    Pheochromocytomas (PCCs) are tumors arising from neural crest-derived chromaffin cells. There are currently few animal models of PCC that recapitulate the key features of human tumors. Since such models may be useful for investigations of molecular pathomechanisms and development of novel therapeutic interventions, we characterized a spontaneous animal model (MENX rats) that develops endogenous PCCs with complete penetrance. Urine was longitudinally collected from wild-type (wt) and MENX-affected (mutant) rats and outputs of catecholamines and their O-methylated metabolites determined by mass spectrometry. Adrenal catecholamine contents, cellular ultrastructure and expression of phenylethanolamine N-methyltransferase (PNMT), which converts norepinephrine to epinephrine, were also determined in wt and mutant rats. Blood pressure was longitudinally measured and end-organ pathology assessed. Compared to wt rats, mutant animals showed age-dependent increases in urinary outputs of norepinephrine (P=0.0079) and normetanephrine (P=0.0014) that correlated in time with development of tumor nodules, increases in blood pressure and development of hypertension-related end-organ pathology. Development of tumor nodules, which lacked expression of PNMT, occurred on a background of adrenal medullary morphological and biochemical changes occurring as early as 1 month of age and involving increased adrenal medullary concentrations of dense cored vesicles, tissue contents of both norepinephrine and epinephrine and urinary outputs of metanephrine, the metabolite of epinephrine. Taken together, MENX-affected rats share several biochemical and pathophysiological features with PCC patients. This model thus provides a suitable platform to study the pathogenesis of PCC for preclinical translational studies aimed at development of novel therapies for aggressive forms of human tumors

    Postmodern Aspects of Mirko Kovač`s Novels

    This paper deals with postmodern techniquesin of the novels of Mirko Kovač, a prominent figure in the postmodern literature. In particular, the study examinesnarrative strategies which deviate from traditional paradigms and yet create a distinct reality of the text. Postmodern principles such as “the poetic narrator ” (and the “poetic reading ” associated with it), temporal distortion, the questioning of historical facts, hybridization of narrative texts, metatextuality and citationality, documentation, fragmentation and text segmentation will be addressed using concrete examples. Mirko Kovač stands out with his distinctive poetics which contains elements of postmodernism, intertwined with traces of other poetics. In this paper we will dwell precisely on the postmodern aspects of his texts. In weaving the fabric of his texts, the author employs narrative strategies that threaten and break the traditional conventional narrative with its prominent elements such as narrative instance, time, space, composition, characters. All canonised forms and values are reviewed and dissolved in a specific way. In his writings, Kovač makes use of specific forms of citation and documentarity. Special attention in our work shall be devoted to Kovač’s novelistic structures, in which he as an author spans a full creative range. We will try to shed light on all phenomena of the narrative text which are developed in terms of the postmodern

    Akademik Mirko Malez, ne samo što je rođen u Ivancu, on je kao čovjek, osobito planinar i zaljubljenik u prirodu, bio duboko vezan uz Ivanec i Ivanščicu, ali i uz cijeli Ivanečki kraj. Ivančani su mu to podjednako vraćali, jednom, zastupljenošću u nekoliko godišta Ivanečkog kalendara, a poslije njegove smrti i dodjelom njegova imena jednoj od četiri ulice ivanečkog cestovnog križa, onog što vodi od gradskog središta prema Lepoglavi. U Ivanečkim kalendarima, svojevremenoj „ivanečkoj enciklopediji u nastavcima“, on se spominje u različitom kontekstu. Spominje se među poznatim ljudima Ivanečkog kraja ili pak u prilozima bibliografiji Ivanca. Nezaobilazan je u objavljenim temama Paleolit, Neolit i špilja Vindija. Posebno valja ukazati na tekstove koje je pisala Malezova suradnica, pokojna Marija Poje, također Ivančanka. Na kraju, Ivanečki kalendar se u svom posljednjem godištu oprostio od Mirka Maleza tekstom uvaženog akademika Milana Heraka.The academician Mirko Malez was not only born in Ivanec, but as a mountaineer and lover of nature, he was deeply attached to Ivanec and Ivanščica, and to the entire area of Ivanec. In return, Ivanec included him in several volumes of Ivanec calendar. After his death, his name was assigned to one of four streets of Ivanec cross road, the one which leads from the centre of town to Lepoglava. In Ivanec calendars, a kind of „encyclopedia of Ivanec” he is mentioned in a different context, i.e. among the famous people of the area of Ivanec as well as in the Annexes to the bibliography of Ivanec. He is inevitable as the author of numerous papers on Paleolithic, Neolithic and Vindija Cave. Special attention should also be paid to the articles of deceased Marija Poje, his collaborator and native of Ivanec as well. Finally, Ivanec calendar bid the last goodbye to Mirko Malez with the paper of distinguished academician Milan Herak

