Islamic Education Movie: Character Learning Through Nussa-Rara Movie
This article examines the Islamic cartoon film Nussa and Rara. The author used Nussa and Rara as a window to see the emergence of Islamic cartoon films. Its strengths are in conveying educational values to children and as a learning medium to build character education for children aged 12 years and under. To explain this phenomenon, the author employed the content analysis method and combined it with relevant data; those were strengthened by many research results and literature studies. Through this method, the author showed that Nussa and Rara movie shares Islamic value in each segment, such as the value of honesty, asking for prayers on time, respecting neighbors, and so on. The author tried to see how every figure, especially Umma as Nussa and Rara’s mother, explains and exemplifies how to be a good Muslim. Through this article, it could be concluded that films are not always judged to have a negative impact on the audience. Even films can contribute as a medium for education and dissemination of Islamic da'wah. Besides, through films, it can also contribute to building good character for children under 12 years of age. Some learning methods that can be used are the many steps or methods that can be planted in children, such as the habituation and storytelling methods. These methods can be used to apply the values of faith, moral values, and worship values. Thus, this article greatly contributes to enriching academic discourse related to character learning for children under 12 years of age. This article concluded that the Nussa and Rara cartoon film really strives to spread Islamic da'wah and build children's character from an early age with daily customs and begin to familiarize children to apply the values of Islamic teachings perfectly

PML-RARA mutations confer varying arsenic trioxide resistance
Acute promyelocytic leukemia (APL) caused by the malignant proliferation of bone marrow-derived cells, which is driven by the chromosomal translocation, leading to the fusion of the promyelocytic leukemia gene (PML) and the retinoic acid receptor-a gene (RARA) (de The et al., 1990; Goto et al., 2011). The oncogenic protein PML-RARA has been found specifically in more than 95% APL patients and is crucial for both the pathogenesis and therapeutics of APL(de The et al., 1990; Zhou et al., 2007; Wang and Chen, 2008). Arsenic trioxide (AS203), an ancient traditional Chinese medicine, has recently been adopted as the first-line treatment for APL patients by the National Comprehensive Cancer Network (Wang and Chen, 2008; Zhu and Huang, 2014), which greatly improved the 5-year overall survival of APL patients (Li et al., 2002; Zhou et al., 2007; Wang and Chen, 2008; Lo-Coco et al., 2013). Mounting biological studies revealed that AS203 cures APL by initiating PML-RARA degradation through specific binding to the PML moiety (Lallemand-Breitenbach et al., 2001; Zhang et al., 2010). Mechanistically, AS203 binds directly to the dicysteine residues C212/213 located in the B2 domain of PML, which induces PML oligomerization, NB formation, SUMOylation, and ultimate degradation of PML (Lallemand-Breitenbach et al., 2008; Jeanne et al., 2010; Zhang et al., 2010).National Natural Science Foundation of China [31470754, 31670786]; National High Technology and Development Program of China [2016YFC1302400]; Doctoral Fund of Ministry of Education of China [20130001130003]SCI(E)中国科学引文数据库(CSCD)LETTER4296-301

Varying responses of PML-RARA with different genetic mutations to arsenic trioxide
Resistance to arsenic and/or all-trans retinoic acid (ATRA) is a challenging problem in the clinical management of acute promyelocytic leukemia (APL). Acquired genetic mutations in the PML moiety of the PML-RARA fusion gene are found in some patients with relapsed/refractory APL. Whether all of the identified point mutations play a role and have a similar function in the mechanisms of arsenic resistance remains unknown. Here we performed in vitro functional analyses and a retrospective analysis of APL patients to investigate the effect of PML-RARA mutations in mediating resistance to arsenic trioxide. Among the 5-point mutations in the PML part of PML-RARA identified in patients with relapsed APL, we found that A216V, S214L, and A216T mutations could attenuate the negative regulation of arsenic on PML-RARA, resulting in the retention of oncoproteins. In contrast, L217F and S220G mutations functioned weakly in this context. Furthermore, we demonstrated that either increasing the concentration of arsenic trioxide or combining it with ATRA could overcome the mutation-triggered arsenic resistance in vitro. In addition to presenting more evidence to reinforce the correlation of genetic mutations in PML-RARA with arsenic efficacy, we provide novel insight into the functional difference of acquired mutations of PML-RARA both in vitro and in the clinical setting. Our findings may help predict the prognosis and select more effective strategies during APL therapy.SCI(E)[email protected]
The Rara of Léogâne, a ritual practice in Haitian communities, has been the subject of several reworkings in the course of social evolution. The events made up of music, dance and ritual procedures are organized as a group and orchestrated by leaders. It is they who first changed the image of this unique cultural practice at the first Rara Festival in 1992. The study of their communications with the media at the time of this first consolidating event, shows a cultural mediation in connection with the public, which found support on the part of all the players in social governance, the mayor’s office, members of parliament and the Ministry of Culture. In his article, the author muses about the processes which have conveyed on Rara a territorial heritage of identity, and he brings to light the importance of the tourist potential latent in this event as a developmental resource in an area torn apart by repeated human confrontations and natural disasters
FIP1L1 presence in FIP1L1-RARA or FIP1L1-PDGFRA differentially contributes to the pathogenesis of distinct types of leukemia
FIP1-like 1 (FIP1L1) is associated with two leukemogenic fusion genes: FIP1L1-retinoic acid receptor alpha (RARA) and FIP1L1-platelet-derived growth factor receptor alpha (PDGFRA). Analyses of a series of deletion mutants revealed that the FIP1 motif in FIP1L1-RARA plays a pivotal role in its homodimerization and transcriptional repressor activity. However, in FIP1L1-PDGFRA, the C-terminal PDGFRA portion possesses the ability of forming a homodimer by itself, making FIP1L1 dispensable for constitutive activation of this kinase. Both the full-length and the C-terminal PDGFRA portion of FIP1L1-PDGFRA could transform the IL-3-dependent hematopoietic cell line, BAF-B03. Moreover, when either the full-length or the C-terminal PDGFRA portion of FIP1L1-PDGFRA was introduced in these cells, they grew in the absence of IL-3. The cells having the C-terminal PDGFRA portion of FIP1L1-PDGFRA, however, were partially IL-3 dependent, whereas the cells having the full-length FIP1L1-PDGFRA became completely IL-3 independent for their growth. Taken together, these results show that FIP1L1 differentially contributes to the pathogenesis of distinct types of leukemia
