    Survival rate and load to failure of premolars restored with inlays: An evaluation of different inlay fabrication methods

    Statement of problem: Studies that evaluate the survival rate and load to fracture of premolars restored with inlays produced using different methods are lacking. Purpose: The purpose of this in vitro study was to compare the survival rate and fracture load of premolars restored with inlays fabricated using different methods. Material and methods: Thirty maxillary premolars were selected, embedded, and prepared to receive inlays fabricated using different methods (n=10): LaCom-digital scanning with Lava C.O.S. scanner (3M ESPE), followed by milling of composite resin block (Lava Ultimate; 3M ESPE) in a milling unit; CeCom-digital scanning with Cerec 3D Bluecam scanner (Dentsply Sirona), followed by milling of a Lava Ultimate block in Cerec (Dentsply Sirona); PresDis-impression with polyvinyl siloxane, inlay made using the lost wax technique, and IPS e.max Press (Ivoclar Vivadent AG) pressed ceramic (lithium disilicate). A dual-polymerizing resin cement system was used to lute the inlays. Inlays were mechanically cycled (2 Hz, 106 mechanical pulses, 80 N) after 24 hours, and the specimens were stored in distilled water at 37°C for 11 months. Then, a fatigue test was conducted using a 10-Hz frequency and 400-N load on the inner inclines of the cusps. The test was complete when the specimen fractured or when the specimen reached 1.5×106 cycles. The specimens that survived fatigue testing were submitted to a single-load fracture test in a universal testing machine and analyzed using a stereoscope for failure classification. Survival rates were estimated using the Kaplan-Meier method and log-rank test (Mantel-Cox). Fracture load data were analyzed using 1-way ANOVA (α=.05). Results: No significant differences were detected among the groups for the survival rate (P=.87) or for the load to fracture (P=.78). Most failures were longitudinal, catastrophic fractures. Conclusions: Premolars restored with inlays fabricated using the tested methods had similar survival rates and loads to fracture

    Effect of the frequency of mechanical pulses for fatigue aging testing on push-out bond strength between glass fiber posts and root dentin

    The aim of this study was to evaluate the influence of the frequency of mechanical pulses during mechanical fatigue aging on bond strength between glass fiber posts and root dentin. Fiber posts were adhesively cemented in 30 bovine roots and the core was built up with composite. All specimens were subjected to mechanical cycling (45 degrees angle; 37 degrees C; 50N; 2x10(6) pulses) at different frequencies: 2, 4, and 8Hz. After the fatigue, each specimen was submitted to push out test. The bond strength was calculated for data analysis (one-way ANOVA, p=0.05). The frequency did not affect the push-out (p=0.7). The main failure mode was between dentin and cement in all groups. The mechanical pulses were not influenced by the bond strength between root dentin and fiber posts. Thus, it is possible to decrease the time-consuming in vitro tests involving posts without damaging the reliability of the test

    Mechanical behavior of a Y-TZP ceramic for monolithic restorations: effect of grinding and low-temperature aging

    This study aimed to investigate the effects of grinding with diamond burs and low-temperature aging on the mechanical behavior (biaxial flexural strength and structural reliability), surface topography, and phase transformation of a Y-TZP ceramic for monolithic dental restorations. Disc-shaped specimens (Zirlux FC, Ivoclar Vivadent) were manufactured according to ISO 6872 (2008) and divided in accordance with two factors: “grinding — 3 levels” and “LTD — 2 levels”. Grinding was performed using a contra-angle handpiece under constant water-cooling with different grit-sizes (extra-fine and coarse diamond burs). LTD was simulated in an autoclave at 134 °C, under a pressure of 2 bar, over a period of 20 h. Surface topography analysis showed an increase in roughness based on surface treatment grit-size (Coarse > Xfine > Ctrl), LTD did not influence roughness values. Both grinding and LTD promoted an increase in the amount of m-phase, although different susceptibilities to degradation were observed. According to existing literature the increase of m-phase content is a direct indicative of Y-TZP degradation. Weibull analysis showed an increase in characteristic strength after grinding (Coarse = Xfine > Ctrl), while for LTD, distinct effects were observed (Ctrl < Ctrl LTD; Xfine = Xfine LTD and Coarse = Coarse LTD). Weibull moduli were statistically similar between all tested groups. Within the limits of this current study, it was feasible to notice that both aging in autoclave for 20 h (LTD) and grinding showed not to be detrimental to the mechanical properties of Zirlux FC Y-TZP ceramic

