A chloride- and calcium-dependent glutamate-binding protein from rat brain. Identification as a ubiquitous constituent of the inner mitochondrial membrane
No full textWe have recently solubilized and enriched a chloride- and calcium-dependent glutamate-binding protein from rat brain (Brose, N., Halpain, S., Suchanek, C., and Jahn, R. (1989) J. Biol. Chem. 264, 9619-9625). The partially purified protein fraction, containing two major protein components of 51,000 Da and 105,000 Da, was used to generate a rabbit antiserum. This serum quantitatively precipitated the binding activity from membrane extracts. Small amounts of the antiserum inhibited glutamate binding when chloride was absent from the incubation medium. Three protein bands were labeled by the serum on immunoblots. From the affinity purified antibody fractions contained in the serum, only the antibodies directed against a 51,000-Da protein were able to immunoprecipitate the binding activity, indicating that this protein is an essential component of the binding site. A survey of a variety of rat tissues by immunoblot analysis revealed a ubiquitous distribution of the protein. After subcellular fractionation of liver and brain, the 51,000-Da protein copurified with mitochondrial markers. Furthermore, exclusive labeling of mitochondria was observed by light and electron microscopy immunocytochemistry. Subfractionation of purified liver mitochondria resulted in a selective association of the protein with inner mitochondrial membranes. Pharmacological characterization of glutamate binding to liver mitochondrial membranes revealed a pattern almost identical to that of the chloride- and calcium-dependent glutamate-binding site in rat brain

lokal yang ada di kawasan sentral sepatu Cibaduyut. Dengan semangat meningkatkan
perekonomian lokal. Veny Brose berusaha mengangkat kembali eksistensi produk
sepatu dengan berbagai model yang sedang tren. Perusahaan produsen sepatu Veny
Brose bermula dari adanya peluang berdasarkan domilisi tempat tinggal yang ada
Cibaduyut, didirikan pada November tahun 2013, dengan kondisi saat itu hanya
memiliki dua orang pegawai, dan kegiatan produksi tersebut dikerjakan di rumah.
Dan saat ini mampu memiliki kurang lebih 15 pegawai serta memiliki tempat
produksi sendiri. Perushaan produsen sepatu Veny Brose masih menggunakan
metode manual atau handmade dengan mempekerjakan tenaga yang ahli
dibidangnya serta menggunakan material yang berkualitas. Kini produknya sudah
dipasarkan skala Nasional hingga Internasional dalam skala kecil melalui ekspor ke
Malaysia, Singapura dan Korea.
Penyusunan Business Plan ini bertujuan untuk merumuskan strategi
pengembangan perusahaan produsen sepatu Veny Brose melalui analisis lingkungan
eksternal dan analisis lingkungan internal, dengan metode analisis SWOT dan
diaplikasikan pada teori-teori perencanaan bisnis diantaranya Design Thinking,
Timmons Model, dan Business Model Canvas.
Hasil perhitungan kelayakan usaha menunjukkan bahwa analisis R/C Ratio
menunjukkan hasil yang positif sehingga termasuk dalam kategori usaha yang layak
untuk dikembangkan. Berdasarkan perhitungan payback period, investasi
perusahaan sepatu Veny Brose dapat diproyeksikan kembali dalam jangka waktu satu
tahun tiga bulan. Perusahaan ini juga menghasilkan NPV positif yang berarti
perusahaan ini layak untuk dikembangkan lebih lanjut. IRR menunjukkan bahwa
produsen sepatu Veny Brose layak untuk diterapkan dan dikembangkan karena
menghasilkan IRR yang lebih besar dari asumsi tingkat diskonto minimum yang
diperluka

No full textTato diplomová práce je zaměřena na zeštíhlení administrativních procesů ve sběru výrobních dat ve společnosti Brose CZ spol. s. r. o. Cílem je snížit pracnost a čas práce v procesu vykazování práce. Teoretická část obsahuje poznatky, které dále slouží jako podklad pro zpracování analytické a projektové části diplomové práce. Analytická část je nejprve zaměřena na analýzu jednotlivých článků v procesu vykazování, a dále na proces jako celek. V analýze byl zjištěn nedostatek v procesu vykazování, konkrétně s výkazy práce, které jsou velmi rozsáhlé a jejich zpracování časově náročné. V projektové části je vytvořen návrh nového výkazu práce, jeho nastavení s pomocí maker a standardy pro práci s ním, díky čemuž se podařilo práci s výkazy zkrátit a zjednodušit. Projekt je finančně i časově zhodnocen. Autorka uvádí i další návrhy, které lze využit v budoucnu.This diploma thesis is focused on lean administration in the process of production data collection in the company Brose CZ spol. s. r. o. The aim of this thesis is to reduce complicated and time-consuming work in process of work report. The theoretical part contains information which also serves as a basis for analytical and project part. The analytical part is first focused on analysis of each part in the process and continue as analysis of all processes. In analytical part have been found out problem with work statements including their big size and long time for their processing. Project part contains a new draft of work statement, its setting with macros, standards how to work with them. These changes will improve whole process, which is timely reduced and work statements are easier for processing. The project is financially and timely evaluated. No full textVeny Brose shoe manufacturing company is a local shoe product manufacturer in the Cibaduyut shoe centre area. The company started from an opportunity based on the domicile of the residence in Cibaduyut, established in November 2013, with conditions at that time only had two employees, and currently able to have approximately 15 employees and have its own production site. Now its products have been marketed on a national to international scale through exports to Malaysia, Singapore and Korea. The preparation of this Business Plan aims to formulate a development strategy for the Veny Brose shoe manufacturer company through external and internal environmental analysis, using the SWOT analysis method. The estimation of business feasibility shows that the R/C Ratio analysis shows positive results so that it is included in the category of businesses that are feasible to develop. Based on the payback period calculation, the investment of the Veny Brose shoe company can be projected to return within a period of one year and three months. The company also generates a positive NPV which means it is feasible to develop further. The IRR shows that it is feasible to implement and develop because it generates an IRR that is greater than the minimum required discount rate assumption

No full textDiplomová práce je zaměřena na projekt zavedení nástrojů štíhlé logistiky ve firmě Brose s.r.o. Teoretická část se soustřeďuje na koncept štíhlé výroby, teorii řízení zásob, na řízení výroby tlakem a tahem a na systém kanban. Tato část je východiskem pro část analytickou, v níž je provedena analýza současného stavu ve vybraných logistických procesech. V projektové části jsou formou případových studií řešeny návrhy pro zlepšení stávajícího stavu, které vedou k redukci plýtvání v dané logistické oblasti. V závěru každé případové studie je provedeno vyhodnocení projektu a srovnání současného a budoucího stavu, ze kterého vyplývají úspory a přínosy pro firmu.The master thesis is focused on a project of implementation of lean logstics tools in com-pany Brose, s. r. o. The theoretical part focuses on a concept of lean production, the theory of inventory management, the pull and push system and kanban system. This part is base for the practical part which concerns on the analysis of current state in selected logistics processes. In project part is focused in form of studies cases on suggestions for improving current state, which leads to reduction of wasting in chosen logistick area.
