Multiple outcomes for PI3K/Akt/mTOR targeting in non-Hodgkin lymphoma
Wachstumsfaktor bedingte Aktivierung des PI3K/Akt/mTOR Signalweg wirkt positiv auf Vermehrung und Überleben. Konstitutive Aktivierung des Signalweges in NHL ist jedoch an Tumorprogression und Therapieresistenz beteiligt. Am Zelllinienmodell wurden zwei mögliche Therapiestrategien der PI3K/Akt/mTOR Inhibition erprobt, PI3K Inhibition mit BKM120 und horizontale Kombination von Zytostatika mit PI3K/Akt/mTOR Inhibitoren Erstens, BKM120 hat Antitumoraktivität in NHL und induziert Zelltod. Auf molekularer Ebene führt BKM120 vermittelte Dephosphorylierung von CDK1 an Y15 zur Aktivierung des M-Phase Komplex CDK1/Zyklin B und Eintritt in die Mitose. Parallel erlaubt die Degradation von Zyklin A und Hochregulation von Zyklin B Progression bis zur Metaphase, hemmt jedoch die Transition in die Anaphase. Anhaltender Metaphasearrest bewirkt programmierten Zelltod über den intrinsischen Signalweg der Apoptose durch Hochregulation der BH3-onlys Puma und Hrk, Aktivierung von Bax/Bak und proteolytische Spaltung von Caspase 9. Verlust von Bax/Bak oder Caspase Inhibition schützt vor BKM120 vermitteltem Zelltod. Bax/Bak defiziente Zellen, welche zusätzlich p53 Mutationen aufweisen, werden polyploid. Die Polyploidie ist ATM-MEK1/2 abhängig und kann mit Caffeine oder U0126blockiert werden. Zur Vermeidung von Polyploidie bedingter Tumorprogression, sollte BKM120 nur in Verbindung mit MAPK/ATM Inhibitoren verwendet werden. Zweitens. Horizontale Kombination PI3K/Akt/mTOR Inhibitoren mit cytotoxischen Substanzen schützt vor Apoptose. Der Schutzeffekt tritt auschließlich bei niedrigen Konzentration auf und ist unabhängig von der Art des Inhibitors bzw. Ebene der Inhibition. Das Onkogen und NFkB Target Pim-2 ist möglicherweise am Schutzmechanismus beteiligt. Durch die PI3K/Akt/mTOR vermittelte Pim-2 Regulation ergibt sich eine neue Rückkopplungsschleife. Im Fazit erschwert die Komplexizität des PI3K/Akt/mTOR Signalweges die Etablierung von Therapien.Growth factor mediated activation of the PI3K/Akt/mTOR pathway positively regulates proliferation and survival. Constitutive activation in NHL, however, is correlated with tumor progression and therapeutic resistance. Therefore, two possible strategies were tested in a cell line model system, Inhibition of PI3K with BKM120 and PI3K/Akt/mTOR Inhibition in addition to cytostatic drug administration. First, it is demonstrated that the pan PI3K inhibitor BKM120 has antitumor activity in NHL and induces cell death. On molecular level, BKM120 mediated dephosphorylation of CDK1 on Y15 causes activation of the M-phase complex CDK1/Cyclin B and entry into mitosis. In parallel, degradation of Cyclin A and Upregulation of Cyclin B enables progression into metaphase but inhibits transition into anaphase. Prolonged metaphase arrest induces programmed cell death via the intrinsic apoptosis pathway by upregulation of the BH3-onlys Puma and Hrk, activation of Bax/Bak and proteolytic cleavage of caspase-9. Loss of Bax/Bak or caspase inhibition protects from BKM120 induced apoptosis. Bax/Bak deficient cells with additional p53 mutation become polyploid. This polyploidy is ATM-MEK1/2 dependent and can be blocked with Caffeine or U0126. To prevent polyploidy related tumor progression, BKM120 should administered only in combination with ATM or MEK inhibitors. Second, combination of PI3K/Akt/mTOR inhibitors with cytotoxic agents protects from apoptosis. The protective effect is only detectable with low PI3K/Akt/mTOR inhibitor concentrations and independent of inhibitor type or cascade level. The oncogene and NFkB target is possibly involved in apoptosis protection and inhibition of NFkB neutralizes the protective effect. PI3K/Akt/mTOR mediated Pim-2 regulation reveals a new feedback loop within the pathway. In conclusion, the complexity of the PI3K/Akt/mTOR pathway impedes therapeutic targeting
Transcriptional control of cellular plasticity in cancer cell senescence
Zelluläre Seneszenz wird als terminaler Zellzyklusarrest definiert, der mit dem Altern und funktionellen Verlust von Geweben verknüpft ist. Eine Seneszenzreaktion wird ebenso durch Onkogene und zytotoxischen Stress verursacht. Die Ausführung des Seneszenzprogramms wird durch eine zeitlich hochdynamische Aktivität von Transkriptionsfaktoren (TF) bedingt. Interessanterweise kann die Zelllinienzugehörigkeit einer Zelle durch die Expression von linien-aberranten TF überschrieben werden.
