    Dissection of Escherichia coli glutamate 5-kinase: Functional impact of the deletion of the PUA domain

    AbstractGlutamate 5-kinase (G5K) catalyzes the controlling first step of the synthesis of the osmoprotective amino acid proline, which feed-back inhibits G5K. Microbial G5K generally consists of one amino acid kinase (AAK) and one PUA (named after pseudo uridine synthases and archaeosine-specific transglycosylases) domain. To investigate the role of the PUA domain, we have deleted it from Escherichia coli G5K. We show that wild-type G5K requires free Mg for activity, it is tetrameric, and it aggregates to higher forms in a proline-dependent way. G5K lacking the PUA domain remains tetrameric, active, and proline-inhibitable, but the Mg requirement and the proline-triggered aggregation are greatly diminished and abolished, respectively, and more proline is needed for inhibition. We propose that the PUA domain modulates the function of the AAK domain, opening the way to potential PUA domain-mediated regulation of G5K; and that this domain moves, exposing new surfaces upon proline binding

    Durante la primera mitad del siglo XVI y en la corte salernitana del último príncipe de la casa Sanseverino y de su esposa, Isabel de Vilamarí (noble señora de origen catalán) se desarrolló un intenso clima intelectual. Allí se congregaron artistas y humanistas italianos y españoles. En este ambiente de intercambio cultural, atento en participar en las ideas de la Reforma que se difundió en Nápoles gracias a B. Ochino y a Valdés, nace el poema De principiis rerum del último académico pontaniano: Escipión Capece. En esta obra no sólo se rastrean motivos lucrecianos y virgilianos sino también el influjo de los tratados cosmológicos de Pontano. En este estudio, la autora propone el análisis de la figura y de la obra de Capece a través de sus lectoras: Isabel de Vilamarí y las mujeres cultas de su corte.During the first half of sixteenth century and in the Salernitan court of the last prince Sanseverino and his wife Isabel de Vilamarí (a lady coming from a noble Catalan family) an intense intellectual climate developed. Italian and Spanish artists and humanists met there. In this environment of cultural exchange, that shared in the Reform ideas divulged in Naples by B. Ochino and Valdés, Scipione Capece (the last member of the Pontanian Academy) writes his poem De principiis rerum. In his book Capece uses Latin literature (Vergil and Lucretius mainly) and Pontano's treatises on cosmology. The author of this paper studies Scipione Capece through his female readership: Isabel de Vilamarí and the learned women from her court

    Impulsivity and compulsivity in Alcohol Use Disorder – implications and comparison with Obsessive-Compulsive Disorder

