    Energieversorgung der Stadtwerke Roth

    Die vorliegende Projektarbeit ist Teil des Gesamtprojektes, welches die regenerative Energieversorgung der Stadtwerke Roth mittels Solar, Wind, Wasserkraft und mit Biogas befeuerten Blockheizkraftwerken sicherstellt. Dabei wird eine Versorgungssystemauslegung mit zugehöriger Speicherdimensionierung durchgeführt. In dieser Arbeit werden die Windkraft und eine Systembewirtschaftungsstrategie genauer beleuchtet. Bei der Standortermittlung für Windenergieanlagen wurde der Suchradius auf den Landkreis Roth ausgeweitet, da im Bereich Stadt Roth keine Möglichkeit für Neuanlagen besteht. Hierbei konnten mögliche Standorte für Windenergieanlagen ermittelt werden. Ob der Standort für eine Windenergieanlage zugelassen ist, wird im Genehmigungsverfahren entschieden. Die durchschnittlichen Windgeschwindigkeiten betragen hier ca. 6,7 m/s. Bei der zukünftigen Energieversorgung werden Neuanlagen im Bereich Windkraft und Photovoltaik benötigt. 92 % dieser übernehmen Windkraftanlagen, 8% übernehmen Photovoltaikanlagen. Die Windkraftanlagen haben hierbei eine Nabenhöhe von 160 m, eine Generatornennleistung von 3 MW und einen Rotordurchmesser von 150 m. Es werden elf Neuanlagen benötigt. Im Gesamtsystem entstehen Stromgestehungskosten von 20,39 ct/kWh. Die Systembewirtschaftungsstrategie errechnet mit Wetterprognosen und Strommarktpreisen eine lukrative Ein- und Verkaufsstrategie für die im Speichersystem gespeicherte Energie. Hierbei ist die Versorgungs-sicherheit der Stadtwerke Roth stets gegeben. Bei einem Strommarktpreis von 150 €/MWh ergäben sich durch die strategische Vermarktung reine Stromgestehungskosten von 7,8 ct/kWh

    Abstract 1917: CHD1 depletion leads to androgen-independent growth of prostate cancer cells lines and regulates the TGFβ system in an EZH2-dependent manner