Die vorliegende Arbeit untersucht Chemotherapie-induzierte Seneszenz (TIS) in Bcl-2 überexprimierenden, deshalb vor Apoptose geschützten, murinen Eµ-Myc B-Zell Lymphomen in An- oder Abwesenheit der Seneszenz-essentiellen Histonmethyltransferase Suv39h1. Analysen auf Transkriptom- und auf Proteinebene ergeben dabei, dass in einer Seneszenz-spezifischen Weise die TF AP-1, PU.1 und C/EBPβ induziert werden, welche normalerweise für die Funktion und Entwicklung myeloischer Zelllinien bedeutend sind. Dementsprechend korreliert der Seneszenzzustand mit Transkripten, Oberflächenmarkern und einer enzymatischen Funktion der myeloischen Linie. Indem die identifizierten TFs heruntergeschaltet oder überexprimiert werden, wird ihre direkte Beteiligung an der Linienuntreue der TIS Lymphome demonstriert.
TIS-Kapazität wird als für den Erfolg von Krebstherapie günstige Eigenschaft betrachtet, da sie zu einem Wachstumsblock führt. Nichtsdestotrotz können sich verweilende TIS Zellen krebsbiologisch auch nachteilig auswirken. Anhand von murinen und humanen, klinisch annotieren Transkriptomdatensätzen kann hier in beiden Spezies ein myeloisch verschobenes, Linienuntreue anzeigendes Genexpressionsprofil mit einer besseren Überlebensprognose korreliert werden. Die vorliegenden Befunde legen nahe, dass die Modulation von TF Aktivitäten in Seneszenz einen potentiellen therapeutischen Angriffspunkt darstellt, um den für den Therapieerfolg nützlichen Zweig des TIS Phänotyps zu befördern.Cellular senescence is regarded as an irreversible cell cycle arrest associated with tissue aging and its functional decline. A senescence response is also evoked by oncogenic and cytotoxic stress. The execution of the senescence program relies on a highly dynamic sequence of transcription factor (TF) activities. Interestingly, cell lineage commitment can be overridden by the expression of lineage-aberrant TFs.
This thesis examines chemotherapy-induced senescence (TIS) in Bcl-2 overexpressing, thus apoptosis-protected, murine Eμ-Myc B-cell lymphomas with or without the senescence-essential histone methyltransferase Suv39h1. Transcriptome as well as protein level analyses reveal senescence-specific induction of the TFs AP-1, PU.1 and C/EBPβ which are typically crucial for myeloid lineage commitment and function. Correspondingly, the senescent state associates with myeloid lineage transcripts, surface markers and enzymatic function, reminiscent of, but not equal to a transdifferentiation phenotype. By knocking down and overexpressing the identified TFs, we demonstrate their direct involvement in the lineage infidelity of TIS lymphomas.
