Human CD8+ T cells mediate protective immunity induced by a human malaria vaccine in human immune system mice
AbstractA number of studies have shown that CD8+ T cells mediate protective anti-malaria immunity in a mouse model. However, whether human CD8+ T cells play a role in protection against malaria remains unknown. We recently established human immune system (HIS) mice harboring functional human CD8+ T cells (HIS-CD8 mice) by transduction with HLA-A∗0201 and certain human cytokines using recombinant adeno-associated virus-based gene transfer technologies. These HIS-CD8 mice mount a potent, antigen-specific HLA-A∗0201-restricted human CD8+ T-cell response upon immunization with a recombinant adenovirus expressing a human malaria antigen, the Plasmodium falciparum circumsporozoite protein (PfCSP), termed AdPfCSP. In the present study, we challenged AdPfCSP-immunized HIS-CD8 mice with transgenic Plasmodium berghei sporozoites expressing full-length PfCSP and found that AdPfCSP-immunized (but not naïve) mice were protected against subsequent malaria challenge. The level of the HLA-A∗0201-restricted, PfCSP-specific human CD8+ T-cell response was closely correlated with the level of malaria protection. Furthermore, depletion of human CD8+ T cells from AdPfCSP-immunized HIS-CD8 mice almost completely abolished the anti-malaria immune response. Taken together, our data show that human CD8+ T cells mediate protective anti-malaria immunity in vivo
Onset of thermal convection in a rectangular parallelepiped cavity of small aspect ratios
Onset of thermal convection of a fluid in a rectangular parallelepiped cavity of small aspect ratios is examined both numerically and analytically under the assumption that all walls are rigid and of perfect thermal conductance exposed to a vertically linear temperature field. Critical Rayleigh number R c and the steady velocity and temperature fields of most unstable modes are computed by a Galerkin spectral method of high accuracy for aspect ratios A x and A y either or both of which are small. We find that if A x is decreased to 0 with A y being kept constant, R c increases proportionally to , the convection rolls of most unstable mode whose axes are parallel to the shorter side walls become narrower, and their number increases proportionally to . Moreover, as A x is decreased, we observe the changes of the symmetry of most unstable mode that occur more frequently for smaller A x . However, if is decreased to 0, although we again observe the increase in R c proportional to , we obtain only one narrow convection roll as the velocity field of most unstable mode for all A. The expressions of R c and velocity fields in the limit of or are obtained by an asymptotic analysis in which the dependences of R c and the magnitude and length scale of velocity fields of most unstable modes on A x and A y in the numerical computations are used. For example, R c is approximated by and in the limits of and , respectively. Moreover, analytical expressions of some components of velocity fields in these limits are derived. Finally, we find that for small A x or A the agreement between the numerical and analytical results on R c and velocity field is quite good except for the velocity field in thin wall layers near the top and bottom walls
Viscosity of peridotite liquid up to 13 GPa: Implications for magma ocean viscosities
The viscosity of synthetic peridotite liquid has been investigated at high pressures using in-situ falling sphere viscometry by combining a multi-anvil technique with synchrotron radiation. We used a newly designed capsule containing a small recessed reservoir outside of the hot spot of the heater, in which a viscosity marker sphere is embedded in a forsterite+enstatite mixture having a higher solidus temperature than the peridotite. This experimental setup prevents spheres from falling before a stable temperature above the liquidus is established and thus avoids difficulties in evaluating viscosities from velocities of spheres falling
through a partially molten sample. Experiments have been performed between 2.8 and 13 GPa at temperatures ranging from 2043 to 2523 K. Measured viscosities range from 0.019 (F0.004) to 0.13 (F0.02) Pa s. At constant temperature, viscosity increases with increasing pressure up to ~8.5
