Pelvic osteolysis is a well-known complication following total hip arthroplasty (THA) [1], both with and without cement [2], [3] and [4]. It has been attributed as the biological response to wear debris of several materials, but polyethylene particles are the most implicated agents [5], [6] and [7]. Research of drugs against age-related diseases
Sarcopenia is a prominent disease in developed societies. It is described as an age-related pathology characterized by a progressive loss of muscle mass and strength. It is the main cause of frailty and disability in the elderly worldwide, affecting up to 50% of 80-year. Currently, no known cure is available for sarcopenia. Our laboratory collaborated with Rejuvenate Biomed, a company specialized in healthy aging, to test the effect of RJx-01, a combination drug, composed of metformin and galantamine, on sarcopenia.
Metformin is a drug used against type 2 diabetes. Galantamine is commonly used in the treatment of Alzheimer’s disease. Both drugs have been linked to a decrease in all-cause mortality. The combination of these molecules has been predicted by Rejuvenate Biomed’s proprietary platform CombinAgeTM to have a synergistic effect on sarcopenia. Indeed, RJx-01 has been proven to ameliorate motor symptoms and muscle structure in C. elegans, in aged mice, as well as in inducible muscle specific OPA1 KO mice, a model of precocious aging. We decided to further characterize RJx-01 by testing the drug on murine models for acute disuse skeletal muscle atrophy. As opposed to chronic models of disuse, we aimed at studying the atrophic process during periods of limb immobilization, mimicking the condition of patients wearing casts or dealing with forced bedrest.
Before proceeding with the RJx-01 tests, we decided to characterize our own model for unilateral immobilization, consisting in a 3D-printed cast. The mounting procedure is non-invasive, simply requiring a light anesthesia and a few drops of superglue. A 2-weeks period of immobilization produced little to no swelling or skin damage and induces the activation of atrophy-related genes, causing a muscle weight loss, cross-sectional area decrease and reduction of muscle force. The contralateral leg did not show signs of overload or hypertrophy when compared to free roaming littermates, offering a good internal control over the immobilized limb. The unmounting procedure is also simple, only requiring the separation of the two halves of the boot with a specialized scissor. Upon cast removal, mice can immediately reload the previously immobilized hindlimb, recovering the normal 4-legged gait. Therefore, the recovery phase can be studied, with mice effectively recovering muscle mass and force in 3 weeks.
We proceeded to test RJx-01 in three groups of wild type mice. We immobilized for 2 weeks a first cohort of aged mice to test the effect of RJx-01 on acute disuse skeletal muscle atrophy as opposed to chronic. We also performed the same experiment in young mice to study possible age-dependent differences in RJx-01. One last cohort of young mice was again immobilized for 2 weeks but allowed to recover for 1 week, to inquire possible effects related to the recovery phase. The preliminary studies showed that RJx-01 treatment resulted in a reduced tendency of percentage muscle loss and had no significant effect on strength after disuse in old mice. In young mice, RJx-01 treatment resulted in a reduced tendency of percentage muscle loss and significantly reduced absolute muscle force loss compared to the untreated group after disuse. RJx-01 had no significant effect on these parameters after one week recovery.
Further, we explored the use of C. elegans to study Parkinson’s Disease (PD). Preliminary tests were executed on C. elegans PD models to select a suitable protocol for future drug testing. C. elegans strains expressing human α-synuclein were investigated, monitoring declines in locomotion and survival, and progression of α-synuclein aggregates formation. One strain of YFP-tagged dopaminergic neurons worms was used to study the natural degeneration of dopaminergic neurons during aging, as well as after exposure to a dopaminergic neurons-selective toxin. These tests led to the writing of replicable protocols suitable for drug testing in C. elegans models

Con Il Libro pel bambino, Don Giulio Tarra risultò il vincitore di due concorsi indetti dal Congresso Pedagogico Italiano nel 1861: quello per un libro di lettura destinato ai bambini sordomuti e l’altro per un libro destinato alle scuole rurali. Fra le opere del Tarra pubblicate da Paolo Carrara è menzionato il volumetto “Lezioni in famiglia, versione da M. Carraud”. Questo contributo intende offrire alcuni elementi d’informazione su Zulma Carraud, autrice delle Historiettes véritables pour les enfants de quatre à huit ans, titolo originale della raccolta di racconti tradotta da Don Giulio. Il libro era uscito a Parigi nel 1864 per i tipi di Hachette. In precedenza lo stesso editore aveva accolto le prime opere di Zulma nelle collane destinate alla scuola primaria. A pochi anni di distanza l’uno dall’altra, la Carraud nella Francia del II Impero e il Tarra nell’Italia postunitaria, entrambi hanno contribuito con i loro “libri di lettura” allo sviluppo dell’editoria per la scuola e per l’infanzia.

With the volume entitled Il Libro pel bambino, Don Giulio Tarra won two literary competitions held in 1861 by the Congresso Pedagogico Italiano: the first was dedicated to books for deaf-mute chidren, the second to books for rural schools. Among Tarra’s works published by Paolo Carrara we find “Lezioni in famiglia, versione da M. Carraud”. This article provides some information about Zulma Carraud, the author of the collection of novels entitled: Historiettes véritables pour les enfants de quatre à huit ans published by Hachette in 1864 and translated into Italian by Don Giulio. Previously the same publisher had collected Zulma’s first works in a series to be used in primary schools. Carraud, during the Second Empire period in France, and Tarra, in post-Unity Italy, both contributed to the development of the publishing industry for schools and children with their educational and entertaining books
Efficacy of Elexacaftor–Tezacaftor–Ivacaftor on chronic rhinosinusitis in cystic fibrosis
Purpose: Our work aims to add evidence on the effectiveness of Elexacaftor-Tezacaftor-Ivacaftor on chronic rhinosinusitis in cystic fibrosis. Materials and methods: We conducted an observational retrospective cohort study at the Cystic Fibrosis Center of a tertiary care hospital to investigate the effect of Elexacaftor-Tezacaftor-Ivacaftor on chronic rhinosinusitis in cystic fibrosis patients, aged 12 or older. The study's endpoints were the change in the occurrence of acute exacerbations of chronic rhinosinusitis, and the variation of the endoscopic and radiologic findings scored using the Lund-Kennedy endoscopic scale, Lund-Mackay, and modified Lund-Mackay radiologic scales, in patients who underwent both pre-treatment and post-treatment examinations. Results: The study population comprised 136 patients, of which 28 underwent both pre-treatment and post-treatment nasal endoscopy and 15 had pre- and post-treatment CT scans. Elexacaftor-Tezacaftor-Ivacaftor provided a significant improvement in chronic rhinosinusitis. The mean number of acute exacerbations of chronic rhinosinusitis per year in the pre-treatment time was 0.55 versus 0.35 during the treatment ( p < 0.0021). The Lund-Kennedy scale had a pre-treatment average score of 4.21 points versus 1.5 points after the start of Elexacaftor-Tezacaftor-Ivacaftor ( p < 0.0001). The average Lund-Mackay and modified Lund-Mackay scores in the pre-treatment time were respectively 14.6 and 16.45 points; and after the start of the therapy, they became 5.87 and 6.73 ( p < 0.0001). Conclusion: Elexacaftor-Tezacaftor-Ivacaftor was associated with fewer acute exacerbations of chronic rhinosinusitis, and a significant improvement of chronic rhinosinusitis evaluated endoscopically and radiologically. To our knowledge, this is the first study investigating the change in the occurrence of acute exacerbation of chronic rhinosinusitis in patients affected by cystic fibrosis in therapy with Elexacaftor-Tezacaftor-Ivacaftor