TIS-capacity is viewed as beneficial to cancer therapy outcome due to its block on proliferation. However, lingering TIS cells can also be detrimental due to the acquisition of latent stemness properties or tumor-protective remodeling of their microenvironment. By interrogating murine and human, clinical course-annotated transcriptome data sets, an association between a myeloid-skewed, lineage infidelity indicating gene expression profile and better tumor prognosis is established in both species. The presented findings suggest that modulation of the senescent TF activities could be therapeutically exploited to foster the cancer patient-beneficial branch of the TIS phenotype

Unterschiedliche Autoregulation am PU.1 Lokus in B -Zellen und myeloischen Zellen
Als Schlüsselfaktor des hämatopoietischen Systems spielt PU.1 eine ent-scheidende Rolle in der Entwicklung der meisten hämatopoietischen Li-nien. Das PU.1 Expressionslevel bestimmt das Differenzierungspotential hämatopoietischer Stammzellen und Vorläufer. In den unterschiedlichen Zelltypen werden verschiedene Expressionsstärken etabliert. Wie diese zelltypischen Expressionslevel vonPU.1 generiert werden, ist bisher weit-gehend unbekannt. In der vorliegenden Doktorarbeit wurde mit Hilfe eines transgenen Maus-modells die cis-regulatorische Einheit von PU.1 definiert, um mit nachfol-genden molekularbiologischen und genomweiten Ansätzen Mechanismen der zellspezifischen Regulation von PU.1 zu aufzuzeigen. Die Definition der cis-regulatorischen Einheit von PU.1 erfolgte mit Hilfe eines transgenen Mausmodells, welches ein humanes PU.1 BAC Kons-trukt trägt. Es konnte gezeigt werden, dass humanes und murines PU.1 substituierbar sind und den gleichen Regulationsmechanismen unterlie-gen. Mit Hilfe genomweite DNaseI hypersensitivity Analysen, Methylie-rungs- und Bindungsstudien konnte ein neuer Regulationsmechanismus beschrieben werden, der eine spezifische kombinatorische Interaktion verschiedener cis-regulatorischer Elemente erfordert. Durch Reportergenassays in verschiedenen Zelltypen war es möglich, einen myeloischen Enhancer zu identifizieren. Es konnte gezeigt werden, dass PU.1 mit zelltyp-spezifischen Transkriptionsfaktoren interagiert, um unterschiedliche Bindungsmuster an seinen regulatorischen Elementen zu etablieren. Dadurch kommt es zu den spezifischen Expressionstärken von PU.1The transcription factor PU.1 occupies a central role in controlling myeloid and early B cell development and its correct lineage-specific expression is critical for the differentiation choice of hematopoietic progenitors. However, little is known of how this tissue-specific pattern is established. We previously identified an upstream regulatory cis-element (URE) whose targeted deletion in mice decreases PU.1 expression and causes leukemia. We show here that the URE alone is insufficient to confer physiological PU.1 expression, but requires the cooperation with other, previously unidentified elements. Using a combination of transgenic studies, global chromatin assays and detailed molecular analyses we present evidence that PU.1 is regulated by a novel mechanism involving cross-talk between different cis-elements together with lineage-restricted autoregulation. In this model, PU.1 regulates its expression in B cells and macrophages by differentially associating with cell-type specific transcription factors at one of its cis-regulatory elements to establish differential activity patterns at other elements
Identification of CLK2 and CLK4 as novel regulators of DNA damage-induced NF-κB activity by chemical dissection
Chemotherapy and radiation are standard-of-care cancer treatments, but their effectiveness is often hampered by therapy resistance within the tumor. Numerous studies have demonstrated that tumor cells can evade cell death triggered by genotoxic therapies by activating IKK/NF-κB pathway, thereby preventing apoptosis. The direct targeting of IKKs using pharmacological interventions is not a viable option due to the significant adverse effects caused by the essential role of IKK/NF-κB signaling in various physiological processes. To circumvent this, previous work by our group identified structurally unrelated small molecule inhibitors, MW01 and MW05, that selectively inhibit IKK/NF-κB solely in response to DNA double strand breaks induced by chemotherapy and radiation. Importantly, these compounds do not interfere with IKK/NF-κB activation triggered by other physiological stimuli.
Initial work began by confirming the genotoxic stress-specific inhibition by the compounds before moving onto target identification studies. Considering the similar cellular effects of both compounds within the DNA damage-induced NF-κB pathway, comparative target identification studies including kinase assay panels, structural derivatization, and molecular signaling characterization were performed, seeking targets shared between both lead compounds. Common regulators shared by other NF-κB stimuli were first excluded as potential targets of the compounds before investigation of the several identified shared targets revealed a previously unknown regulators of genotoxic stress-induced NF-κB activity, Cdc-like kinases (CLK) 2 and 4, as the functional target of MW01 and MW05. Silencing of the CLK2 and 4 revealed that they are essential for DNA damage-induced NF-κB activity and promote the phosphorylation of IKK at Ser-85, a genotoxic stress specific ATM-dependent phosphosite, critically localizing the CLKs within the cascade between ATM and IKK. CLK2 and 4 were also confirmed as the target of active structural derivatives of MW01 and MW05 and were spared by inactive derivatives, confirming CLK2 and 4’s role in genotoxic stress-induced NF-κB. In addition, CLK inhibitor MU-1210 also inhibited NF-κB following DNA damage, suggesting that CLK inhibitors could be used to potentiate the tumor killing effect of standard cancer treatments.