    Geographic profiling in Nazi Berlin: fact and fiction

    Geographic profiling uses the locations of connected crime sites to make inferences about the probable location of the offender’s ‘anchor point’ (usually a home, but sometimes a workplace). We show how the basic ideas of the method were used in a Gestapo investigation that formed the basis of a classic German novel about domestic resistance to the Nazis during the Second World War. We use modern techniques to re-analyse this case, and show that these successfully locate the Berlin home address of Otto and Elise Hampel, who had distributed hundreds of anti-Nazi postcards, after analysing just 34 of the 214 incidents that took place before their arrest. Our study provides the first empirical evidence to support the suggestion that analysis of minor terrorism-related acts such as graffiti and theft could be used to help locate terrorist bases before more serious incidents occur

    Speziation mikrobiologisch alkylierter, leichtflüchtiger Selenverbindungen in Abhängigkeit der geochemischen Verfügbarkeit des Selens

    Selen kann in verschiedenen Oxidationszuständen (+6, +4, ±0, -2) in unterschiedlichen Umweltkompartimenten auftreten. Verbundenen damit sind verschiedene Eigenschaften, wie z. B. die Wasserlöslichkeit, die in direktem Zusammenhang mit der Migrationsfähigkeit sowie der Bioverfügbarkeit steht. Im Rahmen der vorliegenden Arbeit wurden die Verfügbarkeit anorganischer Selenspezies und damit die Mobilisierbarkeit dieser in verschiedenen Laborexperimenten untersucht. Hierbei wurde an Goethit adsorbiertes Selenit sowohl mit einer Reinkultur des aktiv methylierenden Pilzes Alternaria alternata als auch mit einer angereicherten Umweltmischkultur inkubiert und die mikrobiologische Zugänglichkeit anhand der Bildung leichtflüchtiger, alkylierter Selenmetabolite wie z. B. Dimethylselenid und Dimethyldiselenid beobachtet. Zur Analyse dieser wurde eine cryotrapping-cryofocussing-GC-ICP-MS-Kopplung etabliert. Die Anteile der methylierten Selenverbindungen stiegen bei Verwendung von A. alternata mit der Inkubationszeit auf 10 % des gelösten Selens und 1 % des Gesamtselens an. Dieser Trend konnte während der Inkubation der Umweltmischkultur nicht beobachtet werden. Hier lagen die Anteile über den gesamten Untersuchungszeitraum bei ca. 0,5 % des gelösten bzw. 0,1 % des Gesamtselens, inklusive eines leichten Abwärtstrends, welcher wahrscheinlich durch die Nutzung der Alkylselenide als Kohlenstoffquelle hervorgerufen wurde. Weiterhin wurde das reduzierte Eisenselenidmineral Ferroselit eingesetzt, um dessen Stabilität gegenüber der Aktivität des sulfatreduzierenden Bakteriums Desulfovibrio gigas zu untersuchen. Mit zunehmender Inkubationszeit und damit verbundener, zunehmender Reduktion des im Nährmedium vorhandenen Sulfates konnte ein Anstieg leichtflüchtiger Organoselenverbindungen in der Gasphase der Kulturansätze festgestellt werden, die im unteren Nanogrammbereich lagen. Einhergehend damit wurde auch die Zunahme der Gehalte an gelöstem Selen und somit die biologisch bedingte Rücklösung aus der Mineral- in die Wasserphase beobachtet. Es konnte gezeigt werden, dass die Aktivität von Mikroorganismen einen deutlichen Einfluss auf die Stabilität von Oberflächenkomplexen des Selenits als auch von mineralischen Selenidspezies hat.Selenium exists in different oxidation states (+6, +4, ±0, -2) in diverse environmental compartments. Different properties are related to these oxidation states, for example water solubility, which is closely correlated with the migration capability and the bioavailability. In this study, the availability of inorganic selenium species was investigated in different laboratory experiments to get information on the mobility of these species. Hereby selenite adsorbed on goethite was incubated with a pure culture of active methylating fungus Alternaria alternata as well as with an enriched environmental mixed culture. The microbiological availability of inorganic selenite was observed by the formation of volatile, alkylated selenium metabolites, e. g. dimethyl selenide and dimethyl diselenide. A cryotrapping cryofocussing gaschromatography system hyphenated with ICP-MS was established for speciation analysis. The amount of methylated selenium compounds increased to 10 % of the dissolved selenium and 1 % of the total selenium during the incubation of A. alternata. This behavior has not been observed during the incubation of the environmental mixed culture. In the latter experiments the amount of volatiles analysed lay at approximately 0.5 % of the dissolved selenium and 0.1 % of the total selenium during the whole investigation time. Additionally there was a slight decrease in the formation of volatile compounds that may be due to microbial usage of alkyl selenides as carbon sources. Furthermore, the reduced iron selenid mineral ferroselite was used to investigate its stability at different activities of the sulfate reducing bacterium Desulfovibrio gigas. With ongoing incubation time and a related reduction of sulfate an increase of volatile organoselenium compounds in the headspace of the cultures up to nanograms was observable. Along with this an increase in dissolved selenium and therefore a biological induced re-dissolution of selenium out of the mineral into the water phase has been observed. It was possible to show in this study, that the activity of microorganisms has a significant impact on the stability of surface complexes of selenite as well as with mineral selenid species
