    La tesi ha ad oggetto l’istituto del risarcimento punitivo, del quale indaga natura, presupposti applicativi e profili funzionali nell’ambito degli ordinamenti di common law, con particolare riferimento all’ordinamento inglese e a quello statunitense. La parte finale del lavoro si occupa di individuare, in chiave comparatistica, punti di collegamento con le esperienze dei sistemi di responsabilità civile continentali, tentando di avviare una rilettura degli stessi in chiave polifunzionale ed assumendo, a tal fine, come banco di prova il risarcimento del danno non patrimoniale.The thesis deals with punitive damages, investigating their nature, applicative requirements and functions, among common law jurisdictions, with particular reference to England and U.S. law. The final part of the work aims at identifying, in a comparative light, connection points with the law of torts of the European jurisdictions, proposing a multi-functional framework and using, for this purpose, damages for non-pecuniary losses as a test case

    Abstract: A solid-phase assembly of model peptides derived from human parathyroid. hormone-related protein (11-19) containing omega-azido- and omega-yl-alpha-amino acid residues in positions i and i+4 was cyclised in solution by an intramolecular Cu-I-catalyzed azide-alkyne 1,3-dipolar Huisgen cycloaddition These series of heterodetic cyclo-nonapeptides varied in the size of the disubstituted 1,2,3-triazolyl-containing bridge, the location and the orientation of the 1,2,3-triazolyl moiety within the bridge The 1,2,3-triazolyl moiety, presented at either 1,4- or 4,1-orientation, is flanked by side chains containing 1-4 CH2 groups that result in bridges comprised from 4-7 CH2 groups connecting residues 13 and 17 Comprehensive conformational analysis employing CD, NMR and molecular dynamics reveals the conformational propensities of these heterodetic cvclo-nonapeptides Cyclo-nonapeptides containing either the 7 methylene bridge (VII and VIII) or the 4 methylene bridge (II) are unstructured in structure-promoting solvent Cyclo-nonapeptide I in which the 1,4-disubstituted 1,2,3-triazolyl is flanked by 3 and 1 CH2 groups in proximity to the respective residues 13 and 17, is stabilized in a non-canonical structure. All the other heterodetic cyclo-nonapeptides (III-VI) in which the 1,2,3-triazolyl is flanked by a total of 5 or 6 CH2 groups nicely accommodate alpha-helical structures and reproduce very closely the helical structure stabilized by the analogous cyclo-nonapeptide in which Lys(13) and Asp(17) are bridged by the isosteric lactam These studies suggest that the bioorthogonal i-to-(i+4) side-chain-to-side-chain cyclization via the prototypic "click reaction" offers a new and powerful approach for generating stable helix mimetic structures

    Stacking Interactions between Adenines in Oxidized Oligonucleotides

    The effects of stacking interactions on the oxidation potentials of single strand oligonucleotides containing up to four consecutive adenines, alternated with thymines and cytosines in different sequences and ratios, have been determined by means of differential pulse voltammetry. Voltammetric measurements point toward the establishment in solution of structured oligonucleotide conformations, in which the nucleobases are well stacked altogether. Molecular dynamics simulations confirm that finding, indicating that single strands assume geometrical parameters characteristic of the B-DNA form. The analysis of the voltammetric signals in terms of a simple effective tight binding quantum model leads one to infer a robust set of parameters for treating hole transfer in one-electron-oxidized DNA containing adenines and thymines

    Hemopressin, a bioactive nonapeptide derived from the α1 chain of hemoglobin, was recently shown to possess selective antagonist activity at the cannabinoid CB(1) receptor [Heimann, A. S., et al. (2007) Proc. Natl. Acad. Sci. U.S.A. 104, 20588-20593]. CB(1) receptor antagonists have been extensively studied for their possible therapeutic use in the treatment of obesity, drug abuse, and heroin addiction. In particular, many compounds acting as CB(1) receptor antagonists have been synthesized and subjected to experiments as possible anti-obesity drugs, but their therapeutic application is still complicated by important side effects. Using circular dichroism and nuclear magnetic resonance spectroscopy, this work reports the conformational analysis of hemopressin and its truncated, biologically active fragment hemopressin(1-6). The binding modes of both hemopressin and hemopressin(1-6) are investigated by molecular docking calculations. Our conformational data indicate that regular turn structures in the central portion of hemopressin and hemopressin(1-6) are critical for an effective interaction with the receptor. The results of molecular docking calculations, indicating similarities and differences in comparison to the most accepted CB(1) pharmacophore model, suggest the possibility of new chemical scaffolds for the design of new CB(1) antagonist lead compounds