MW01 and MW05 were tested in co-treatment with DNA damaging agents, in the context of on-going DNA damage, and in patient derived glioblastoma cells to assess their potential clinical applications. Critically, neither MW01 nor MW05 exhibit general toxicity; instead, they notably enhance apoptosis specifically in tumor cells following genotoxic stress. In BRCA1-deficient cells and in co-treatment with PARP inhibitor Olaparib, both characterized by on-going DNA damage, MW01 and MW05 potentiated DNA damage and p53 levels, suggesting that the compounds unbalance the NF-κB/p53 axis in favor of apoptosis. This approach introduces a novel therapeutic strategy to curb NF-κB activity induced by DNA damage in cancer cells without impacting its essential functions in healthy cells.Chemotherapie und Bestrahlung gehören zu den Standard-Krebstherapien, ihre Wirksamkeit wird jedoch häufig durch das Auftreten von Therapieresistenzen im Tumor beeinträchtigt. Zahlreiche Studien haben gezeigt, dass Tumorzellen dem durch genotoxische Therapien ausgelösten Zelltod entgehen können, indem sie den IKK/NF-κB-Signalweg aktivieren und so die Apoptose verhindern. Das direkte Angreifen von IKKs durch pharmakologische Interventionen ist keine praktikable Option, da die wesentliche Rolle der IKK/NF-κB-Signalübertragung bei verschiedenen physiologischen Prozessen erhebliche negative Auswirkungen erwarten lässt. Um dies zu umgehen, hat unsere Gruppe strukturell nicht verwandte niedermolekulare Inhibitoren, MW01 und MW05, identifiziert, die die einzigartige Fähigkeit besitzen, die Aktivierung von IKK/NF-κB ausschließlich als Reaktion auf durch Chemotherapie und Bestrahlung verursachte DNA-Doppelstrangbrüche selektiv zu hemmen. Wichtig ist, dass diese Verbindungen die durch andere normale physiologische Reize ausgelöste IKK/NF-κB-Aktivierung nicht beeinträchtigen.
Die ersten Arbeiten begannen mit der Bestätigung der genotoxischen Stress-spezifischen Hemmung durch die Verbindungen, bevor die Studien zur Identifizierung der Targets fortgesetzt wurden. In Anbetracht der ähnlichen zellulären Wirkungen beider Verbindungen innerhalb des durch DNA-Schäden induzierten NF-κB-Stoffwechsels wurden vergleichende Studien zur Identifizierung von Zielmolekülen durchgeführt, einschließlich Kinase-Assay-Panels, struktureller Derivatisierung und molekularer Signalcharakterisierung, um gemeinsame Zielmoleküle der beiden Leitverbindungen zu finden. Gemeinsame Regulatoren anderer NF-κB-Stimuli wurden zunächst als potenzielle Ziel-Proteine der Verbindungen ausgeschlossen, bevor die Untersuchung der verschiedenen identifizierten gemeinsamen Ziel-Enzyme einen bisher unbekannten Regulator der durch genotoxischen Stress induzierten NF-κB-Aktivität, die Cdc-ähnlichen Kinasen 2 und 4 (CLK2 und 4), als funktionelles Ziel von MW01 und MW05 ergab. CLK2 und 4 wurden auch als Ziel-Kinasen aktiver Strukturderivate von MW01 und MW05 bestätigt und blieben von inaktiven Derivaten verschont, was die Rolle von CLK2 und 4 bei der durch genotoxischen Stress induzierten NF-κB bestätigt. Darüber hinaus hemmte ein externer, strukturell unähnlicher CLK-Inhibitor, MU-1210, ebenfalls NF- κB nach DNA-Schäden, was darauf hindeutet, dass CLK-Inhibitoren zur Verstärkung der tumortötenden Wirkung von Standard-Krebstherapien eingesetzt werden könnten.
