    以十三個荔枝品種爲材料,進行RAPD的片段分析。經篩選200個逢機引子後,選出23個引子所產生的259個條帶(RAPD markers),做爲十三個荔枝品種間的親緣關係評估之依據。並利用Jaccard氏的計算方法,求得不同樣品間的相似度;並利用UPGMA方法,建立其親緣關係圖。結果顯示參試的十三個荔枝品種可區分爲兩大群,其中玉荷包與三月紅最爲相似,並與竹葉、新興、糖薄、高雄早生等品種叢聚在指數0.53處,而沙坑小核、尚書淮、糯米滋、桂味、桂枝及黑葉等品種則成另一叢聚在指數0.52處,其中桂味與桂枝的相似度100%,顯示這兩個品種可能爲同種異名。另外,就引子所產生條帶數而言,在23個引子中,以UBC-199產生9個條帶數爲最少,其次爲OPE-06及OPG-01產生10個條帶數,而OPG-08產生26個條帶數爲最多,平均每個引子可以產生15個條帶,從這些引子中篩檢出鑑別玉荷包品種的專一性條帶7條、鑑別糯米滋品種的專一性條帶6條、鑑定沙坑小核品種的專一性條帶7條、及鑑定高雄早生品種的專一性條帶6條、一個尚書淮品種的專一性條帶、四個新興品種的專一性條帶、一個竹葉品種的專一性條帶及兩個桂味及桂枝品種共有的專一性條帶,這些品種專一性的條帶將可供作不同荔枝品種鑑別用。The random amplified polymorphic DNA (RAPD) markers were used to identify the genetic relationship among introduced lychee cultivars in this study. Among 200 different decamer primers, 23 primers were selected; the selected primers amplified a total of 259 DNA bands. The total number of amplified bands per primer varied from 9 (UBC-199) to 26 (OPG-08), on an average 11.3 bands were induced by each primer. Jaccard's similarity coefficients for interval measure within and between cultivars were used to produce a cluster diagram using the unweighted pair-group method (UPGMA). The results showed that eighteen samples from thirteen lychee cultivars could be divided into two major groups: one included Yu ho pao, San yue hong, Zhu yeh, Xin xing, Liu yue xue, Tang bo and Gao xiong zao sheng; the other included Sha keng xiao ho, Shang shu huai, No mi zi, Kwai wei, Kwai zi and Hei yeh. The Jaccard's similarity coefficient of Kwai wei and Kwai zi was 1.00, implying that Kwai wei and Kwai zi may be the same cultivar. From 259 RAPD marker bands, we discovered 8 specific bands for Yu ho pao, 6 for No mi zi, 7 for Sha keng xiao ho, 6 for Gao xiong zao sheng, 1 for Shang shou huai, 4 for Xin xing, 2 for Hei yeh, 1 for of Zhu yeh and 2 for Kwai wei (Kwai zi). Those specific single bands of different cultivars will be recovered from agarose gel and cloned into TA vector system for sequencing, in order to develop new specific primer sets for the use of identifying different cultivars and hybrid posterity

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    本研究利用快速增殖cDNA (rapid amplification of cDNA ends, RACE)技術,從愛文芒果果皮之訊息RNA(mRNA)中選殖類胡蘿蔔素(phytoene desaturase, PDS)基因之互補DNA(cDNA)以瞭解其果實發育期間色素基因表現情形。PDS基因全長爲1,882bp,包含一個長達1,644bp的解讀框架(open reading frame, ORF)可轉譯成548個胺基酸,5'及3'端未轉譯區(untranslated region, UTR)分別爲36bp及202bp,由序列比對分析結果顯示,該基因的胺基酸序列與其他植物物種具有50.0-74.1%的相似性。PDS基因經逆轉錄及DNA擴增後利用管家基因(housekeeping gene) actin之相對表現量,偵測其在不同果實發育時期之相對表現量,顯示愛文芒果PDS基因在果實發育期間爲穩定表現之基因,PDS基因從幼果期至黃熟果均有表現,與actin基因的相對表現量在幼果期(stage1)最高,隨著果實發育,該基因的表現量呈現降低的趨勢。而果實在成熟過程中,總胡蘿蔔素的含量會在果實達到生理成熟期後才會快速地大量增加,而該基因的表現量卻未隨總胡蘿蔔素的增加而增加,顯示在果實後熟過程中,PDS基因並未對類胡蘿蔔素生成有直接的影響。由於在果實發育過程中,芒果PDS基因的表現量呈現下降之趨勢,該基因的表現模式應爲負調節(down-regulated)方式。Mango (Mangifera indica) is a tropical fruit containing carotenoids. In order to understand the expression of phytoene desaturase (PDS), a key enzyme for the biosynthesis of β-carotene, during fruit development, we havecloned and characterized a cDNA encoding PDS from young mango fruits, cv. Irwin. Mango PDS cDNA contains 1,882bp, with 1,644bp open reading frame encoding for a polypeptide of 548 amino acid residues. The 5' and 3' untranslated regions (UTR) were 36bp and 202bp, respectively. The deduced amino acid sequence of this PDS has 50.0-74.1% of similarity to PDS from other plant species. The relative expression levels and carotenoids content of the Irwin mango PDS at the six fruit development stages and in mature leaves were detected. The result showed that the expression level of PDS gene at different fruit development stages was reduced and the carotenoids increased rapidly, when fruit are maturing. These findings suggest that mango PDS is down regulated during the fruit ripening process

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
