No full textBackground: In recent years, considerable knowledge has been gained on the molecular mechanisms underlying retinal cell fate specification. However, hitherto studies focused primarily on the six major retinal cell classes (five types of neurons of one type of glial cell), and paid little attention to the specification of different neuronal subtypes within the same cell class. In particular, the molecular machinery governing the specification of the two most abundant neurotransmitter phenotypes in the retina, GABAergic and glutamatergic, is largely unknown. In the spinal cord and cerebellum, the transcription factor Ptf1a is essential for GABAergic neuron production. In the mouse retina, Ptf1a has been shown to be involved in horizontal and most amacrine neurons differentiation.Results: In this study, we examined the distribution of neurotransmitter subtypes following Ptf1a gain and loss of function in the Xenopus retina. We found cell-autonomous dramatic switches between GABAergic and glutamatergic neuron production, concomitant with profound defects in the genesis of amacrine and horizontal cells, which are mainly GABAergic. Therefore, we investigated whether Ptf1a promotes the fate of these two cell types or acts directly as a GABAergic subtype determination factor. In ectodermal explant assays ... Full text linkThe adult frog retina retains a reservoir of active neural stem cells that contribute to continuous eye growth throughout life. We found that Yap, a downstream effector of the Hippo pathway, is specifically expressed in these stem cells. Yap knock-down leads to an accelerated S-phase and an abnormal progression of DNA replication, a phenotype likely mediated by upregulation of c-Myc. This is associated with an increased occurrence of DNA damage and eventually p53-p21 pathway-mediated cell death. Finally, we identified PKNOX1, a transcription factor involved in the maintenance of genomic stability, as a functional and physical interactant of YAP. Altogether, we propose that YAP is required in adult retinal stem cells to regulate the temporal firing of replication origins and quality control of replicated DNA. Our data reinforce the view that specific mechanisms dedicated to S-phase control are at work in stem cells to protect them from genomic instability

No full textFace à l’œuvre essentiellement romanesque de Philippe Forest, commencée à la mort de son enfant de quatre ans et qui se poursuit dans l'énonciation et la circonscription sans fin de cette mort, la parole critique rencontre une double résistance : celle de la compassion, au risque de se perdre dans une sympathie muette qui compromet l'analyse, immédiatement inspirée par une œuvre éminemment personnelle dans son témoignage et sa souffrance ; celle aussi de la portée éthique d'un tel projet. Comment écrire sur l'enfant mort de quelqu'un d'autre ? Face au paradoxe d'un deuil intime qui se donne à lire, tenir un discours critique qui rende justice à l'écriture sans renoncer à creuser la douleur du texte relève de la gageure.Le livre de Maïté Snauwaert ne cherche pas à masquer ces écueils, il les envisage avec sensibilité et rigueur critiques. Elle livre ici un essai riche et profus, qui puise ses analyses aux sources multiples de la littérature comme de la psychologie ou de la sociologie, du romanesque comme de l'essai de poétique, des grandes figures occidentales (Barthes, Benjamin, Agamben, Ricoeur, Kierkegaard) à celles de l'Orient qui siègent haut dans le panthéon personnel de Forest (Kobayashi Issa, Kenzaburô Ôé)

No full textLa France est le premier pays d’Europe à avoir légiféré sur l’interdiction des signes religieux à l’école. Ce mémoire propose d’expliquer comment la loi du 15 mars 2004 s’est imposée comme une nécessité en France. L’étude porte d’abord sur la construction du port du foulard islamique dans les écoles comme problème public et politique en retraçant l’ « affaire du voile » de 1989 à 2003. Une analyse en profondeur de la Commission Stasi éclaire ensuite les modalités de la production d’un consensus entre ses membres et démontre le rôle essentiel que la Commission Stasi a joué dans la production de la loi en permettant l’adhésion de l’opinion publique.France was the first European country to pass a law prohibiting Islamic headscarves in its public schools. The purpose of this thesis is to explain how the law voted in March, 15th 2004 has become a necessity in France. The first part, based on the period from 1989 to 2003 will demonstrate how the Islamic headscarf in schools has turned into a public and political problem known as the headscarf “affaire”. Then, the second part will provide an in depth analysis about the “Commission Stasi” revealing the production of a consensus among its members and showing its influence on the public opinion and its leading role in the production of the law