Parallel zu den Studien zur Identifizierung der Zielmoleküle wurden MW01 und MW05 auch bei der gleichzeitigen Behandlung mit DNA-schädigenden Substanzen, im Zusammenhang mit laufenden DNA-Schäden und in Glioblastomzellen von Patienten getestet, um ihre potenziellen klinischen Anwendungen zu bewerten. Kritisch anzumerken ist, dass weder MW01 noch MW05 eine allgemeine Toxizität aufweisen; stattdessen verstärken sie insbesondere die Apoptose in Tumorzellen nach genotoxischem Stress. In BRCA1-defizienten Zellen und bei gleichzeitiger Behandlung mit dem PARP-Inhibitor Olaparib, die beide durch anhaltende DNA-Schäden gekennzeichnet sind, verstärkten MW01 und MW05 den γH2AX-Wert, einen Marker für DNA-Schäden, und den p53-Wert, was darauf hindeutet, dass die Verbindungen die NF-κB/p53-Achse zugunsten der Apoptose aus dem Gleichgewicht bringen. Dieser Ansatz stellt eine neuartige therapeutische Strategie dar, um die durch DNA-Schäden in Krebszellen induzierte NF-κB-Aktivität zu bremsen, ohne ihre wesentlichen Funktionen in gesunden Zellen zu beeinträchtigen
Modulierung der NF-KB-Aktivität in T-Zellen durch den Carmal1-Bcl10-Malt1 Komplex
Das Schicksal aktivierter T-Zellen wird durch eine Vielzahl NF-kappaB regulierter Ziel-Gene bestimmt, wobei aktivierende und deaktivierende Signale für die Ausbalancierung einer adäquaten T-Zell Antwort benötigt werden. Im Rahmen dieser Arbeit konnte gezeigt werden, dass die negativ-regulatorische Modulierung des Carma1-Bcl10-Malt1 (CBM)-Proteinkomplexes für die Steuerung der NF-kappaB Aktivität in T-Zellen von großer Bedeutung ist. Überraschenderweise ist die Bildung des CBM-Komplexes abhängig von IKKbeta, einer Kinase, die zuvor ausschließlich mit CBM-nachgelagerten Effektorfunktionen in Verbindung gebracht wurde. IKKbeta übernimmt eine duale Funktion bei der Regulation des CBM-Komplexes: Obwohl IKKbeta zunächst für die Bildung des CBM-Komplexes benötigt wird, führt die Phosphorylierung der CBM-Komplexkomponente Bcl10 durch IKKbeta bereits kurze Zeit nach Beginn der T-Zell Aktivierung zu einer Dämpfung der Signalübertragung. Biochemische Analysen zeigen, dass die Phosphorylierung von Bcl10 die Proteinaffinitäten innerhalb des CBM-Komplexes beeinflusst, wodurch es zu einer Umlagerung des Komplexes mit negativ-regulatorischem Effekt kommt. Weiterführende Experimente haben aufgedeckt, dass Bcl10 im Zuge anhaltender T-Zell Stimulation lysosomal degradiert wird. Die Degradation von Bcl10 führt zum Zerfall des CBM-Komplexes und unterbindet die weitere Signalübertragung trotz persistenter Stimulation. Die Tatsache, dass beide in dieser Arbeit identifizierten negativ-regulatorischen Mechanismen am CBM-Komplex angreifen, unterstreicht die Bedeutung dieses Komplexes für die Signalübertragung in T-Zellen. Weiterhin besteht aufgrund der präsentierten Daten Anlass zur Annahme, dass in aktivierten T-Zellen ein vielfältig positiv und negativ regulierter Multikomponentenkomplex gebildet wird, der eine nicht-hierarchische Signalübertragung unterstützt.A multitude of NF-kappaB regulated target genes determines the fate of activated T cells, whereas activating and de-activating signals are crucial for balancing adequate T cell responses. The presented data illustrate that negative-regulatory modulation of the Carma1-Bcl10-Malt1 (CBM)-complex is of great importance for the control of NF-kappaB activity in T cells. Surprisingly IKKbeta, a kinase that so far was thought to be involved in CBM-downstream effector functions, is needed for CBM-complex formation. IKKbeta exhibits a dual function regulating the CBM-complex: while initially being essential for the formation of the CBM-complex, phosphorylation of the CBM-complex component Bcl10 by IKKbeta shortly after the onset of T cell activation leads to a damping of signal transduction. Biochemical analysis reveal that Bcl10 phosphorylation influences the intermolecular protein affinities of the CBM-complex components causing a remodeling of the complex with a negative-regulatory effect. Further experiments uncover that upon persistent T cell activation Bcl10 is degraded by the lysosome. Bcl10 degradation promotes the collapse of the CBM-complex and thereby interferes with ongoing signal transduction despite persistent stimulation. Considering the fact that both negative-regulatory processes affect CBM-complex activity underscores the important role of this complex in T cell signal transduction. Moreover, the presented data demonstrate that formation of a multi-component signaling complex in activated T cells facilitates versatile positive, negative and non-hierarchical regulation