    Effect of Grinding and Multi-Stimuli Aging on the Fatigue Strength of a Y-TZP Ceramic

    Abstract This study aimed to investigate the effect of grinding and multi-stimuli aging on the fatigue strength, surface topography and the phase transformation of Y-TZP ceramic. Discs were manufactured according to ISO-6872:2008 for biaxial flexure testing (diameter: 15 mm; thickness: 1.2 mm) and randomly assigned considering two factors “grinding” and “aging”: C- control (as-sintered); CA- control + aging; G- ground; GA- ground + aging. Grinding was carried out with coarse diamond burs under water-cooling. Aging protocols consisted of: autoclave (134°C, 2 bars pressure, 20 hours), followed by storage for 365 days (samples were kept untouched at room temperature), and by mechanical cycling (106 cycles by 20 Hz under a load of 50% from the biaxial flexure monotonic tests). Flexural fatigue strengths (20,000 cycles; 6 Hz) were determined under sinusoidal cyclic loading using staircase approach. Additionally, surface topography analysis by FE-SEM and phase transformation analysis by X-ray Diffractometry were performed. Dixon and Mood methodology was used to analyze the fatigue strength data. Grinding promotes alterations of topographical pattern, while aging apparently did not alter it. Grinding triggered t-m phase transformation without impacting the fatigue strength of the Y-TZP ceramic; and aging promoted an intense t-m transformation that resulted in a toughening mechanism leading to higher fatigue strength for as-sintered condition, and a tendency of increase for ground condition (C &lt; CA; G = GA). It concludes that grinding and aging procedures did not affect deleteriously the fatigue strength of the evaluated Y-TZP ceramic, although, it promotes surface topography alterations, except to aging, and t-m phase transformation.</jats:p

    Mechanical behavior of yttria-stabilized tetragonal zirconia polycrystal: Effects of different aging regimens

    Abstract This study aimed to characterize and compare the effect of different aging regimens on surface characteristic (topography and roughness), structural stability (phase transformation) and mechanical performance (Weibull analysis) of a Y-TZP ceramic. Discs (15 × 1.2 mm; VITA In-Ceram YZ) were prepared according to ISO 6872-2015 for biaxial flexural strength testing and randomly assigned into five groups (n = 30): as-sintered, no aging treatment (CTRL); 20 h in autoclave at 134°C, 2 bar pressure (AUT); intermittent mechanical loading at 20 Hz/106 load pulses (MechLoad); AUT followed by MechLoad (AUT+MechLoad); and storage in distilled water at 37°C, for 1 year (STO). The following analyses were performed: roughness (n = 30), surface topography (n = 2), phase transformation (n = 2) and biaxial flexure strength (n = 30). Phase transformation (increase of m-phase content) was shown to be a spontaneous, unavoidable and time-dependent process, occurring even under ambient conditions (dry storage after 1 year = 6.0% increase), and is considerably accelerated in the presence of moisture (STO = 17.6%; AUT= 63.1%; and AUT+MechLoad = 59.9%). For roughness parameters, only Ra was affected by aging, and the highest values were observed for AUT+MechLoad (0.25 ± 0.07 µm). For Weibull analysis, structural reliability (Weibull moduli) and characteristic strength were not impaired after aging, and some aging conditions led to increased values (highest weibull moduli in AUT, and highest characteristic strength in STO). Phase transformation proves to be a time-dependent spontaneous mechanism that is accelerated in the presence of different stimuli. However, none of the aging regimens had a negative effect on the characteristic strength and structural reliability of Y-TZP ceramic