GPa but then decreases between ~8.5 and 13 GPa. The change in the pressure dependence of viscosity is likely associated with structural changes of the liquid that occur upon compression. By combining our results with recently published 0.1 MPa peridotite liquid viscosities [D.B. Dingwell, C. Courtial, D. Giordano, A. Nichols, Viscosity of peridotite liquid, Earth Planet. Sci. Lett. 226 (2004) 127–138.], the experimental data can be described by a non-Arrhenian, empirical Vogel-Fulcher-Tamman equation, which has been modified by adding a term to account for the observed pressure dependence of viscosity. This equation reproduces
measured viscosities to within 0.08 log10-units on average. We use this model to calculate viscosities of a peridotitic magma ocean along a liquid adiabat to a depth of ~400 km and discuss possible effects on viscosity at greater pressures and temperatures than experimentally investigated
Compressibility of pyrochlore-type MgZrSi2O7 determined by in situ X-ray diffraction in a large-volume high pressure apparatus
The pyrochlore-type MgZrSi2O7 has been investigated by in situ synchrotron energy-dispersive X-ray diffraction measurements up 24.3GPa at room temperature in a multianvil high pressure apparatus. The pressure has been calculated using an internal gold metal pressure calibrant. No phase transformation was observed in this study. A third-order BirchMurnaghan equation of state was used to fit the pressurevolume data. The fitting gives a volume of V-0=802.8 +/- 0.2 angstrom(3), an isothermal bulk modulus of K-T=249 +/- 12GPa, and first pressure derivative of K-T=3.2 +/- 1.3. If K-T is fixed as 4, K-T is obtained as 241.8 +/- 2.6GPa. Compared with zirconate or titanate pyrochlores, MgZrSi2O7 shows a smaller compressibility due to a larger cation radii ratio (r(Mg, Zr)/r(Si)).Physics, MultidisciplinarySCI(E)EI2ARTICLE11-73
<Article>The Original Map of Arrowsmith as a Main Source of 'Shintei Bankoku Zenzu' 「新訂万国全図」(Revised Map of the World)
Modern geography in Japan, the author considers, emerged in the early 1800's. 'Shintei Bankoku Zenzu', a representative work of the era, was an integrated result of the wide collecting of Chinese and European sources and, in addition, of the scientific investigation for proper land of Japan and of the explorations which they had been attempting so as to obtain geographical informations of northern terra incognita. In it is observed not only their passive acceptance of Chinese and European knowledge, but also the active engagement accompanied by their own 'langage de la géographie'. The author intends in this article to throw light on the date of publishing of A. Arrowsmith's map, which was a main source of 'Shintei Bankoku Zenzu', investigating into the cartographic elements of an extant Japanese translation of that map of the part north to Japan, that is the Sea of Okhotsk and Kamchatka, with its explanation, then to set forth the fact that scholars in Japan had already realized a remarkable 'langage de la géographie' on the world more than a half century before the Meiji 明治 Restoration
Oral vancomycin induces sustained deep remission in adult patients with ulcerative colitis and primary sclerosing cholangitis
Ulcerative colitis (UC) is a chronic inflammatory bowel disease of unknown etiology. The treatment of UC is challenging, especially when it is associated with primary sclerosing cholangitis (PSC), a chronic inflammatory disease of the bile ducts that affects around 5% of patients with UC, and leads to an increased risk of cholangiocarcinoma and colorectal cancer. Microbiota is considered to play an important role in the pathogenesis of UC, although the efficacy of antibiotics in this context is only limited and transient. Several studies have investigated the use of antibiotics for the treatment of PSC in adult and pediatric populations, with conflicting results. In this brief report, we describe the effect of oral vancomycin treatment in three patients with UC and PSC refractory to conventional and biologic therapies. All three patients achieved clinical remission and mucosal healing with vancomycin 500mg twice a day administered orally. Maintenance treatment with oral vancomycin was well tolerated and led to sustained clinical and endoscopic remission in all three patients. Oral vancomycin also improved liver function tests in two patients who did not have pre-existing cirrhosis