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Identifizierung und Charakterisierung neuer Interaktionspartner von E2F3
Der pRB/E2F-Signalweg ist ein zentraler Regulator der Proliferationskontrolle in Säugerzellen, der in fast allen auftretenden Tumoren dereguliert ist. Durch unterschiedliche Mutationen in Komponenten dieses Signalwegs kommt es letzten Endes zu einer erhöhten Aktivität der E2F-Transkriptionsfaktoren und somit zu einer verstärkten Transkription von E2F-Zielgenen in diesen Tumoren. Um die molekularen Mechanismen der Rolle von E2F3 in der Zellzykluskontrolle und der Tumorigenese besser zu verstehen, wurden in dieser Arbeit per GST-Pulldown mit anschließender Massenspektrometrie neue potenzielle Interaktions-partner von E2F3 identifiziert. Ein identifizierter Interaktionspartner war die SNF2-ähnliche Helikase HELLS. HELLS interagiert in vitro und in vivo spezifisch mit der Marked Box-Domäne von E2F3, aber nicht mit anderen untersuchten E2F-Transkriptionsfaktoren, wie durch GST-Interaktionsstudien und Ko-Immunpräzipi-tationsexperimente demonstriert werden konnte. Durch Chromatin-Immunpräzipitation konnte zusätzlich gezeigt werden, dass E2F3 für die Rekrutierung von HELLS an E2F-regulierte Promotoren wie z. B. CDC6 oder p107 verantwortlich ist. Die shRNA-vermittelte Depletion von HELLS führte zu einer stark verminderten Induktion von allen untersuchten E2F-Zielgenen nach Serumstimulation und einem verspäteten Eintritt in die S-Phase der HELLS-depletierten Zellen, was zeigt, dass HELLS essenziell für die Induktion von E2F-Zielgenen ist. Bei der immunhistochemischen Untersuchung der E2F3- und HELLS-Expression in humanen Prostatakarzinomen zeigte sich, dass sowohl E2F3 als auch HELLS in späten aggressiven Stadien dieser Tumore sehr stark exprimiert sind, jedoch nur sehr schwach in den weniger aggressiven Tumoren. Diese Versuche zeigen, dass es sich bei HELLS um einen neuen Bestandteil des pRB/E2F-Signalwegs handelt, der eventuell in der Entstehung gewisser Tumorarten eine Rolle spielt und somit ein neues potenzielles Ziel für neuartige Krebstherapien darstellt.The pRB/E2F pathway is a key regulator of proliferation in mammalian cells and is commonly mutated in human tumors. These mutations in the components of the pRB/E2F pathway lead to deregulated activity of the E2F transcription factors resulting in increased expression of E2F target genes. To further understand the molecular mechanisms of E2F3 in cell cycle control and its role in tumorigenesis new interaction partners for E2F3 were identified in the course of this thesis with the help of a GST-Pulldown approach coupled to mass spectrometric analysis. One of the identified interaction partners was the SNF2-like helicase HELLS. With the help of GST-interaction studies and Co-Immunoprecipitation assays it could be demonstrated that HELLS interacts specifically with E2F3 via its Marked Box domain but does not bind to the other investigated E2F transcription factors. HELLS could be detected at E2F target genes like p107 and CDC6 in vivo with the help of Chromatin-Immunoprecipitation assays. Furthermore, the forced recruitment of E2F3 to E2F target genes led to an enhanced binding of HELLS to these promotors suggesting that HELLS is recruited to E2F target genes via protein-protein interaction with E2F3. The shRNA-mediated depletion of HELLS led to a strongly reduced induction of E2F target genes and a delay in S-phase entry, showing that HELLS is essential for the induction of E2F target genes. During the immunohistochemical analysis of human prostate cancer specimens it became evident that both E2F3 and HELLS are strongly expressed in the more aggressive late stages but only weakly expressed in the early stages of this tumor type. These findings demonstrate that HELLS is a new component of the E2F/pRB pathway which might play a role in the development of certain tumors and might represent a new target for novel cancer therapies
