Characterization of Neoparamoeba pemaquidensis strains: PCR-RFLP of the internal transcribed spacer region from the amoeba and endosymbiont
No full textNeoparamoeba pemaquidensis continues to be an ongoing problem for commercial finfish aquaculture and has also sporadically been associated with mass mortalities of commercially relevant marine invertebrates. Despite the ubiquity and importance of this amphizoic amoeba, our understanding of the biology as it applies to host range, pathogenicity, tissue tropism, and geographic distribution is severely lacking. This may stem from the inability of current diagnostic tests based on morphology, immunology, and molecular biology to differentiate strains at the subspecies level. In the present study, we developed a polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method based on the internal transcribed spacer (ITS) region that can accurately differentiate amoeba strains of N. pemaquidensis. The investigation focused on the complications of the amoeba ITS microheterogeneity in the development of a subspecies marker and the use of the endosymbiont, Ichthyobodo necator related organism (IRO), ITS region as an alternative marker. The combination of host amoeba and endosymbiont ITS PCR-RFLP analyses was successfully used to correctly identify and characterize an N. pemaquidensis isolate from an outbreak of amoebic gill disease in Atlantic salmon Salmo salar from the west coast of North America (Washington State, USA).Charles G. No full textAu cours de l’unification de l’Italie, l’éducation physique a fait une contribution essentielle à la préparation des nouveaux citoyens de la nouvelle nation en termes de caractère moral, éthique, idéologique, y compris les aspects physiques. Toutefois, il a fallu attendre la loi De Sanctis en 1878, pour qu’elle devienne une discipline obligatoire. Tout au long de ce texte, sera présenté comment le changement de la nouvelle capitale de Florence à Rome entraîna de nombreux problèmes concernant les installations sportives et la difficulté de promouvoir l’activité physique, non seulement dans la capitale mais aussi dans toute la nation. À la fin du 19e siècle, la création des Écoles Normales pour former des professeurs d’éducation physique et la création d’un certain nombre de fédérations et associations témoignait de l’intérêt quant à la promotion du sport, mais son introduction était encore loin de celles d’autres pays européens comme l’Allemagne, la France ou la Suisse. SALT project: preliminary physical and mineralogical characterization of Alpine sulphates for geological and geotechnical modeling
No full textThe excavation of underground infrastructures in geological sequences containing sulphates may cause swelling, water weakening, creep and karst. All these phenomena have been observed in tunnels for decades, but related technical problems still remain. This is especially true for geologically complex domains as Alpine environments, where the mechanical behviour of sulphates needs to be evaluated in relation to the structural complexity of the fracture network and of the 3D geometries of the orogenic context. This study, that is developed in the framework of the SALT project, a multi scale and interdisciplinary investigation of sulphate salts in the Alpine region, proposes a description of the main features of outcropping sulphates in Western Alps, providing useful preliminary information to afford technical issues related to sulphates during the realization of infrastructures in the Alps. © 2024 The Author(s).C.