    O Pan-Africanismo na obra O Quilombismo de Abdias Nascimento

    The general objective of this research is to describe how Pan-Africanism is incorporated in the concept of quilombismo, created by Abdias Nascimento. To achieve this objective, at first, a bibliographic research is carried out on the history and concept of Pan-Africanism and its main aspects. Subsequently, we will demonstrate Nascimento's trajectory, emphasizing moments that were important for his contact with the Pan-Africanist movement, such as his self-exile, between the years of 1968-1981. During which time he participated in several Pan-African congresses and meetings, which can be found in biographies, sociological studies, collections and in his own books. In a second moment, wishing not only to summarize the importance of the intellectual as a militant, but also to analyze within a theoretical-scientific perspective, his work O Quilombismo: documents of a pan-Africanist militancy (1980) shows us important aspects about the incorporations and transformations that occurred with the author's thought in contact with the Pan-Africanist ideology, and how these notions were applied to the understanding of the reality of black people in the Brazilian context. In this way, it was perceived that Nascimento's relationship with Pan-Africanism radicalized his voice for the causes of Afro-Brazilians, ending up revealing the reality of Africans and Afro-descendants in Brazil. Therefore, the intellectual production of this author is an important contribution to the recognition and appreciation of the history, cultural manifestations and memories of the Afro-Brazilian population. In addition, it was found that the intellectual developed the project of the National Quilombist State, an alternative model for a free, fair, egalitarian and sovereign Brazilian society reorganizationO objetivo geral da presente pesquisa ? descrever como o pan-africanismo est? incorporado no conceito de quilombismo, este criado por Abdias Nascimento. Para atingir esse objetivo, em um primeiro momento ? realizada uma pesquisa bibliogr?fica sobre a hist?ria e conceito de pan-africanismo e suas principais vertentes. Posteriormente, demonstraremos a trajet?ria de Nascimento, dando ?nfase a momentos que foram importantes para o seu contato com o movimento pan-africanista, como, por exemplo, seu autoex?lio, entre os anos de 1968-1981. Per?odo no qual participou de diversos congressos e encontros pan-africanos, que podem ser encontrados em biografias, estudos sociol?gicos, acervos e nos seus pr?prios livros. Em um segundo momento, desejando n?o apenas a se resumir em expor a import?ncia do intelectual como militante, mas tamb?m analisar adentro de uma perspectiva te?rico-cient?fica, a sua obra O Quilombismo: documentos de uma milit?ncia pan- africanista (1980) nos mostra aspectos importantes sobre as incorpora??es e transforma??es que ocorreram com o pensamento do autor em contato com a ideologia pan-africanista, e como essas no??es foram aplicadas ao entendimento acerca da realidade do negro no contexto brasileiro.     Polishing of Ground Y-TZP Ceramic is Mandatory for Improving the Mechanical Behavior

    Abstract It evaluated the effect of aging by Low Temperature Degradation (LTD), executed after post- processing surface treatments (polishing, heat treatment and glazing), on the surface characteristics (micromorphology and roughness) and on the structural stability (phase transformation and mechanical behavior-flexural strength and structural reliability) of a ground yttrium-stabilized tetragonal zirconia polycrystal (Y-TZP) ceramic. Discs of Y-TZP (VITA In-Ceram YZ) were manufactured (ISO:6872-2015; 15 mm in diameter and 1.2 ± 0.2 mm in thickness) and randomly assigned into 10 groups according two factors: “aging” in 2 levels (with or without) and “surface treatment” in 5 levels (Ctrl: as-sintered; Gr: grinding with coarse diamond bur; Gr + HT: grinding plus heat treatment; Gr + Pol: grinding plus polishing; Gr + Gl: grinding plus glazing). Roughness (n=30), biaxial flexural test (n=30), phase transformation (n=2), and surface topography (n=2) analyses were performed. Aging led to an intense increase in monoclinic (m) phase content for all the tested conditions, being the as-sintered samples (Ctrl= 65.6%) more susceptible to the t-m phase transformation. Despite of increasing the m-phase content, aging was not detrimental for characteristic strength (except to the grinding condition). There was no significant reduction in the Weibull modulus after surface treatments. Additionally, heat treatment and glazing after grinding led to a decrease in characteristic strength, while polishing presented the highest characteristic strength values. Thus, polishing is mandatory after grinding the Y-TZP ceramic, while performing glazing or heat-treatment alone after grinding lead to the worst mechanical performance.</div

    Silicone Disclosing Material used after Ceramic Surface Treatment Reduces Bond Strength

