (β = 10.655, p = 0.002; β = -1.056, p = 0.013; β = -1.751, p = 0.000008, respectively); ii) patients with worse-than-median levels of the three factors showed higher risk for major cardiovascular events than others (HR: 2.22, 95%CI = 1.23-4.02, p = 0.008). Both findings were independent of LDL-c, blood pressure or ongoing therapies. Conclusion: Our findings suggest interwoven actions of poor glycemic control, low grade inflammation and low HDL-c on atherosclerotic processes in type 2 diabetes. © 2013 Elsevier Ireland Ltd

The aim of this article is to define the cultural profile of Luigi Bacci: Spanish teacher, author of a large number of Hispanic studies and protagonist of many cultural initiatives to strengthen ties between Italy and Spain. The study of his relationships with politicians, the approach to some of his works and the study of documents from various archives have enabled us to approach one of those agents in cultural relations that have an incredible passion for the other country.

Book reviewe: Michele Bacci, "The Many Faces of Christ. Portraying the Holy in the East and West, 300 to 1300"

    No full text
This new book by Michel Bacci is dedicated to the image of Christ, and more specifically to how existing visual tradition relates to the issue of the authentic face of Jesus. It explores how an iconography that was considered typical for centuries came into being and the various strategies underlying this process. The study demonstrates that the iconic representation of Christ portrayed as a handsome young man with a beard and long hair was neither the first nor the only option considered by Late Antique artists and medieval churchmen for the image of Jesus. Hence, the author of The Many Faces of Christ challenges the common belief that the appearance and physiognomic traits of divine figures were from the outset fixed in art and canonized by tradition. As a consequence, the standard narrative of established long-lasting patterns and singular archetypal models is substituted in the book by a story of change and experimentation in which Christian phenomena at times find direct counterparts in other religious traditions