    Trabalho Final do Mestrado Integrado em Medicina apresentado à Faculdade de MedicinaRecently, impulsivity and compulsivity have been defined as orthogonal and positively related constructs that present a relatively high prevalence, which justify its study. Alcohol use disorder/AUD and obsessive-compulsive disorder/OCD are examples of these types of perturbations, which demonstrate similarities and differences in the initial impulse, deregulation of inhibitory systems and perpetuation of behaviours. The objectives of this study were to compare the levels of impulsivity and compulsivity in patients with AUD and OCD, and in the first ones, to comprehend the role of dimensions of impulsivity and compulsivity in the severity of the disease.We performed an observational cross-sectional study. We recruited 40 patients diagnosed with OCD and 32 patients with AUD, who responded to a sociodemographic inquiry and the questionnaires Autism Spectrum Quotient for Adults, Obsessive Compulsive Inventory – Revised, Barratt Impulsiveness Scale and the Depression, Anxiety and Stress Scale 21. Additionally, the patients with AUD responded to Parkinson’s Disease Impulsive-Compulsive Disorders Questionnaire – Current Short and Alcohol Use Disorders Identification Test.Patients with AUD presented higher scores of impulsivity and of obsessive-compulsive symptoms than patients with Obsessive-Compulsive Disorder. In patients with AUD, the severity consumption was positively correlated with the motor and non-planning dimensions of impulsivity; these explained 28.1% and 15.9% of the variance of the symptoms’ severity (p<.05). Most patients with AUD reported suffering from symptoms of deregulation of impulse control, some of them explained by the levels of impulsivity (compulsive shopping) and of OC symptoms (compulsive shopping and hypersexuality).Although compulsivity has a high importance in the development of AUD, the largest report of it in this disease in comparison with OCD is due to socially prescribed perfectionism and auto-occultation, characteristic of patients with this last disease. Results support the orthogonal relation between compulsivity and impulsivity in AUD and also the fact of difficulty of reflection before acting and disorganization being predictors of alcohol consumption severity. The higher levels of obsessive-compulsive symptoms and its role in explaining impulse control disorders (hypersexuality and compulsive shopping), in AUD patients, suggest these people not only have higher predisposition to initiate dysfunctional behaviours (alcohol consumption) but also difficulty to stop those behaviours. Additionally, these results highlight the diversity of AUD symptoms.AUD is phenotypically heterogeneous, encompassing manifestations of impulsivity and compulsivity spectrum. The conceptualization of AUD as an impulsivity/compulsivity disorder has diagnostic, therapeutic and sociocultural implications.A impulsividade e a compulsividade têm sido definidas recentemente como construtos ortogonais e positivamente relacionados, nomeadamente nas perturbações da impulsividade/compulsividade, que apresentam uma prevalência relativamente elevada, o que justifica o seu estudo. São exemplos deste tipo de perturbações, a Perturbação por Uso de Álcool/PUA e a Perturbação Obsessivo-Compulsiva/POC, que apresentam entre si semelhanças e diferenças no impulso inicial, desregulação dos sistemas inibitórios e perpetuação de comportamentos. Os objetivos deste estudo foram comparar níveis de impulsividade e de compulsividade entre doentes com PUA e POC e, nos primeiros, compreender o papel de dimensões da impulsividade e da compulsividade na gravidade da doença.Realizámos um estudo observacional, correlacional e transversal. Recrutámos 40 doentes com o diagnóstico de POC e 32 com o diagnóstico de PUA, que responderam a um inquérito sociodemográfico e aos questionários Barratt Impulsiveness Scale, Obsessive-Compulsive Inventory – Revised e Escala de Ansiedade, Depressão e Stresse 21. Adicionalmente, os doentes com PUA responderam aos questionários Parkinson’s Disease Impulsive-Compulsive Disorders Questionnaire – Current Short e Alcohol Use Disorders Identification Test.Os doentes com PUA apresentaram pontuações mais elevadas de impulsividade e de sintomas obsessivo-compulsivos do que os doentes com POC. Nos doentes com PUA, a gravidade dos consumos correlacionou-se positivamente com as dimensões motora e não-planeamento da impulsividade; estas explicaram 28,1% e 15,9% da variância da gravidade dos consumos (p<0,05). A maioria dos doentes com PUA relatou sofrer de sintomas de desregulação do controlo dos impulsos, alguns dos quais foram explicados pelos níveis de impulsividade (compras compulsivas) e de sintomas obsessivo-compulsivos (compras compulsivas e hipersexualidade).Apesar de a compulsividade ter uma elevada importância no desenvolvimento da PUA, o maior relato da mesma nesta doença em relação à POC dever-se-á ao perfeccionismo socialmente prescrito e auto-ocultação característicos destes últimos doentes. Os resultados suportam a relação ortogonal da impulsividade e compulsividade na PUA e ainda o facto de a dificuldade em refletir antes de agir e a desorganização serem preditores da gravidade dos consumos alcoólicos. Os níveis elevados de sintomas obsessivo-compulsivos e o seu papel na explicação das perturbações do controlo do impulso (hipersexualidade e compras compulsivas), na PUA, indicam que estas pessoas têm maior predisposição para iniciar comportamentos disfuncionais (consumo de bebidas alcoólicas), e maior dificuldade em pará-los. Adicionalmente, estes resultados apontam para a diversidade sindromática da PUA.A PUA é uma perturbação fenotipicamente heterogénea, abarcando manifestações do espetro impulsivo e compulsivo. A sua conceptualização como perturbação impulsiva/compulsiva tem implicações diagnósticas, terapêuticas e socioculturais
