    © 2021, The Author(s), under exclusive licence to Springer Nature Limited.The use of prime editing—a gene-editing technique that induces small genetic changes without the need for donor DNA and without causing double strand breaks—to correct pathogenic mutations and phenotypes needs to be tested in animal models of human genetic diseases. Here we report the use of prime editors 2 and 3, delivered by hydrodynamic injection, in mice with the genetic liver disease hereditary tyrosinemia, and of prime editor 2, delivered by an adeno-associated virus vector, in mice with the genetic eye disease Leber congenital amaurosis. For each pathogenic mutation, we identified an optimal prime-editing guide RNA by using cells transduced with lentiviral libraries of guide-RNA-encoding sequences paired with the corresponding target sequences. The prime editors precisely corrected the disease-causing mutations and led to the amelioration of the disease phenotypes in the mice, without detectable off-target edits.     The present invention discloses a wide band optical amplifier with a common mid-stage device. In the wide band optical amplifier according to the present invention, the signals that enter the amplifier are first split into a couple of sub-band signals. The sub-band signals are amplified independently at different optical amplification sections, but they share a common mid-stage device, which reduces the complexity and the cost of the amplifier. The sub-band signals propagate count-directionally in the common mid-stages device to reduce the nonlinear interactions between the signals in different sub-bands. A wide band optical amplifier incorporating a common mid-stage device with directionality is also disclosed

    Myocardial infarction is diagnosed when blood levels of biomarkers are increased in the clinical setting of acute myocardial ischemia. Among the biomarkers, troponin I is the preferred biomarker indicative of myocardial necrosis. It is tissue specific for the heart. Myocardial infarction is rarely reported following seizure. We report a case of elevated troponin I in a patient after an episode of generalized tonic-clonic seizure. The diagnosis was type 2 myocardial infarction

    Various efforts to overcome the current crisis of shipbuilding industry have been being made in every area, including development, administration, and customer management. For overcoming the crisis, this study proposes a methodology to reconsider the supply management competency, which consists of production, procurement, and logistics, by the virtualization of Plan Do See (PDS) cycle using simulation technology and introduces a case study to verify the proposed methodology. The keys to the methodology are a comprehensive simulation framework including systematic analysis for the index investigated through simulation and a simulation platform for constructing a smooth input/output environment by avoiding dependencies on existing solutions.