    Abstract Introduction: Therapy of metastasized prostate cancer (PCA) largely depends on androgen deprivation therapy (ADT). Despite initial response, tumors inevitably reach castration resistance (CRPC), a stage at which chemotherapy and second-generation antiandrogens (e.g. Enzalutamide) represent the main therapeutic options. Acquired resistance to these drugs is an emerging problem, raising a need for new approaches. Deletion or mutation of the Chromodomain Helicase DNA binding protein 1 (CHD1) gene, encoding an ATP-dependent chromatin remodeler, is more prevalent in CRPC compared to earlier tumor stages. Therefore, we investigated the effects of CHD1 depletion in androgen-sensitive LNCaP cells. Experimental procedures: LNCaP cells were grown in media with either 10% fetal bovine serum (FBS), 10% charcoal-stripped serum (CSS) or 10% CSS plus 1nM testosterone. Growth was visualized by image-based cell cytometry. After 10 days of growth, RNA sequencing (RNA-seq) was performed to investigate changes in gene expression (between the different growth conditions with and without CHD1 depletion). Results were verified by qRT-PCR and Western blot. The identified treatment options were tested with small-molecule inhibitors in different concentrations. Finally, we performed chromatin immunoprecipitation followed by next-generation sequencing (ChIP-seq) in order to identify the epigenetic mechanisms underlying the differential regulation. Results: We identified a correlation between androgen-independent growth and CHD1 depletion which could be compensated by addition of 1 nM testosterone. Moreover, we identified CHD1-dependent regulation of Enhancer Of Zeste Homolog-2 (EZH2), which could be confirmed at the protein level. Treatment with 1 µM JQ-EZ (an EZH2 inhibitor) showed a stronger effect on the proliferation of CHD1-knockdown cells compared to controls. ChIP-seq and RNA-seq analysis helped to identify a subset of EZH2-bound genes depending on CHD1 knockdown and androgen deprivation. Among those genes, we identified the I-SMAD genes SMAD6 and SMAD7 which are regulators of TGFβ and BMP signaling to be upregulated after androgen-deprivation and downregulated with CHD1 knockdown. We also showed differential regulation of PMEPA1, a TGFβ-dependent inhibitor of PCA bone metastasis. Coherently, we found upregulation of mesenchymal markers such as CDH2, pointing towards a role in EMT. Conclusion: We identified a new regulatory mechanism for CRPC harboring a CHD1-deletion leading to a more mesenchymal state in an EZH2-dependent manner. First experiments with an EZH2 inhibitor in our system indicate a possible therapeutic use.     This dissertation explores the utilization of photographic archives in the realm of contemporary art photography, with specific attention directed towards the long-term archival art projects conducted by two American conceptual artists, Taryn Simon (b. 1975) and Evan Roth (b. 1978). Centering on these two case studies that bridge the divide between physical and digital archival contexts, this analysis aims to examine the manner in which Simon and Roth navigate a spectrum of photographic and archival formats, materials, and conceptual frameworks in both tangible and virtual domains. My argument, based in a monographic approach, is firmly anchored in the selection of Simon and Roth as central figures of study. These artists skillfully harness the concept of the photo-archive not only to contemplate memory and historical narratives but also to expand the parameters of contemporary art photography. In doing so, they make substantial contributions to the ongoing discourse surrounding "post-photography." This becomes particularly relevant in an era characterized by the ubiquity of photographic imagery, driven by the pervasive influence of digital transformations on our daily lives. This dissertation aims to elucidate how the artists' unique methodologies in curating and presenting photo-archives as contemporary art result in distinctive visual and performative experiences for viewers in exhibition contexts. Through this analysis, it explores the experiential dimension of photography within the framework of Simon and Roth's creative works and exhibitions. It is my contention that Simon and Roth's artistic projects serve a dual purpose, effectively engaging audiences through aesthetically compelling photographic compositions and elaborate site-specific installations while also provoking intellectual and critical contemplation. Their projects prompt reflection on contemporary aspects of our image-centric culture, spanning visual knowledge creation, categorization, preservation, and distribution, as well as the shaping of collective memory and identity through photographic archives. Additionally, their artworks explore the material and immaterial aspects of photography and archives, illuminating these dynamics within our visual culture. This dissertation examines selected archival artworks by Simon and Roth in the context of their curatorial and institutional settings. It emphasizes the significant impact of these factors on the audience's reception of the works and concludes that these institutional and curatorial contexts greatly contribute to our understanding of the multifaceted expressions and possibilities of photography within contemporary art practice.Cette thèse explore l'utilisation des archives photographiques dans le domaine de la photographie contemporaine, en accordant une attention particulière aux projets d'art archivistique à long terme menés par deux artistes conceptuels américains, Taryn Simon (née en 1975) et Evan Roth (né en 1978). Elle examine la dimension expérientielle de la photographie dans les oeuvres créatives et les expositions de Simon et Roth. Leurs projets engagent le public à travers des compositions photographiques esthétiquement captivantes et des installations élaborées tout en suscitant une réflexion intellectuelle et critique. Leurs projets incitent à la réflexion sur les aspects contemporains de notre culture centrée sur l'image, couvrant la création, la catégorisation, la préservation et la distribution des connaissances visuelles, ainsi que la formation de la mémoire collective et de l'identité à travers les archives photographiques. De plus, leurs oeuvres d'art explorent les aspects matériels et immatériels de la photographie et des archives, éclairant ainsi ces dynamiques au sein de notre culture visuelle. Cette thèse examine en détail certaines oeuvres sélectionnées de Simon et Roth dans le contexte de leurs cadres muséographique et institutionnel.     The Name Simon

    Speculation about the “change” in the name of the apostle Peter will undoubtedly always be in order. In this respect the recent article of Cecil Roth of Oxford raises an interesting point. He suggests that the apostle's name Peter prevailed in time over Simon because of a current tendency of contemporary Judaism to avoid the use of the name Σίμων or װעמש. The latter name was “commonly or even methodically modified or eliminated, for some reason or the other, among the Jews at the beginning of the Christian era. One finds it difficult to avoid the conclusion that the modification of the name of the Apostle by the elimination of ‘Simon’ was connected with this and due to the same cause, whatever that may have been.” Roth offers parallels of persons whose name was Simon, but who were known more usually by a patronymic or a nickname (ben Sira, ben Zoma, ben Azzai, ben Nanos, bar Kochba), and suggests that it was a peculiarly “patriotic” name, borne by great national and revolutionary leaders such as Simon Maccabee, Simon the High Priest (Sir 50.1–21), Simeon ben Šeṭaḥ (politician-Rabbi of the second century B. C), Simon the rebel (Josephus, JW 2.4,2 #57; Ant. 17.10,6 #273), Simon the son of the founder of the Zealots, Judah the Galilean (Josephus, Ant. 20.5,2 #102), Simon bar Giora (leader in the First Revolt), Simon bar Kochba (leader of the Second Revolt).</jats:p