Molecular and metabolic determinants of metastasis development and progression
MACC1, ein Hauptregulator von Metastasen, ist an zahlreichen Kennzeichen von Krebs beteiligt, einschließlich dereguliertem Metabolismus. Dennoch ist seine Rolle im Krebsstoffwechsel unklar. In der vorliegenden Arbeit wurde eine systematische Analyse von MACC1-getriebenen metabolischen Netzwerken durchgeführt. MACC1 erhöhte die GLUT1 auf Zellmembrane, was zu einer erhöhten Glukoseanreicherung, einem erhöhten Glukosefluss und somit zu einer erhöhten Zellproliferation führte. Außerdem, reduzierte MACC1 den Glutaminfluss unabhängig von der Nährstoffverfügbarkeit. Bei Glucoseentzug erhöhte MACC1 die Pyruvataufnahme und zeigte aber die geringe Auswirkungen auf den Pyruvatfluss. In vivo, MACC1 zeigte erhöhte Aufnahme von 18F-FDG und 18F-Glutamat in Lebermetastasen. Zusammengefasst zeigen diese Ergebnisse, dass MACC1 mehrere Wirkungen auf den Krebs-Metabolismus zeigt, was es attraktiv macht, seine Wirkungen in Krebsmodellen weiter zu untersuchen.
Metastasierung ist die Haupttodesursache bei Darmkrebs. Fünfzehn bis zwanzig Prozent der Darmkrebs Patienten im Stadium II entwickeln im Verlauf der Erkrankung Metastasen, jedoch bleiben die Kriterien der Wahrscheinlichkeit mit welcher Patienten von Chemotherapie profitieren werden ungenau. Hier wurde das Potenzial der Plasma-Metabolomik zur Vorhersage von Metachronmetastasen untersucht. Plasma metabolische Profile wurden wesentlich unterschiedlich zwischen nicht-metastasierten und metachron metastasierten Patienten gefunden. Wie Klassifikationsmodelle aus Entscheidungsbäumen und Support-Vektor-Maschinen gezeigt haben die Plasmametaboliten haben die Fähigkeit nicht-metastasierte von metachron metastasierten Darmkrebs Patienten zu unterscheiden, mit einer durschnittlichen Vorhersagegenauigkeit von 0,75 bzw. 0,82 für jede der Methoden, angemessen. Zusammen, zeigen diese Ergebnisse, dass Plasmametaboliten das Potenzial haben, Darmkrebs Patienten gemäß ihrem Metastasierungsrisiko nichtinvasiv zu stratifizieren.MACC1, a master regulator of metastasis, is involved in most hallmarks of cancer, including deregulated metabolism. Here, a systematic analysis of MACC1-driven metabolic networks by elucidation of cell nutrient preferences, environment dependent alterations of nutrient utilization, metabolic pathway functionality and metabolic tracing using 13C-labeled metabolic substrates had been performed. MACC1 was found to enhance surface GLUT1 thus leading to increased glucose depletion, glucose flux and hence increased cell proliferation. Besides, MACC1 was found to reduce glutamine flux independent of nutrient availability. Upon glucose deprivation MACC1 was found to enhance pyruvate uptake exhibiting minor effects on pyruvate flux. In vivo, MACC1 increased uptakes of 18F-FDG and 18F-glutamate in liver metastatic lesions. Together, these findings demonstrate that MACC1 exhibits multiple effects on cancer metabolism, thus making it attractive to further study its effects in cancer models.
Metastasis is the main cause of death from colorectal cancer (CRC). Fifteen to twenty percent of stage II CRC patients develop metastasis during the course of disease, however the criteria of likely benefitting patients from chemotherapy remain imprecise. Here, the potential of plasma metabolomics to predict metachronous metastasis was assessed. Plasma metabolic profiles were shown to be significantly different between non-metastasized and metachronously metastasized CRC patients. As demonstrated by supervised classifications using decision trees and support vector machines plasma metabolites have the power to distinguish non-metastasized from metachronously metastasized CRC patients giving average prediction accuracy of 0.75 and 0.82 for each of the methods, respectively. Together, these results demonstrate that plasma metabolites have the potential to non-invasively stratify CRC patients according to their metastasis risk