    Purpose: To evaluate the effect of a silicone disclosing procedure performed at different timepoints on the shear bond strength (SBS) of cements (self- adhesive composite cement, self-etch composite cement, resin-reinforced glass-ionomer cement) to different substrates (zirconia, lithium disilicate, bovine dentin). Materials and Methods: The substrate/cement combinations were assigned to two groups (n = 15) according to the timepoint, at which the vinyl polyether silicone disclosing agent was applied: after (experimental groups, EXP) or before (control groups, CTRL) specific micromechanical treatments of the substrate surface. To increase standardization, the cements were applied into rubber rings (2.2 mm diameter x 1.0 mm thickness) positioned on the substrate surface. After luting procedures, all specimens were stored in 37°C distilled water for 24 h, then subjected to SBS testing using a wire loop of 0.2 mm diameter at a crosshead speed of 1 mm/min until failure. Failure analysis was performed for all tested specimens. SBS data were submitted to Weibull analysis. Results: The silicone disclosing procedure performed after micromechanical surface treatment reduced the characteristic shear bond strength to zirconia and lithium disilicate when compared to CTRL. However, for dentin specimens, there was no significant difference between CTRL and EXP for any of the cements investigated. Failure analysis showed a predominance of interfacial failures. Conclusion: The silicone disclosing procedure performed after the micromechanical treatment of ceramic surfaces negatively affected the cement bond strength. Therefore, after using it to check the fit of a prosthesis, clinicians should carefully clean the ceramic surface

    Environmental toxicity, redox signaling and lung inflammation:the role of glutathione

    Glutathione (gamma-glutamyl-cysteinyl-glycine, GSH) is the most abundant intracellular antioxidant thiol and is central to redox defense during oxidative stress. GSH metabolism is tightly regulated and has been implicated in redox signaling and also in protection against environmental oxidant-mediated injury. Changes in the ratio of the reduced and disulfide form (GSH/GSSG) can affect signaling pathways that participate in a broad array of physiological responses from cell proliferation, autophagy and apoptosis to gene expression that involve H(2)O(2) as a second messenger. Oxidative stress due to oxidant/antioxidant imbalance and also due to environmental oxidants is an important component during inflammation and respiratory diseases such as chronic obstructive pulmonary disease, idiopathic pulmonary fibrosis, acute respiratory distress syndrome, and asthma. It is known to activate multiple stress kinase pathways and redox-sensitive transcription factors such as Nrf2, NF-kappaB and AP-1, which differentially regulate the genes for pro-inflammatory cytokines as well as the protective antioxidant genes. Understanding the regulatory mechanisms for the induction of antioxidants, such as GSH, versus pro-inflammatory mediators at sites of oxidant-directed injuries may allow for the development of novel therapies which will allow pharmacological manipulation of GSH synthesis during inflammation and oxidative injury. This article features the current knowledge about the role of GSH in redox signaling, GSH biosynthesis and particularly the regulation of transcription factor Nrf2 by GSH and downstream signaling during oxidative stress and inflammation in various pulmonary diseases. We also discussed the current therapeutic clinical trials using GSH and other thiol compounds, such as N-acetyl-l-cysteine, fudosteine, carbocysteine, erdosteine in environment-induced airways disease

    No preconscious attentional bias towards itch in healthy individuals

    Rapidly attending towards potentially harmful stimuli to prevent possible damage to the body is a critical component of adaptive behavior. Research suggests that individuals display an attentional bias, i.e., preferential allocation of attention, for consciously perceived bodily sensations that signal potential threat, like itch or pain. Evidence is not yet clear whether an attentional bias also exists for stimuli that have been presented for such a short duration that they do not enter the stream of consciousness. This study investigated whether a preconscious attentional bias towards itch-related pictures exists in 127 healthy participants and whether this can be influenced by priming with mild itch-related stimuli compared to control stimuli. Mild itch was induced with von Frey monofilaments and scratching sounds, while control stimuli where of matched modalities but neutral. Attentional bias was measured with a subliminal pictorial dot-probe task. Moreover, we investigated how attentional inhibition of irrelevant information and the ability to switch between different tasks, i.e., cognitive flexibility, contribute to the emergence of an attentional bias. Attentional inhibition was measured with a Flanker paradigm and cognitive flexibility was measured with a cued-switching paradigm. Contrary to our expectations, results showed that participants attention was not biased towards the itch-related pictures, in facts, attention was significantly drawn towards the neutral pictures. In addition, no effect of the itch-related priming was observed. Finally, this effect was not influenced by participants' attentional inhibition and cognitive flexibility. Therefore, we have no evidence for a preconscious attentional bias towards itch stimuli. The role of preconscious attentional bias in patients with chronic itch should be investigated in future studies.Applied Ergonomics and Desig
