Strategic Reforms for Accelerated Agricultural Growth in Pakistan
Agricultural growth rates in the 1960s, 1970s, 1980s and 1990s show that strong growth during the 1960s was driven by several factors, including greater certainty in the use of irrigation water (as a result of an agreement with India), the introduction of productivityenhancing fertiliser-seed packages, the introduction of tubewells and the electrification of rural areas, and policy changes that improved the profitability of farming. Growth during the 1970s dropped to 2.3 percent as a result of the uncertainty created by land reforms in 1972 and 1977, severe climatic shocks, a cotton virus that depressed production for most of the decade, and political instability. The recovery in the 1980s and early 1990s can be attributed to the introduction of new cotton varieties and improved management techniques, as well as to a gradual improvement in economic incentives. Closer inspection of the nature and sources of this growth raises concerns about its sustainability and casts doubt on the ability of the sector to grow by more than 3–4 percent a year in the future. Many of the past sources of agricultural growth in Pakistan appear to have been fully exploited. Strategy for the future must effectively address the followings. Allowing the market to Operate, policy reforms that support the ongoing structural adjustment should be given top priority. To address the crisis in irrigation management market-determined incentives must be allowed to determine resource allocation within the irrigation system. Reform in extension should include establishing closer links with research institutions and reducing the number of front-line extension workers and replacing them with fewer, bettertrained workers who are more responsive to the needs of farming systems. Full-fledged land reform is difficult to enact and can be considered only after a comprehensive study of costs and benefits. Some important measures can be implemented immediately, however. Foremost is providing security of tenure to many farmers, especially tenants-at-will, thereby improving responsiveness to incentives and creating better incentives for long-term investments.

Disruption of brainstem monoaminergic fibre tracts in multiple sclerosis as a putative mechanism for cognitive fatigue:a fixel-based analysis
In multiple sclerosis (MS), monoaminergic systems are altered as a result of both inflammation-dependent reduced synthesis and direct structural damage. Aberrant monoaminergic neurotransmission is increasingly considered a major contributor to fatigue pathophysiology. In this study, we aimed to compare the integrity of the monoaminergic white matter fibre tracts projecting from brainstem nuclei in a group of patients with MS (n = 68) and healthy controls (n = 34), and to investigate its association with fatigue. Fibre tracts integrity was assessed with the novel fixel-based analysis that simultaneously estimates axonal density, by means of 'fibre density', and white matter atrophy, by means of fibre 'cross section'. We focused on ventral tegmental area, locus coeruleus, and raphe nuclei as the main source of dopaminergic, noradrenergic, and serotoninergic fibres within the brainstem, respectively. Fourteen tracts of interest projecting from these brainstem nuclei were reconstructed using diffusion tractography, and compared by means of the product of fibre-density and cross-section (FDC). Finally, correlations of monoaminergic axonal damage with the modified fatigue impact scale scores were evaluated in MS. Fixel-based analysis revealed significant axonal damage - as measured by FDC reduction - within selective monoaminergic fibre-tracts projecting from brainstem nuclei in MS patients, in comparison to healthy controls; particularly within the dopaminergic-mesolimbic pathway, the noradrenergic-projections to prefrontal cortex, and serotoninergic-projections to cerebellum. Moreover, we observed significant correlations between severity of cognitive fatigue and axonal damage within the mesocorticolimbic tracts projecting from ventral tegmental area, as well as within the locus coeruleus projections to prefrontal cortex, suggesting a potential contribution of dopaminergic and noradrenergic pathways to central fatigue in MS. Our findings support the hypothesis that axonal damage along monoaminergic pathways contributes to the reduction/dysfunction of monoamines in MS and add new information on the mechanisms by which monoaminergic systems contribute to MS pathogenesis and fatigue. Proteomic characterization of Corynebacterium pseudotuberculosis biofilm / Syaida Anati Abd Rashid
Corynebacterium pseudotuberculosis is a pathogenic intracellular parasite causing caseous lymphadenitis (CLA) mostly in sheep and goats, thus, leading to significant economic losses. The morphology, biochemical composition, and antimicrobial susceptibility pattern of C. pseudotuberculosis biofilm have previously been addressed. However, little is known about the whole-cell proteome expression in C. pseudotuberculosis biofilm. The present study was performed to determine proteome profiles of planktonic and biofilm fractions of C. pseudotuberculosis using 1D-SDSPAGE and to identify differentially expressed proteins during the biofilm formation by C. pseudotuberculosis using LC-MS/MS. The FESEM image showed that heterogenous C. pseudotuberculosis biofilm was successfully formed within 24h. The treatment with antimicrobials substantially inhibited the viability of the C. pseudotuberculosis biofilm. The percentage of biofilm inhibition was found to be in the range between 12.92% to 87.44%. Both planktonic and biofilm fractions showed the expression of nine protein bands ranging between 33.7 kDa and 150 kDa. However, a protein band of 48.3 kDa appeared in the planktonic fraction but not in the biofilm fraction

Improving MHC-I ligand identifications from LC-MS/MS data by incorporating allelic peptide motifs
MHC class I (MHC-I)-bound ligands play a pivotal role in CD8 T cell immunity and are hence of major interest in understanding and designing immunotherapies. One of the most commonly utilized approaches for detecting MHC ligands is LC-MS/MS. Unfortunately, the effectiveness of current algorithms to identify MHC ligands from LC-MS/MS data is limited because the search algorithms used were originally developed for proteomics approaches detecting tryptic peptides. Consequently, the analysis often results in inflated false discovery rate (FDR) statistics and an overall decrease in the number of peptides that pass FDR filters. Andreatta et al. describe a new scoring tool (MS-rescue) for peptides from MHC-I immunopeptidome datasets. MS-rescue incorporates the existence of MHC-I peptide motifs to rescore peptides from ligandome data. The tool is demonstrated here using peptides assigned from LC-MS/MS data with PEAKs software but can be deployed on data from any search algorithm. This new approach increased the number of peptides identified by up to 20-30% and promises to aid the discovery of novel MHC-I ligands with immunotherapeutic potential
